Development of Monoclonal Antibodies Specific to Galectin of Pine Wood Nematode, Bursaphelenchus xylophilus (Steiner and Buhrer) Nickle and Their Utilization for Detection of Pine Wood Nematodes (소나무재선충[Bursaphelenchus xylophilus (Steiner and Buhrer) Nickle]의 GaLectin에 대한 특이적인 단클론 항체 제작과 진단에의 활용)

  • Currently, there is no available tool that rapidly diagnoses pine wood nematode (PWN)-infected pine trees in the field. In this study, we synthesized and purified PWN Galectin, which might be an antigen specific to PWN, using the Baculovirus expression system. We used PWN Galectin as an antigen for generating 1,464 fusion hybridoma cell lines secreting monoclonal antibodies (Mabs). Among them, we selected 62 fusion hybridoma cell lines showing high reactivity to PWN Galectin. We further selected 12 fusion hybridoma cell lines showing high reactivity to the standard PWN-infected pine tree phosphate buffered saline (PBS) extract. Additionally, two fusion hybridoma cell lines showing no or extremely low reactivity were used as controls. The selected fusion hybridoma cell lines were subjected to limiting dilutions for selecting and establishing Mab-secreting cell lines showing higher reactivity to the standard PWN-infected pine tree extract than to the standard normal pine tree PBS extract. Moreover, the selected fusion hybridoma cell lines were further selected based on their higher reactivity to PWN protein extracts than to three non-pathogenic nematode protein extracts. The Mab-secreting cell lines established in this study could be used to develop rapid diagnostic tools that can be used in the field or in laboratories for detecting PWN-infected pine trees or PWN.

Risk Assessment of Pine Tree Dieback in Uljin and Bonghwa (울진·봉화 일대 금강소나무 고사 피해 특성 분석)

  • Tree dieback in Geumgang pine forest has occurred in Uljin and Bonghwa since the 2010s. In order to identify status of tree dieback and prevent further damages, a monitoring project for tree dieback in Geumgang pine forest had been launched by Southern regional office of forest service in 2020. This study was conducted to understand the characteristics of tree dieback occurrence and assess the high risk areas using the occurrence data in the project. Pine tree dieback occurred frequently in areas with mountain ridges in high elevation, dry south-facing slopes, mature stands, and high temperature rise in winter. Furthermore, the result of risk assessment showed that 6.2 percent(5,294ha) of Geumgang pine forest(85,000 ha) in total study area are at high risk of tree dieback. As the pine trees in the high risk area are prone to experience the dieback due to temperature and drought-related extreme weather events, regular forest management activities are needed to reduce the drought stress of pine trees. Forest health management for the pine forest with high protection priority can be also useful strategy to counter the risk of decline. This results can be used as the basic information for the adaptive forest management to climate change.

  • Ring-width chronologies of Scots pine (Pinus sylvestris L.) and Norway spruce (Picea abies (L.) Karst.) from two experimental forest plots in Kaunas, Lithuania were developed to study tree growth-climate relationship in different geohydrological conditions using response function analysis. The tree ring-width chronologies of Scots pine ranged from 1883~1987 A.D. and 1864~1989 A.D., and Norway spruce 1838~1987 A.D. and 2870~1989 A.D., respectively. The response function analysis has vividly demonstrated that the growth of Scots pine is favoured by warm summer and Norway spruce by cool and moist summer. Spring temperature has shown direct relationship with tree growth of both the species. There also exists notable intraspecies analogies in growth responses except some minor differences.

  • Noticed that large number of the Korean white pine (Pinus koraiensis S. et Z.) has lost its typical excurrent tree from by removal of apical shoot in the process of cone harvesting, the investigation and analysis for the species were made on the basis of the relationship among DBH, total height, merchantable height, and merchantable volume, compared with normally grown Japanese larch (Larix leptolepis Gordon). One hundred sample trees for each species were selected in the same aged stands for the study. Results have indicated that the pine has shown abnormal height growth pattern mainly by the disturbance of external artificial force. The pine's forked and/or crooked stems by the reason lave led the result of less merchantable volume in the same DBH classes. Consequently, the pine would have undesirable tree form in terms of timber production, unless free from apical shoot disturbances. Considered to the merit of the Korean white pine that can produce timber and pine nuts, management alternatives for the pine stand were suggested to achieve maximum rate of return for the silvicultural investment.

