Prediction of the Concentration of Diphenylhydantion in the Brain Using a Physiological Pharmacokinetic Hybrid Model

  • A physiological pharmacokinetic hybrid model was developed in order to predict the disposition kinetics of diphenylhydantoin (DPH) in the brain from the plasma conentration data of DPH. The model was constructed under the assumptions of well-stirred, plasma flow-limited and lienar tissue diposition kinetics of DPH. DPH was administered intravenously to the rats at a dose of 10 mg/kg together with/without sodium salicylate (SA;10 mg/kg) and the DPH concentrations in the plasma and brain were determined. Plasma protein binding of DPH concentrations in the plasma and brain were determined. Plasma protein binding of DPH was also determined using equilibrium dialysis technique. Then the model was tested for its predictability of DPH concentrations in the brian from the plasma data of DPH. It was found that the predicted values of DPH concentrations in the brian were in fair agreement with the experimental values in the rats of both treatments. The 2-fold increase in the brain concentration of DPH by SA-coadinistration was predicted well from the plasma concentration and plasma free fraction ($f_p$) data of DPH using the model. Therefore, the hybrid model was concluded to be very useful for the prediction of the concentrations of DPH in the brain from the plasma concentration data. Finally, DPH concentrations in the human brian was calculated using this model from plasma DPH data in the literature, yet the scale-up of this model to the human is not convinced.

Effects of Coptidis Rhizoma on Lowering Lipid and Oxidative Stress

  • The effects of hwangryun(Coptidis Rhizoma) on lowering lipid and oxidative stress in the induced obesity rat was observed. The concentration of plasma triglyceride in hwangryun treatment groups showed the low values compared to the control group, and as the increased hwangryun, the concentration of triglyceride decreased. The concentration of plasma total cholesterol decreased in all hwangryun treatment groups. However the concentration of plasma HDL-cholesterol and LDL-cholesterol showed no significantly difference in all the treatment groups. Thiobarbituric acid reactive substance(TBARS) concentration in plasma and liver showed low values in all hwangryun treatment groups compared to the control group. Glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) activities showed no significantly difference in all the treatment groups. However catalase(CAT) activity showed a tendence to increase in hwangryun groups, and in 200mg/kg hwangryun treatment group showed significantly a high value than the control group. Summarizing the results above, hwangryun has the functional materials that lowering lipid and works with oxidative stress.

Effect of Vitamin E Supplementation on Plasma Antioxidant Vitamins and Immunity Status of Crossbred Cows

  • Twenty crossbred (HF${\times}$Tharparkar) dry pregnant cows were divided into four equal groups. They were supplemented with 1,000 I.U. $\alpha$-tocopheryl acetate from 0 (group I), 15 (group II), 30 (group III) and 60 (group IV) days before parturition to 1month of lactation. All the cows were kept under similar feeding and management conditions. Blood plasma samples collected on specific days were analyzed for $\alpha$-tocopherol, retinol, total antioxidant activity (FRAP), immunoglobulin and calcium. Plasma $\alpha$-tocopherol concentration at 30 days prepartum averaged 3.5, 4.1, 4.4 and $3.9{\mu}g/ml$ and decreased by 50.0, 41.4, 34.1 and 33.3 percent on the day of parturition in the four respective groups. After calving, plasma vitamin E started to recover earlier in groups II, III and IV as compared to group I. Mean plasma $\alpha$-tocopherol concentration at 21 days postpartum was significantly higher in groups II, III and IV (2.9, 3.5 and $3.1{\mu}g/ml$) compared to group I ($1.9{\mu}g/ml$) cows. Plasma retinol concentration also showed a substantial decrease in all the groups on the day of calving but recovered to its normal value at 3 weeks postpartum. Plasma total antioxidant activity averaged 901, 895, 859 and $875{\mu}mol/l$ in the four respective groups on 30 days prepartum and decreased on the day of calving in all the groups, but the decrease was less in groups III and IV. Plasma immunoglobulin concentration was higher in group IV, followed by groups III, II and I, respectively, showing better immune status of vitamin E supplemented cows due to less oxidative stress. Supplementation of vitamin E resulted in higher plasma calcium concentration. The data showed that vitamin E supplementation should be started at least 30 days prepartum to reduce oxidative stress in periparturient cows.

대두올리고당이 흰쥐의 장내 균총, 지질 대사 및 면역 능력에 미치는 영향 (Effects of Soyoligosaccharides and Varing Dietary Fats on Intestinal microflora. Lipid Profiles and Immune Responses in Rats)

