Although many diseases could be treated by the development of modern medicine, there are some incurable diseases including brain cancer, Alzheimer disease, etc. To study human brain cancer, various animal models were reported. Among these animal models, mouse models are valuable tools for understanding brain cancer characteristics. In spite of many mouse brain cancer models, it has been difficult to find a new target molecule for the treatment of brain cancer. One of the reasons is absence of large animal model which makes conducting preclinical trials. In this article, we review a recent study of molecular characteristics of human brain cancer, their genetic mutation and comparative analysis of the mouse brain cancer model. Finally, we suggest the need for development of large animal models using somatic cell nuclear transfer in translational research.
포유동물의 난자는 제 2 감수분열 중기에서 배란되어 수정된다. 일반적으로 포유동물에서의 수정은 inositol triphosphate (IP3) 또는 cyclic adenosine diphosphoribose (cADPr)에 의해 조절되어지는 세포질내 칼슘변화에 의해 일어난다. 난활성이 일어나면 세포질내 고농도의 MPF (maturation promotion factor)는 감소하고, 전핵이 형성되고, cytoskeleton이 재구성되고, 단백질합성이 조절되어진다. 이러한 모든 과정이 정상적으로 일어난 후 수정란은 종특이한 발달경로로 배발달을 시작한다. 본 논문에서는 돼지난자의 인위적인 난활성 유도방법에 대해서 설명하고자 한다. 신호전달(signal transduction) 경로에 의한 하나의 기전이 돼지난자에서 확인되었는데, 이 경로의 자극은 난활성으로 이루어진다. 또한 전기자극에 의한 난활성방법에 의해 12일까지 정상배 발달이 이루어진다고 보고되고 있다. 향후 연구에서는 정자가 어떠한 기전에 의해서 배발달을 시작하게 하는지를 밝혀내야 한다고 본다.
Tissue plasminogen activator (tPA) plays important roles in the brain after excitotoxic injury. This study was conducted to produce transgenic pig harboring human tissue plasminogene activator (htPA) gene. Recombinent htPA(rhtPA) genes containing bovine-β-casein promoter (bBC) were prepared for microinjection and testified the expression level of htPA protein from the Chinese hamster ovary (CHO) cell lines before NDA microinjection into the porcine pronuclei. Recent studies showed that tight junctions (TJs) integrity and assembly are required for blastocyst development in mouse and pig models. However, the biological functions of TJs associated with embryo implantation and maintenance of pregnancy were not investigated yet. To examine whether disrupted TJs affect further embryo development, we employed RNAi approach and inhibitor treatment. The embryos were injected with Cxadr (Coxsackievirus and adenovirus receptor) siRNA for knock down (KD) and treated with Adam10 (A Disintegrin and Metalloproteinase specific inhibitor 10; GI254023X; SI). We compared blastocyst development and paracellular sealing assay using FITC dextran uptake between control and KD or SI embryos. Finally, we transferred control and Cxadr KD or Adam 10 SI treated blastocyst to uteri of recipients. Cxadr KD and Adam 10 SI showed lower blastocyst development and more permeable to FITC-dextran. Moreover, we observed that half of KD and inhibited embryos failed to maintain pregnancies after the second trimester. Our findings suggested that TJs integrity is required for the maintenance of pregnancy and can be used as a selective marker for the successful application of assisted reproduction technologies.
Miniature pig (minipig) has been considered as an important laboratory animal in the developmental biotechnology researches with respect to xenotransplantation, stem cell, somatic cell nuclear transfer and embryo transfer. Given that the laboratory minipigs are normally housed at an indoor facility, they pass the time with lying or sleeping unless it is feeding time. Therefore, it is necessary to provide environmental enrichments to satisfy their innate needs and to lessen atypical behaviors caused by stress, on the purpose of welfare. We quantitatively investigated the type of preferable enrichment for the laboratory minipigs as well as its effect on their daily life. They presented a great interest to the pliable pail but a rapid loss of attraction to non-preferable enrichments. When the daily life of the single housed minipigs was quantified based on duration of playing or resting, they were more actively engaged in lively activities in the presence of enrichments. In addition, the provision of enrichments could effectively alleviate the conflicts during group housing when new pen mate was introduced, resulting in reduction of wound cases. We believe the considerations of animal welfare are essential to the conduct of better research because animals in the non-stressful environment will be more physiologically stable and provide more reliable results in the animal experiments.

