Inhibitory effects of ultraviolet-C light and thermal treatment on four fungi isolated from pig slaughterhouses in Korea

  • Pig slaughterhouses harbor high humidity because of the necessary cleaning that takes place simultaneously with slaughter, which facilitates the existence of mold. Due to the enclosed space, there are several limitations to the control of mold growth with respect to cleaning, ventilation, and drying. In this study, the prevalence of fungi was investigated in four pig slaughterhouses in Korea. Four fungi (Aspergillus niger, Penicillium commune, Penicillium oxalicum, and Cladosporium cladosporioides) were detected with the highest frequency. These four strains were subjected to various treatments to reduce their growth. The fungi were inoculated onto stainless steel (SS) chips and treated with ultraviolet (UV)-C irradiation and hot water. Individual treatments with UV-C (15, 30, 90, 150, 300, and 600 mJ/cm2), and hot water (60, 65, 70, and 83℃) were performed to sanitize the SS chips. Simultaneous cleaning with 60℃ hot water and more than 150 mJ/cm2 of UV-C reduced the fungal incidence by > 6.5 Log from 6.6-7.0 Log CFU/cm2 (initial count). Our results demonstrate that a combined treatment of UV-C and hot water is the most economical and convenient way to prevent microbiological contamination of small tools (such as knives and sharpeners) and steel surfaces in slaughterhouses.

Studies on Leptospiral Antibody in Korean Cattle and Pigs (Leptospira 속균(屬菌)에 대한 한우(韓牛)와 돈(豚)의 혈중항체조사(血中抗體調査))

  • Investigation of leptospiral antibody in Korean cattle and pigs was carried out from February to October, 1971. Ten different living antigens, namely L. icterohaemorrhagiae, L. canicola, L. antumnalis, L. hebdomadis, L. australis A, L. pomona, L. pyrogenes, L. grippotyphosa, L. bataviae and L. javanica, were used. A total of 590 Korean cattle and 460 pig blood samples collected from Seoul Majang-dong slaughterhouse were tested by the rapid microscopic agglutination test. Throughout the studies the following results were obtained and summarized. 1. Of 590 serum samples of Korean cattle 51 were positive(8.64%). 2. Of 460 serum samples of pigs 27 were positive (5.87%). 3. Of 51 positive cattle samples, 29(4.92%) showed antibody to a serotype of L. icterohaemorrhagiae and 18(3.0%) to L. canicola, and 4 (0.68%) to L. pomona. Eight of L. icterohaemorrhagiae positive samples showed a cross reaction to L. canicola. 4. Of 27 positive pig samples, 14(3.04%) showed antibody to L. ictereohaemorrhagiae and 7(1.52%) to L. grippotyphosa. 4(0.87%) to L. canicola, 2(0.43%) to L. pomona. Two of L. canicola positive samples showed a cross reaction to L. grippotyphosa. 5. Serum samples of seven pigs, showing antibody positive to L. grippotyphosa were first observed in Korea. 6. Infection rate of bovine and porcine leptospirosis, in Korea, appeared to be lower than that of Japan, Taiwan, Thailand and the Philippines.

Biochemical Methane Potential of Animal Manure and Cultivated Forage Crops at the Reclaimed Tideland (가축분뇨와 간척지 사료작물의 메탄발생량)

  • Anaerobic biodegradability(AB), which can be determined with the ultimate methane yield by the decomposition of organic materials, is one of the important parameters for the design and the operation of anaerobic digestion plant. In this study, Biochemical Methane Potential(BMP) test has been carried out to evaluate the methane yield of animal manures, such as pig and cattle slurries, and different forage crops cultivated at the reclaimed tideland, such as maize, sorghum, barley, rye, Italian ryegrass(IRG), rape, rush and sludge produced from slaughterhouse wastewater treatment plant(SWTP). In the ultimate methane yield of animal manure, that of pig slurry(no used a EM) was 407 $mlCH_4/gVS_{fed}$ higher than 242 $mlCH_4/gVS_{fed}$ of cattle slurry. The ultimate methane yield of spike-crop rye was 442.36 $mlCH_4/gVS_{fed}$ the highest among different forage crops, the other showed the value above a methane yield of 300 $mlCH_4/gVS_{fed}$. The forage crop could be used as a good substrate to improve the methane production in anaerobic co-digestion together with animal manure.

  • This study was carried out to examine properties of water resistant plywood by using serum protein adhesive which is natural, environment-friendly and human-friendly. For the preparation of the serum protein adhesive, pig blood from slaughterhouse was centrifuged and serum was separated from corpuscles and concentrated to 30% by dry weight basis. This concentrated serum protein was modified with PF resin (50% NVC) with the ratio of 9 : 2.5. Plywood made by this modified serum protein gave 1.21 N/$mm^2$ of dry bonding strength, 0.80 N/$mm^2$ of wet boil bonding strength, 0% of cyclic delamination test value, and 0.025 ppm of HCHO emission, which met the excellent super $E_0$ grade and water resistant plywood.

