Changes in the Concept of Nutrients and Transition of Plant Breeding (영양소의 변천과 식물육종의 추이)

  • During the first half of twentieth century, even though the importance of non-calorie essential micronutrients of 13 vitamins and 17 minerals has been known to alleviate nutritional disorder; the primary objective of agriculture and plant breeding programs has been to increase the productivity and seed yields, and macronutrients of proteins, fats, and carbohydrates made up the bulk of foodstuff which were used primarily as an energy source. In the last decade it has been found that non-essential micronutrients encompass a vast group of phytochemicals including antioxidants that are not strictly required in the diet but when present at sufficient levels work as health-promoting chemicals. Nowadays agricultural crops are grown for health rather than for food or fiber, and modifying the nutritional compositions of plant foods has become an urgent health issue. To ensure an adequate intake of essential vitamins and minerals, and to increase the consumption of health-promoting phytochemicals, the researches on plant secondary metabolism have been made. The attempt to improve nutritional quality of crops has been blocked by a lack of basic knowledge of plant metabolism. The advent of genomics era enabled new approaches to make crossing regardless of species, family, or phylum barriers, and the accumulation in our basic knowledge on plant secondary metabolism during the coming decade would be tremendous. As the major staple crops contain insufficient amount of many micronutrients, fortification strategy will be a necessary practice. Elevated intake of specific vitamins, C, E, and $\beta$-carotene, mineral selenium, antioxidants, and phytochemicals significantly reduces the risk of chronic disease such as cancer, cardiovascular disorder, diabetis, and other degenerative disease associated with aging. As the attempt to improve the nutritional quality of crops requires the basic knowledges on plant metabolism, plant biochemistry, human physiology, and food chemistry, strong interdisplinary collaboration among plant biotechnologists, human nutritionists, and food scientists will be needed. Inhibition of cancer, cardiovascular disease, and other degenerative disorder may be the biggest goal facing nutritional plant breeders. But the assumption that simply increasing dietary level of any compound will necessarily improve human health is a dangerous idea because many plant secondary products and dietary contaminants have paradoxical (hermetic) effects. Before biotechnical manipulation is undertaken to elevate or reduce any individual constituent of crops, the contribution of the micronutrient to human health must first be investigated.

소형 펀치 시험에 의한 강용접부의 파괴강도 평가에 관한 연구 1

  • It was reported that the toughness for welded region was influenced by various factors such as the gradient for prior austenite grain size, the variation of microhardness and the characteristic microstructure depending on distance from the fusion boundary. Therefore, in order to evaluate the fracture strength of the weldment in which the microstructures change continuously, it is important to assess the peculiar strength of each microstructure in welded region. It was known that the small punch(SP) test technique which was originally developed to study the irradiation damage effect for the structures of nuclear power plant was also useful to investigate the strength evaluating of nonhomogeneous materials. In this paper, by means of a small punch test technique the possibility of evaluating strength of parent and welded region in SS41 and SM53B steels was investigated. The obtained results are summerized as follows: 1) The small punch test which showed markedly the ductile-brittle transition behavior in this experiment may be applied to evaluation for the fracture strength of welded region. 2) It was shown that the ductile-brittle regime lied in Region III(plastic membrane stretching region) of the flow characteristics observed in SP test. 3) The SP test technique which shows a more precipitous energy change transition behavior than the other test technique is able to estimate the more precise transition temperature. 4) It could be seen that in comparision with the structure of parent the structure of weld HAZ in SS41 steel was improved while it in SM53B steel was deteriorated.