  • The aim of this study was to evaluate the effects of soyoligosaccharides on intestinal microflora transit time lipid profiles and immune responses. Forty-eight male rats of Sprague-Dawley strain were divided into six groups(SYS: soybean oil-sucrose SYO: soybean oil-oligo SES; esame oil-sucrose SEO:sesame oil-oligo BFS : beef tallow-sucrose BFO : beef tallow-oligo) soyoligo-groups(SYO, SEO, BEO) were fed the diet containing 5% soyoligosaccharides but control groups not fed them and all groups were fed 20% fats for 4 weeks. The number of fecal bifidobacteria tended to increase after soyoligosaccharides feeding. The significant increase was found in SEO group compared to SES in that of fecal bifidobacteria,. Stool wet weight dry fecal weight and stool water content were increased significantly in soyoligo-group(SYO, SEO, BFO) compared to control groups(SYS, SES, BFS) and gastrointestinal transit time(TT) tended to increase. Soyoligosaccharides lowered plasma cholesterol in SEO group significantly more than did other groups. soyoligo-groups(SYO, SEO, BFO) tended to decrease liver triglycerides compared to control groups. Plasma IgG concentration increased in beef tallow groups(BFS, BFO) than that in other dietary fat groups. Soyoligosaccharides and kinds of fats had effects on plasma C3 concentration and mitogenesis of the spleen cells. Soyoligosaccharides had no effects on plasma leptin and insulin concentration but kinds of fats did so that sesame groups were decreased significantly in plasma leptin concentration but increased significantly in plasma insulin concentration compared to other dietary fat groups. In conclusion dietary soyoligosaccharides changed the composition of the intestinal microflora beneficially with sesame oil and partially had effect on plasma and liver lipid profiles .(Korean J Nutrition 33(6) : 597∼612, 2000)

플라즈마/광촉매 결합시스템에 의한 톨루엔 제거특성 (Removal Characteristics of Toluene by the Combined Plasma/Photocatalyst System)

  • The main purpose of this study is to analyze the characteristics of toluene removal by plasma, photocatalyst, and plasma/photocatalyst system with the major parameters such as flow rate, inlet toluene concentration and applied voltage, etc., experimentally. In the combined plasma/photocatalyst process, rates of toluene conversion are represented as 99% at flow rate 250, 500 mL/min while, below 97% at flow rate 1000 mL/min due to the low residence time(reaction time) at the same applied voltage 4173 voltage and toluene inlet concentration 50 ppm. The intermediate products are detected by GC/MS analysis showing the small amounts of benzoic acid, benzyl alcohol and residual ozone concentration $0.04{\sim}0.05$ ppm generated by plasma process in the present system.

  • This study was conducted to investigated the possibility of inactivating wilt germs (Fusarium oxysporum f. sp. radicis lycopersici) using Dielectric Barrier Discharge (DBD) plasma in a hydroponic system. Recirculating hydroponic cultivation system for inactivation was consisted of planting port, LED lamp, water tank, and circulating pump for hydroponic and DBD plasma reactor. Two experiments were conducted: batch and intermittent continuous process. The effect of plasma treatment on Total Residual Oxidants (TRO) concentration change, Fusarium inactivation and growth of lettuce were investigated. In the batch experiment, most of the Fusarium was inactivated at a TRO concentration of 0.15 mg/L or more at four-day intervals. There was no change in lettuce growth after two times of plasma treatment for one week. The intermittent continuous experiment consisted of 30-minute, 60-minute, and 90-minute plasma treatment in 2 day intervals and 30-minute treatment a one-day; most of the Fusarium was inactivated only by treatment for 30-minute every two days. However, if inactivation under $10^1CFU/mL$ is required, it will be necessary to treat for 60 minutes in 2 day intervals. The plasma treatment caused no damage to the lettuce, except the 30 min plasma treatment ay the one-day interval. It was considered that the residual TRO concentration was higher than that of the other treatments.

관상동맥협착 환자의 혈장 총 Homocysteine, 엽산 및 비타민 $B_{12}$ 농도 (Plasma Total Homocysteine, Folate and Vitamin $B_{12}$ Concentrations in Patients with Coronary Stenosis)

  • Recently elevated plasma homocysteine concentration is considered an independent risk factor for atherosclerosis and thrombosis with coronary artery disease. Folate and vitamin $B_{12}$ are cofactors and closely related with metabolism of homocysteine. The purpose of this study is to evaluate the correlation between homocysteine and folate and vitamin $B_{12}$ in patients with ischemic heart disease. Twenty-six patients, in whom coronary angiographic finding revealed more than $50\%$ of stenosis at least in one coronary vessel were enrolled as the patient group, and thirty subjects, in whom angiographic finding revealed in not significant stenosis, but complained of chest pain, were selected as the control group. Fasting venous blood was obtained and measured the concentration of plasma total homocysteine, folate and vitamin $B_{12}$ by high performance liquid chromatography and fluorescence detection method. We examined the correlation between homocysteine and folate and/or vitamin $B_{12}$ in the control group and the patient group, respectively. Compared with the control group, the patient group had relatively higher plasma total homocysteine concentration ($10.7\pm4.2\;vs\;9.6\pm3.5$ umol/L), but showed no significant difference. Folate and vitamin $B_{12}$ concentration are low in the patient group, but showed no significant difference between patient and control group. Plasma total homocysteine concentration showed negative correlation with folate and vitamin $B_{12}$ in both the control group and the patient group, and showed significantly negative correlation in patient group {r = -0.550 (p < 0.01) vs r = -0.609 (p < 0.01)}. We knew that the plasma total homocysteine concentration were relatively elevated in patient group compared with the control group. Because plasma total homocysteine concentrations are closely negative correlated with folate and vitamin $B_{12}$ in the patient group, folate and vitamin $B_{12}$ supplement can lower the mortality and morbidity of ischemic heart disease. Effects of Refeeding with a Protein-Free Diets Supplemented with Various Essential Amino Acids on the Plasma Insulin-Like Growth Factor-I Concentration in Fasting Young Chickens