Comparative Genomics Approaches to Understanding Virulence and Antimicrobial Resistance of Salmonella Typhimurium ST1539 Isolated from a Poultry Slaughterhouse in Korea

  • Non-typhoidal Salmonella (NTS) is one of the most frequent causes of bacterial foodborne illnesses. Considering that the main reservoir of NTS is the intestinal tract of livestock, foods of animal origin are regarded as the main vehicles of Salmonella infection. In particular, poultry colonized with Salmonella Typhimurium (S. Typhimurium), a dominant serotype responsible for human infections, do not exhibit overt signs and symptoms, thereby posing a potential health risk to humans. In this study, comparative genomics approaches were applied to two S. Typhimurium strains, ST1539 and ST1120, isolated from a duck slaughterhouse and a pig farm, respectively, to characterize their virulence and antimicrobial resistance-associated genomic determinants. ST1539 containing a chromosome (4,905,039 bp; 4,403 CDSs) and a plasmid (93,876 bp; 96 CDSs) was phylogenetically distinct from other S. Typhimurium strains such as ST1120 and LT2. Compared to the ST1120 genome (previously deposited in GenBank; CP021909.1 and CP021910.1), ST1539 possesses more virulence determinants, including ST64B prophage, plasmid spv operon encoding virulence factors, genes encoding SseJ effector, Rck invasin, and biofilm-forming factors (bcf operon and pefAB). In accordance with the in silico prediction, ST1539 exhibited higher cytotoxicity against epithelial cells, better survival inside macrophage cells, and faster mice-killing activity than ST1120. However, ST1539 showed less resistance against antibiotics than ST1120, which may be attributed to the multiple resistanceassociated genes in the ST1120 chromosome. The accumulation of comparative genomics data on S. Typhimurium isolates from livestock would enrich our understanding of strategies Salmonella employs to adapt to diverse host animals.

Selection of Early Cleaved Embryos and Optimal Recipients to Improve Efficiency of Pig Cloning

  • Early cleavage is a reliable prognostic tool for successful embryo transfer in assisted reproduction because early cleaved embryo show better pregnancy rate after transfer. There for, preparation of good embryo recipient is important factor to optimize efficiency of pig cloning. The present study was performed to evaluate the effect of early cleavage on the in vivo development of cloned embryos and to analyze breed, parity and estrous synchrony to optimize recipient for pig cloning. In vitro matured porcine oocytes derived from local slaughterhouse and fibroblasts derived from miniature pig fetuses were used for somatic cell nuclear transfer (SCNT). Reconstructed embryos were transferred to recipient pigs on the same day of SCNT or after 1~2 days of in vitro culture for selecting early cleaved embryos. Breed, parity and date of standing estrous of recipients were recorded for analysis. After 25~35 days after embryo transfer pregnancy was diagnosed using ultrasonography, and pregnant recipients were monitored till delivery. Between purebred and crossbred, no significant difference was founded in both pregnancy and delivery rates. However, early cleaved embryos showed significantly higher pregnancy (46.2%) and delivery (12.8%) rates compared to non-selectively transferred group (24.8% and 4.5%, respectively). The results also showed that the recipients showing standing estrous on the same day of SCNT and less than 4 parities were most suitable for pig cloning.

Comparision of carcass characteristics, meat quality, and fatty acid profiles between Duroc and corssbred pigs (Duroc × Korean native pig) (두록과 교잡돈(두록×재래돼지)의 도체 및 육질 특성과 지방산 조성 비교)

  • This study was conducted to develop new crossbred pig using Korean native pig and Duroc. Fifteen pigs of pure Duroc (D) and 26 crossbred gilts (15 of DK1 and 11 of DK2) were reared until $195{\pm}5$ days old, then slaughtered at local slaughterhouse. Pork loin was gathered and vacuum packed from left carcass after 24 h of slaughter to analyze meat quality traits, such as color, pH, water holding capacity (WHC), cooking loss and shear force (SF), and free fatty acid composition. Live weight and carcass weight of Duroc and DK1 were $119.1{\pm}8.7/82.91{\pm}6.1kg$ and $116.3{\pm}6.3/80.91{\pm}4.4kg$, respectively, and it was significantly higher than those of DK2 ($104.7{\pm}8.4/71.36{\pm}5.7kg$) (p<0.05).There was no significant difference in proximate composition between animal groups, however DK2 showed significantly lower shear force (SF) and higher water holding capacity (WHC) than other groups (p<0.05). The redness of DK2 also showed significantly higher than Duroc (p<0.05), however there was no significant difference in lightness and yellowness (p>0.05). DK1 showed the highest monounsaturated fatty acids (MUFAs) contents of $51.45{\pm}2.0%$ and DK2 showed the highest polyunsaturated fatty acids (PUFAs) contents of $8.98{\pm}1.4%$, however there was no significantly difference in PUFA/SFA ratio between pig groups (p>0.05). Duroc and DK2 contain significantly higher amount of linoleic ($7.99{\pm}1.2$ and $8.11{\pm}1.3%$, respectively) and linolenic acid ($0.43{\pm}0.1$ and $0.44{\pm}0.1%$, respectively) than DK1, and DK1 contains significantly higher amount of oleic acid ($47.32{\pm}1.8%$) than others (p<0.05).

