천마의 젖산발효에 따른 이취성분 및 Parishin 유도체의 변화 (Changes of Off-Odor Constituent and Parishin Derivatives of Fermentation of Gastrodia elata Rhizome by Lactic Acid Bacteria Strains)

  • Gastrodia elata Blume often has been used for the treatment of headaches, convulsions, hypertension, and neurodegenerative diseases. The main active constituents are gastrodin, 4-hydroxybenzyl alcohol, vanillyl alcohol, 4-hydroxybenzylaldehyde and parishin A, B, C and E. Because Gastrodia elata has also unacceptable off-odor (swine barnyard-like) for food, there is a need to reduce it as well as allow for greater utilization as a functional food materials. In this study, a major off-odor producing substance of Gastrodia elata was fractionated by steam distillation and silica gel column chromatography. The substance was identified as p-cresol(4-methyl phenol) by GC-MS analysis and comparison of the retention time with that of an authentic compound in GC. The content of p-cresol in fermented Gastrodia elata was decreased. A fermented sample of Latobacillus sakei for 2 days was reduced to 54.7%, when compared with a unfermented sample. The five parishin derivatives in Gastrodia elata were identified by HPLC-MS analyses, and a comparison of HPLC retention times with those of authentic compounds. When compared with parishin derivatives of an unfermented Gastrodia elata, those of Gastrodia elata fermented by L. sakei, increased to 18.3% for 2 days. Increases of about 14.0~38.4% of the total phenolic compounds and 57.4~77.3% total flavonoids were found in fermented extracts, by 3 lactic acid bacteria strains. They were compared with $97.1{\pm}2.9{\mu}g/g$ and $40.9{\pm}2.0{\mu}g/g$ in the unfermented control, respectively. The extracts of Gastrodia elata Blume that were fermented by lactic acid bacteria had higher DPPH free radical scavenging activity and FRAP reducing power than the unfermented control.

섬개야광나무 잎 추출물 및 분획물의 생리활성 평가 및 활성 물질 분리 (Biological Activities and Separation of Active Substance of Extract and Fractions from Cotoneaster wilsonii Nakai Leaf)

  • Background: Previously, studies have observed that the leaf extract of Cotoneaster wilsonii Nakai has potent antioxidant and anti-inflammatory activites. Therefore, further research was conducted to separate the active antioxidant and anti-inflammatory compounds in the leaf of the C. wilsonii. Methods and Results: The anti-oxidant effects were evaluated by analyzing the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, estimating the totla phenolic content (TPC) and total flavonoid content (TFC), and using reducing power assay method. The ethyl acetate (EtOAc) fraction demonstrated the highest DPPH radical scavenging activity (IC50; 9.69 ㎍/㎖), and the highest TPC and TFC (345.98 mg·GAE/g, and 74.23 mg·QE/g). Moreover, it indicated the highest effects on nitric oxide production. Therefore, the active compound was separated using the EtOAc fraction, determined on the basis of spectral data, and identified as (+)-catechin. Conclusions: The first compound separated was made from the leaf of C. wilsonii was (+)-catechin, which produced potent anti-oxidant and anti-inflammatory effects.

석화의 캘러스 유도 및 메틸 자스모네이트 처리에 의한 항염증 활성 증진 (Callus Induction and Increase in Anti-Inflammatory Activity by Treatment of Methyl Jasmonate in Adenium obesum)

