미토콘드리아막의 용질투과성에 미치는 양이온의 영향 (Effect of cation on solute permeability of mitochondrial membrane)

  • Mitochondria were isolated from Chlorella cells effects of cation on solute permeability of mitochondrial membrane were investigated using P$^{32}$ as a tracer. It was strikingly increased uptake of phosphate for NaCl, KCl, while evidently decreased phosphate uptake of mitochondrial membrane for $MgCl_2$, $CaCl_2$. This consider that uptake of monovalent cation were increased, but uptake of divalentcation were decreased for permeability of mitochondrial membrane as if the permeability of protoplasmic membrane.

Effects of Ethanol on $Na^+-dependent$ Solute Uptake in Rabbit Renal Brush-Border Membrane Vesicles

  • This study was undertaken to examine the effect of ethanol on $Na^+-dependent$ transport systems (glucose, phosphate, and dicarboxylate) in renal brush-border membrane vesicles (BBMV). Ethanol inhibited $Na^+-dependent$ uptakes of glucose, phosphate, and succinate in a dose-dependent manner, but not the uptakes of $Na^+-dependent.$ The $H^+/TEA$ antiport was reduced by 8% ethanol. Kinetic analysis showed that ethanol caused a decrease in $V_{max}$ of three transport systems, leaving $K_m$ values unchanged. Ethanol decreased phlorizin binding, which was closely correlated with the decrease in $V_{max}$ of $Na^+-glucose$ uptake. These results indicate that ethanol inhibits $Na^+-dependent$ uptakes of glucose, phosphate, and dicaboxylate and that the reduction in $V_{max}$ of $Na^+-glucose$ uptake is caused by a decrease in the number of active carrier proteins in the membrane.

인광석 처리 토양에서 담배의 인산 흡수와 생육에 미치는 인산 가용화균의 효과 (Effect of Phosphate Solubilizing Fungi on P Uptake and Growth of Tabacco in Rock Phosphate Applied Soil)

  • 인광석을 처리한 토양에서 인산 가용화균 Penicillium oxalicum CBPS-3F-Tsa이 식물의 인산 흡수와 생육에 미치는 효과를 온실조건에서 조사하였다. 인광석을 처리한 토양에서 담배 식물체중의 인산 농도는 증가하였으며, 인산 가용화균을 접종한 경우에는 인산 농도가 더욱 증가하였다. 식물체의 총 인산 흡수량도 인산 가용화균을 접종한 경우 증가하였다. 담배 식물의 생육 또한 인광석과 인산 가용화균을 동시에 처리한 경우 인광석만 처리한 경우에 비하여 통계적으로 유의성 있게 증가하였는데, 이러한 인산 가용화균의 생육 촉진 효과는 인산 흡수에 미치는 영향과 밀접한 관계를 보였다. 따라서 인산 가용화균과 인광석 처리에서 나타난 식물의 인산 흡수량 및 생육 증대 효과는 결국 접종한 인산 가용화균에 의한 난용성 인광석의 가용화에 따른 결과로 판단된다.

무산소조 NO3-N 농도 변화에 따른 DNPAOs에 의한 인 제거 (The Removal of the Phosphorus by DNPAOs According to the Loading of the Influent NO3-N in Anoxic Zone)

  • This study was conducted to investigate the ratios of phosphorus release to COD uptake, phosphorus release to nitrate removal, and phosphorus uptake to phosphorus release by DNPAOs(denitrifying phosphate accumulating organisms). In case $I{\sim}IV$, influent 1 were fed with synthetic wastewater with influent 2 $NO_3^--N$ injection to anoxic zone and the case V were fed with municipal wastewater with side stream oxic zone instead of influent 2 $NO_3^--N$ injection. As a result, the ratio of phosphorus release to carbon uptake was increased in accordance with nitrate supply. The DNPAOs simultaneously took up phosphate and removed nitrate from the anoxic reactor. In case $I{\sim}IV$, with above 20 mg/L of sufficient $NO_3^--N$ supply, phosphate was taken up excessively by the DNPAOs in anoxic condition. The large amount of both uptake and release of phosphorus occurred above 20 mg/L of nitrate supply, achieving the ratio of phosphorus uptake to phosphorus release to be 1.05. In case V, phosphate luxury uptake was not occurred in system due to 6.98 mg/L of insufficient $NO_3^--N$ supply and the ratio of phosphorus uptake to phosphorus release was 0.98. Consequently, if nitrate as the electron acceptor was sufficient in anoxic zone, the ratio was found to be high.