Production of Embryos by Intracytoplasmic Sperm Injection (ICSI) in Pig

  • The objective of this study was to assess the development of porcine follicular oocytes fertilized by ICSI. Cumulus-oocyte-complexes (COCs) were collected by aspiration from follicles of 2-7 mm in diameter from a local slaughterhouse ovaries. Oocytes matured for 40-44 h were centrifuged at 12,000g for 6 min and then injected with sperm prepared by swim-up procedure in the presence or absence of 5 mM dithiothreitol (DTT). Injected oocytes were cultured in NCSU 23 medium during 6 to 8 days. IVF controls were compared to those of resulting embryos. The results obtained were as. follow: 1, The rates of cleavage and development rates into blastocyst by ICSI were not significantly (P<0.05) different between with (53.0% and 19.7%) or without (48.3% and 23.8%) centrifugation, respectively. 2. The cleavage and developmental rates to blastocyst after ICSI with or without 5mM DTT treated-sperm were not significantly (P<0.05) different (60.4% vs 16.4% and 48.5% vs 22.2%, respectively). 3. The cleavage and the developmental rates to blastocyst were not significantly (P<0.05) different between the zygotes obtained by IVF (51.8% vs. 22.4%) and ICSI (51.4% vs. 21.6%). 4. The number of blastomere in blastocyst stages after IVF or ICSI was not significantly different (46.7 $\pm$2.9 and 41.9$\pm$4.6).

In this study, Biochemical methane potential (BMP) test has been carried out to evaluate the methane yields of animal manures such as pig and cattle slurries, and different forage crops cultivated at the reclaimed tideland such as maize, sorghum, barley, rye, Italian ryegrass (IRG), rape, rush, and waste sludge produced from slaughterhouse wastewater treatment plant (SSWTP). In the ultimate methane yield and biodegradability of animal manure, those of pig slurry were 345 $mlCH_4/gVS_{fed}$ and 44.7% higher than 247 $mlCH_4/gVS_{fed}$ and 46.4% of cattle slurry (Cat. 2). The ultimate methane yield and biodegradability of spike-crop rye (Rye 1) were 442.36 $mlCH_4/gVS_{fed}$ and 86.5% the highest among different forage crops, those of the other forage crops ranged from 306.6 to 379 $mlCH_4/gVS_{fed}$ of methane yield with the AB having the range of about 60 to 77%. Therefore the forage crops could be used as a good substrate to increase the methane production and to improve the biodegradability in anaerobic co-digestion together with animal manure.

LIMK1/2 are required for actin filament and cell junction assembly in porcine embryos developing in vitro

  • Objective: This study was conducted to investigate the roles of LIM kinases (LIMK1 and LIMK2) during porcine early embryo development. We checked the mRNA expression patterns and localization of LIMK1/2 to evaluate their characterization. We further explored the function of LIMK1/2 in developmental competence and their relationship between actin assembly and cell junction integrity, specifically during the first cleavage and compaction. Methods: Pig ovaries were transferred from a local slaughterhouse within 1 h and cumulus oocyte complexes (COCs) were collected. COCs were matured in in vitro maturation medium in a CO2 incubator. Metaphase II oocytes were activated using an Electro Cell Manipulator 2001 and microinjected to insert LIMK1/2 dsRNA into the cytoplasm. To confirm the roles of LIMK1/2 during compaction and subsequent blastocyst formation, we employed a LIMK inhibitor (LIMKi3). Results: LIMK1/2 was localized in cytoplasm in embryos and co-localized with actin in cell-to-cell boundaries after the morula stage. LIMK1/2 knockdown using LIMK1/2 dsRNA significantly decreased the cleavage rate, compared to the control group. Protein levels of E-cadherin and β-catenin, present in adherens junctions, were reduced at the cell-to-cell boundaries in the LIMK1/2 knockdown embryos. Embryos treated with LIMKi3 at the morula stage failed to undergo compaction and could not develop into blastocysts. Actin intensity at the cortical region was considerably reduced in LIMKi3-treated embryos. LIMKi3-induced decrease in cortical actin levels was attributed to the disruption of adherens junction and tight junction assembly. Phosphorylation of cofilin was also reduced in LIMKi3-treated embryos. Conclusion: The above results suggest that LIMK1/2 is crucial for cleavage and compaction through regulation of actin organization and cell junction assembly.
