The activities of enzymes of carbohydrate metabolism have been determinated in the host cytosolic and bacteroid fractions of cowpea (Vigna unguiculata) nodules formed with B. japonicum I 16 and in roots of nodulated cowpeas. The host cytosolic fraction of the nodules contained the enzymes of glycolytic pathway and the pentose phosphate pathway, whereas the bacteroids had only limited capacity for carbohydrate metabolism and appeared to be insufficient for the complete glycolytic pathway as well as starch synthesis and degradation. In a time-course study, using plants grown in a glasshouse, the acetylene-reducing activity (ARA) of the nodules increased in parallel with the total N content of plants and protein of the nodules until approximately 8 weeks after planting. Subsequently, the weight and size of the nodules and the weight of the plants continued to increase, but there was a sharp decrease in the ARA and the total N content of the plants.
Among the proteins secreted from Lactobacillus acidophilus KCTC 3151, a 36 kDA and 24 kDa protein, whose amounts were relatively abundant, were purified and their N-terminal amino acid sequences determined. The N-terminal amino acid sequence of 36 kDa protein exhibited high homology with thymidine phosphorylase and glyceraldehyde-3-phosphate dehydrogenase. The N-terminal amino acid sequence of the 24 kDa protein did not show significant homology with proteins in Protein Data Base nor Gene Bank. Nucleotide sequence of the gene encoding 36 kDa protein indicates that the protein possesses the domains for a-helical, phosphate binding and pyrimidine binding sites, which are also shown in thymidine phosphorylases. Also, the protein contains conserved domains of dehydrogenase II and III. However, the activity of thymidine phosphorylase or glyceraldehyde-3-puospnate dehydrogenase could not be detected in the purified fractions of the 36 kDa protein.
To explore the action mechanism of Ijintang in the brain, the authors investigated the effects of Ijintang fractions on MgNaK ATPase and MgCa ATPase in rat brain synaptosomes prepared from cerebral cortex. The activities of MgNaK ATPase and MgCa ATPase were assayed by the level of inorganic phosphate liberated from the hydrolysis of ATP. Fraction WH-95-7 at the concentration of $10^{-2}%$ decreased the activity of MgNaK ATPase about 34.1% and also reduced the activity of MgCa ATPase about 49.3% But, other fractions (WB-95-7, WC-95-7, MB-95-7, MC-95-7, MH-95-7) did not significantly changed the activities of the MgNaK ATPase and MgCa ATPase The decreased activity of MgNaK ATPase by WH-95-7 will decrease the rate of $Ca^{2+}$ efflux, probably via an Na-Ca exchange mechanism and will increase the rate of $Ca^{2+}$ entry by the depolarization of nerve terminals. The reduced activity of MgCa ATPase by WH-95-7 will result in the decreased efflux of $Ca^{2+}$. As a conclusion, it can be speculated that lithium elevates the intrasynaptosomal $Ca^{2+}$ concentration via inhibition of the activities of MgNaK ATPase and MgCa ATPase. and this increased $[Ca^{2+}]i$ will cause the release of neurotransmitters.
Remediation of metal(loid)s-contaminated sites is crucial to protect human and ecosystem. Solidification and stabilization of metal(loid)s by the binder amendment is one of the cost-effective technologies. In this study, metal (loid)s in various field-contaminated soils obtained from steel-making, metal refinery and mining tillage were immobilized by the application of single binders such as diammonium phosphate (DAP), lime, and ladle slag. The efficiency of solidification and stabilization was evaluated by Toxicity Characteristic Leaching Procedure (TCLP) and the Standard, Measurements and Testing programme of European Union (SM&T) extraction processes. In terms of TCLP extraction, the binder was effective in order of lime > DAP > ladle slag. All binders were highly effective in the immobilization of Pb, Zn, Cu, Ni, and Cd. The increased immobilization efficiency is attributed to the increase in the Step III and IV fractions of the SM&T extraction. Lime and ladle slag were highly effective in the immobilization of the metal(loid)s, however, As release increased with DAP due to competition between the phosphate originated from DAP and arsenate. A further study is needed for the better immobilization of multi metal(loid)s using binary binders.
