옥수수(Zea may L.)재배 토양에서 제지슬러지퇴비의 시용효과: 무기태인산을 중심으로(II) (Influence of paper Mill Sludge Compost to Corn(Zea may L.) cultivated Soil(Incase of Inorganic Phosphorus))

  • 본 연구는 제지슬러지퇴비를 농업분야에 활용할 때 시용효과를 분석하고자 수행하였고, 대상작물은 옥수수였다. 퇴비는 사질토양과 일정비율로 혼합하였으며, 시용효과를 분석하기 위해 인산의 가용화, 작물체의 양분흡수. 이에 따른 토양중 무기인산의 변화를 평가했다. 포트 시험은 비닐하우스에서 이루어졌으며, 인산의 분획물과 작물의 수량을 조사하였다. 포토실험에서 시간에 따른 변화중 Saloid-P의 양은 다소 증가하는 경향이었으나, Fe-P는 감소하였고, 시용량이 많았던 100Mg/ha 처리구에서는 감소의 폭이 뚜렷하였다. 제지슬러지퇴비를 시용한 포트에서 옥수수를 재배한 결과, 퇴비의 시용량이 많을수록 수량은 증가하였으며, 질소의 흡수는 증가한 반면, 식물체중 인산의 농도는 감소하는 경향이었다. 그러나 퇴비의 시용량 증가로 식물체중 인산의 농도가 감소하는 경향이었음에도 불구하고 인산의 결핍은 일어나지 않았다.

대두(大豆)에 대한 인광석분말(燐鑛石粉末)의 시용효과(施用效果) (Effect of Application of Phosphate Rock Powder on Soybean Cultivation)

  • 토양중(土壤中) 유효(有效) 인산함량(燐酸含量)이 상이한 밭토양에 콩을 재배하여 인광석(燐鑛石)을 원료로하여 제조된 인산질비료(燐酸質肥料)와 인광석분말(燐鑛石粉末)의 직접시용 효과를 비교 관찰하였고 시용(施用)한 인산성분(燐酸成分)이 토양중에서의 반응(反應)과 변화(變化)를 통하여 인산(燐酸)의 존재형태(存在形態)를 파악하였다. 신개간지(新開墾地) 밭토양에서 콩의 수량(收量)은 1년차에서 인광석분말(燐鑛石粉末)을 400kg과 600kg/ha으로 다량(多量) 시용한 처리가 과석(過石) 보다 증수하였으나 2년차에는 인광석분말(燐鑛石粉末)을 200㎏으로 연용(連用)한 처리를 제외하고는 감수(減收)하였다. 저(低) 인산(燐酸) 밭토양에서 콩의 수량(收量)은 인광석분말(燐鑛石粉末)을 다량(多量) 시용한 처리에서 증수(增收)하였고 잔효(殘效)도 인정되었다. 식물체(植物體) 및 인산(燐酸) 및 칼슘함량은 인광석분말(燐鑛石粉末)의 시용량(施用量)이 많을수록 높았다. 수확기(收穫期) 저(低) 인산(燐酸) 밭토양의 인산형태별(燐酸形態別) 함량(含量)은 시용량(施用量)이 증가할수록 인광석분말(燐鑛石粉末)이 과석(過石) 및 용성린비(熔成燐肥) 보다 많았으며, 인산형태(燐酸形態)는 1년차에는 T-P가 가상 높고 Org-P, Fe-P, Ca-P, Al-P순(順)으로 낮아졌고 2년차는 1년차와 동일(同一)하였다.

