CALCIUM EFFECTS OF VISUAL ADAPTATION IN A VERTEBRATE RETINA (I)

  • Calcium has a variety of functions in neuron and muscle cells and blood clotting, especially in the visual system where dark adapted rods cotransport with Na$^+$ into the cell. An influx of Ca$^{2+}$ flows out of the cell through the Na$^+$ - Ca$^{2+}$ exchanger. By using a modified Ussing chamber in order to bring in vivo environment close, we have concluded that Ca$^{2+}$ blocks the activity of guanylate cyclase; in consequence, having an effect on the amplitude of electroretinogram (ERG). We suggest that Ca$^{2+}$ moves between the photoreceptor and the vitreous humor by way of certain Ca$^{2+}$ transport mechanisms. Also, the effect of Zn$^{2+}$ in Ca$^{2+}$ - free ringer solution caused an elevation of amplitude in ERG and a reduction of threshold.

  • Light regulates a variety of genes in higher plants. The expression of light-induced plant genes is regulated at the level of transcription via red- light photomorphogenic receptor, phytochrome, as well as unknown blue light photoreceptor(s). Ribulose-5-phosphate carboxylase/oxygenase (Rubisco) small subunit (SSB) and light harvesting chlorophyll a/b (Cab) protein are those of the best understood genes regulated by light. 5'-upstream flanking sequence (- -400) of Rubisco SSB and Cab genes sis known as a light responsive, enhance-like element. It responses to red and blue light in transgenic plant system as a tissue specific manner. Phytochrome gene is also regulated by light. In contrast to most of the light regulated plant genes, it is negatively controlled by red light. Search for the cis- and trans-acting factors responsible for the light signal is in progress to understant photomorphogenesis and development in higher plants.

  • With a remarkable advance in CMOS (complimentary metal-oxide-semiconductor) process technology, a variety of vision sensors with signal processing circuits for complicated functions are actively being developed. Especially, as the principles of signal processing in human retina have been revealed, a series of vision chips imitating human retina have been reported. Human retina is able to detect the edge and motion of an object effectively. The edge detection among the several functions of the retina is accomplished by the cells called photoreceptor, horizontal cell and bipolar cell. We designed a CMOS vision chip by modeling cells of the retina as hardwares involved in edge and motion detection. The designed vision chip was fabricated using $0.6{\mu}m$ CMOS process and the characteristics were measured. Having reliable output characteristics, this chip can be used at the input stage for many applications, like targe tracking system, fingerprint recognition system, human-friendly robot system and etc.

  • G Protein-coupled receptors (GPCRs) mediating wide ranges of physiological responses is one of the most attractive targets for drug development. Rhodopsin, a dim-light photoreceptor, has been extensively used as a model system for structural and functional study of GPCRs. Fish have rhodopsin finely-tuned to their habitats where the intensity and the wavelength of lights are changed depending on its water-depth. To study the detailed molecular characteristics of GPCR architecture and to understand the fishery light-sensing system, genes encoding rod opsins were isolated from fishes living under different photic environments. Full-length rod opsin genes were obtained by combination of PCR amplification and DNA walking strategy of genomic DNA isolated from olive flounder, P. olivaceus, Japanese eel, A. japonica, and Common carp C. carpio. Deduced amino acid sequences showed a typical feature of rod opsins including the sites for Schiffs base formation (Lys296) and its counter ion (Glu113), disulfide formation (Cys110 and Cys187), and palmitoylation (Cys322 and Cys323) although Cys322 is replaced by Phe in Japanese eel. Comparison of opsins by amino acid sequence alignment indicated the closest similarity between P. olivaceus and H. hippoglossus (94%), A. japonica and A. anguilla (98%), and C. carpio and C. auratus (95%), respectively.

Construction of a New Agrobacterium tumefaciens-Mediated Transformation System based on a Dual Auxotrophic Approach in Cordyceps militaris

  • Cordyceps militaris is a significant edible fungus that produces a variety of bioactive compounds. We have previously established a uridine/uracil auxotrophic mutant and a corresponding Agrobacterium tumefaciens-mediated transformation (ATMT) system for genetic characterization in C. militaris using pyrG as a screening marker. In this study, we constructed an ATMT system based on a dual pyrG and hisB auxotrophic mutant of C. militaris. Using the uridine/uracil auxotrophic mutant as the background and pyrG as a selection marker, the hisB gene encoding imidazole glycerophosphate dehydratase, required for histidine biosynthesis, was knocked out by homologous recombination to construct a histidine auxotrophic C. militaris mutant. Then, pyrG in the histidine auxotrophic mutant was deleted to construct a ΔpyrG ΔhisB dual auxotrophic mutant. Further, we established an ATMT transformation system based on the dual auxotrophic C. militaris by using GFP and DsRed as reporter genes. Finally, to demonstrate the application of this dual transformation system for studies of gene function, knock out and complementation of the photoreceptor gene CmWC-1 in the dual auxotrophic C. militaris were performed. The newly constructed ATMT system with histidine and uridine/uracil auxotrophic markers provides a promising tool for genetic modifications in the medicinal fungus C. militaris.