Identification of Transition Characteristics and Bio-concentration Factors of Heavy Metal (loid)s in the Selected Perennial Root Medicinal Plants

  • This study was conducted to identify transition characteristics of arsenic (As), cadmium (Cd), and lead (Pb) and to calculate bio-concentration factors (BCF) in the three perennial root medicinal plants, namely Codonopsis lanceolata (Deoduck), Platycodon grandiflorum (Balloon flower) and Panax ginseng (Korean ginseng) grown in major medicinal plant producing districts in Korea. Average BCF values ranged from 0.009~0.029 in As, 0.334~1.453 in Cd, and 0.021~0.023 in Pb in three perennial root medicinal plants. The BCF values increased in the order of ginseng (0.029) > deodeok (0.012) > balloon flower (0.009) for As, balloon flower (1.453) > deodeok (0.685) > ginseng (0.334) for Cd, and ginseng (0.023) > deodeok (0.022) > balloon flower (0.021) for Pb. The BCF values calculated in this study will be useful for predicting the uptake of heavy metal (loid)s. Further study on uptake and accumulation mechanism of toxic metal (loid)s by agricultural products is required to assess the human health risk associated with soil contamination.

Ecological characteristics and distribution of Chuiangne (Pyrus ussuriensis var. acidula) at habitats (자생 취앙네(Pyrus ussuriensis var. acidula) 분포와 생태적 특성)

  • Five major naturally populated habitats around the Mt. Daeduk, Gangwon province for wild Chuiangne (Pyrus ussuriensis var. acidula) are mostly located on the southeast side of the mountains at 1,216-1,306m in altitude at sea level and are in very good sunney areas. The total of 96 taxa dividing into 40 families, 79 genera 84 species and 12 varieties have in these areas. Most of these plants are heliophytes, which love sunshine, Compositaes such as Artemisia stolonifera, Synurus deltoides, or Rosaceaes like Potentilla fragarioides var. major are also shown. These areas have been unknown destroyed, so it is possibly thought the second transition has been progressed. The index of species diversity of plant groups in these areas is 4.715-5.270 based on the Shannon-Wiener's method. The wild Chuiangne with 34.15%, which are 5-10㎝ DBH, have been found in habitats. The vegetation around the habitats must be managed by the man in order to keep sound growth of the wild Chuiangne.

Brassinosteroids-mediated regulation of ABI3 is involved in high-temperature induced early flowering in plants

  • The interplay of plant hormones is one of the essential mechanisms for plant growth and development. A recent study reported that Brassinosteroids (BR) and ABSCISIC ACID (ABA) interact antagonistically in early seedling developments through the BR-mediated epigenetic repression of ABSCISIC ACID-INSENSITIVE 3 (ABI3). However, the other physiological roles of the BR-mediated regulation of ABI3 and ABA responses beyond early seedling developments remain largely unknown. Here, we showed that the activation of BR signaling by high temperatures promotes flowering time through the suppression of ABI3 expressions. Elevated ambient temperature induced early flowering in wild type Col-0 plants, but not in BR-defective bri1-116 mutant plants. Conversely, a hyper BR biosynthetic dwf4-D mutant displayed more sensitive thermomorphic long shoot elongation and early flowering. Both expression patterns and physiological responses supported the biological roles of ABI3 in the regulation of floral transition and reproduction under high temperature conditions. Finally, we confirmed that the lowered expressions of the transcript and protein levels of ABI3 brought on by elevated temperature were correlated with warmth-induced early flowering phenotypes. In conclusion, our data suggest that the BR- and warmth-mediated regulation of ABI3 are important in thermomorphic reproductive phase transitions in plants.

Ecological Characteristics and Distribution of Plant Resources of Pyrus and Malus sp. in Jindong Valley, Gangwon Province

  • Four major naturally populated areas around the Jindong valley in Inje-Gun, Gangwon province for wild Pyrus ussuriensis, Malus sieboldii, and Malus baccata are mostly located on the southeast side of the mountains at 630-745m in altitude at sea level and are in very good sunny areas. The total of 77 taxa dividing into 32 families, 58 genera, 65 species, and 12 varieties has been inhabited in these areas. Most of these plants are heliophytes, which love sunshine, but some of rhizo-plants such as Sasa borealis, climbing plants such as Actinidia arguta and Celastrus orbiculatus, or naturalized plants like Bidens frondosa are also shown. These areas have been continually destroyed, so it is possibly thought that the second transition has been progressed. The index of species diversity of plant groups in these areas is 0.672∼1.465 based on the Shannon-Wiener's method, but its index for the area that rhizo-plants like Sasa borealis are growing as an under planting decreases relatively. In this study, the oldest plant of Pyrus ussuriensis in Korea, which is 120cm in diameter at breast height (DBH), 19m in tree height and 25m in width, has been found.

