Background: This study determined the effects of mulching, an environment friendly organic cultivation method, on antioxidant compound contents and growth in Codonopsis lanceolata, commonly known as Deodeok. Methods and Results: C. lanceolata was treated by mulching with several different methods (a non-woven fabric, biodegradable film, or rice husks) and also treated with hand weeding. A non-treatment plot was used as a control. The growth and levels of weed control in C. lanceolata were better in plants cultivated under mulching treatments (non-woven fabric, biodegradable film, and rice husks) than in those under non-mulching treatments (hand weeding and non-treatment). The contents of antioxidant compounds, such as total flavonoids, phenolics, and anthocyanins, were highest under the biodegradable film treatment, followed by the non-woven fabric treatment, rice husks treatment hand weeding, and non-treatment. There were identifiable differences in DPPH and ABTS activity in comparison to antioxidant compound content by solvent fractions. Mulching treatments resulted in higher DPPH scavenging activity in water and ethyl ether fractions and ABTS scavenging activity in n-butanol fractions than in other fractions, as opposed to hand weeding and non-treatment groups, although total activity of DPPH and ABTS did not increase with mulching treatments. Conclusions: Mulching C. lanceolata with biodegradable film and non-woven fabric is an effective method for improving plant growth and inhibiting the occurrence of weeds as well as for increasing antioxidant compound content and altering antioxidant activity.

Bacillus subtilis BD0310 isolated from tea leaves was used for the development of a biofungicide against Pestalotiopsis longiseta causing gray blight of tea plants. The optimum growth conditions were investigated for the mass cultivation of the microbial agent. The optimum temperature and cultivation time were determined as $12{\sim}24$ hours at $30^{\circ}C$ and the optimum initial pH was pH 7.0 in nutrient broth. Among the tested carbon sources of fructose, galactose, glucose, glycerol, inositol, lactose, maltose, sorbitol and starch, maltose and inositol were found to highly increase antifungal activity of the microbial agent against P. longiseta. Yeast extract and tryptone apparently increased antifungal activity of the microbial agent among the tested nitrogen sources of casein, tryptone, malt extract, yeast extract and $(NH_4)_2SO_4$. The results will make a contribution to mass production of the antagonistic bacterium Bacillus subtilis BD0310 for development of a microbial agent controlling gray blight of tea plants.
Phenotypic characteristics, pathogenicity and fungicides resistance of fifty one isolates of Botrytis cinerea obtained from various host plants were observed and determined. The relationships between these characteristics were also investigated on the basis of isolation host plants. The isolates of B. cinerea varied in the capacity of sclerotia formation and sporulation. The pathogenicity of 44 isolates from tomato, cucumber, and strawberry was significantly stronger with 3.2 cm in average diameter of necrotic lesions on cucumber leaves than that of seven isolates from other host plants such as orange, gerbera, ginseng, kiwi, grape, pear and from butter with 1.8 cm in average diameter of necrotic lesions. Benomyl resistance of 12 isolates from tomato plants was much higher with the $EC_{50}$, 562 ppm than that of 19 isolates from various host plants. Diethofencarb resistance, however, of 11 isolates from strawberry plants was highest with the $EC_{50}$, 210 ppm among isolates from other host plants. Polygalacturonase activity varied among isolates in the range of 0 to 103 unit and that of isolates from tomato, cucumber and strawberry was slightly lower than that of isolates from other host plants. No significant relationship between pathogenicity and fungicides resistance, polygalacturonase activity was found among 51 isolates of B. cinerea. Isozyme patterns of polygalacturonase produced from two strongly and weakly pathogenic isolates (FC122, KC6) were slightly different depending upon carbon sources during cultivation.