The process of embryo implantation requires physical contact and physiological communication between the conceptus trophectoderm and the maternal uterine endometrium. During the peri-implantation period in pigs, the conceptus undergoes significant morphological changes and secretes estrogens, the signal for maternal recognition of pregnancy. Estrogens secreted from the conceptus act on uterine epithelia to redirect $PGF_2{\alpha}$, luteolysin, secretion from the uterine vasculature to the uterine lumen to prevent luteolysis as well as to induce expression of endometrial genes that support implantation and conceptus development. In addition, conceptuses secrete cytokines, interferons, growth factors, and proteases, and in response to these signals, the uterine endometrium produces hormones, protease inhibitors, growth factors, transport proteins, adhesion molecules, lipid molecules, and calcium regulatory molecules. Coordinated interactions of these factors derived from the conceptus and the uterus play important roles in the process of implantation in pigs. To better understand mechanism of implantation process in pigs, this review provides information on signaling molecules at the conceptus-uterine interface during early pregnancy, including recently reported data reported.
Imprinted genes are essential for fetal development, growth regulation, and postnatal behavior. However, little is known about imprinted genes in livestock. We hypothesized that certain putatively imprinted genes affected normal peri-implantation development such as embryo elongation, initial placental development, and preparation of implantation. The objective of the present study was to investigate the mRNA expression patterns of several putatively imprinted genes during the porcine peri-implantation stages from day 6 to day 21 of gestation. Imprinted genes were selected both maternally (Dlk1, IGF2, Ndn, and Sgce) and paternally (IGF2r, H19, Gnas and Xist). Here, we report that the maternally imprinted gene IGF2 was expressed from day 6 (Blastocyst stage), but Dlk1, Ndn, and Sgce were not expressed in this stage. These genes were first expressed between days 12 and day 14. All the maternally imprinted genes studied showed significantly high expression patterns from day 18 of embryo development. In contrast, paternally imprinted genes IGF2r, H19, Gnas, and Xist were first expressed from day 6 of embryo development (BL). Our data demonstrated that the expression of H19 and Gnas genes was significantly increased from day 14 of the embryo developmental stage, while IGF2r and Xist only showed high expression after day 21. This study is the first to show that the putatively imprinted genes were stage-specific during porcine embryonic development. These results demonstrate that the genes studied may exert important effects on embryo implantation and fetal development.
A field trial was performed to evaluate the effects of hormone treatment on estrus induction, ovulation, embryo transfer and reproductive performance in post-weaning sows. This trial involved 61 mixed breed sows of varying parity on a commercial pig farm. Sows were allocated to one of five trials: control group involved 25 sows that were treated with a single intramuscular injection of 5 ml physiological saline, 6 sows received 1,500 IU PMSG on the day of weanning and 500 IU HCG at the onset of estrus in trial I, 7 sows received 750 IU PMSG on the day of weanning and 500 IU HCG at the onset of estrus in trial II, 5 sows were treated with the same as trial II on day 28 after weanning in trial III. and 18 sows were treated with 10 mg PGF$_2$$\alpha$ plus 2 mg estradiol benzoate on day 31 after weanning in trial IV. Ovarian responses were checked by laparotomy and ova were recovered by oviducal flushing between 40 and l00hrs after mating. Fertilized ova were transferred into the oviduts of recipient sows synchroni- zed. The results obtained were summarized as follows: 1. Percentages of sows detected in standing estrus following treatment were 86~100% among trial groups. The interval from treatment to standing estrus(6l.7$\pm$0.5lhrs) in lOmg PGF$_2$$\alpha$ and 2mg estradial henzoate treated group was significantly earlier than in other trial groups(P<0.05). 2. Average number of ovulations was 11.5~37.8 among trial groups. The ovulation rate in 1,500 IU PMSG and 500 IU RCG treated group (37.8$\pm$ 19.87) was significantly different from other trial groups(P<0.05). 3. Ova were recovered by oviducal flushing between 40~ l00hrs after mating and recovery rates of ova wore 91.4% between 40~59hrs. 4. Fertilized ova were transferred into the oviducts of 8 recipient sows synchronized with 7 to 17 ova per animal. Three of the recipients were pregnant and delivered 25 piglets. 5. Four of the donor sows in those embryo collection was not successful were pregnant following oviducal flushing and delivered 23 piglets. 6. Recurrence of estrus and farrowing performance of experimental sows were observed following the experiment was no difference among trial groups, respectively.