MET1-Dependent DNA Methylation Represses Light Signaling and Influences Plant Regeneration in Arabidopsis

  • Plant somatic cells can be reprogrammed into a pluripotent cell mass, called callus, which can be subsequently used for de novo shoot regeneration through a two-step in vitro tissue culture method. MET1-dependent CG methylation has been implicated in plant regeneration in Arabidopsis, because the met1-3 mutant exhibits increased shoot regeneration compared with the wild-type. To understand the role of MET1 in de novo shoot regeneration, we compared the genome-wide DNA methylomes and transcriptomes of wildtype and met1-3 callus and leaf. The CG methylation patterns were largely unchanged during leaf-to-callus transition, suggesting that the altered regeneration phenotype of met1-3 was caused by the constitutively hypomethylated genes, independent of the tissue type. In particular, MET1-dependent CG methylation was observed at the blue light receptor genes, CRYPTOCHROME 1 (CRY1) and CRY2, which reduced their expression. Coexpression network analysis revealed that the CRY1 gene was closely linked to cytokinin signaling genes. Consistently, functional enrichment analysis of differentially expressed genes in met1-3 showed that gene ontology terms related to light and hormone signaling were overrepresented. Overall, our findings indicate that MET1-dependent repression of light and cytokinin signaling influences plant regeneration capacity and shoot identity establishment.

Evolution of Plant RNA Viruses and Mechanisms in Overcoming Plant Resistance (식물 RNA 바이러스의 진화와 병저항성 극복 기작)

  • Plant RNA viruses are one of the most destructive pathogens that cause a significant loss in crop production worldwide. They have evolved with high genetic diversity and adaptability due to the short replication cycle and high mutation rate during genome replication, which are characteristics of RNA viruses. Plant RNA viruses exist as quasispecies with high genetic diversity; thereby, a rapid population transition with new fitness can occur due to selective pressure resulting from environmental changes. Plant resistance can act as selective pressure and affect the fitness of the virus, which may lead to the emergence of resistance-breaking variants. In this paper, we introduced the evolutionary perspectives of plant RNA viruses and the driving forces in their evolution. Based on this, we discussed the mechanism of the emergence of variant viruses that overcome plant resistance. In addition, strategies for deploying plant resistance to viral diseases and improving resistance durability were discussed.

  • In order to develop the evaluation methods of degradation for the Ni-Cr-Mo-V steel, which is in use for turbine rotor in nuclear power plant, the degraded materials were prepared by simulated degradation methods. The result of impact test and fatigue crack growth test shows that the FATT(Fracture Appearance Transition Temperature) and fatigue crack growth rate increased with the increase of degradation. And the result of new electrochemical polarization test method was suggested for the evaluating FATT, fatigue crack growth exponent and coefficient C values based on the results of relationship between corrosion current density(Icorr) & FATT, and the m & C and Icorr.

  • We study on the implementation of reconfigurable position control system which is based on Open Control Platform(OCP) for Unmanned Expedition Vehicles(UEV) in ubiquitous environment. The control system uses hierarchical control structure and OCP structure which contains three layers such as core OCP, reconfigurable control API(Application Programmer Interface), generic hybrid control API. The goal of our research is to implement an UEV control system using advanced software technology. As a specific control problem, we study a transition management problem between PID control and neural network control depending on fault or parameter change of the plant, i.e., UEV. The concept of the OCP-based software-enabled control can provide synergy effect by the integration of software component, middleware, network communication, and control, and thus can be applied to various systems in ubiquitous environment.