This study was conducted to evaluate the quality variation of Lithospermum radix on the pigment contents and antioxidant activities according to different growth stages and areas of cultivation. Acetylshikonin contents showed the tendency to decrease gradually from the middle of July (0.28%) to the end of August (0.05%) and then the content was increased again to the end of October (0.25%). Shikonin content was detected as small amount of about 0.009% during the period. The weight of plants was increased from the end of September to the end of October and showed the highest value as 19.8 g on October 25. ROS scavenging activity was the highest in the early of October as $IC_{50}$ value of 0.11 ${\mu}g/mL$. Lithospermum radix of September showed lower ROS scavenging activities than those of other growth stages as $IC_{50}$ value of 1.02 and 0.49 ${\mu}g/mL$ on September 9 and September 27, respectively. Among 17 areas cultivated Lithospermum radix, 10 areas (59%) showed 0.05-0.10% of acetylshikonin contents and 3 areas (18%) were measured to contain 0.16-0.26% of acetylshikonin.
Protopectinase (PPases) are heterologous group of enzymes that degrade pectin from the insoluble protopection which is constituent of the middle lamella and primary cell wall of higher plants by restricted depolymerization. From the previous report[6], enzymatically separated plant cells, which are produced from plant tissues by PPases treatment, showed well-conserved cellular components with their rigid cell wall and this characteristic is applicable to preparation of novel food material. The purpose of this study is to investigate the effect of medium composition of PPase production from Bacillus subtilis EK11 which was selected as a PPase producer. Various carbon sources and concentrations on PPase production were studied and corn starch at 0.7% was the most effective for production of PPase. Among the nitrogen sources, yeast extract was the most effective for PPase production and the effect of (NH4)2SO4 was notable as inotganic nitrogen source. Inorganic compounds such as KH2PO4, K2HPO4, Na3-citrate.2H2O and MgSO4 were optimized for PPase production. PPase activity was inhibited by the adition of Ba2+ or Zn2+. The optimal medium for PPase production was devised: 0.7% corn starch, 0.3% yeast extract, 1.4% KH2PO4, 0.6% K2HPO4, 0.1% Na3-citrate.2H2O and 0.02% MgSO4. PPase production by using the optimum medium was carried out with shaking cultivation at 37$^{\circ}C$. The maximum PPase activity of 256unit/ml could be obtained after the cultivation for 48hrs. The activity was increased about 2.2timesthan the activity, 112 unit/ml, in basal medium.
The hairy root was induced form potato tuber disc by infection of a. rhizogenes. The detection of the agropine and mannopine by paper electrophoresis confirmed that induced hairy root was transformed by A. rhizogenes. The activity of peroxidase was the highest at 5 weeks and isozyme pattern of peroxidase revealed 3 cathodic bands and 2 anodic bands and new C4 band(pI 4.6) was observed at 7 weeks after cultivation in hairy root was isoelectric focusing. To study the effect of Ca2+ on cell wall formation in hairy root, channel blocker of Ca2+ was treated. The activity of $\beta$-glucan synthetase II(GS II) related to cell wall synthesis was inhibited by about 50% in diltiazem and flunarizine treatment than that of control, but stimulated in CaCl2 treatment. Therefore these results showed that Ca2+ might be an effective factor in the cell wall formation. The activity of GS II by NaF treatment was increased by about 30%. This result suggested that the activity GS II is changed through phosphorylation process.

Background: Ginseng (Panax ginseng Meyer) is an invaluable medicinal plant containing various bioactive metabolites (e.g., ginsenosides). Owing to its long cultivation period, ginseng is vulnerable to various biotic constraints. Biological control using endophytes is an important alternative to chemical control. Methods: In this study, endophytic Trichoderma citrinoviride PG87, isolated from mountain-cultivated ginseng, was evaluated for biocontrol activity against six major ginseng pathogens. T. citrinoviride exhibited antagonistic activity with mycoparasitism against all ginseng pathogens, with high endo-1,4-${\beta}$-D-glucanase activity. Results: T. citrinoviride inoculation significantly reduced the disease symptoms caused by Botrytis cinerea and Cylindrocarpon destructans and induced ginsenoside biosynthesis in ginseng plants. T. citrinoviride was formulated as dustable powder and granules. The formulated agents also exhibited significant biocontrol activity and induced ginsenosides production in the controlled environment and mountain area. Conclusion: Our results revealed that T. citrinoviride has great potential as a biological control agent and elicitor of ginsenoside production.
