Preparation for Protein Separation of an Ion-Exchange Polymeric Stationary Phase Presenting Amino Acid and Amine Units Through Surface Graft Polymerization

  • Ion-exchange polymeric stationary phases presenting amino acid and amino groups were prepared by the surface grafting of glycidyl methacrylate onto a silica gel surface and subsequent amination. Three kinds of amino acids-L-arginine (Arg), D-lysine (Lys), and D-histine (His)-were used in this study. An ion-exchange polymeric stationary phase presenting ethylene diamine (EDA) was also prepared by surface graft polymerization. Separation of the model proteins bovine serum albumin (BSA), chick egg albumin (CEA), and hemoglobin (Hb) was performed using the amino acid- and amine-derived columns. In separating the CEA/BSA mixture, the resolution time of BSA was longer than that of CEA when using the EDA column, whereas the resolution time of BSA was shorter than that of CEA when using the Arg, Lys, and His columns. In the separation of the Hb/BSA mixture, the resolution time of BSA was longer than that of Hb in the EDA column, whereas the resolution time of BSA was shorter than that of Hb in the amino acid columns (D-Lys, L-Arg, and D-His).

  • Three different style capillary columns, a packed capillary with temporary quartz wool frit, a packed capillary with immobilized frit, and an immobilized packed-capillary, were easily prepared with a commercially available (S)-N-(3,5-dinitro-benzoyl)leucine-N-phenyl-N-alkylamide derived chiral stationary phase. Liquid chromatographic chiral separations of some racemic amino acid derivatives on these columns were performed and the results were compared to each other. The packed capillary with immobilized frit showed some merits in chiral chromatography.

  • The efficiency of complex slurry preparation route for the development of high performance RS-SiCf/SiC composites has been investigated. The green bodies for RS-SiC and RS-SiCf/SiC composite materials prior to the infiltration of molten silicon were prepared with various C/SiC complex matrix slurries, which associated with both different sizes of starting SiC particles and blending ratios of starting SiC and carbon particles. The reinforcing materials in the composite system were uncoated and C coated Tyranno SA SiC fiber. The characterization of RS-SiC and RS-SiCf/SiC composite materials was examined by means of SEM, EDS and three point bending test. Based on the mechanical property-microstructure correlation, process optimization methodology is discussed.

Preparation of Ag, Pd, and Pt50-Ru50 colloids prepared by γ-irradiation and electron beam and electrochemical immobilization on gold surface

  • PVP-protected Ag, Pd and $Pt_{50}-Ru_{50}$ colloids were prepared independently by using ${\gamma}$-irradiation and electron beam (EB) at ambient temperature. UV-visible spectra of these colloids show the characteristic bands of surface resonance and give evidence for the formation of nanoparticles. Transmission electron microscopy (TEM) experiments were used to know the morphology of nanoparticles prepared by ${\gamma}$-irradiation and EB. The size of Ag, Pd, and $Pt_{50}-Ru_{50}$ nanoparticles prepared by ${\gamma}$-irradiation was ca. 13, 2-3, 15 nm, respectively. While, the size of Ag, Pd, and $Pt_{50}-Ru_{50}$ nanoparticles prepared by EB was ca. 10, 6, and 1-3 nm, respectively. Cyclic voltamograms (CV) were recorded for the Au electrodes immobilized with these nanoparticles. CVs indicated the modifications in the surface as a result of immobilization.

  • The various size and morphology microcapsules were prepared to produce smell sweet by heating condensation reaction of melamine and formaldehyde using 5-types of surfactants such as the laurylbenzenesulfonic acid sodium salt (SDS), polyvinylpyrrolidon (PVP), polyvinyl alcohol (PVA), Span-80 and 2-acrylamido-2-methyl-1-1 propanesulfonic acid (AMP). As result it was found that the size and morphology of microcapsule is intimately associated with a kind of surfactants. In order to prepare microcapsule with antibacterial, the silver nanoparticle was prepared by gamma-irradiation. microcapsule with silver nanoparticle was prepared.

