The production of extracellular mannitot by an efficient mannitol-producing bacterium, Lactobacillus sp. KY-107 was studied in shake flask culture using the modified MRS medium. Maximum mannitol production was obtained with fructose as the sole carbon source. Within 95 hours of incubation, a final concentration of 70g/L of mannitol from 100g/L fructose was obtained with an indicated yield of 86% based on fructose consumed. However, higher concentrations of fructose could not effectively be transformed to mannitol due to a lack of osmotolerance. The strain produced no other polyols such as glycerol and sorbitol as by-products. Yeast extract was best nitrogen source and high levels of inorganic phosphate up to 10g/L did not show any detrimental effect for mannitol formation. Manganese ion played important role in both cell growth and mannitol production. The optimum culture temperature and initial pH were $35^{\circ}C$ and 6-8, respectively.
Fungi, such as the wood-rotting Polyporus brumalis, are excellent sources of pharmaceutically interesting natural products such as sesquiterpenoids. In this study, we investigated the biosynthesis of P. brumalis sesquiterpenoids on modified medium. Ten additional species of white rot fungi were inoculated in medium containing nutrients such as $C_6H_{12}O_6$, $C_4H_{12}N_2O_6$, $KH_2PO_4$, $MgSO_4$, and $CaCl_2$ at $28^{\circ}C$ for 25 days. After 10 days of incubation, eudesmane-type sesquiterpenes, ${\beta}$-eudesmane and ${\beta}$-eudesmol, were only synthesized during the growth phase of P. brumalis. Experiments excluding one nutrient at a time were conducted to determine the effects of inorganic nutrients on sesquiterpene biosynthesis. In conclusion, GC-MS analysis showed that biosynthesis of sesquiterpenes was differentially regulated by inorganic nutrients such as $MgSO_4$, $C_4H_{12}N_2O_6$, and $KH_2PO_4$. We found $MgSO_4$ supplementation to be vital for eudesmane-type sesquiterpene biosynthesis in P. brumalis; nitrogen ($C_4H_{12}N_2O_6$) and phosphate ($KH_2PO_4$) inhibited the synthesis of P. brumalis metabolites. Magnesium is a known cofactor of sesquiterpene synthase, which promotes ${\beta}$-eudesmol synthesis. To mechanistically understand eudesmane-type sesquiterpene biosynthesis in P. brumalis, further research into the genes regulating the dynamics of such biosynthesis is warranted.
The solubility and physicochemical stability of caroverine hydrochloride (CRV), an antispasmodic, in buffered aqueous solutions were studied using a reverse phase high performance liquid chromatography. The solubilty of the drug at pH 2.76-5.40 was similar at the range 31.9-36.2 mg/ml $(34^{circ}C)$, but, at the pH higher than 6.0, markedly decreased. The use of polyethylene glycol 400 as a cosolvent did not increase the solubility at any compositions examined. Moreover. increasing molar concentration of aqueous phosphate buffer from 0 to 0.5 M remarkably decreased the solubility. The degradation of CRY followed the apparent first-order kinetics. The degradation was accelerated with decreasing pH and increasing storage temperature. The half-lives for the degradation of CRY (1.0 mg/ml) at pH 1.28. 4.01 and 5.93 $(45^{\circ}C)$ were 2.8, 31.4 and 124 hr. respectively. The pHs of incubated solutions were to some extent lowered perhaps due to the formation of acidic degradation products. The addition of disodium edetate (0.01%) to the CRY solution (pH 4.95) retarded 2.5 times the degradation rate at $45^{\circ}C$, but the use of sodium bisulfite (0.1%) accelerated 2.9 times the rate. The activation energy for the CRY solution (20 mg/ml. pH 5.4) containing 0.01% EDTA was calculated to be 5.98 kcal/mole. When the solution was stored under nitrogen displacement in ampoule, there was no significant degradation even after 3 months at $40^{\circ}C$, indicating that protection from oxidation by air (oxygen) is essential for the complete stabilization of CRY solution.

