Peanut is an allotetraploid derived from a single recent polyploidization. Polyploidization has been reported to have caused significant loss in genetic diversity during the domestication of cultivated peanuts. Single nucleotide polymorphism (SNP)-based markers such as cleaved amplified polymorphic sequences (CAPS) derived from next-generation sequencing (NGS) have been developed and widely applied for breeding and genetic research in peanuts. This study aimed to identify the genetic diversity and population structure using 30 CAPS markers and 96 peanut accessions from five different origins. High genetic dissimilarities were detected between the accessions from Korea and those from the other three South American origins generally regarded as the origin of peanuts, while the accessions from Brazil and Argentina presented the lowest genetic dissimilarity. Based on the results of the present study, accessions from Korea have unique genetic variation compared to those from other countries, while accessions from the other four origins are closely related. Our study identified the genetic differentiation in 96 peanut accessions from five different origins, and this study also showed the successful application of SNP information derived from re-sequencing based on NGS technology.
Levels of genetic diversity, population genetic structure, and gene flow in Hosta yingeri, a herbaceous perennial endemic to Taehuksan, Sohuksan, and Hong Islands, were investigated. Starch gel electrophoresis was conducted on leaves for 101 plants collected from three populations. Although the distribution of thespecies is restricted in the islands, it maintains high levels of genetic variatin; 64% of polymorphic loci in at least one population (Ps), the mean number of alleles per locus (Ap) of 1.92, and the mean effective number of alleles per locus (Aep) of 1.52. Overall, mean genetic diversity (Hep=0.250) was substantially higher than mean estimate for species with very similarlife history traits (0.102). Large populaton size, the persistence of multiple generations within populations, high fecundity, predominantly outcrossing breeding system, large size of pollinator visitation areas may be explanatory factors contributing the higher level of genetic diversity maintained within populations. Analysis of fixation indices showed an overall slight excess of heterozygotes (mean FIS=-0.066) relative to Hardy-Weinberg expectations, which may in part be due to the near self-incompatible breeding system in the species. Significant differences in allele frequencies among populaitns were found for 14 out of 16 polymorphic loci (P<0.05). Slightly more than 80% of the total variation in the species was common to all populations (GST=0.198). As expected, indirect estimate of the number of migrants per generation (Nm=0.45, calculated from mean GST) and nine private alleles found in the three populations indicate that gene movement among three isolated island populations was low.

To offer the basic data of vegetation restoration by buried seed of soil, this research was conducted at bare land and abandoned field in Yesan-gun. Germination plants of buried seed were consisted of 40 taxa (37 species, 3 varieties) in bare land, and 41 taxa (37 species, 4 varieties) in abandoned field. Classification of germination plants by family was the most in Gramineae, and emergent frequency of plots was the highest of Digitaria ciliaris. The soil depth of the most plants appearance was 2~5cm in bare land and 5~10cm in abandoned field, and the soil depth of the most population appearance was 0~2cm both in bare land and in abandoned field. Population number of buried seed germination was decreased according to soil depth. Crepidiastrum sonchifolium was a plant that population number of buried seed germination is the most. Similarity index was 0.33 in aerial part plants and buried seed plants, and 0.55 in bare land and abandoned field.
Root discs of 4-year-old ginseng, Panax ginseng C. A. Meyer, were inoculated with the higher($10^8$ colonyforming units(CFU)/ml) and lower($10^6\;or\;10^5$ CFU/ml) initial inoculum levels of a plant-growth promoting rhizobacterium(PGPR), Paenibacillus polymyxa GBR-1 to examine rot symptom development and bacterial population changes on the root discs. At the higher inoculum level, brown rot symptoms developed and expanded on the whole root discs in which the bacterial population increased continuously up to 4 days after inoculation. In light and electron microscopy, ginseng root cells on the inoculation sites were extensively decayed, which were characterized by dissolved cell walls and destructed cytoplasmic contents. However, no rot symptoms were developed and the bacterial population increased only during the initial two days of inoculation at the lower inoculum level($10^6$ CFU/ml) of P. polymyxa GBR-1. At the lower inoculum level($10^5$ CFU/ml), boundary layers with parallel periclinal cell divisions, structurally similar to wound periderm, were formed internal to the inoculation sites, beneath which the cells were intact containing numerous normal-looking starch granules and no disorganized cell organelles, suggesting that these structural features may be related to the suppression of symptom development, a histological defense mechanism.

