Physiological Activities of Roots Extracts from Calystegia japonica

  • This study was investigated to evaluate the contents of polyphenols and flavonoids, and physiological activities of various extracts from Calystegia japonica roots for making good use of their functional materials. The roots of C. japonica were extracted with water (WE), ethanol (EE) and hot water (HWE) by different methods. Among these extracts, the highest extracting yield was 30.30% of HWE, while the highest contents of total polyphenols and flavonoids were 40.85% and 6.40% of WE, respectively. The nitrite scavenging abilities were ranged from 31.31% (HWE) to 37.46% (EE) at pH 1.2 and 1.0 mg/ml concentration. In the measurements of electron donating abilities, EE showed the highest effect as 91.83% at 0.3 mg/ml assay concentration, and the electron donating ability was decreased as the extract concentration was increased. In the results of superoxide dismutase (SOD)-like activity, HWE showed the highest effect as 7.15% at 1.0 mg/ml. The tyrosinase inhibition activities of WE and EE were 15.28% and 14.97%, respectively. The xanthine oxidase inhibitory effects were ranged from 97.50 to 99.28% at 1.0 mg/ml. These results indicate that C. japonica extract has a good antioxidant effects and could be useful for developing functional products.

Protocatechuic Acid Content and Physiological Activities of Chaenomeles sinensis Extracts Prepared with Different Methods (모과의 추출조건에 따른 Protocatechuic Acid 함량 변화 및 활성 평가)

  • The aim of this study was to investigate the potential of Chaenomeles sinensis as a content of protocatechuic acid and physiological activities. The analyzed the content of protocatechuic acid of Chaenomeles sinensis water extract (CSW) according to different extract temperature and time for extraction. It was analyzed as 0.14±0.00 mg/g in the extract at 100℃ for 3 h. Protocatechuic acid content increased as extraction time and temperature increased. The activity of the CSW at 100℃ for 3 h was measured. As a result of measuring DPPH, ABTS radical scavenging activity and SOD activity, antioxidant activity was found and α-glucosidase inhibition activities. Cell viability was shown for RAW 264.7 cells, and NO (nitric oxide) production was also inhibited. Finally, content of protocatechuic acid increased with increasing extraction time and extraction temperature, and exhibited antioxidant activity, inhibition of NO production, and anti-diabetic activity. It is believed that the Chaenomeles sinensis is likely to be developed as a functional material.

Assessment of the Physiological Activities of Flower Extracts from White Lotus (백련 꽃 추출물의 생리활성 효과)

  • This study was conducted to gather basic data on the physiological activities of flower extracts from 4 cultivars of white lotus to promote the increase of public consumption. Four cultivars of white lotus: c.v. 'Garam', 'Choeue', 'Baekhwageonryeon', and 'Seungdal' were harvested on 14, August, 2007, and the physiological activities of flower extracts obtained by hot water extraction and ethanol extraction were examined. Total phenol content was highest in the 'Garam' flower hot water extracts, $144.2{\mu}g{\cdot}mL^{-1}$, and lowest in the 'Seungdal' flower ethanol extracts, $63.4{\mu}g{\cdot}mL^{-1}$. DPPH radical scavenging activity was highest at 2,000ppm flower extract of 'Garam' obtained by hot water extraction and ethanol extraction of 90.9% and 83.0%, respectively. Total flavonoid content in the flower extracts of 'Garam' obtained by hot water extraction and ethanol extraction showed the highest levels at $34.1{\mu}g{\cdot}mL^{-1}\;and\;33.9^{\mu}g{\cdot}mL^{-1}$, respectively. Nitrite scavenging activity at 1,000ppm flower extract was highest in 'Garam'. Extracts obtain by hot water and ethanol showed 91.3% and 80.4% activity, respectively. No significant difference in tyrosinase inhibition activity was observed among the cultivars or as a result of extraction method. Overall anti-microbial activity of the flower extracts was slightly higher in 'Garam' than in the other cultivars.