The carbon dioxide, nitrogen, and phosphate removals from wastewater using microalgae have extensively been studied. Especially, the green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. These hydrocarbons suggest that the possibility of renewable biofuels to be converted into useful fuels such as gasoline by simple catalytic cracking. The poor recovery (18 - 32%) of hydrocarbon from B. braunii culture in two-phase bubble column seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. In order to overcome low recovery efficiency, two-stage extraction culture system including culture vessel and two-phase separator is now under development, resulted improving contact between solvent phase and cell suspension. Hydrocarbon recovery using this process was more than two times as that using two-phase extraction culture.
In the medium containing 7% rice powder, 0.15% $NH_4NO_3$ and 0.1% $MnSO_4$ with initial pH 6.0, the red pigment production by Monascus anka albidus was observed. The mycelium were cultured at $30^{\circ}C$ for 5 days with reciprocal shaking (130 rpm). As a carbon source, glutinous rice gave the highest production of pigment. Ammonium nitrate and $KH_2PO_4$ as a nitrogen source and phosphate source, respectively, stimulated best the production of the red pigment. The optimum C:N ratio was found to be 18:1. The production of the pigment by the strain was 2.6 mg/ml in a flask, but 1.8 mg/ml in 5l fermentor, respectively.
In this study, algae cultivation using the photobioreactor has been applied to remove the nitrogen and phosphorus compounds in the wastewater of the livestock industry. The optimal ratio of nitrate and ortho-phosphate concentration was found for the enhancement of removal efficiency. To achieve the high density culture of algae, the photobioreactor consisted of optical fibers wes developed to get the sufficient light intensity. The light could be illuminated uniformly from light source to the entire reactor by the optical fibers. The structured kinetic model was proposed to describe the growth rate, consumption rate of nitrates and ortho-phosphates in algae culture. The self-organizing fuzzy logic controller incorporated with genetic algorithm was constructed to control the semi-continuous wastewater treatment system. The proposed fuzzy logic controller was applied to maintain the nitrated concentration at the given set-point with the control of wastewater feeding rate. The experimental results showed that the self-organizing fuzzy logic controller could keep the nitrate concentration and enhance algae growth.

Hydroponic farming is a method to grow a plant without soil. Plants can be grown on water or hydroponic growing media, and they are fed with mineral nutrient solutions, which are fertilizers dissolved into water. Hydroponic farming has the advantage of increasing plant productivity over conventional greenhouse farming. Previous studies of hydroponic nutrient wastewater from acyclic hydroponic farms pointed out that hydroponic nutrient wastewater contained residual nutrients, and they were drained to a nearby river bank which causes several environmental issues. Also, previous studies suggest that excessive use of the nutrient solution and disposal of used hydroponic growing media and crop wastes in hydroponic farms are major problems to hydroponic farming. This study was conducted to determine the impact of hydroponic nutrient wastewater, used hydroponic growing media, and crop wastes from acyclic hydroponic farms on the surrounding environment by analyzing water quality and soil analysis of the above three factors. Three soil cultivation farms and several hydroponic farms in the Gangwon C region were selected for this study. Samples of water and soils were collected from both inside and outside of each farm. Also, a sample of soil and leachate from crop waste piles stacked near the farm was collected for analysis. Hydroponic nutrient wastewater from acyclic hydroponic farm contained an average of 402 mg/L of total nitrogen (TN) concentration, and 77.4 mg/L of total phosphate (TP) concentration. The result of TP in hydroponic nutrient wastewater exceeds the living environmental standard of the river in enforcement decree of the framework act on environmental policy by 993.7 times. Also, it exceeds the standard of industrial wastewater discharge standards under the water environment conservation act by 6~19 times in TN, and 2~27 times in TP. Leachate from crop waste piles contained 11,828 times higher COD and 395~2662 times higher TP than the standard set by the living environmental standard of the river in enforcement decree of the framework act on environmental policy and exceeds 778 times higher TN and 5 times higher TP than the standard of industrial wastewater discharge standards under the water environment conservation act. For more precise studies of the impact of hydroponic nutrient wastewater, used hydroponic growing media, and crop wastes from acyclic hydroponic farms on the surrounding environment, additional information regarding a number of hydroponic farms, arable area(ha), hydroponic farming area, seasonal, weather, climate factor around the river, and the property of the area and farm is needed. Analysis of these factors and additional water and soil samples are needed for future studies.