  • Background: Callus cultivation has the advantage of producing a large amount of tissue of a plant in a laboratory regardless of the environment, for extracting an active substance. In the present study, callus formation was induced in the leaves of the succulent plant Adenium obesum (Forssk.) Roem & Schult. After callus cultivation, anti-inflammatory activity tests were conducted, because leaves and stems of A. obesum have been reported to possess biological activity. Methods and Results: In order to induce callus formation, various concentrations of plant growth factors, such as kinetin, naphtha-leneacetic acid (NAA), 6-benzyladenine (BA), and indole-3-acetic acid (IAA) were added to MS solid medium. The maximum callus proliferation was induced by mixed medium consisting of NAA ($2mg/{\ell}$) and BA ($1mg/{\ell}$). In addition, an elicitor was added to the medium under optimal conditions for initiating suspension culture. After suspension culturing, the activities of the callus extracts were compared and analyzed. The cytotoxicity and anti-inflammatory activity tests revealed that the anti-inflammatory activity of the callus extract and the content of phenolic compounds were elevated after treatment of the callus culture with the elicitior. Conclusions: A. obesum callus might be considered as potential source of biologically active anti-inflammatory material.

효모에 의한 phenol 성 물질의 자화에 관한 연구 (Studies on the Utilization of Phenolic Substance by Yeast)

  • 분리된 효모에 의한 aromatic ring compound의 전환반응 실험에 앞서 phenol 자화능에 대한 환경 인자의 영향을 조사하기 위하여 phenol을 유일한 탄소원으로 사용하여 분리된 효모균을 30시간, 3$0^{\circ}C$에서 진탕배양하여 균체증식량을 측정하였다. 1) pH3.5~4.5 온도 35$^{\circ}C$에서 가장 자화능이 우수하였으며 phenol의 처음 농도 1000ppm 까지 는 쉽게 이용할 수 있었고 2000ppm에서는 5 일간 배양시켰을 때 비로소 균체증식이 나타났다. 2) pH 5.0, 배양온도 3$0^{\circ}C$에서 본 균의 maximum specific growth rate는 0.27hr-1 이었으며, phenol 을 1000ppm씩 단계적으로 첨가하면서 phenol 소비율에 대한 균체수율 정수 Y값을 구한 결과 Y=3.2였다. 3) Yeast extract를 필수영양원으로 첨가할 때 0.005~0.01% 농도에서 가장 효과적이었으며, 또 산화환원소 효소 조해제인 HgCl$_2$, phenylhydrazine 을 명명 $10^{-5}$M, $10^{-3}$M을 배지내에 첨가하면 분리된 효모균의 증식을 거의 볼 수 없었다. 4) Phenol의 자화에 명종 실소원의 효과를 비교 해 본 결과 urea가 가장 효과적이었고, 이 효모균은 phenol 외에 catechol, resorcinol, benzidine 을 자화할 수 있다

한국내 고추나물의 하이퍼리신 유전자(HyH)의 탐색과 유럽의 서양고추나물과 비교 (Detect of Hypericin (HyH) gene in Hypericum erectum in Korea and Comparison of H. perforatum in Europe)

  • 하이퍼리신(Hypericin, HyH)은 예로부터 성 요한의 풀(St. John's Wort)로 널리 알려져 있는 서양고추나물(Hypericum perforatum)에서 추출되는 약리성분이다. 서양고추나물은 국내에서는 자생하지 않으나 같은 속의 고추나물(H. erectum) 등이 이속에 속하며 우리나라에 자생한다 . 고추나물을 조직배양으로 RNA를 추출하고 cDNA를 합성한 후 Hyp 유전자를 추출하여 서열화한 결과, 전체 크기는 732 bp로 나타났으며 서양고추나물의 HyH-1과 거의 99.8% 서열 일치를 나타내었다. 이 서열의 152개 아미노산 역시 서양고추나물의 항산화효소와 98% 일치하였으며 자작나무와 능금속 식물에서 유발되는 알레르기 유발유전자와 약 60% 일치를 나타내었다. 본 연구 결과 우울증치료제로 사용되는 하이퍼리신 추출에 우리나라 자생종인 고추나물이 이용될 수 있을 것으로 사료된다.