Nutritional Compositions and Physiological Activities of Chungbuk New Mulberry Cultivar 'Cheongsu' (충북 신품종 오디 '청수'의 영양성분 및 생리활성 분석)

  • Mulberry (oddi) is one of the most popular functional foods with many physiological components. This study investigated and compared the nutritional compositions and physiological activities of four mulberry fruits from Morus albo L. including Cheongilppong, Iksuppong, Suwonppong and Cheongsuppong (a new cultivar from Chungcheongbuk-do). To analyze the nutrient contents, mulberry fruits were freeze-dried. The results showed that the proximate compositions of the four mulberry cultivars ranged from 9.61~14.11% for moisture, 8.28~11.90% for crude protein, 3.70~4.86% for crude ash, 4.28~5.54% for crude lipid and 7.46~10.78% for crude fiber. The above proximate contents of mulberry cultivars were not significantly different. However, Cheongsuppong had the highest content of reducing sugar, 74.7%. Chungsuppong and Iksuppong showed higher contents of total polyphenol and anthocyanin than other mulberry cultivars, while all mulberry cultivars showed high antioxidant activities. Hypoglycemic effect had a slightly higher level in Suwonppong and Cheongsuppong than in the other samples. Taken together, the new cultivar "Cheongsu" oddi can be suggested as a potential source of functional food.

  • Chemical compositions and physiological activities of Platycodon grandiflorum roots grown for 4 and 24 years were investigated. Chemical compositions of P. grandiflorum roots grown for 24 years were moisture 82.7%, crude protein 1.6%, crude lipid 2.1% crude ash 0.7%. total sugar 6.0% total dietary fiber 2.3% crude saponin 3.4mg% and ascorbic acid 3.0 mg%. Contents of each components in 24 years old roots were higher than that in 4 years old roots and the most abundant mineral was potassium being 879.9mg/kg. Physiological activities of solvent extract from P. grandiflorum roots grown for 4 and 24 years were high in order of ethanol methanol and water extract. The electron-donating abilities and nitrite-scavenging effect of solvent extract of 24 years were high in order of ethanol methanol and water extract. The electron-donating abilities and nitrite-scavenging effect of solvent extract of 24 years old roots were higher than those of 4 years of old roots in the range of 10~40% respectively.

Quality Characteristics and Physiological Activities of Takju with Whole Chestnut (통밤을 첨가한 탁주의 품질특성 및 생리활성)

  • This study investigated the quality characteristics and physiological activities of takju prepared with whole chestnut (15%, 30% and 50% with steamed rice) during fermentation. The pH level began to decrease after the secondary brewing stage. The total acid as well as the organic acid content increased during fermentation. Lactic acid and succinic acid were the main organic acids in takju fermented with whole chestnut. The level of total sugar and reducing sugars in takju fermented with whole chestnut increased at the first brewing stage and then slowly decreased after 4 days of fermentation. Also, the total number of viable cells and microbial populations such as lactic acid bacteria and yeast in the treatments increased to about 108 CFU/mL after 1 days of fermentation and then decreased gradually afterward. The ethanol content in takju fermented with whole chestnut rapidly increased during the initial 4 days of fermentation, to a maximum content of 18.2% after 8 days. The colour values of the treatments did not show any significant differences between 0% and 15% chestnut content. However L value decreased, while the a and b values increased when the whole chestnut content above 30%. The total polyphenol level, electron donating ability, nitrite-scavenging ability and ferrous ion effect also increased as the ratio of whole chestnut increased. Sensory scores of takju fermented with 15% chestnut were greater than those of takju prepared by other treatments.

Comparison of Antioxidant and Physiological Activities in Various Solvent Extracts of Ixeris repens (갯씀바귀(Ixeris repens) 추출물의 항산화성 및 생리활성)

  • Ixeris repens is a type of halophyte that grows in high salinity sands found in coastal sand dunes and sandy shores. This study was conducted to investigate the contents, antioxidant potency, and physiological activities of I. repens. In analyses of general composition, carbohydrate, protein, ash, and moisture content were 57.42%, 10.48%, 11.99% and 10.29%, respectively. Potassium, calcium, sodium, and magnesium were its most prevalent minerals. The solvents used to extract I. repens were 70% ethanol, 80% methanol, and distilled water. Among the resultant extracts, ethanol and methanol extracts displayed higher total polyphenol and flavonoid contents than the water extract. ABTS (IC50, 0.12 mg/mL) and FRAP (0.77 mM) radical scavenging activity were highest in the water extract, while methanol extract exhibited the strongest DPPH radical scavenging activity (IC50, 1.32 mg/mL), NO scavenging activity (IC50, 4.10 mg/mL), and reducing power (EC50, 0.14 mg/mL). Tyrosinase, elastase, and α-glucosidase inhibitory activities were highest in the ethanol extract. The ethanol extract also possessed the most potent acetylcholinesterase inhibitory activity. These results indicate that I. repens may be useful as an antioxidant, and a functional substance in food and pharmaceutical materials.