Effects of Epidermal Growth Factor on the Apoptosis and Implantation Related Genes in Bovine Embryos Developing in vitro

  • Epidermal growth factor (EGF) induces well-documented mitogenic and differentiating effects on murine and bovine preimplantation embryos. However, the effects of EGF on apoptosis and implantation-related gene expression in bovine embryos developing in vitro have not been evaluated. The objective of this study was to determine the effects of exogenous EGF in the presence and absence of BSA on the preimplantation development of bovine embryos. In addition, we measured cell number, apoptosis, and expression of apoptosis and implantation-related genes of the blastocysts that developed in these culture conditions. In vitro produced bovine embryos were randomly cultured in the same medium containing 0 or 10 ng/ml EGF in the presence and absence of 0.8% BSA. More 2-cell embryos developed into blastocysts at day 7 when BSA was present than when BSA was absent. The addition of 10 ng/$m\ell$ EGF into the medium did not significantly increase the developmental rate and the cell numbers per blastocyst. However, addition of EGF in the presence of 0.8% BSA significantly reduced the degree of apoptosis in the blastocysts (P<0.01). To investigate whether EGF modulates mRNA expression of apoptosis-related genes, mRNA was prepared from single blastocysts and each preparation was subjected to RT-PCR for Bcl-2 and Bax transcripts. EGF did not alter the relative abundance of Bax gene expression in the presence of BSA, but increase Bcl-2 (P<0.01) The relative abundance of Interferon tau expression was increased by EGF treatment in the presence of BSA. These results suggest that EGF and BSA synergistically enhance Bcl-2 and interferone tau gene expression, which may result in a net increase in viability in bovine embryos.

  • The optical near-field patterns, propagation loss and mode sizes of x-cut $Ti:LiNbO_3$ optical waveguide which was fabricated by Ti-diffusion varying with Ti strip thickness in wet oxygen atmosphere were discussed at optical wavelength 1550nm. As Ti thickness increased from $760{\AA}$, the insertion loss of waveguide was decreased. But at Ti thickness $1500{\AA}$, mode sizes are widely broadened. The Ti thickness of below $1100{\AA}$ and above $1500{\AA}$ showed negative effects to propagation loss and fiber coupling. The best Ti thickness for fabricating low propagation loss and good fiber coupling was inferred to be between $1100{\AA}-1500{\AA}$ in our conditions. And for Ti thickness $1150{\AA}$, its propagation loss, horizontal/vertical mode sizes were showed 1.61 dB/cm, $11.9/8.9{\mu}m$ for TM, 0.22 dB/cm, $12.0/9.1{\mu}m$ for TE respectively.

  • Objectives : The purpose of this study was to investigate the interpretation of the Pyeongwi-san(PWS) prescription in order to obtain the evidence for clinical applications. Methods and Result : We interpreted on the PWS-related contents based on 15 classic books, analyzed it according to followed categories ; origin, indication and precaution, usage, meaning of name, physiological and pathological situation of PWS, explain about each herbs. Conclusion : 1. PWS was first mentioned in the Bakjebang, its application was enhancing appetite. Application of PWS not only expanded digestive disease, infectious disease, and pain in articular, but also used to toniyfing drug. 2. Generous application method of PWS was ‘Sujunbok(水煎服)’. Pill preparation(丸劑) and decoction with salt(鹽湯劑) were also used 3. "Pyeongwi(平胃)“ in PWS means "flatten the stomach". There are some theory about methodology to "Pyeongwi (平胃)“, one is "drain the pathogen in stomach(敦阜說)”, other is "tonifying stomach's healthy qi(卑監說)“, the other is compromise theory. 4. Most medical literature related application situation of PWS to dampness(濕邪) in stomach. In Gangsulwongobangsunju, there are more detailed explain focused on metal qi(金氣), and fire qi(火氣) 5. Atractylodes japonica Koidz(蒼朮) drys dampness and fortifys the spleen, Magnolia officinalis Rehder(厚朴) eliminates dampness and treats fullness, Citrus reticulata Blanco(陳皮) moves and drains qi, Glycyrrhiza uralensis Fisch(甘草) tonify and harmonize in herbal formula

Preparation and characterization of CdS nanoparticle on the surface of silica nanoparticles (실리카 나노입자 표면에 CdS 나노입자의 제조 및 평가)

  • Poly(vinylpyrrolidone) stabilized cadmium sulfide (CdS) nanoparticles were loaded onto the surface of silica ($SiO_2$) nanoparticles by using ${\gamma}$-irradiation. TEM micrograph reveals the presence of ~20nm sized CdS nanoparticles on the surface of $SiO_2$ nanoparticles. XRD patterns confirm the crystalline. PL spectra of the simple PVP-stabilized CdS nanoparticle and $SiO_2$@CdS composite confirm the differences in the emission characteristics between them. Two prominent emission peaks were noted around 550 nm and 600 nm for PVP-stabilized CdS nanoparticles). The emission peaks noted for the PVP-stabilized CdS nanoparticles were found to be blue shifted for $SiO_2$@CdS composites. Besides, an additional emission peak around 450 nm was noticed for the $SiO_2$@CdS composite. The presence of CdS nanoparticles influence the emission characteristics and induce quantum confinement effect.