This paper is aim to report the core aspects of Codex guideline for organically grown food which is finalized by FAO/WHO Codex Alimentarius Commission on May 2000 in 28th session of the Codex committee on food labelling. The chapter of animal production had discussed for a long time before it was finalized in Ottawa/canada as well as use of GMO(Genetically modified organism), manure from factory farming, animal welfare, and fodder inputs consisting of at least 85% for ruminants and 80% for non-ruminants. As the guideline for Codex set out the several things which is very difficult for Asian country, Organic farmer in Korea should pay an attention to maintain/increase the fertility and biological activity by cultivation of legumes, green manures or deep-rooting plants in an appropriate multi-annual rotation programme, and incorporation in the soil of organic material from holding producing in accordance with the guidelines. Pest, diseases and weeds should be controlled by choice of appropriate varieties, appropriate rotation, mechanical cultivation, diversified ecosystems, flame weeding, animal weeding and steam sterilization. The use of plant growth hormone, GMO and manure from industrial management system are not allowed, and closed recycling system, rotation, resistant seeds again pest and disease should be practiced in organic farming. But these are not unfortunately practiced in the country. In the conclusion it was strongly suggested to enact the Basic Standard for Korean organic agriculture which contains the core principles of Codex guidelines, and to try the importation of the most advanced theory, skills and technology from leading country in organic farming.
This study aimed to investigate the efficiency of arbuscular mycorrhizal fungi (AMF) in enhancing plant performance and bioactive compound concentrations in tomatoes (Lycopersicon esculentum Mill.). This factorial pot experiment included nine replications over 120 days of cultivation. Three AMF species (Rhizophagus prolifer, Claroideoglomus etunicatum, and Acaulospora mellea) were utilized as inoculum, while non-mycorrhizal controls with or without synthetic NPK fertilizer were compared. Interestingly, C. etunicatum KS-02 inoculations effectuated the best fruit growth and weight, which were statistically higher than those of the control without AMF. However, only fruit fresh weight was higher in plants inoculated with C. etunicatum KS-02 than those treated with the synthetic NPK fertilizer. In addition, C. etunicatum KS-02 inoculations induced a ≥ 11% increase in DDPH (1,1-diphenyl-2-picrylhydrazyl) activity, lycopene content, and carotenoid content compared to the control. This study is the first to report Claroideoglomus species' effectiveness in promoting growth, fruit yield, and bioactive compound production in L. esculentum Mill. These findings substantiate the significant potential of C. etunicatum KS-02 for tomato cultivation without the adverse effects of excessive synthetic fertilizer use.
Orchids are evolutionally known to be the most advanced plants in the order Liliales, and comprise approximately 1,000 genera and 35,000 species world-wide. In Korea, more than 110 species of Orchidaceae have been reported to be cultivated or to be collected in the wild. Orchids aye mostly dependant on orchid mycorrhizae(OM) throughout or in part of their life cycle. The OM endomycorrhizae belonging to basidiomycetes or rarley ascomycetes are needed for orchid seed germination. Various fungi, including plant pathogenic, antagonistic and symbiotic fungi, were isolated from the roots of orchid native to Korea. The OM fungi collected from the roots of Cymbidium goeringii were three species of Rhizoctonia namely, R. repens (anamorph state of Tulsanella repens), R. endophytica (Ceratobasidium cornigerum), and an unidentified species (possibly an anamorph of T. calospora). These symbiotic fungi induced peloton in the cortical cells of orchid roots, and differed biologically and in 18s rDNA sequences from plant pathogenic Rhizoctonia species. Also, the mycorrhyzal fungi enhanced the orchid root absorption of nitrogen sources and minerals from the soil. The activity of mycorrhizal fungal hyphae in the roots caused prevention from pathogenic fungi. In nature, the peloton is observed in the cortical cells of Cymbidium goeriingii roots, indicating mycorrhizal colonization in the native orchid roots. On the other hand, pathogenic fungi such as Fusarium and/or Rhizoctonia species are mostly isolated from commercial orchid plants. These suggest that application of symbiotic mycorrhizal fungi should be needed for orchid cultivation in nurseries and at the time of transplanting.