Reversed Effects of Phosphate Fertilizer on Reducing Phytoavailability of Cadmium in Mine Tailing Affected Soil

  • To reduce effectively cadmium (Cd) phytoavailability by phosphate in mine tailing affected soil, fused and super phosphate (FSP), a main phosphate fertilizer in South Korea, was selected as phosphate source and then applied at the rates of 0, 78, 390, and 780 $P_2O_5kg\;ha^{-1}$. FSP did not decrease Cd extractability and radish Cd uptake, but increased significantly. The effect of Cd supplement and soil negativity increase through FSP application was not significant. Soil pH decreased markedly with increasing FSP application, which increased significantly soil Cd extractability and radish Cd uptake. As a result, phosphate fertilizer for reducing Cd phtyoavailability in heavy metals contaminated soil should be carefully selected as alkaline type.

Cu(II)를 이용하여 표면개질된 활성탄의 인산염 제거효율 향상 (Enhancement of phosphate removal using copper impregnated activated carbon(GAC-Cu))

  • The adsorption process using GAC is one of the most secured methods to remove of phosphate from solution. This study was conducted by impregnating Cu(II) to GAC(GAC-Cu) to enhance phosphate adsorption for GAC. In the preparation of GAC-Cu, increasing the concentration of Cu(II) increased the phosphate uptake, confirming the effect of Cu(II) on phosphate uptake. A pH experiment was conducted at pH 4-8 to investigate the effect of the solution pH. Decrease of phosphate removal efficiency was found with increase of pH for both adsorbents, but the reduction rate of GAC-Cu slowed, indicating electrostatic interaction and coordinating bonding were simultaneously involved in phosphate removal. The adsorption was analyzed by Langmuir and Freundlich isotherm to determine the maximum phosphate uptake(qm) and adsorption mechanism. According to correlation of determination(R2), Freundlich isotherm model showed a better fit than Langmuir isotherm model. Based on the negative values of qm, Langmuir adsorption constant(b), and the value of 1/n, phosphate adsorption was shown to be unfavorable and favorable for GAC and GAC-Cu, respectively. The attempt of the linearization of each isotherm obtained very poor R2. Batch kinetic tests verified that ~30% and ~90 phosphate adsorptions were completed within 1h and 24 h, respectively. Pseudo second order(PSO) model showed more suitable than pseudo first order(PFO) because of higher R2. Regardless of type of kinetic model, GAC-Cu obtained higher constant of reaction(K) than GAC.

Verification of Enhanced Phosphate Removal Capability in Pure Cultures of Acinetobacter calcoaceticus under Anaerobic/Aerobic Conditions in an SBR

  • Laboratory experiments were conducted using pure cultures of Acinetobacter under an-aerobic/aerobic cyclic conditions to explain the release and uptake of soluble phosphate in an activated sludge process showing enhanced biological phosphate removal (EBPR). Under anaerobic/aerobic cyclic conditions in a Sequencing Batch Reactor (SBR), COD uptake concurrent with soluble phosphate release by Acinetobacter was not significant during the anaerobic periods, indicating that EBPR would not be established in pure cultures. However Acinetobacter cells accumulated higher phosphate content (5.2%) in SBR than that obtained (4.3%) from batch experiments. These results suggest that Acinetobacter sp. may not follow the proposed pattern of behavior of poly-P bacteria in EBPR activated sludge Plants.

Hydrogen Peroxide-induced Alterations in Na+-phosphate Cotransport in Renal Epithelial Cells

