Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.
The pathogenic effort of high glucose, possibly in concert with fatty acids, is mediated to vascular complications of diabetes via increased production of reactive oxygen species(ROS), reactive nitrogen species(RNS), and subsequent oxidative stress. This study was carried out to investigate the suppressive effect of buchu(Allium tuberosum) on oxidative stress in streptozotocin(STZ)-induced diabetes in Sprague Dawley male rats. The effect of buchu supplementation (10%) on lipid peroxidation, and antioxidative defense system in blood and liver was compared among normal rats fed basal diet(normal) and diabetic rats fed basal diet(DM-control) or 10% buchu-supplemented diet(DM-buchu). Diabetes was experimentally induced by the femoral muscle injection of 50 mg STZ per kg of body weight. Animals were sacrificed after 4 wks of experimental diets feeding. The induction of diabetes by STZ elevated the level of lipid peroxidation represented by thiobarbituric acid-reactive substances(TBARS) and conjugated dienes in plasma, LDL, liver, and erythrocytes. 10% buchu-supplemented diet significantly reduced the levels of conjugated dienes in erythrocytes(p<0.05) and lowered TBARS in liver and LDL to the levels of control. Induction of diabetes by STZ elevated Mn-superoxide dismutase(Mn-SOD) activity and lowered activities of glutathionine reductase(GSH-red) and glutathionine peroxidase(GSH-px). Catalase activity was not affected by the induction of diabetes by STZ. However, buchu supplementation to diabetic rats significantly elevated catalase activity(p<0.05) and slightly elevated GSH-px and GSH-red activities in liver. GSH levels of blood and liver were lowered or not changed by induction of diabetes by STZ, respectively, while buchu supplementation to diabetic rats significantly elevated hepatic GSH level (p<0.05). In conclusion, it can be concluded that buchu might be a food source to attenuate oxidative stress in diabetic patients by inhibiting lipid peroxidation, by increasing hepatic GSH level, and by inducing anti-oxidative enzyme systems.
Effects of siho(Bupleuri Radix) extracts on lipid composition and antioxidant system were investigated in rat fed high fat diet. body weight gain, concentration of plasma triglyceride, total cholesterol, LDL-cholesterol showed a tendency to decrease in siho(Bupleuri Radix) extracts groups. However, the concentration of HDL-cholesterol showed no significantly different among treatments. Plasma and liver TBARS concentration showed a low values in siho(Bupleuri Radix) extracts groups. Activities of SOD, GSH-Px and CAT showed a tendency to increase in siho(Bupleuri Radix) extracts groups.
Objective : Effects of Gamibangpungtongseungsan on lipid composition and antioxidant system were investigated in rat fed high fat diet. Methods : We fed control group rats high fat diet and administered normal saline for 8 weeks, and experimental group rats high fat diet and administered extract of Gamibangpungtongseungsan for 8 weeks. Results : Body weight gain, concentration of plasma triglyceride, total cholesterol, LDL-cholesterol showed a tendency to decrease in Gamibangpungtongseungsan groups. However, the concentration of LDL-cholesterol showed a tendency to increase in Gamibangpungtongseungsan groups. Plasma and liver TBARS concentration showed a low values in Gamibangpungtongseungsan groups. Activities of SOD, GSH-Px and CAT showed a tendency to increase in Gamibangpungtongseungsan groups. Conclusions : These results indicate that Gamibangpungtongseungsan has a deep inhibitory effect on lipid composition and antioxidant system.
The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. This study was performed to investigate the effect of Puerariae radix-ethanol extracts rich in isoflavone on the antio-xidative system of rats. For this purpose, first, Puerariae radix was extracted with ethanol, and its total isoflavone and puerarin contents were analysed. Second, female Sprague Dawley rats were fed for 6 weeks with four diets which were based on AIN96G diet and supplemented with Puerariae radix-ethanol extracts to contain isoflavone. The isoflavone contents of four experimental diets were 0 mg, 500 mg, 1,000 mg, 2,000 mg per kg diet, respectively (control, P0.05%,P0.1%, P0.2%). Liver and erythrocyte activities of antioxidative enzyme such as superoxide dismutase (SOD), catalase,glutathione peroxidase (GSHpx) were measured. Also, plasma and liver malondialdehyde (MDA) concentrations, liver glutathione (GSH) and oxidized glutathione (GSSG) concentrations were measured. The total isoflavone content of Puerariae radix-ethanol extract was 3067.6 mg per 100 g extract and the content of puerarin was 2557.4 mg per 100 g extract. The erythrocyte activities of GSH-Px and catalase were higher in group P0.1% and P0.2%. But SOD activity of erythocyte did not show any difference by the Puerariae radix-ethanol extract supplementation in diet. The activity of SOD in liver increased significantly by the supplementation of extract, showing highest level in P0.1% group. The liver GSH concentration increased significantly in group of P0.05%, P0.1%, and P0.2% compared with control group (p <0.05). The GSSG concentration in liver showed no difference by the supplementation of Puerariae radix extract from the control group, except P0.2% group. The plasma MDA concentration did not show any significant differences by the extract supplementation. But the liver MDA concentration decreased by the extract supplementation, showing the lowest level in P0.1 % diet group. These results suggest that the supplementation of Puerariae radix-ethanol extract can inhibit lipid peroxidation in liver and enhance the antioxidative defense competence of rats.