  • The effect of refeeding with various single essential amino acids on the recovery of plasma insulin-like growth factor-I (IGF-I) concentration in fasted young chickens was examined. Young chickens (29 days of age) were divided into 15 experimental groups. Chickens in one group were fed on the commercial diet ad libitum for 4 days. The remaining 56 chickens in 14 experimental groups were fasted. After 2 days of fasting, 52 chicks in 13 fasted groups were refed with one of the following experimental diets for 2 days. Eleven experimental diets were protein-free diets supplemented with one of 11 essential amino acids (Arg, Gly, His, Ileu, Leu, Met, Phe, Lys, Thr, Trp, Val). The remaining 2 experimental diets were a protein-free diet containing 11 essential amino acids and a protein-free diet not supplemented with amino acids. Birds in the remaining fasted group continued to be fasted for 2 days. Fasting for 2 days markedly reduced plasma IGF-I concentration. When fasted chickens were refed the protein-free diet containing either Gly alone or all essential amino acids, plasma IGF-I concentration was recovered to the level similar to that of fed chickens. Protein-free diet alone, however, failed to restore the reduced IGF-I concentration in plasma. Body weight loss modulated by feeding with protein-free diets supplemented with various single essential amino acids was associated with changes in plasma IGF-I concentrations. We concluded that body weight loss by feeding with a protein-free diet was lower than that of fasted chickens and that body weight loss associated with the decrease in plasma IGF-I concentration was modulated by feeding with protein-free diets containing various single essential amino acids.

EFFECT OF ABOMASAL INFUSION OF ALANINE AND ASPARTIC ACID ON GROWTH HORMONE SECRETION IN SHEEP

  • Effects of animo acids infusion into the abomasum on plasma growth hormone (GH) concentration were investigated using three wethers of 54 kg of average body weight. Wethers were infused with either 3.25 mmol/kg BW/day of sodium chloride solution (control), 3 mmol/kg BW/day of alanine (Ala), or 3 mmol/kg BW/day of aspartic acid (Asp) continuously for five days through an abomasum cathether in a $3{\times}3$ Latin square desing. On the day of starting infusion (day 0) and day 4 blood samples were collected from a jugular vein every fifteen minutes for six hours after feeding, and their GH concentrations were measured. Blood samples were also collected immediately before starting infusion (day 0), and before feeding of day 1, day 2 and day 4, and their plasma free amino acid concentrations were measured. In the animals infused with Ala, plasma free Ala concentration was increased by Ala infusion and it continued for four days. Plasma GH concentration of these animals increased on day 0, but this phenomenon disappeared on day 4. In the animals infused with Asp, the increase in plasma Asp concentration was observed only on day 1. Plasma GH concentration of these animals was not affected by Asp infusion. These results suggest that continuous Ala infusion stimulates GH secretion for a short period, but the effect would not last long, and that continuous Asp infusion does not affect plasma GH concentration.

Development and Evaluation of an Oral Controlled Release Delivery System for Melatonin

  • Sugar spheres loaded with melatonin (MT) were coated with $Aquacoat^{\circledR}$ to control the release rate of MT over 8 hours. A zero-order release pattern over 8 hours was obtained with 20% coating on 8-10 mesh beads in USP basket dissolution studies. MT in 20% coated beads was quite stable at room temperature with less than 5% MT degraded during 6 months' storage. Dissolution profiles were also unchanged after 6 months. An oral preparation containing MT-loaded uncoated beads for immediate release and 20% coated beads with $Aquacoat^{\circledR}$ for controlled release over 8 hours was evaluated in six human subjects. When total 0.5 mg MT as low dose (immediate release portion of MT, 0.1 mg) was administered to four subjects, average peak plasma MT concentration was reached at about 600 pg/ml and maintained at about 10 pg/ml over 8 hours. Plasma MT concentration-time profiles were similar in shape to computer-simulated profiles. However, maximal plasma MT concentrations were three times greater compared to computer simulated curve. These results suggest that MT dose, ratio of immediate and controlled release MT, and pharmacokinetic parameters selected are adjusted to mimic endogenous MT concentration-time curve. In another study, 0.2 mg MT having 10% of immediate release portion and 80% controlled release portion produced plasma MT concentration-time curve which is more similar to endogenous profiles. A low bioavailability (<20%) may result from extensive first pass metabolism and remaining amounts of MT from controlled beads. A good correlation between plasma MT concentration and urinary excretion rate of 6-sulphatoxymelatonin (6-STMT), a major metabolite of MT was observed. As plasma MT concentration increased, urinary excretion rate of 6-STMT increased concomitantly. The linear relation between plasma MT and urinary excretion rate of 6-STMT was statistically significant. This result suggests that urinary 6-STMT may be used as an index of circadian rhythms of MT in humans.
