Factors Affecting Efficiency of Shoot Induction in Citrus junos Sieb.

  • To enhance the shoot induction efficiency from nodal stem of yooza, the culture conditions such as basal medium, carbohydrate source, solidifying agent and the optimum concentration of plant growth regulators for shoot induction were investigated. The nodal explants were cultured better on MS medium than on MT, SH, B5 or W media in considering of shoot induction rate and mean shoot length. Solidifying agent in medium was better with 0.8% agar than with 0.3% agar, 1.2% agarose or 0.2% gelrite. Carbohydrate source in shoot induction medium was efficient with 30 g/L sucrose. The optimum concentrations of plant growth regulators were determined that 0.1 mg/L NAA as auxin was effective on the shoot induction, and 1.0 mg/L BAP as cytokinin induced multiple shoots efficiently. Shoot induction was the most effective on MS medium supplemented with 4 mg/L $GA_3$ in yooza.

In vitro shoot proliferation of Alnus japonica (Thunberg) Steudel

  • In vitro proliferation system was achieved by using nodal segment excised from greenhouse grown juvenile stock plants of Alnus japonica. Stem explants were cultured on MS medium supplemented with different plant growth regulators of cytokinin and/or their combinations. The most effective cytokinin source was the combination of zeatin 2.0 mg/L and TDZ 0.05 mg/L producing the average number of shoots (16.8 $\pm$ 3.6). In addition, healthy roots were formed after small clumps of shoots were transferred to half strength of MS medium containing IBA 0.02 mg/L with optimal rooting capacity. Soil acclimatization was successfully conducted in cell tray containing artificially mixed soil with 92 % survival rate.

  • The effects of various dehiscent application such as dehiscent materials (big chaffs, vermiculite etc.), growth regulators and agricultural chemicals (plant protector.) on stimulation of dehiscence and shortening of dehiscent period were investigated Results obtained were as follows : 1. The moisture content of endosperm and seed coat at 10 day after dehiscent application amounts between 40% and 50%. 2. Endosperm diameter was increased with time of stratification, and the embryo growth showed in linear function, 3. Non-dehiscent seed showed also normally development of embryo, and the property of dehiscence dependent from physico-chemical nature of ginseng seed coat. 4. The best dehiscent materials were big chaffs and followed vermiculite, sand and sand with big chaffs. 5. The effect of dehiscence of ginseng seed showed higher activity in fungi than in bacteria in general. 6. Agricultural chemicals ( plant Protector) reduced the dehiscent rate of ginseng seed 7. The best timing of dehiscent treatment was between August 1 and August 10 but the smaller amount of dehiscent rate after August 10 dehiscent appllication indicated that big chaffs and growth regulator treatment may be controlled shortening of dehiscent period of ginseng seed.

Effect of Growth Regulators, Genotypes and Cutting Position on Rooting and Root Growth of Chrysanthemum zawadskii H. (九折草 揷穗로부터 發根 및 根生長에 미치는 생장조절물질, 九折草 種 및 揷穗位置의 효과)

  • This study was conducted to determine the effect of growth regulators, genotype, and cutting position on the rooting and root growth from cutting of Chrysanthemum zawadskii H.. Rooting rate of Keungugeolcho in the treatement of IBA 500 and 1000 ppm was the better than those of other treatments of IAA, NAA and Rooton. Rooting rate differed depending on the genotype. Hangryobonggugeolcho was better than Keungucheolcho in rooting rate. The treatment of rooton remarkably induced many roots from the cuttings of eight accessions of Chrysanthemum zawadskii H.. Also, rooting rate and number of root differed depending on cutting position. When cuttings including shoot tip were cultured on tray containing bed soil, rooting rate and number of root induced from cuttings with shoot tip was higher than when cuttings without shoot tip and with lateral axillary bud were cultured.

Effect of Growth Regulators and Antioxidant Mixture on the Anther Floating Cultures of Rice (벼의 약 부유배양에서 캘러스 유도 및 식물체 분화에 미치는 생장조절제와 항산화제의 영향)

  • To investigate the effect of growth regulators and antioxidant in anther floating cultures of rice, anthers were cultured in liquid media supplemented with different growth regulators, and the effect of antioxidant mixture : (Sigma Chemical Co.) on callus induction and plant regeneration were investigated. N6 medium with 1 mg/L 2,4-D and 1 mg/L kinetin was the best for rice anther floating cultures, which showed 48.5% of callus induction and 6.8% of green plant regeneration. The callus induction was not affected by antioxidant mixture in liquid medium, and antioxidant mixture (250 mg/L) was effective for the reduction of brownish callus and improvement of plant regeneration Antioxidant mixture showed better effectiveness when it was supplemented to both media for callus induction and plant regeneration.