Antioxidant Enzyme Activity and Meat Quality of Meat Type Ducks Fed with Dried Oregano (Origanum vulgare L.) Powder

  • One-day-old Cherry valley meat-strain ducks were used to investigate the effect of supplemental dried oregano powder (DOP) in feed on the productivity, antioxidant enzyme activity, and breast meat quality. One hundred sixty five ducks were assigned to 5 dietary treatments for 42 days. The dietary treatment groups were control group (CON; no antibiotic, no DOP), antibiotic group (ANT; CON+0.1% Patrol), 0.1% DOP (CON+0.1% DOP), 0.5% DOP (CON+0.5% DOP), and 1.0% DOP (CON+1.0% DOP). Upon feeding, 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity of oregano extracts was higher than that of tocopherol, although it was less than that of ascorbic acid. As a result of in vivo study, DOP in the diet showed no effects on final body weight, feed intake, or feed conversion ratio. However, dietary 0.5% and 1% DOP supplementation caused a significant increase in the serum enzyme activity of superoxide dismutase (SOD) compared with CON and ANT, while glutathione peroxidase (GPx) in tissue was increased as compared to ANT (p<0.05). Cooking loss from ducks fed with DOP decreased compared with the control ducks. Thiobarbituric acid reactive substance (TBARS) values of duck breast meat at 5 d post slaughter was found to be significantly reduced in ducks whose diets were supplemented with 0.5% and 1% DOP (p<0.05). These results suggest that diets containing 0.5% and 1% DOP may beneficially affect antioxidant enzyme activity of GPx and SOD, improve meat cooking loss, and reduce TBARS values in breast meat at 5 d of storage in ducks.

Ericaceous식물의 allelopathy와 경쟁력에 의한 침엽수 갱신 저해 (Competitive Ability and Allelopathy of Ericaceous Plants as Potential Causes of Conifer Regeneration Failures)

  • 온대 삼림지대에서 산화 적지나 개벌 적지에 왕성하게 번성하는 것이 ericaceous속이다. ericaceous속의 식물들은 성장이 빨라서 다른 종과의 경쟁에서 유리하고 또한 allelopathy 효과 때문에 침엽수 갱신에 영향을 미친다. Kalmia가 번성하는 지역에서는 삼림 갱신이 잘 되지 않거나 조림한 침엽수의 생장이 저조하였다. 이러한 생상 저해 현상은 서유럽의 Calluna - Sitca spruce - Scots pine 생태계와 미국의 북서 태평양 지역의 Gaultheria - cedar/hemlock 생태계에서도 나타나고 있다. 삼림이 파괴된 다음에 Kalmia - black spruce 생태계의 다양한 변화는 침엽수 생장을 저해하거나 갱신을 방해하는 원인으로 생각되어지고 있다. 양분경쟁에 대한 논쟁의 여지는 있으나 Kalmia는 black spruce 생장을 억제하는 주원인으로 작용한다. 여러 곳의 야외, 실내 및 온실에서 한 실험 결과는 이러한 결론을 뒷받침해 주고 있다. 본 논문에서는 Kalmia 잎으로부터 8가지의 폐놀물질을 분리동정해 냈으며 이들 중 4가지는 black spruce에 고도의 독성을 나타냈다. Kalmia 우점지역에서 allelopathic 영향을 적게 하므로써 black spruce의 갱신을 증진시키기 위한 여러 가지 방법에 대한 고찰을 하였다.

카르노스산에 의한 화이트소시지의 항산화 및 항균 효과 (Antioxidant and Antibacterial Effects of Carnosic Acid on White Sausage)

