Nicotiana tabacum과 Petunia inflata의 전기적 원형질체융합에 의한 속간 체세포 잡종의 생성 (Intergeneric Somatic Hybrids by Electrofusion of Protoplasts Between Nicotiana tabacum and Petunia inflata)

  • Leaf mesophyll protoplasts of Nicotiana tabacum (nitrate reductase deficient mutant) were fused with cell suspension protoplasts of albino Petunia inflata in an electric field. Hybrid cell colonies were selected for nitrate reductase proficiency and chlorophyll synthesis. Five hybrid plant lines, regenerated from the selected calli lines, were analysed by electrophoresis, number of chromosomes and morphological characters. Somtic hybrid plants showed both parent patterns in the isozymesof isoleucine aminopeptidase and esterase. The hybrids had the expected chromosome number of 62 and exhibited an intermediate floral morphology when compared with the parents, but plant height and leaf arrangement were similar to N. tabacum.

한국산 차나무과의 화분분류학적 연구 (A Palynotaxonomic Study on the Korean Theaceae)

  • Pollen grains of six geners and six species from Korean Theaceae were observed with light and scanning electron microscopes. Pollen morphology divided the investigated species into the tricolporoidate Theoideae and the tricolporate Ternstroemiodideae. On the basis of surface sculpturing pattern, further palynological relationships and classificatin were understood. Some of the nomenclatural problems were discussed along with the palynological results.

Morphological and Molecular Characterization of Thamnocalamus falconeri Hook f. ex. Munro

  • The economy of India and so also of many Asian countries depends on bamboos and their uses are not only in domestic items but also in rural housing and raw materials to several industries and germplasm characterization is an important link between the conservation and utilization of plant genetic resources. Classical taxonomic studies of the bamboos are based on floral morphology and growth habit, which can cause problems in identification due to erratic flowering coupled with different biotic agencies and environmental factors. Identification and genetic relationships among accessions of Thamnocalamus falconeri were investigated using morphology and random amplified polymorphic DNAs (RAPD) technique. Analysis started by using 51 vegetative characters and forty two 10-mer primers that allowed us to distinguish different genotypes hailing from different eco- zones of Garhwal Himalayas (India). The selected primers (12) were used for identification and for establishing a profiling system to estimate genetic diversity. A total of 79.33% polymorphism was estimated by using 12 selected primers. The genetic similar analysis was conducted based on binary digits i.e. presence (1) or absence (0) of bands, which revealed a wide range of variability among the species whereas genetic relatedness was quite high based on vegetative characters. Cluster analysis clearly showed two major clusters for both of the markers viz. morphology and RAPD belonging to 10 accessions of T. falconeri. Two major clusters were further divided into minor clusters. Cluster based on RAPD marker showed grouping of accessions of closed locality whereas analogy was reported for vegetative traits. The RAPD technique has the potential for use in species identification and genetic relationships studies of bamboo for breeding program.

한국산 열당과 종자 형태의 분류학적 적용 (Systematic application of seed morphology In Korean Orobanchaceae)

  • 한국산 열당과의 종자형태를 조사하기 위하여 5속 5종 1 변종과 히말라야의 Boschniakia himalaica Hook. f. & Thomson in Hook. f.(총 7 분류군)의 종자를 주사전자현미경을 이용하여 관찰하였다. 한국산 열당과는 종자 크기, 종피의 구조, 과실 당 종자의 수에 따라 두 가지 유형이 구별 되었다. - Type I: 크기가 1mm 미만, 종피 구조는 망상, 과실 당 종자의 개수는 약 1,000개 이상. Type I은 외종피의 내부 표면 무늬에 따라 세 가지 하위구분으로 나뒤어 질 수 있다; (1) Subtype Ia: 망상 -불규칙한 줄무늬형(Ia')과 깊은 망상(Ia")을 포함한다. (2) Subtype Ib: 유공상, (3) Subtype Ic: 평활상. Type II: 크기가 1mm 이상, 종피구조는 불규칙하게 돌출된 모양(creasted), 과실 당 종자의 개수는 약 1,000개 미만(Lathraea japonica Miq.). 한국산 열당과에 대한 종자 형질의 분류학적 적용에 관하여 간략하게 검토하였다.

Effect of Inherent Anatomy of Plant Fibers on the Morphology of Carbon Synthesized from Them and Their Hydrogen Absorption Capacity

  • Carbon materials were synthesized by pyrolysis from fibers of Corn-straw (Zea mays), Rice-straw (Oryza sativa), Jute-straw (Corchorus capsularis) Bamboo (Bombax bambusa), Bagass (Saccharum officinarum), Cotton (Bombax malabaricum), and Coconut (Cocos nucifera); these materials were characterized by scanning electron microscope, X-ray diffraction (XRD), and Raman spectra. All carbon materials are micro sized with large pores or channel like morphology. The unique complex spongy, porous and channel like structure of Carbon shows a lot of similarity with the original anatomy of the plant fibers used as precursor. Waxy contents like tyloses and pits present on fiber tracheids that were seen in the inherent anatomy disappear after pyrolysis and only the carbon skeleton remained; XRD analysis shows that carbon shows the development of a (002) plane, with the exception of carbon obtained from bamboo, which shows a very crystalline character. Raman studies of all carbon materials showed the presence of G- and D-bands of almost equal intensities, suggesting the presence of graphitic carbon as well as a disordered graphitic structure. Carbon materials possessing lesser density, larger surface area, more graphitic with less of an $sp^3$ carbon contribution, and having pore sizes around $10{\mu}m$ favor hydrogen adsorption. Carbon materials synthesized from bagass meet these requirements most effectively, followed by cotton fiber, which was more effective than the carbon synthesized from the other plant fibers.

금강초롱꽃 (Hanabusaya asiatica)과 근연분류군의 비교연구 1. 외부형태 및 해부학적 형질 (Comparative studies on the Hanabusaya asiatica and its allied groups 1. External morphology and anatomical characters)

  • 조사된 금강초롱꽃과(科) 근연 분류군 4속 9분류군에 대하여 외부형태학적 형질을 재검토하고 해부학적 형질을 통하여 종간 분류 및 금강초롱꽃속(屬)이 특산속으로서 구별이 가능한가를 알아보았다. 연구결과, 외부형태에 있어서는 뿌리의 모양, 잎의 형태, 엽병의 날개 유무, 화관의 형태, 꽃받침의 형태, 엽병의 날개 유무, 화관의 형태, 꽃받침의 형태, 화서의 형태, 주두의 형태 등이 식별 형질로서 가치가 인정되었으며 해부학적 형질에서는 횡단면의 내부구조 보다는 표피의 돌출정도와 돌출 수가 종간 구별을 위한 형질로 생각되어진다. 이번 조사를 통하여 금강초롱꽃속(屬)은 외부형태 및 해부학적 형질에서 유사 분류군들과 잘 구분되어 특산속으로서 잘 지지되었으며 기존의 학자들(Fedorov, 1957; Nakai, 1952; Hong, 1983)에 의해 분류형질로 인식되어온 엽서와 화서의 형태, 화관의 형질 들이 이번 연구에서도 속, 종을 구별하는데 매우 유용한 형질로 나타났다.