Extractions fro fertilizer and soil samples were performed to yield the operationally defined fractions 'soluble' chromate (extractable with $NH_4NO_3$), 'exchangeable' chromate (extractable with phosphate buffer pH 7.2), and these results were compared with the data obtained by extractions with ammonium sulfate, borate buffer pH 7.2, saturated borax pH 9.6, and polyphosphate (Graham's salt). In order to maintain the pH of extractant solution about constant, the concentration of extractant buffer had to be raised to at least 0.5 M. The results strongly depended on the kind of extractant, and the solid: liquid ratio. For most of the samples investigated, the extraction efficiency increased in the order borate-sulfate-nitrate-phosphate. Whereas the recovery of $K_2CrO_4\;and\;CaCrO_4$ added to the samples of basic slags prior to the extraction was about complete, the recovery of added $PbCrO_4$ was highly variable. In soil extracts, the color reaction was interfered from co-extracted humics, which react with the chromate in weak acid solution during the time period necessary for color reaction (1 hour). However, this problem can be overcome by standard addition and subtraction of the color of the extractant solution. In soil extract of about pH < 7, organic material reduced chromate during the extraction period also, and standard addition of soluble chromate is recommended to prove recovery and the stability of chromate in the samples. In admixtures of soils and basic slags, results for hexavalent chromium were lower than from the mere basic slags. This effect was more pronounced in phosphate than in nitrate extracts. As a proficiency test, samples low in organic carbon from contaminated sites in Hungary were tested. The results from $NH_4NO_3$ extracts satisfactorily matched the results of the Hungarian labs obtained from $CalCl_2$ extractants.
Hexane, Hexane/diethylether and chloroform fractions from Panax ginseng C.A. Meyer stroungly inhibitied human platelet aggregation induced by a high dose of thrombin (2$\mu$/ml). Chloroform fraction more strongly inhibited the platelet aggregation than the other two fraction among them. There were fatty acid ester and phosphate ester instead of polyacethylene compounds in the chloroform fraction.
일산화탄소를 유일한 탄소 및 에너지원으로 이용하여 성장한 Acinetobacter sp. 1의 일산화탄소 산화를 전자전달계를 조사하였다. 이 세균은 a, b, c, 그리고 o형의 사이토크롬을 소유하고 있었고, UQ 10은 이 세균의 생리적인 전자수용제로 작용할 수 없음이 밝혀졌다. 이 세균에 존재하는 사이토크롬들 중에서 b와 o는 일산화탄소 산화와 연관되어 있으나 a와 c는 관계가 없었으며, NAD(P)도 이 세균의 일산화탄소 산화와 무관한 것으로 밝혀졌다.
Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.
시설재배지내 염류와 인산이 집적된 토양(Total-P; $2140mg\;kg^{-1}$)에서 이온간 경쟁에 의한 인산의 유효도를 증진효과 조사하기 위해 현장 작물재배시험을 실시하였다. 규산의 인산유효도 증진효과를 알아보기 위해 규산을 처리하지 않은 삼요소구(NPK)를 대비구로 하여, 인산무시용 조건에서 규산질비료를 세수준(0, 2, $4Mg\;ha^{-1}$) 처리하여 비교하였다. 인산을 시용한 삼요소구에 비해 인산을 무처리하고 규산을 처리시 배추의 생육과 수량은 크게 증대되었다. 특히 인산을 무처리하고 규산을 $4Mg\;ha^{-1}$ 처리시 삼요구에 비해 약 25%의 수량증대가 확인되었다. 식물체내 인산함량은 처리간 유의적인 차이는 없었으나 규산 처리량 증가에 따른 수량증대로 인산 흡수량이 유의적으로 증가하였다. 규산질비료 시용량 증가에 따라 토양 중 유효인산함량의 유의적 증가가 하였으며, 토양의 유효규산과 식물체의 규산함량간($r=0.76^{**}$), 그리고 식물체 규산함량과 배추 수량간($r=0.78^{**}$)에는 고도의 정의 상관관계가 확인되어 시용규산이 토양 내 인산의 유효도 증진과 작물의 인산흡수량 증가에 직접적 효과가 있는 것으로 조사되었다. 규산질비료 처리량이 증가함에 따라 식물체의 인산 흡수량 증가로 인해 시험 후 토양 중 Total-P 함량은 다소 감소하였으나, 유효인산 함량에는 큰 차이를 발견할 수 없었다. 시험 전 토양과 비교할 때 배추수확 후 토양 내 전 인산 함량 감소는 주로 Ca-P 함량 감소로부터 기인된 것으로 조사되었다. 결과적으로 인산 집적지 토양에 규산질비료의 적절한 시용은 인산의 유효도를 증진시켜 작물의 수량을 증대시키고, 인산시비량 감축에 따른 인산집적을 크게 감소시킬 수 있을 것으로 평가되었다.