  • Ophthalmic Laser System is widely used in Selective Laser Trabeculoplasty of Open Angle Glaucoma and Ocular Hypertension. Versatile ophthalmic laser system is a medical device with technology that checks the condition of the treatment area by irradiating a continuous laser pulse on the treatment area, and monitoring the microbubble reaction caused by the temperature increase of the melanosome through the ultrasonic signal and the optical signal sensor. It performs selective laser treatment without damaging the photoreceptor by controlling the wavelength of the laser when microbubbles are detected. This study aims to suggest a guideline for evaluating safety, performance and clinical effectiveness of Versatile Ophthalmic Laser System in accordance with the growing technology. International Standards, Regulations, and Clinical Trial Protocols were investigated and analyzed for this study. As a result of this study, the safety, performance and clinical effectiveness test guideline for Versatile Ophthalmic Laser System were proposed. This guideline will ensure the safety and efficacy of Medical device, and furthermore it is expected to be able to promote the development of technology development by supporting a clinical trial plan.

Chemical Coupling between Horizontal Cells in the Catfish Retina

  • The effects of GABA and glutamate on the horizontal cells were explored by an intracellular recording method to discern the mechanisms of receptive field formation by chemical coupling in the catfish outer retina. The results suggest that the horizontal cells of the catfish retina might use GABA as their transmitters and that the GABAergic system contributes to the formation of receptive fields of the horizontal cells. GABAC receptors may be involved in a chemical coupling between horizontal cells and concerned with the depolarizing actions by GABA on horizontal cells in the catfish retina. Since the chloride equilibrium potential is more positive than the dark membrane potential in horizontal cells, GABA released from a horizontal cell may depolarize the neighboring horizontal cells. Thus a chemical coupling between horizontal cells may be formed. $GABA_A$ receptors also may be involved in the negative feedback mechanism between photoreceptor and horizontal cell. And glutamate may be involved in connecting positive and negative feedback systems since it potentiated the GABA's actions. Therefore, it is presumed that large receptive fields in the catfish retina are formed not only by electrical coupling but also by chemical coupling between horizontal cells. And information travels laterally by pathways involving both electrical coupling composed of gap junctions and chemical coupling in the retinal network.

We have measured the Ca$\^$++/, K$\^$+/, and Na$\^$+/ concentration in dark and light adapted bullfrog's (Rana catesbeiana) vitreous humor. The calcium concentration of the light adapted bullfrog's vitreous humor was higher than that of the dark adapted bullfrog's vitreous humor This means that ion activity between the photoreceptor and vitreous humor side is light dependent and we have found that a Ca$\^$++/ channel and Ca$\^$++/K$\^$+/ exchanger exist in the vitreous humor side. Taken together permeability of Ca$\^$++/, K$\^$+/ and K$\^$+/ ion internal limiting membrane faced in the vitreous humor side has light-dependent activity during the illumination.

Deposition Behavior and Photoelectrochemical Characteristics of Chlorophyll a Langmuir-Blodgett Films

  • The deposition behavior and photoelectric response characteristics of chlorophyll a(Chl a) monolayers and multilayers were investigated under various film fabrication conditions. Chl a LB films were deposited onto quartz and pretreated ITO glass substrates under several fabrication conditions, including surface pressure and number of layers. The absorption spectra of Chl a in a solution state and solid-like state (LB films) were fairly consistent with each other, and two absorption peaks were found at 678 and 438nm, respectively. The prepared Chl a LB films were set into an electrochemistry cell equipped with a Pt plate as the counter electrode, and the photoelectric response characteristics were obtained and analyzed relative to the light illumination. By considering the resulting photocurrents, the optimal fabrication conditions for Chl a LB films were determined as 20mN/m of surface pressure and 20 layers. The action spectrum of the Chl a LB films was obtained in the visible region, and was found to be in good agreement with the absorption spectrum. The possible application of the proposed system as a constituent of an artificial color recognition device was suggested based on combining with the photoelectric conversion property of another light-sensitive biological pigment.

A Study on the Development of Electrophotographic Photoreceptor by Dye Sensitized System (II) (색소증감에 따른 전자사진 감광체 개발에 관한 연구)

  • 본 연구는기존의 보고에서 보다 감광화효율을 더욱 향상시키기 위하여 결정형이 다른 전하발생물질 (CGM:Charge Generation Material)과 전하수송물질(CTM:Charge Transport Material)을 사용하여 감광체를 만들었고, 이들의 정전특성을 비교검토하였다. 전하발생물질로서 결정형이 다른 $\alpha$-, $\beta$-, x-형 무금속 프탈로시아닌 (H$_{2}$Pc)을 사용한 결과, x-H$_{2}$Pc를 사용한 감광체의 감도는 E$_{1}$2/의 값이 2.62 $\mu$J/$\textrm{cm}^2$로 가장 두수하게 나타났다. 전하발생층(CGL:Charge Generation Layer)에 첨가되는 CGM-CTM, CGM-CTM-ZnO로 구성된 감광체보다 전하발생층에 전하발생계만으로 구성된 감광체의 경우의 전하유지율 (80%) 및 감도(E$_{1}$2/=2.83$\mu$J/$\textrm{cm}^2$)면에서 우수함을 보여주었다. 한편 binder로서 PVB-co-PVA-co-PVA$_{c}$[poly(viny1 butyral-co-viny1 alcohol-co-viny1 acetate)]를 사용했을 때는, CGM-CTM으로 구성된 감광체보다 CGL에 CGM-CTM으로 구성된 감광체의 경우의 전하유지율(71%) 및 감도(E$_{1}$2/=2.62$\mu$J/$\textrm{cm}^2$)면에서 우수함을 보여주었다.
