소의 히아론산 분해효소(PH-20)의 Pichia pastoris에서의 생산 최적화 (Optimization of Bovine Testicular PH-20 hyaluronidase Production in Pichia pastoris)

  • 소 고환 유래의 hyaluronidase 효소 PH-20을 pPIC9 expression vector를 사용하여 Pichia pastoris에서 발현하였다. 생산된 재조합 단백질 rPH-20b는 75 kDa의 분자량을 보였고 7460 units/L의 효소활성을 보였다. 세포내보다 세포외의 효소활성이 두배 높았다. pH 의 경우 buffer를 사용 안 한 경우가, 또한 $30^{\circ}C$ 배양 조건에서 높은 활성을 보였다. 1 M sorbitol 삼투압조건과 0.3% 메탄올의 생산유도인자를 사용시 성장과 생산에 유리하였으며 0.4 M 아르기닌을 첨가시 재조합 단백질의 분해가 감소하였다.

Gene Cloning, High-Level Expression, and Characterization of an Alkaline and Thermostable Lipase from Trichosporon coremiiforme V3

  • The present study describes the gene cloning and high-level expression of an alkaline and thermostable lipase gene from Trichosporon coremiiforme V3. Nucleotide analysis revealed that this lipase gene has an open reading frame of 1,692 bp without any introns, encoding a protein of 563 amino acid residues. The lipase gene without its signal sequence was cloned into plasmid pPICZαA and overexpressed in Pichia pastoris X33. The maximum lipase activity of recombinant lipase was 5,000 U/ml, which was obtained in fed-batch cultivation after 168 h induction with methanol in a 50 L bioreactor. The purified lipase showed high temperature tolerance, and being stable at 60℃ and kept 45% enzyme activity after 1 h incubation at 70℃. The stability, effects of metal ions and other reagents were also determined. The chain length specificity of the recombinant lipase showed high activity toward triolein (C18:1) and tripalmitin (C16:0).

Pichia pastoris에서 사람 락토페린 N-lobe의 발현과 항균활성 (Expression of human lactoferrin N-lobe in Pichia pastoris and its antibacterial activity)

  • 락토페린(LF)는 철이온과 결합하는 당 단백질로서 항균, 항바이러스, 항진균 등의 기능을 가지고 있으며, 생체의 각종 체액으로부터 분비되는 다기능성 단백질이다. 본 연구에서는 사람의 락토페린(hLF)으로부터 유래된 N-lobe의 유전자를 분리하고 산업용 균주로서 많이 사용되는 메탄올자화 효모인 Pichia pastoris에서 발현시켰다. 재조합 사람 락토페린 N-lobe (rhLF-N)는 배양액으로 분비 발현되었으며, 3L 발효조에서 약 $458{\mu}g/ml$이 수준으로 생성되었다. rhLF-N을 정제한 다음 SDS-PAGE와 western blot으로 분석하여 분자량 35 kDa 단백질을 확인하였으며, hLF에 대한 항체를 이용하여 면역확산법으로 면역성을 확인하였다. rhLF-N의 mRNA 발현양상을 qRT-PCR로 분석한 결과 메탄올 첨가에 의한 발현 유도 후 2-3일째에 발현율이 가장 높았으며, 4일째에는 점차적으로 감소하였다. 정제한 rhLF-N을 이용하여 항균활성을 조사한 결과 Staphylococcus aureus, E. coli, Pseudomonas aeruginosa, Burkholderia cepacia, Salmonella typhimurium과 같은 병원성 균에 대해 광범위한 항균활성을 보였으나, LF유래 항균 peptide들과 항균활성을 비교하였을 때, 항균력이 상대적으로 매우 떨어지는 것으로 나타났다. 비록 본 연구에서 발현한 rhLF-N은 항균력은 떨어지나, hLF에 비해 그 크기가 작고 배양조건 연구로 P. pastoris에서 대량 생산이 가능하며, 배양액으로 분비시킬 수 있기 때문에 정제 비용 등을 고려 할 때 산업적 응용에는 보다 유리할 것으로 사료된다.

Antibacterial Activity of Recombinant Pig Intestinal Parasite Cecropin P4 Peptide Secreted from Pichia pastoris

  • Cecropins (Cec) are antibacterial peptides and their expression is induced in a pig intestinal parasite Ascaris suum by bacterial infection. To explore the usefulness of its activity as an antibiotic, CecP4 cDNA was prepared and cloned into the pPICZ B expression vector and followed by the integration into AOX1 locus in Pichia pastoris. The supernatants from cell culture were collected after methanol induction and concentrated for the test of antimicrobial activity. The recombinant P. patoris having CecP4 showed antimicrobial activity when tested against Staphyllococcus aureus in disc diffusion assay. We selected one of the CecP4 clones (CecP4-2) and performed further studies with it. The growth of recombinant P. pastoris was optimized using various concentration of methanol, and it was found that 2% methanol in the culture induced more antibacterial activity, compared to 1% methanol. We extended the test of antimicrobial activity by applying the concentrated supernatant of CecP4 culture to Pseudomonas aeruginosa and E. coli respectively. Recombinant CecP4 also showed antimicrobial activity against both Pseudomona and E. coli, suggesting the broad spectrum of its antimicrobial activity. After improvements for the scale-up, it will be feasible to use recombinant CecP4 for supplementation to the feed to control microbial infections in young animals, such as piglets.

