Changes of Physiological Activity by Drying Temperature in Leaf of Eriobotrya japonica (건조온도에 따른 비파 잎의 생리활성 변화)

  • In DPPH radical and nitric oxide scavenging ability, the extract of lowest temperature condition(40-15H ; 15 hours at $40^{\circ}C$ for drying) exhibited highest activity. The total phenolic and flavonoid contents of each extracts were found to be $120.4{\sim}193.3\;mg$/g and $86.91{\sim}94.55\;mg$/g respectively. It is shown that 40-15H was the highest content in each compound. In antimicrobial activity, a lower drying temperature conditions were found to be more strong activities in disc diffusion assay and each extracts showed MIC of identical level against every tested microbial strains. However 100-2H(2 hours at $100^{\circ}C$ for drying) was exhibited MIC of slightly low level against some strains. And every extract showed fine cell viabilities($101.7{\sim}122.9%$) against RAW 264.7 cell. In anti-proliferation activity against AGS, each extract showed a similar inhibition activity. However in anti-proliferation activity against HeLa, a lower drying temperature conditions showed more strong activities.

The Effect of Lifestyle on Mibyeong for University Employees (교직원의 생활습관이 미병에 미치는 영향)

  • This study aimed to identify the effects of lifestyle on Mibyeong, and provide basic data for health promotion activities for management of Mibyeong. A total of 405 data were analyzed for Daejeon University employees from July, 2015 to Jan, 2016. In this study, we collected the data about sex, age, BMI, dietary habit, physical activity, smoking, drinking, and Mibyeong index. There was a difference between Mibyeong groups according to irregular eating, eating of night snack, and smoking for men, while overeating, irregular eating, and physical activity for women. Logistics regression analysis, adjusting for sex and age, was used to estimate related factors of Mibyeong. Compared to the healthy group, Mibyeong 2 group tended to overeat 4 times a week with odds ratio of 3.52, eat irregularly with odds ratio of 2.67, lack of physical activity with odds ratio of 3.30, and to smoke with odds ratio of 3.07. This study suggests that lifestyle, particularly dietary habit, physical activities, and smoking, might be significantly associated with Mibyeong. Good lifestyle could help prevent Mibyeong.

  • The Purpose of this study is to examine residents' subjective satisfaction of the elderly housing environment and to suggest some major factors which can be used as basic design guidelines for the elderly housing. as the number of the elderly is growing in Japan For this purpose. data were collected through a questionnaire survey method, Some questions on their hope for the conditions of the elderly housing were asked to 265 old People living in the suburbs of Chiba city. Japan. Based on the findings of the statistical factor analysis, the major factors which affected on the elderly housing environment of the study can be summarized as follows ;(1)Factor1 is correlated with the space design element represented by the elderly dwelling performance and overall efficiency for the dwelling exterior space design (2)Factor2 is relative to the interior environmental element with Physiological and sensitive performance for borne services system. (3)Factor3 can be explained the moving and communication performance for Physical uneasiness of outdoor activities and request of public cultural spaces. (4)Factor4 is relative to the mental rest element with safety accidents for moving activities etc. and psychological isolation for the elderly.

Immunosuppressive Characteristics of Oligomycin Derivatives Produced by Streptomyces lydicus MCY-524

  • A strain producing immunosuppressive substances was isolated from a soil in Cheju island. By morphological, cultural, and physiological studies, the strain was identified as Streptomyces lydicus MCY-524. Cultured broth was purified by silica gel, sephadex LH-20 and preparative HPLC and gave two immunosuppressive compounds, MCH-22 and MCH-32. They dramatically suppressed the B cell activation with lipopolysaccharide, T cell activation by mixed lymphocyte response, and primary T-dependent antibody response at a final concentration of 1 ${\mu}g$/ml. They also markedly suppressed the proliferation of lymphocytes induced by lipopolysaccharide, pokeweed mitogen, and concanavaline A at the same concentration. Their suppressive activities, which were comparable to those of cyclosporin A, suggested that they were potent and broad immunotoxic agents on the immune functions of murine lymphocytes.

  • Chlorella ellipsoidea cells were cultured in an iron, copper, zinc, manganese, molybdenum or boron-free medium. Physiological activities such as growth rate, reproduction, endogenous and glucose respiration, photosynthetic activity and biosythesis of chlorophyll of the micro-element definition cells were measured. It generally, growth rate, respiratory and photosynthetic activities, and biosynthesis of chlorophyll of the micro-element deficient cells decreased more or less, compared with those of the normal cells. The growth of the algal cells in an iron-free medium were retarded severely with the chlorosis, and the photosynthetic activity of the cells decreased remarkably even though the low content of chlorophyll in the cells owing to the iron-deficiency is considered. Therefore, it is deduced that iron takes part in the photosynthetic process itself, possibly by its participation in the photo phosphorylation coupled with electron transport. Respiratory activity of boron-deficient cells showed the most severe decrease whereas those of the molybdenum-deficient cells showed very slight decrease in spite of severe growth retardation.