This paper describes the variations in eight agronomic traits in three unadapted local landraces and an inbred cultivar of corn. To compare the agronomic traits in field evaluation with molecular marker evaluation the genotypes of the plant introduction were also checked by 4 microsatellite-SSR loci. The variations of the eight agronomic traits were higher in the local landrades than in the inbred line. which was substantiated by the high genetic variation in the landrades with microsatellite-SSR loci. The level of genetic variation was also different between landraces. Since the genetic evaluation can be easily quantified by the analysis of microsatellite-SSR loci. the threshold level of genetic homogeneity in the population for parental lines in breeding program can be determined and the effort of maintaining the landrace population would be alleviated. As an example in our analysis. the entry from Whachon should not need the same number of selfing generations as the other two landraces to get the level of inbred state. Since this line showed lowest intra-genetic variation within the population.

The results are summarized as follows: 1. Population density (PD) was extremely decreased by the times of year. 2. Pasture productivity was highest at 3 year old pasture, was lowest at 5 year old pasture and was recovered at 7 year old pasture. 3. Recovery of pasture productivity was followed the increase of dry weight of plant (DW/pl.) and number of tillers per plant (NT/pl.) 4. The population density of 7 year old pasture was 14 plants per square meter. 5. The complete genome sequence of a Slovak SL-1 isolate of Tomato mosaic virus (ToMV) was determined from the next generation sequencing (NGS) data, further confirming a limited sequence divergence in this tobamovirus species. Tomato genotypes Monalbo, Mobaci and Moperou, respectively carrying the susceptible tm-2 allele or the Tm-1 and Tm-2 resistant alleles, were tested for their susceptibility to ToMV SL-1. Although the three tomato genotypes accumulated ToMV SL-1 to similar amounts as judged by semiquantitative DAS-ELISA, they showed variations in the rate of infection and symptomatology. Possible differences in the intra-isolate variability and polymorphism between viral populations propagating in these tomato genotypes were evaluated by analysis of the capsid protein (CP) encoding region. Irrespective of genotype infected, the intra-isolate haplotype structure showed the presence of the same highly dominant CP sequence and the low level of population diversity (0.08-0.19%). Our results suggest that ToMV CP encoding sequence is relatively stable in the viral population during its replication in vivo and provides further demonstration that RNA viruses may show high sequence stability, probably as a result of purifying selection.
Sugarcane mosaic virus (SCMV) is one of the most damaging viruses infecting sugarcane, maize and some other graminaceous species around the world. To investigate the genetic diversity of SCMV in Iran, the coat protein (CP) gene sequences of 23 SCMV isolates from different hosts were determined. The nucleotide sequence identity among Iranian isolates was more than 96%. They shared nucleotide identities of 75.5-99.9% with those of other SCMV isolates available in GenBank, the highest with the Egyptian isolate EGY7-1 (97.5-99.9%). The results of phylogenetic analysis suggested five divergent evolutionary lineages that did not completely reflect the geographical origin or host plant of the isolates. Population genetic analysis revealed greater between-group than within-group evolutionary divergence values, further supporting the results of the phylogenetic analysis. Our results indicated that natural selection might have contributed to the evolution of isolates belonging to the five identified SCMV groups, with infrequent genetic exchanges occurring between them. Phylogenetic analyses and the estimation of genetic distance indicated that Iranian isolates have low genetic diversity. No recombination was found in the CP cistron of Iranian isolates and the CP gene was under negative selection. These findings provide a comprehensive analysis of the population structure and driving forces for the evolution of SCMV with implications for global exchange of sugarcane germplasm. Gene flow, selection and somehow homologous recombination were found to be the important evolutionary factors shaping the genetic structure of SCMV populations.
