This study was performed to determine the composition of phenolic substances contained in the leaves of Cirsium setidens (Compositae), validate the high-performance liquid chromatography (HPLC) method, and determine the in vivo sedative effect of the main component pectolinarin. Six phenolic compounds isolated from C. setidens were spectroscopically identified as chlorogenic acid (1), hyperoside (2), 3,4-di-O-caffeoylquinic acid (3), caffeic acid methyl ester (4), linarin (5), and pectolinarin (6) and then used as standard compounds for HPLC analysis. HPLC proved to be precise, accurate, and sensitive for the simultaneous analysis of the phenolic substances. In particular, six compounds showed good regression ($R^2$ > 0.999) within test ranges and recovery was in the range of 95.4 - 104.8%. The content of pectolinarin was considerably higher (156.48 mg/g) than those of other phenolic substances including the other flavone glycoside, linarin (18.99 mg/g). The contents of other phenolic substances, in order, were chlorogenic acid (8.41 mg/g), 3,4-di-O-caffeoylquinic acid (5.74 mg/g), hyperoside (4.33 mg/g), and caffeic acid methyl ester (0.51 mg/g). Oral administration with compound 6 (10 and 20 mg/kg) enhanced the sleeping time induced by pentobarbital in mice, indicating that it has a sedative effect.

Brown rice flour (BRF) was treated with different carbohydrases (Viscozyme, Termamyl, Celluclast, AMG, Ultraflo, and Pentopan), and then aqueous alcoholic extracts (70% ethanol) from the treated RBF were examined for their phenolic compositions and antioxidant activities (ABTS and DPPH radical scavenging activity). All the carbohydrases tested induced significant increases in ABTS radical scavenging activity (2.1-3.0 times). Moreover, These enzymes increased the amount of extractable free phenolic acids by 10-15 times, especially for ferulic and p-coumaric acid. Among the enzymes tested, Pentopan which was active in arabinoxylan hydrolysis appeared to be most effective in increasing the free phenolic acid content and ABTS radical scavenging activity than other enzymes. Enzymatic hydrolysis of cell wall polysaccharides in BRF could be used as an effective procedure for raising the amount of extractable phenolic acids and thus increasing the antioxidant activity of BRF extract.
In order to verify the effect of using functional cosmetic ingredients for skin beauty, the composition and content of some phenolic acid and flavonoids in the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds were analyzed, and the skin care antioxidant activity was investigated. The results are as follows. Total polyphenol and flavonoid contents were found to be higher in pericarp extract than seed extract in both hot water and 80% methanol extract. In the analysis of the composition and content of phenolic acid and flavonoids, two kinds of phenolic acids were identified in the hot water extract of pericarp, 6 kinds of phenolic acids were identified in the 80% methanol extract, and one kind of flavonoid. Two types of phenolic acids were identified in the hot water and 80% methanol extract of seeds, respectively. DPPH and ABTS radical scavenging activities tended to increase in proportion to the treatment concentration in both hot water and 80% methanol extracts, and antioxidant activity was found to be high. Therefore, from the above results, it was found that the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds contained various kinds of phenolic acid and flavonoids, and the antioxidant activity was also high. It is believed to be of value as a natural antioxidant in skin care cosmetic ingredients.
The aqueous ethanolic whole plant extract of Rosa arabica was found to contain the new natural dimeric phenolic compound, ellagic acid 3,3'-dimethyl ether $4-O-{\alpha}-rhamnopyranoside$, 9, along with ten known phenolic metabolites (1-8, 10 and 11). Structures of all compounds (1-11) were established by routine methods of analysis and confirmed by FAB-MS, $^1H\;and\;^{13}C$ NMR spectral analysis.
Six phenolic acids were isolated from Jindalrae flowers (Rhododendron mucronulatum Turcz.), an edible plant in Korea. These compounds were identified as chlorogenic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, caffeic acid, ferulic acid, and p-coumaric acid on the basis of IR, UV, $^{1}H$ and $^{13}C$ NMR, FAB-MS, ES-MS and/or El-MS data. Chlorogenic acid (0.2 g) present in both ethyl acetate and ethyl ether fractions comprised up to 38.5% of the total phenolic acid amount (0.52 g) finally recovered by means of polyamide C-200 column chromatography, preparative TLC, recrystallization, and Sephadex LH-20 column chromatography The antioxidant activities were measured in an ethanol solution of linoleic acid in the presence of ferric thiocyanate. The antioxidant efficiency increased in the order of p-coumaric acid<${\alpha}-tocopherol$
The free phenolic fraction from Korean white ginseng (Panax ginseng C.A. Meyer) was studied by GLC/MS as trimethylsilyl and methyl derivative. Five phenolic compounds such as 2,6-ditert butyl p-crestal, phloroglucinol, protocatechuic acid, isoferulic acid, quinic acid were identified newly. And additionally 13 organic acids and hydrocarbons were also identified in the fraction.