  • 로즈마리의 주요성분으로는 carnosic acid, carnosol, rosmarinic acid, ursolic acid 등의 폴리페놀(polyphenolic)로 구성되어 있다. 식물기원의 페놀혼합물은 항산화 작용과 항균작용을 포함한 유용한 기능과 영양적인 효과에 주목을 끌고 있다. 본 연구는 로즈마리 추출물의 하나인 카르노스산을 화이트소시지 원료에 첨가한 후 실온 저장기간 동안 pH, TBARS, VBN, 전자공여능 및 총균수의 변화를 조사하였다. 카르노스산의 첨가는 무첨가구인 대조구에 비해 낮은 pH, TBARS값, 그리고 VBN값을 보여 주었으며, 높은 항산화 작용과 항균작용을 나타내었다. 이들의 활성은 카르노스산의 농도가 높을수록 증가하였다. 또한 카르노스산의 병원성균에 대한 항균활성을 측정한 결과, 두 병원성균(E. coli KCCM 11234와 S. enteritidis KCCM 12021)에 대하여 강한 항균활성을 보여주었다. 따라서 화이트소시지에 있어서 카르노스산은 천연보존제로서 충분히 이용 가능한 결과를 나타내었다.

Effects of Lotus (Nelumbo nucifera) Leaf Hot Water Extracts on the Quality and Stability of Eggs using Ultrasonication Treatment during Storage

  • This study was performed to investigate the effects of lotus leaf hot water extracts treatment on the quality and stability of eggs using impregnation treatment through ultrasonication during storage. A total of 480 eggs were categorized into four treatment groups (n=30 each)-non-treated (CON), soaked for 30 min in lotus leaf hot water extracts without ultrasonication (T1), sonicated in distilled water (T2), and sonicated in lotus leaf hot water extracts (T3)-and stored for 15 d at 30℃. The egg weight, Haugh unit (HU), egg grade, albumen height, yolk color, eggshell thickness, eggshell breaking strength, and weight loss were measured for egg quality assessment. 2-Thiobarbituric acid reactive substance (TBARS) and volatile basic nitrogen (VBN) contents were measured as stability indicators. Additionally, total phenolic contents (TPC), total flavonoid contents (TFC), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity were evaluated. The HU, egg grade, albumen height, and yolk color of T3 were significantly higher than those of CON (p<0.05). No significant differences in eggshell thickness and eggshell breaking strength are observed among the groups. The weight loss of T3 was significantly lower than that of the other groups during storage (p<0.05). The application of lotus leaf hot water extracts also significantly reduced TBARS and VBN (p<0.05). The TPC, TFC, and DPPH radical scavenging activity of T3 were significantly higher than those of the other groups (p<0.05). These results suggest that lotus leaf hot water extracts may be useful as a natural ingredient for improving the quality and stability of eggs during storage.

밤나무 잎의 항미생물 효과 및 활성물질 (Active Compounds and Antimicrobial Effects from Castanea crenata Leaf)

  • 밤나무 잎에 함유된 기능성 물질 검색의 일환으로 항미생물 효과를 검정하고 활성물질의 분석을 시도하였다. 예비실험을 통해 추출물의 활성을 확인한 후 시료를 n-hexane, EtOAc, MeOH로 순차 추출하여 정제한 다음 활성이 강한 MeOH 분획의 0.2 g, 0.5 g에 해당하는 추출물로 13종의 미생물에 대해 활성을 측정한 결과, Cram 양성 세균 중에서 S. aureus와 S. epidermidis는 대조구로 사용한 benzoic acid보다 강한 활성이 나타났고, M. luteus와 L. mesenteroides, B. subtilis에서도 활성이 인정되었으며, Gram 음성 세균인 E. coli, S. typhimurium, P. aeruginosa에서도 강한 활성이 나타나 밤나무 잎 추출물은 비교적 넓은 항균 spectra와 활성이 있음이 확인되었다. 이에 활성물질을 규명하고자 시료를 solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, HPLC 등으로 정제하고 활성이 나타난 산성 획분과 페놀성 획분의 물질을 분리한 후 MS, $^1H-NMR,\;^{13}C-NMR$ 등으로 분석한 결과, 산성 획분에서는 stigmast- 5-en-3-ol($\beta$-sitosterol)이 활성을 갖는 물질로 나타났다.