Confirmation of Fruiting Bodies of Tricholoma matsutake Occurring from Matsutake-infected Pine Trees Using Microsatellite Markers (Microsatellite marker를 이용하여 송이감염묘로부터 발생한 송이 확인)

  • Tricholoma matsutake is commercially important wild edible mushroom. The only method for cultivation of T. matsutake has been to use matsutake-infected pine trees. In this study, twenty-one fruiting bodies were collected from the matsutake-infected pine trees in the Hongcheon experimental site in 2020. The fruiting bodies from the existing production area of matsutake-infected pine tree and from its transplantation site were found to be identical through monitoring and analysis of 13 microsatellite markers. Transplanted matsutake-infected pine trees succeeded in fairy-ring formation and matsutake fruiting in 2010 and 2017 to 2020. In conclusion, the matsutake mycelium of transplanted matsutake-infected pine tree originates from the existing matsutake-infected pine tree production.

Carbon Storage of Exotic Slash Pine Plantations in Subtropical China

  • Exotic conifer trees have been extensively planted in southern China because of their high apparent growth and yield. These fast-growing plantations are expected to persist as a considerable potential for temporary and long-term carbon sink to offset greenhouse gas emissions. However, information on the carbon storage across different age ranges in exotic pine plantations is often lacking. We first estimated the ecosystem carbon storage across different age ranges of exotic pine plantations in China by quantifying above- and below-ground ecosystem carbon pools. The carbon storage of each tree component of exotic pine (Pinus elliottii) increased significantly with increasing age in Duchang and Yiyang areas. The stem carbon storage except <10 years in Ji'an areas was the largest component among all other components, which accounts for about 50% of the total carbon storage followed by roots (~28%), branches (~18%), and foliage (~9%). The mean total tree carbon storage of slash pine plantations for <10, 10-20 and 20-30 years across three study areas was 3.69, 13.91 and $20.57Mg\;ha^{-1}$, respectively. The carbon stocks in understory and forest floor were age-independent. Total tree and soil were two dominant carbon pools in slash pine plantations at all age sequences. The carbon contribution of aboveground ecosystem increased with increasing age, while that of belowground ecosystem declined. The mean total ecosystem carbon storage of slash pine plantations for <10, 10-20 and 20-30 years across China was 30.26, 98.66 and $98.89Mg\;ha^{-1}$, respectively. Although subtropical climate in China was suitable for slash pine growth, the mean total carbon stocks in slash pine plantations at all age sequences from China were lower than that values reported in American slash pine plantations.

Topographic and Meteorological Characteristics of Pinus densiflora Dieback Areas in Sogwang-Ri, Uljin (울진 소광리 산림유전자원보호구역 내 금강소나무 고사지역의 지형 환경 특성 분석)

  • Korean Red Pine (Pinus densiflora) has been protected and used as the most ecologically and socio-culturally important tree species in Korea. However, as dieback of Korean red pines has occurred in the protected area of the forest genetic resources. The aims of this study is to identify causes for dieback of pine tree by investigating topographical characteristics of pine tree dieback and its correlation to meteorological factors. We extracted the dead trees from the time series aerial images and analyzed geomorphological characteristics of dead tree concentration area. As a result, 1,956 dead pine trees were extracted in the study region of 2,600 ha. Dieback of pine trees was found mostly in the areas with high altitude, high solar radiation, low topographic wetness index, south and south-west slopes, ridgelines, and high wind exposure compared to other living pine forest area. These areas are classified as high temperature and high drought stress regions due to micro-climatic characteristics affected by topographic factors. As high temperature and drought stress are generally increasing with climate change, we can evaluated that a risk of pine tree dieback is also increasing. Based on these geomorphological characteristics, we developed a pine tree dieback risk map using Maximum Entropy Model (MaxEnt), and it can be useful for establishing Korean red pine protection and management strategies.

