Preparation and Properties of Phytosphingosine Ascorbate with Retaining Skin Development Effects (피부 활성을 갖는 Phytosphingosine Ascorbate의 합성)

  • In the human skin, vitamin C (L -ascorbic acid) that is well known as the activated materials has effects that is skin anti-aging and wrinkle repair by giving impetus to collagen biosynthesis and anti-oxidation, and that is the sun screen, a wound recovering, inhibition melanogenesis and so on. In spite of its effects, vitamin C has the defects of the skin stimulation and easily oxidized instability by water, air, heat and light. For solving their matters, many investigation is advanced and its results are synthesized the various vitamin C derivatives. And yet they have not solved the unstable property of vitamin C and were still insufficient for the comparing with the effect of the pure vitamin C itself. In this study, in order to prepare vitamin C derivative of being improved the stability and to apply vitamin C effect in the skin, we prepared new vitamin C derivative, phytosphingosine ascorbate, by using phytosphingosine, one of sphingolipids, which have a distinguished skin affinity. Phytosphingosine ascorbate can be prepared as the ionic bond between amine group (-NH$_2$) of phytosphingosine and hydroxy group (-OH) of vitamin C by way of the relatively simple reaction. So the structure and properties of the synthesized phytosphingosine ascorbate was confirmed the use of elemental analysis (C 58.3 : H 9.3 : N 2.8 : O 29.5), MALDI TOF-MS (Mw=492.58), Ultraviolet spectra (268.5nm), lH NMR, FT-IR spectra, thermal analysis (m.p=l54$^{\circ}C$), HPLC and so on. And we could confirm the anti-bacterial and anti-oxidation effects. Based on these results, we could confirm to prepare a new material that was expected of both effects of vitamin C and phytosphingosine and that is improved properties of vitamin C.

Phytosphingosine promotes megakaryocytic differentiation of myeloid leukemia cells

  • We report that phytosphingosine, a sphingolipid found in many organisms and implicated in cellular signaling, promotes megakaryocytic differentiation of myeloid leukemia cells. Specifically, phytosphingosine induced several hallmark changes associated with megakaryopoiesis from K562 and HEL cells including cell cycle arrest, cell size increase and polyploidization. We also confirmed that cell type specific markers of megakaryocytes, CD41a and CD42b are induced by phytosphingosine. Phospholipids with highly similar structures were unable to induce similar changes, indicating that the activity of phytosphingosine is highly specific. Although phytosphingosine is known to activate p38 mitogen-activated protein kinase (MAPK)-mediated apoptosis, the signaling mechanisms involved in megakaryopoiesis appear to be distinct. In sum, we present another model for dissecting molecular details of megakaryocytic differentiation which in large part remains obscure.

Study on phytosphingosine and Phytosphingosine-1-phosphate as a cosmetic ingredient (Phytosphingosine과 Phytosphingosine-1-phosphate의 화장품 소재 특성 연구)

  • In this study, it studies about Phytosphingosine (PhS) and Phytosphingosine-1-phosphate(PhS1P), and it tries to confirm the effect through anti-inflammatory, anti-melanin, MMP-1 revelation inhibition, and Western blot analysis experiment after grasping toxicity about 3 cells by using B16F10 melanin cell, RAW264.7 macrophage, and HDF fibroblast in order to find out whether it is possible to use as cosmetic material or not by studying biological activity in terns of skin care. As a result of this experiment, it confirmed that toxicity about B16F10, RAW264.7, HDF cell is low, and PhS1P appeared stronger inhibition activity than PhS in anti-inflammatory NO inhibitory activity experiment. MMP-1 revelation was greater in PhS1P, and it confirmed that the mechanism is due to reduction in ERK activity. On the other hands, melanin generation inhibitory activity is better than arbutin, and it confirmed that the mechanism is due to inhibition of revelation of MTF and Tyrosinase. In a nutshell, PhS and PhS1P that are bioactive substance may confirm the possibility to be used as functional cosmetic for wrinkle and skin improvement of whitening cosmetic.

  • A short and efficient synthesis of (2S,3S)-safingol from commercially available D-ribo-(2S,3S,4R)-phytosphingosine is described. The highlights of the synthesis are a stereoselective one-pot transformation of a diol into an epoxide under phase transfer catalytic conditions and a regioselective epoxide-opening reduction with a hydride reagent.

  • The effects of agitation rate and agitation time on the synthesis of sphingosome were studied. As increase in the agitation rates 4,000, 6,000, 8,000, 10,000 and 12,000 rpm the viscosity of sphingosome were decreased. The most sufficient agitation rate was 8,000rpm for which micell viscosity and stability. The effect of agitation time on the sphingosome viscosity and stability was investigated by changing the agitation times 2, 4, 6, 8 and 10 minute at 8,000rpm. 4 minute was the most sufficient for the viscosity and stability. The sphingolipid activity of cutaneous wound healing in impaired mice was examined. As a result, we could prove that phytosphingosine-HCl medically worked on wound healing well. For the phytosphingosine-HCl, it was found that the experimentally determined medical action more effective than that of tetra-acetylphytosphingosine.