Comparison of Antioxidant and Physiological Activities of Various Solvent Extracts from Hizikia fusiformis (추출용매에 따른 톳(Hizikia fusiformis) 추출물의 항산화 및 생리활성 비교)

  • The seaweed Hizikia fusiformis is rich in protein, carbohydrates, vitamins, and minerals. This study investigated the antioxidant and physiological activities of H. fusiformis extracts prepared with 70% ethanol, 80% methanol, or distilled water. The extraction yields of these various solvent extracts were as follows: ethanol extract, 15.26%; methanol extract, 17.95%; and water extract, 45.62%. The methanol extract showed the highest total polyphenol content (24.06 mg GAE/g), but total flavonoid content was similar in all extracts. ABTS (2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) radical scavenging activity was highest in the ethanol extract (IC50: 0.90 mg/mL), while the methanol extract exhibited the strongest DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity (IC50: 8.09 mg/mL), reducing power (EC50: 0.40 mg/mL), and ferric reducing antioxidant power (0.28 mM). By contrast, tyrosinase and α-glucosidase inhibitory activities were higher in the ethanol extract than in the other extracts. The high BACE1 (β-secretase) inhibitory activity was observed in the ethanol extract (IC50: 1.03 mg/mL). These results indicate that H. fusiformis ethanol extracts may be useful for their antioxidant and functional properties in food and pharmaceutical materials.

DIFFERENTIATION OF BASIC EMOTIONS BY EEG AND AUTONOMIC RESPONSES (뇌파 및 자율신경계 반응특성에 의한 기본정서의 구분)

  • The discrete state theory on emotion postulated that there existed discrete emotions, such as happiness, anger, fear, disgust, and so forth. Many investigators who emphasized discreteness of emotions have suggested that discrete emotions entailed their specific activities in the autonomic nervous system. The purposes of this study were to develop a model of emotion-specific physiological response patterns. The study postulated six emotions (i.e., happiness, sadness, anger, disgust, fear, and surprise) as the basic discrete emotions. Thirty eight college students participated in the present study. Twelve slides (2 for each emotion category) were presented to the subjects in random order. During resting period of 30 s prior to the presentation of each slide, four presentation of each slide, four physiological measures (EEG, ECG, EDA, and respiration) were recorded to establish a baseline. The same physiological measures were recorded while each slide was being presented for 60 s (producing an emotional sate). Then, the subjects were asked to rate the degree of emotion induced by the slide on semantic differential scales. This procedure was repeated for every slide. Based upon the results, a model of emotion-specific physiological response patterns was developed: four emotion (fear, disgust, sadness, and anger) were classified according to the characteristics of EEG and autonomic responses. However, emotions of happiness and surprise were not distinguished by any combination of the physiological measures employed in this study, suggesting another appropriate measure should be adopted for differentiation.

Cosmeceutical effect from native medicinal plants of blue belt Ulleung islands (청정해역 울릉도자생약초를 이용한 화장품 약리활성 연구)

  • Objectives : The purpose of this research was total polyphenol contents, anti-oxidant activities, anti-inflammatory activities and anti-wrinkle activities of Ulleung islands plants for application as a cosmeceutical ingredients Methods : We were experimented total polyphenol contents, anti-oxidant activities, anti-inflammatory activity and anti-wrinkle activities. Results : In the physiological activities, most Ulleung islands plants is showed the highest anti-oxidant, anti-inflammatory activity in ABTS+ radical cation scavenging activity, hydrogen peroxide($H_2O_2$) scavenging activity, nitric oxide scavenging activity. All experiment of the water and ethanol extract from the Ulleung islands plants ingredients were gradually increased as well. Conclusions : These results suggest that extracts from Ulleung islands plants can be used in natural ingredient in food or cosmetic industry.