Antioxidative and anti-α-amylase activities of four wild plants consumed by pastoral nomads in Egypt

  • In this study, four plants grown naturally in Egypt that are usually consumed by pastoral nomads were screened for their total phenolic contents, antioxidative, and anti-$\alpha$-amylase activities. Dried powder of plant's part was extracted in absolute or 70% ethanol. A polar extract of Panicum turgidum (PTPE) had the highest total polyphenol content {92.5 mg gallic acid equivalents (GAE)/g}, followed by an alkaloid extract of Withania somnifera (WSAlk; 77.5 mg GAE/g), and an ethanol extract of Leptadenia pyrotechnica (LPEE; 59.1 mg GAE/g). By employing different assays such as DPPH radical scavenging, reducing power, $Fe^+$ chelating, $H_2O_2$ scavenging and total antioxidant capacity, it was shown that PTPE, WSEE (ethanol extract of W. somnifera), WSAlk and LPEE had promising antioxidant activity, though, their potency varied according to the different tests. WSAlk had the highest level of $\alpha$-amylase inhibition (40.2%) in vitro, followed by WSEE (30.5%). Therefore, it can be concluded that these plants, especially extracts of PTPE, and WSAlk are beneficial to physiological health, and could be used in food and pharmaceutical industries to prepare dietary supplements, functional foods or food preservatives.

Elucidation of the Regulation of Ethanol Catabolic Genes and ptsG Using a glxR and Adenylate Cyclase Gene (cyaB) Deletion Mutants of Corynebacterium glutamicum ATCC 13032

  • The cyclic AMP receptor protein (CRP) homolog, GlxR, controls the expression of several genes involved in the regulation of diverse physiological processes in Corynebacterium glutamicum. In silico analysis has revealed the presence of glxR binding sites upstream of genes ptsG, adhA, and ald, encoding glucose-specific phosphotransferase system protein, alcohol dehydrogenase (ADH), and acetaldehyde dehydrogenase (ALDH), respectively. However, the involvement of the GlxR-cAMP complex on the expression of these genes has been explored only in vitro. In this study, the expressions of ptsG, adhA, and ald were analyzed in detail using an adenylate cyclase gene (cyaB) deletion mutant and glxR deletion mutant. The specific activities of ADH and ALDH were increased in both the mutants in glucose and glucose plus ethanol media, in contrast to the wild type. In accordance, the promoter activities of adhA and ald were derepressed in the cyaB mutant, indicating that glxR acts as a repressor of adhA. Similarly, both the mutants exhibited derepression of ptsG regardless of the carbon source. These results confirm the involvement of GlxR on the expression of important carbon metabolic genes; adhA, ald, and ptsG.

Screening of Cytotoxicity and Antimicrobial Effects of Hexane Extracts from Cornis fructus (산수유 헥산 추출물의 항균효과 및 세포독성)

  • Cornis fructus was extracted by successive extraction and then fractionated with hexane extract to get active fractions. This study was performed to determine the cytotoxic effect of hexane extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. Hexane extract showed cytotoxic effect against A549, B16 melanoma and MDA-MB-231. Futher fractionation with hexane extract was performed to obtain effective fraction, fraction 3 showed the cytotoxic effect against A549 and MDA-MB-231. In antimicrobial test of each fraction of hexane extract, fraction 5 showed antimicrobial activities against P. putida and P. aeruginosa.

Tribulus terrestris Suppresses the Lipopolysaccharide-Induced Inflammatory Reaction in RAW264.7 Macrophages through Heme Oxygenase-1 Expressions

  • The fruit of Tribulus terrestris L. (Zygophyllaceae) is an important source of traditional Korean and Chinese medicines. In this study, NNMBS223, consisting of the ethanol extract of T. terrestris, showed potent anti-inflammatory activities in RAW264.7 macrophages. We investigated the effect of NNMBS223 in suppressing the protein expression of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2 and production of iNOS-derived nitric oxide (NO), COX-2-derived prostaglandin E2 (PGE2) in lipopolysaccharide (LPS)-stimulated macrophages. In addition, NNMBS223 induced expression of heme oxygenase (HO)-1 through nuclear translocation of nuclear factor E2-related factor 2 (Nrf2) in macrophages. The effects of NNMBS223 on LPS-induced production of NO and PGE2 were partially reversed by the HO activity inhibitor tin protoporphyrin (SnPP). These findings suggest that Nrf2-dependent increases in expression of HO-1 induced by NNMBS223 conferred anti-inflammatory activities in LPS stimulated RAW264.7 macrophages.

Growth Arrest and Apoptosis of Human Uterine Cervical Carcinoma Cells Induced by Trichosanthes Semen Extract (과루인이 자궁경부암세포의 성장억제 및 세포고사에 미치는 영향)

  • To investigate the effects of Trichosanthes semen extract on the growth and apoptosis of human uterine cervical carcinoma cells. Effects of Trichosanthes semen extract on the growth of ME-180 cells were assayed by MTT assay. Apoptosis induced by Trichosanthes semen extract was detected by fluorescent microscopy, DNA fragmentation analysis and flow cytometry. Caspase-3 and caspase-8 activities were assayed. Trichosanthes semen extract induced ME-180 cells to die in a dose- and time-dependent manner. ME-180 cells treated with Trichosanthes semen extract exhibited typical characteristics of apoptosis. The population of Sub-G1 cells increased significantly, and the cells represented the reduced size, condensed chromatin and apoptotic bodies. They showed the decreased mitochondrial membrane potential and increased activities of caspase-3 and caspase-8. The results suggest that Trichosanthes semen extract induced ME-180 cell apoptosis and the activation of caspase and mitochondrial pathway were involved in the process of Trichosanthes semen extract-induced apoptosis.