This study was carried out to investigate the free amino acid and derivatives and phenolic acid and flavonoid compositions and contents of hot water and 95% ethanol extracts of Imperata cylindrica Beauvois var. koenigii root. The purpose of this study is to obtain basic data and to confirm their applicability as functional cosmetic and functional food ingredients on extracts. 15 kinds and 9 kinds of free amino acids were detected in both extracts, respectively. 5 kinds and 4 kinds of protein free-amino acid derivatives were detected in both extracts, respectively. Total phenol contents were 116.50 ± 0.06 and 140.10 ± 0.04 mg% in both extracts, respectively. The total flavonoid contents were 31.80 ± 0.03 and 43.90 ± 0.05 mg% in both extracts, respectively. 5 kinds and 6 kinds of phenolic acids were detected in both extracts, respectively. In the case of flavonoid analysis, no flavonoid compound was detected in the hot water extract while taxifolin was identified in the 95% ethanol extract. Based on the above results, the hot water and 95% ethanol extracts of I. cylindrica Beauvois var. koenigii root are rich in free amino acids, amino acid derivatives, phenolic acids and flavonoids which confirm their potential for applications in cosmeceuticals, nutricosmetics formulations and functional foods.
This study was carried out to investigate the possible determination of total phenolic compounds by Folin-Denis method from Panax ginseng C.A. Meyer and their extraction conditions. It was possible to apply the method for determination of total phenolic compounds from Korean red ginseng. But 3 kinds of amino acids such as tyrosine, cystein and tryptophan, and 3 kinds of vitamins such as ascorbic acid, pyridoxin HCI and thiamine HCI affected strongly the colorization by the method. Effective alcoholic solvent for the extraction was 60% ethanol, appropriate extraction temperature, time and times were 40-80。C, 1-2 hours and 3 times, respectively.
Enhanced production of individual phenolic compounds by subcritical water hydrolysis (SWH) of pumpkin leaves was investigated at various temperatures ranging from 100 to $220^{\circ}C$ at 20 min and at various reaction times ranging from 10 to 50 min at $160^{\circ}C$. Caffeic acid, p-coumaric acid, ferulic acid, and gentisic acid were the major phenolic compounds in the hydrolysate of pumpkin leaves. All phenolic compounds except gentisic acid showed the highest yield at $160^{\circ}C$, but gentisic acid showed the highest yield at $180^{\circ}C$. The cumulative amount of individual phenolic compounds gradually increased by 48.1, 52.2, and $78.4{\mu}g/g$ dry matter at $100^{\circ}C$, $120^{\circ}C$, and $140^{\circ}C$, respectively, and then greatly increased by $1,477.1{\mu}g/g$ dry matter at $160^{\circ}C$. The yields of caffeic acid and ferulic acid showed peaks at 20 min, while those of cinnamic acid, p-coumaric acid, p-hydroxybenzoic acid, and procatechuic acid showed peaks at 30 min. Antioxidant activities such as 2,2-diphenyl-1-picrylhydrazyl and ferric reducing antioxidant power values gradually increased with hydrolysis temperature and ranged from 6.77 to 12.42 mg ascorbic acid equivalents/g dry matter and from 4.25 to 8.92 mmol $Fe^{2+}$/100 g dry matter, respectively. Color $L^*$ and $b^*$ values gradually decreased as hydrolysis temperature increased from $100^{\circ}C$ to $140^{\circ}C$. At high temperatures ($160^{\circ}C$ to $220^{\circ}C$), L* and b* values decreased suddenly. The $a^*$ value peaked at $160^{\circ}C$ and then decreased as temperature increased from $160^{\circ}C$ to $220^{\circ}C$. These results suggest that SWH of pumpkin leaves was strongly influenced by hydrolysis temperature and may enhanced the production of phenolic compounds and antioxidant activities.