Cloning of a Gene Encoding Dextranase from Lipomyces starkeyi and its Expression in Pichia pastoris

  • A gene(lsd1) encoding dextranase from Lipomyces starkeyi KSM22 has been previously cloned, sequenced, and expressed in Saccharomyces cerevisiae. The gene consisting of 1,824 base pairs and encoding a protein of 608 amino acids was then cloned into and secretively expressed in Pichia pastoris under the control of the AOX1 promoter. The dextranase productivity of the P. pastoris transformant(pPIC9K-LSD1, 134,000 U/I) was approximately 4.2-fold higher than that of the S. cerevisiae transformant(pYLSD1, 32,000 U/I) cultured in an 8-1 fermentor. Over 0.63 g/l of active dextranase was secreted into the medium after methanol induction. The dextranase of the P. pastoris transformant, as analyzed by SDS-PAGE and Western blotting, showed only one homogeneous band. This dextranase of the P. pastoris transformant showed a broad band near 73 kDa. Rabbit monoclonal antibodies against a synthetic LSD1 peptide mix also recognized approximately 73 kDa.

Production of Recombinant Humanized Anti-HBsAg Fab Fragment from Pichia pastoris by Fermentation

  • In this report, we describe the high-yield secretory expression of the recombinant human anti-HBsAg Fab fragment from Pichia pastoris that was achieved by co-integration of the genes encoding the heavy and light chains (both under the control of alcohol oxidase promoter) into the genome of the yeast cells. The fed-batch fermentations were carried out in a 5 L scale. Both chains of the Fab were successfully expressed upon methanol induction. The absorbance ($OD_{600}$) of the broth can reach 350~500 at the end of fed-batch phase. After the induction, the expression level of the recombinant Fab (soluble) reached 420~458 mg/L. The recombinant Fab fragment was purified from the crude culture supernatant by ion exchange chromatography and the purity of the recombinant Fab fragment was over 95%. The affinity activities of the crude fermentation supernatant and the purified Fab were analyzed by indirect ELISA, which showed that the purified recombinant Fab fragment had high affinity activity with hepatitis B surface antigen.

Co-Expression of Protein Tyrosine Kinases EGFR-2 and $PDGFR{\beta}$ with Protein Tyrosine Phosphatase 1B in Pichia pastoris

  • The regulation of protein tyrosine phosphorylation is mediated by protein tyrosine kinases (PTKs) and protein tyrosine phosphatases (PTPs) and is essential for cellular homeostasis. Co-expression of PTKs with PTPs in Pichia pastoris was used to facilitate the expression of active PTKs by neutralizing their apparent toxicity to cells. In this study, the gene encoding phosphatase PTP1B with or without a blue fluorescent protein or peroxisomal targeting signal 1 was cloned into the expression vector pAG32 to produce four vectors. These vectors were subsequently transformed into P. pastoris GS115. The tyrosine kinases EGFR-2 and $PDGFR{\beta}$ were expressed from vector pPIC3.5K and were fused with a His-tag and green fluorescent protein at the N-terminus. The two plasmids were transformed into P. pastoris with or without PTP1B, resulting in 10 strains. The EGFR-2 and $PDGFR{\beta}$ fusion proteins were purified by $Ni^{2+}$ affinity chromatography. In the recombinant P. pastoris, the PTKs co-expressed with PTP1B exhibited higher kinase catalytic activity than did those expressing the PTKs alone. The highest activities were achieved by targeting the PTKs and PTP1B into peroxisomes. Therefore, the EGFR-2 and $PDGFR{\beta}$ fusion proteins expressed in P. pastoris may be attractive drug screening targets for anticancer therapeutics.

Attenuated Secretion of the Thermostable Xylanase xynB from Pichia pastoris Using Synthesized Sequences Optimized from the Preferred Codon Usage in Yeast

  • Xylanase has been used extensively in the industrial and agricultural fields. However, the low-yield production of xylanase from native species cannot meet the increasing demand of the market. Therefore, improving the heterologous expression of xylanase through basic gene optimization may help to overcome the shortage. In this study, we synthesized a high-GC-content native sequence of the thermostable xylanase gene xynB from Streptomyces olivaceoviridis A1 and, also designed a slightly AT-biased sequence with codons completely optimized to be favorable to Pichia pastoris. The comparison of the sequences' expression efficiencies in P. pastoris X33 was determined through the detection of single-copy-number integrants, which were quantified using qPCR. Surprisingly, the high GC content did not appear to be detrimental to the heterologous expression of xynB in yeast, whereas the optimized sequence, with its extremely skewed codon usage, exhibited more abundant accumulation of synthesized recombinant proteins in the yeast cell, but an approximately 30% reduction of the secretion level, deduced from the enzymatic activity assay. In this study, we developed a more accurate method for comparing the expression levels of individual yeast transformants. Moreover, our results provide a practical example for further investigation of what constitutes a rational design strategy for a heterologously expressed and secreted protein.

  • The cDNA of endo-1,$4-{\beta}-xylanaseA$, isolated from Phaenerocheate chrysosporium was expressed in Pichia pastoris. Using either the intrinsic leader peptide of XynA or the ${\alpha}$-factor signal peptide of Saccharomyces cerevisiae, xylanaseA is efficiently secreted into the medium at maximum concentrations of 1,946 U/L and 2,496 U/L, respectively.

  • 본 연구에서는 산업적으로 중요성이 날로 증가하고 있는 PLC의 대량 생산을 위해서 B. cereus ATCC10987에서 분리된 PLC 유전자를 pPICZa C에 삽입한 P. pastoris system을 개발하여 extracellular PLC를 생산하였다. 또한, 공업적 이용을 위해 여러 조건에서 B. cereus와 P. pastoris에서 생산된 extracellular PLC의 활성도를 측정하여 특성을 살펴보았다.