Chlorella의 인산화합물 및 유기물함량에 미치는 중금속의 영향 (Effects of heavy metals on contents of various phosphate compounds and organic compounds in chlorella cells)

  • The effects of heavy metals on the growth rate and phosphate metabolism of Chlorella elliposidea cells were investigated. Chlorella cells were cultured in the media treated with Hg(0.3, 0.7, 0.9 ppm), Cd(1, 5, 15ppm), and Zn(1, 5, 50ppm) for 6days. Aliquots cells were taken out at the inoculation and at intervals during the culture, and measured packed cell vlolume and optical density. The inhibitions of heavy metals on the growth rate and chlorophyll contents were traced. Also after 6 days culture, the amounts of inorganic phosphate and organic compounds of various fractions in Chlorella cells were observed. The turbid effects of heavy metals on the growth rate and chlorphyll contents of Chlorella cells were in order of Hg>Cd>Zn. Because heavy metals depressed the biosynthesis of inorganic polyphosphates and nucleic acids and turn over of inorganic phosphates, the amounts of various phosphate compounds were decreased. The inhibitory effect of photosynthesis by heavy metals resulted in lower contents of carbohydrate. Due to the turbidity of biosynthesis of amino acids by heavy metals, contents of protein were reduced in comparison with those of control. It is suggested conciusively that the minimum concentrations affected by heavy metals on the growth rate and phosphate metabolism of Chlorella cells were 0.7 ppm Hg, 15ppm Cd, 50ppm Zn.

  • 옥수수 글루텐을 식물성 효소인 파파인으로 가수분해하여 글루텐 펩타이드를 생산하였다. 생산된 글루펜 펩타이드류는 인이온 존재하에서 인이온과 칼슘이온과의 침전형성을 방지하여 칼슘의 용해성을 증진시켰다. 칼슘이온의 용해도는 8.3mg의 펩타이드 존재시 대조구에 비해 5.2배 증대되었다. 산성 아미노산의 함량이 매우 높은 이들 펩타이드류를 Delta pak 칼럼을 이용하여 분획하였으며 이들 분획 가운데 3번째 분획이 가장 높은 칼슘 용해성을 보였다.

  • 1.Each cells homogenized from Undaria were reacted in reaction micture to persue the phosphate metabolism in Undaria cell. Aliquots of the cells were taken out at the begin-ning and at intervals during the reaction, and analyzed for the content of total-P in various fractions of the cell constituents. 2.The P-contents in fraction of polyphosphate "B" decreased remarkably, while that in fraction of RNA polyphosphate "C" showed slow increase. 3.As well as in Chlorella cells, inorganic phosphates in DNA-P, protein-P, and lipid-P were transferred from polyphosphate, RNA-P turnovered from inorganic phosphate that is in cytoplasm, and RNA polyphosphate complex from polyphosphate, and it was suggested that inorganic phosphates in polyphosphate "B" could transformed into polyphosphate "A" & "C", and polyphosphate "C" into polyphosphate "A".

  • Starting from the February 2008 till the end of November 2014, abundance of phytoplankton and their composition living in the coastal waters of Kyoungju city were investigated. Environmental and biological properties were also observed at 10 different stations on a seasonal basis. Due to the environmental variables, fluctuating pattern was appeared during the entire period of observation with different degree, as compared to those found in other costal waters in the East Sea. The concentration of phosphate was turned out to be very low which was even less than threshold level in the study area. Phytoplankton community structure was dominated by diatoms (both micro- and nanoplankton fractions) for several years and seasonal succession was also relied on the diatoms. The importance of dinoflagellates in the community was relatively low. Abundance of phytoplankton was heavily affected by physical factors in the surface water, however, affected more by chemical factors including nutrients in the deep water. For periods of this study, the phosphate concentrations was observed in an extremely low, which indicates that the main limiting factor affecting phytoplankton growth could be phosphate.

Anti-obesitic Effect of Orostachys japonicus in Rats Model fed a Hyperlipidemic Diet