  • This study was undertaken to examine the effect of oxidants on membrane transport function in renal epithelial cells. Hydrogen peroxide ($H_2O_2$) was used as a model oxidant and the membrane transport function was evaluated by measuring $Na^+$-dependent phosphate ($Na^+$-Pi) uptake in opossum kidney (OK) cells. $H_2O_2$ inhibited $Na^+$-Pi uptake in a dose-dependent manner. The oxidant also caused loss of cell viability in a dose-dependent fashion. However, the extent of inhibition of the uptake was larger than that in cell viability. $H_2O_2$ inhibited $Na^+$-dependent uptake without any effect on $Na^+$-independent uptake. $H_2O_2$-induced inhibition of $Na^+$-Pi uptake was prevented completely by catalase, dimethylthiourea, and deferoxamine, suggesting involvement of hydroxyl radical generated by an iron-dependent mechanism. In contrast, antioxidants Trolox, N,N'-diphenyl-p-phenylenediamine, and butylated hydroxyanisole did not affect the $H_2O_2$ inhibition. Kinetic analysis indicated that $H_2O_2$ decreased Vmax of $Na^+$-Pi uptake with no change in the Km value. Phosphonoformic acid binding assay did not show any difference between control and $H_2O_2$-treated cells. $H_2O_2$ also did not cause degradation of $Na^+$-Pi transporter protein. Reduction in $Na^+$-Pi uptake by $H_2O_2$ was associated with ATP depletion and direct inhibition of $Na^+$-$K^+$-ATPase activity. These results indicate that the effect of $H_2O_2$ on membrane transport function in OK cells is associated with reduction in functional $Na^+$-pump activity. In addition, the inhibitory effect of $H_2O_2$ was not associated with lipid peroxidation.

Hydrogel 키토산비드를 이용한 수중의 양이온 중금속과 음이온의 제거 효율 평가 (Removal of both cation and anion pollutant from solution using hydrogel chitosan bead)

  • Cu(II) can cause health problem for human being and phosphate is a key pollutant induces eutrophication in rivers and ponds. To remove of Cu(II) and phosphate from solution, chitosan as adsorbent was chosen and used as a form of hydrogel bead. Due to the chemical instability of hydrogel chitosan bead (HCB), the crosslinked HCB by glutaraldehyde (GA) was prepared (HCB-G). HCB-G maintained the spherical bead type at 1% HCl without a loss of chitosan. A variety of batch experiment tests were carried out to determine the removal efficiency (%), maximum uptake (Q, mg/g), and reaction rate. In the single presence of Cu(II) or phosphate, the removal efficiency was obtained to 17 and 16%, respectively. However, the removal efficiency of Cu(II) and phosphate was increased to 50~55% at a mixed solution. The maximum uptake (Q) for Cu(II) and phosphate was enhanced from 11.3 to74.4 mg/g and from 3.34 to 36.6 mg/g, respectively. While the reaction rate of Cu(II) and phosphate was almost finished within 24 and 6 h at single solution, it was not changed for Cu(II) but was retarded for phosphate at mixed solution.

Arbuscular mycorrhizal fungus인 Glomus manihot 균 접종이 고추의 성장과 영양분 흡수에 미치는 영향 (Effects of Arbuscular Mycorrhizal Fungus, Glomus manihot, on Plant Growth and Nutrient Uptake of Pepper Seedlings)

  • 균귄 곰팡이(arbuscular mycorrhizal fungus, AMF)의 하나인 Glomus manihot을 이용하여 고추의 성장과 무기이온 흡수 및 고추의 치명적 질병인 역병 방제에 미치는 효과를 조사한 결과 매우 유의적인 결과를 얻을 수 있었다. AMF 균주 처리에 의해 질소, 인산, 칼리의 흡수와 초기 중량이 증가되었음을 파종 2주만에 확인할 수 있었으며 고추 품종에 따른 차이는 크게 나타나지 않았다. 이는 Glomus manihot이 고추의 발아와 동시에 부리에 공생하게 되어 나타나는 결과였으며 따라서 파종 시기에 맞춰 AMF를 투입하는 것이 중용한 것으로 판단된다. 야외에서 6주간 경작한 결과, 중량은 AMF와 불용성 인산을 동시에 처리한 결과 40% 가까운 증가 효과를 나타내었다. 이러한 결과는 불용성 인산을 사용 가능케 하는 AMF의 특징에 의한 것으로 토양 내 문제점이 되고 있는 인산 과용에 의한 불용성 인산염의 제거에도 매우 좋은 효과를 나타내리라 판단되었다. 또한 Phyto-phtohra capsici에 의한 고추 역병에 대해서도 Glomus manihot의 접종이 그 피해를 경감시킬 수 있는 것으로 나타났다. 따라서 고추 품종에 상관없이 파종 초기에 Glomus manihot을 접종한다면 고추의 성장과 질병 방제효과를 통해 보다 좋은 품질의 고추를 생산할 수 있을 것으로 판단되었다.