To reduce both the floatation of the seedling of rice and the failure in standing in the paddy field when the barley straw was applied to paddy field before planting the rice, we tested the effect of rice rooting with plowing methods and irrigation rates for 2 years from 2003 to 2004. This study was carried out in paddy field with Fluvio-Marine deposit in Jeonbug series and the operating accuracy and the change of soil physico-chemical properties depending on plowing methods and irrigation rates following the barley straw applying were examined. There was a less floatation of barley straw in the dry-rotaryI+water-rotaryI(DRI+WRI) plot than in the plowing+water-rotary(PL+WRI) plot. The ratio of miss-planted and floating seedling also decreased by 1.7%, 2.6% in the DRI+WRIplot compared with PL+WRI plot. The soil physical property was improved with the decreasing soil hardness, bulk density and increasing soil porosity after the application of barley straw, especially enhanced greatly in the increase of porosity, gaseous phase and with the decrease of soil hardness, bulk density of subsurface soil in DRI+WRI plot. And the change of soil chemical property were increased the content of total carbon$^{\circ}{\S}$nitrogen$^{\circ}{\S}$organic matter and available phosphate while decreased the content of exchangeable cations and available silicate after the application of barley straw. Also the content of organic matter, available phosphate and cation exchangeable capacity were increased, whereas caron/nitrogen ratio was decreased in DRI+WRI plot compared with PL+WRI plot. The number of panicles, spikelets per square meter were increased and 1,000 grains weight of hulled rice was gained more in DRI+WRI plot at irrigation rate of $500ton\;ha^{-1}$, in DRI+WRII plot at irrigation rate of $700ton\;ha^{-1}$. So the rice yields were increased by 7%, in DRI+WRI and 5% in DRI+WRII plot, respectively compared with PL+WRI plot. The result of this study indicated that the most appropriate plowing method with barley straw application on rice cultivation at double cropping in normal paddy field plain land was DRI+WRI.

Soybean meal was fermented by Aspergillus usami in order to reduce phytate content. Aflatoxin B1 was not detected in the fermented soybean meal. The contents of crude protein, crude fiber, ether extract and crude ash were slightly increased following fermentation with a concomitant reduction in nitrogen free extract. Though the fermentation partly degraded proteins in the soybean meal, there was small difference in amino acid composition between the soybean meal and the fermented soybean meal. The results showed that the fermentation did not affect nutritional value of protein in soybean meal. Approximately 55% of phosphorus extracted by trichloroacetic acid was inositol hexaphosphate (phytate) in the soybean meal. The content of inositol tetra to hexaphosphates was not detected in the fermented soybean meal. These results indicated that the fermentation almost completely eliminated phytate in soybean meal. Phytase activity was not detected in the unfermented soybean meal. However, the enzyme activity in the fermented soybean meal was 167.7 U/g. When the fermented soybean meal in supplemented in formula feeds, phytase in the fermented soybean meal might partly degrade the phytate in other ingredients in the digestive tract. The fermented soybean meal is possibly used as a phytate-free protein source of feed, which contains high available phosphorus.
Culture conditions of yellow pigment in Monascus sp. were studied. According to the studies of culture conditions optimum condition was found to be pH 4.5, 3 days of incubation with 3% of sucrose as carbon source, 0.2 % of yeast extract as nitrogen source and 75m1 of medium in the 500m1 erlenmyer flask by rotary shaking (rpm 180) at 180 r.p.m. Effective levels of inorganic compounds were found to be 0.25 % of potassium phosphate monobasic and 0.1 % of Magnesium sulfate.