  • We investigated the effect of Orostachys japonicus extracts on serum lipids, leptin and insulin level in hyperlipidemic rats. Also, diacylglycerol acyltransferase (DGAT) activity and thiobarbituric acid reactive substance (TBARS) were assessed. Inhibitory effect of DGAT related to triglyceride synthesis emerged approximately 96% in EtOAc fraction and showed 90% and 67%, respectively, in CHCl3 and BuOH fractions. Furthermore, the EtOAc and BuOH fractions inhibited 81% and 77%, respectively, in glycerol-3-phosphate acyl transferase (GPAT). Hyperlipidemia and obesity marker, contents of leptin and insulin on serum of hyperlipidemic rats, decreased 50% and 25%, respectively, compared with control group in treated EtOAc fraction. The oxidative stress marker, a concentration of TBARS, showed decrease of approximately 30% in treated EtOAc fraction. Moreover, high density lipoprotein (HDL)-cholesterol contents on serum of rats fed a hyperlipidemic diet were increased 10% and low density lipoprotein (LDL)-cholesterol decreased 50% as well as triglyceride amount of feces multiplied approximately two times more than control group in treated EtOAc fraction. The data suggest that the fractions of O. japonicus may be a potent biomaterial for treatment of hyperlipidemia or obesity.

지렁이(지룡)의 해열성분에 관한 연구 (Studies of Antipyretic Component of the Earthworm)

  • In order to confirm the exact antipyretic component in the earthworm, etherial extract of American earthworm(Red Worm) was fractionated into five fractions by using silica gel column chromatography and thin layer chromatography. The fraction including free fatty acids was found to possess artipyretic response and standard arachidonic acid showed marked antipyretic response on typhoid vaccinated rabbits. Arachidonic acid was identified from the free fatty acid fraction of the earthworm by using gas liquid chromatography. Thus it was considered that the antipyretic activity of the free fatty acid may be due to the presence of arachidonic acid. Lipid-free earthworm powder was extracted with phosphate buffer (pH, 8.0, 0.1M) and all the proteins was salted out by ammonium sulfate. The crude precipitate was dialyzed and the impure proteins were eliminated at pH 5.4 and 4.6. The remaining protein solution was fractionated with various concentrations of acetone. The acetone fractions were identified by using S.D.S. polyacrylamide gel electrophoresis and disc gel electrophoresis. The precipitate at 85% acetone concentration and the remaining proteins in the supernatant did not exhibit the antipyretic activity.

SLS에 의한 PCL/TCP 복합체 제작공정변수의 최적화 (Processing Optimization of PCL/TCP Composites Produced by Selective Laser Sintering)

  • This article investigates the fabrication of polycaprolactone (PCL) composites filled with different volume fractions (10-30%) of tricalcium phosphate (TCP) by selective laser sintering (SLS) for tissue engineering scaffolds. Optimal processing parameters for each composition were developed by design of experiments (DOE). Specimens for compressive testing for each composition were fabricated and tested. The results showed that the compressive modulus increases as a function of TCP volume fraction. The experimentally measured compressive moduli were compared with moduli predicted by Halpin's theoretical model and were found to be in excellent agreement. This result proved that experimentally determined processing parameters for each composition were well optimized.

Chlorella의 생리적, 생화학적 제활성에 미치는${\gamma}$-선의 영향

  • The sensitivities of Chlorella ellipsoidea to ${\gamma}$-ray from Cobalt-60 were determined by measuring the photosynthetic and respiratory activities and the changes in phosphate contents in various fractions of the irradited cells, which were further grown in a standard medium after irradiation, were compared to those of non-irradiated normal cells. The photosynthetic and repiratory activities of the cells were almost inversely proportional to the dose of ${\gamma}$-ray irradiated and the photosynthetic activity was more sensitive than the respiratory activity of the cells. The most sensitive to ${\gamma}$-ray was growth activity, followed by photosynthesis, exogenous and endogenous respirations of the cells in decreasing order. Chlorella cells were so resistant to ${\gamma}$-ray comapred with other organisms that about 280,000 r dose of ${\gamma}$-ray irradiaton was necessary to reduce as much as half the subsequent photosynthetic activity. When the irradiated algae were further cultured in a standard medium, the phosphate contents in the fraction of DNA, RNA and phosphoprotein decreased considerably compared with those of non-irradiated normal cells, while the phosphate contents in the fraction of polyphosphates increased than those of control. Therefore, it was deduced that ${\gamma}$-ray inhibited the synthesis of DNA from polyphosphates, that the growth of Chlorella cells were consequently retarded.