Studies on Feed Values of Oak(Quercus serrata Thunb.) and Pine (Pinus densiflora) Browses (졸참나무지엽과 소나무지엽의 사료가치에 관한 연구)

  • Chemical composition, mineral compounds, cellular constituents, digestibility for dry matter, total digestible nutrients, and tannin constituents were investigated to evaluate feed value for oak and pine browses to use effectively browses as forest by products. The results of this study were as follows : Crude protein was 6.00∼7.89% for oak browse and 5.15∼6.06% for pine browse sampled through July to October, It gradually decreased in oak browse but not in pine browse as the growth of tree was progressed stages. Crude fat content of oak and pine browse were 1.90∼2.68% and 6.30∼6.33%. Crude fiber content was 33.3∼35.2% for oak browse and 33.7∼34.8% for pine browse and they tended to increase according to the growth stage of trees. Crude ash was 3.13∼2.78% for oak browse and 2.11 ∼1.93% for pine browse, respectively. Ca, Mg, Mn and Cu content were higher in oak browse than those in pine browse, but P, K, Na, Fe, and Zn content were similar each other. Mineral contents were decreased as the growth of tree was progressed(P<.05). Acid detergent fiber content of pine and oak browse were 47.7∼52.0% and 46.1∼48.1%, and they increased as the tree grew. Neutral detergent fiber content of pine and oak browse were 64.1∼67.9% and 65.0∼66.5%. NDF content was increased according to the tree growth in pine browse but it was the same in oak browse(P<.05). in vitro digestibility of dry matter was 51.7∼48.4% for pine browse that decreased according to the tree growth(p<.05) and 53.0∼5l.4% for oak browse. Total digestible nutrients of oak browse was 50.9∼52.4% and that of pine browse was 47.7∼51.1% which decreased as the tree grew. Oak browse showed the same relative feeding value(RFV) as pine browse, and it decreased as the growth of tree was progressed(P<.05). Tannin contents was 2.96% for oak browse, 6.27% for pine browse. Tannin contents decreased when browses were dried.

Consideration of Silvicultural Practice by Taking Community Type of Pinus densiflora Stand (식생형을 고려한 소나무 임분의 조림적 고찰)

  • In Korea, Red pine(Pinus densiflora) stands at a very important place, historically, culturally, and emotionally and it is one of the tree species that can produce domestic timber as forest resources economically. The growing space for Red pine forest in Korea is gradually reducing while the space for deciduous tree forests including oak series is gradually increasing. Thus, it is required that the research work should be done on renewal for pine forest retention and its related forest management practices. This research aims at developing stable, sustainable management forests and inducing nature renewal by grasping growth environment and succession process through the pattern of stand and structure analysis of the red pine(Pinus densiflora) for central region. The pine forests in central region were classified into four communities, such as Acer pseudo-sibolianum, Quercus acutissima, Lindera erythrocarpa, and Pinus densiflora and they are showing different characteristics by pattern according to their growing district. There appeared a lot of red pines in the upper layer, but there existed high weight of broad-leaved forest tree species, such as oak series in the middle layer and the appearance of the red pine was meager in the lower layer. Therefore, it appears that the red pine has a high influential ecological strength in terms of correlativity; however, if the red pine in tree layer is dead by aging in nature state with the lapse of time, it is considered that oak species, such as Quercus mongolica, Quercus variabilis, and Quercus serrata will be dominant below sub-tree layer. In addition, there appear differences in characteristics and growth pattern of the red pine forest depending on stand pattern, so it is considered that a differential application method should be employed in the pine forest management.

A Comparison of Litterfall Dynamics in Three Coniferous Plantations of Identical Age under Similar Site Conditions

  • This study was carried out to evaluate litterfall dynamics in three adjacent coniferous tree plantations (larch: Larix leptolepis; red pine: Pinus densiflora; rigitaeda pine: P. rigitaeda) planted in the same year (1963), and growing under similar environmental conditions in the Sambong Exhibition Forests, Hamyang-gun, Gyeongsangnam-do. Litter was collected monthly between July 2006 and June 2008. Needle, broad leaf and total litter inputs followed a similar monthly pattern in the three coniferous plantations. The amounts of needles, flowers, and miscellaneous litter were significantly lower in the larch than in the two pine plantations, while branch litter was significantly higher in the larch than in the two pine plantations. Average total litterfall for two years was significantly higher for the pine (5,475 kg $ha^{-1}\;yr^{-1}$ for red pine and 5,290 kg $ha^{-1}\;yr^{-1}$ for rigitaeda pine) plantations than for the larch (3,953 kg $ha^{-1}\;yr^{-1}$ plantation. Needle litter comprised about 73.1% of total litterfall for the rigitaeda pine, 70.8% for the red pine and 62.9% for the larch plantations. Our results demonstrate that litterfall inputs can be affected by tree species.