The effect of dietary methionine level on lipid peroxidation of rats was studied. Rats were fed vitamin E- selenium- deficient diet or diet supplemented with various levels (0.3, 0.6, 0.9%) of methionine. In rat fed MF diet, body weight gain and feed efficiency ratio were decreased compared with those of control rats, but reversed by supplementation with 0.3 and 0.6% methionine. Lipid peroxide levels in plasma and hepatic mitochondrial fraction of MF group rats were significantly higher than those of control rats. However, supplementation with 0.6% methionine modified this increment. GSH-Px activity was decrased to varying degrees in erythrocyte and hepatic mitochondrial fraction from rats fed MF diet. Methionine supplementation did not affect induction of this enzyme activity. Examination of hepatocytes by electronmicroscopy showed that Influence of vitamin E, selenium, and methionine deficiency was mainly characterized by lipid droplets, swollen mitochondria and microvilli destruction. Supplementation with various levels of dietary methionine modified these changes to some extent. The results of this experiment indicated that MF diet causes significant change in lipid peroxide level, GSH-Px activity and morphology of rats which these changes may lessen by supplementation with 0.6% methionine.

The effect of commercial deer antler drink (provided by Chung-yang Deer Farm) on blood glucose level, plasma lipids and antioxidants state in type 2 diabetic patients were studied. Ten patients with type 2 diabetes participated in the study and consumed 2 pouches (200 mL) of deer antler drink every day for 3 weeks. No significant differences were observed in levels of triglycerides, total cholesterol, low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C). However, oxidized LDL measured as conjugated dienes decreased in the patients after the trial. Plasma tocopherols and carotenoids levels showed no significant changes. No significant differences were observed in erythrocyte SOD, catalase and GSH-Px in the each group. No significant differences were observed in plasma TRAP. The results would suggest that deer antler drink influences conjugated dienes but long-term intervention trial may be necessary to see further beneficial effect of deer antler drink in diabetic patients.

To investigate antioxidative and antiaging effects of yellow and black soybeans in vivo system, male SD rats (n=24) were fed the diets containing casein (C), yellow soybean (YS) or black soybean (BS) for 8 weeks. Experimental groups showed the preventive effect on lipid and protein oxidation, especially protein oxidation of plasma was significantly inhibited in BS group. SOD and catalase activities were not different among the groups, but hepatic glutathione peroxidase (GSH-px) activity was significantly lowered in YS and BS groups compared with control group. The contents of superoxide anion radical in cytosol were significantly lowered in experimental groups compared with control group. And hydroxyl radical was slightly lowered in soybean groups. Lipofuscin accumulation on the heart and eyes of rats was effectively inhibited by yellow soybean and black soybean, respectively. In conclusion, these results suggest that yellow soybean and black soybean have the antioxidative/antiaging effect in vivo system and they have similar activities despite different color of seed coat.

The effects of mungbean sprout juice on cadmium-induced hepatotoxicity in rats were investigated. Sprague-Dawley rats weighing about 90g were divided into 4 groups and raised for 6 weeks. ; control group(CON), mungbean sprouts juice-administered group(MSJ), cadmium-administered group(CAD) fed water containing 40 ppm cadmium chloride and mung bean sprouts juice and cadmium-administered group(MCD). The diet was supplied every day for the measurement of feed efficiency ratio(FER) and net weight gain was measured every 3 days. The activities of serum glutamic oxaloactic transaminase(GOT) and glutamic pyruvic transaminase(GPT), superoxide dimutase(SOD), catalase and glutathione peroxidase(GSH-Px) in the liver and the hepatic contents of glutathione were examined. The contents of Cd in liver and kidney of the rats were determined by using ICP(Inductively Coupled Plasma Emission Spectrophotometer). Growth rate and FER were decreased in CAD group, compared with CON group but the changes were not significant in MCD group. The activities of serum GOT and GPT, SOD, catalase and GSH-Px in the liver were increased by Cd administration, but the alterations were decreased by supplementation with mungbean sprouts juice. The level of glutathione decreased in CAD group(26.8$\pm$9.0mg/g liver), whereas it increased in MCD group(36.4$\pm$15.8mg gliver). The content of Cd in the liver and kidney in MCD group(9.57 ppm, 4.88 ppm) was decreased, compared with CAD group(12.81 ppm, 5.46 ppm). This result suggested that mungbean sprout juice has a lowering effect on the accumulation of Cd in the liver and kidney and it is believed that the juice has some protective effects to Cd-induced hepatotoxicity in rats, but the mechanism of these effects was obscure.
