Antioxidant activity of Cinchona officinalis stem bark extracts

  • Stem bark of Cinchona sp. (Rubiaceae) is one of the well known drugs for its therapeutic values in traditional as well as modern medicine. Even though a lot of work has been carried out on quinoline alkaloids of Cinchona, its phenolic constituents received very little attention. In the present study, we evaluated antioxidant properties of C. officinalis stem bark methanolic extract and water extract containing phenolic compounds (total phenolics 21.37, 5.18% w/w respectively in the two extracts) in different in vitro and ex vivo models viz., antiradical activity by DPPH reduction, superoxide radical scavenging activity in riboflavin/light/NBT system, nitric oxide radical scavenging activity in sodium nitroprusside/Greiss reagent system and inhibition of lipid peroxidation induced by iron-ADP-ascorbate in liver homogenate and haemolysis of erythrocytes induced by phenylhydrazine in erythrocyte membrane stabilization study. Both the extracts exhibited very good antioxidant activity in all the models tested. The phenolic compounds including tannins present in the stem bark seem to offer protection from the oxidative damage.

Anti-oxidative effects of Mori Folium, Mori Ramulus and Mori Cortex Radidus (상엽(桑葉), 상지(桑枝) 및 상백피(桑白皮)의 항산화 활성 연구)

  • Objective : This study was designed to investigate the anti-oxidative potentials to wide the usage of Morus alba L. derived from medicinal herbs on cosmoceutical fields. Methods : Anti-oxidative potentials were investigated by using several kinds of assays including electron donating ability, and the total contents of phenolic compounds were also measured. In some cases TLC (Thin Layer Chromatography) method were used to identify the patterns of phenolics in herbal extracts. Results : Aurones which are sub-types of flavonoids were observed in methanol extract of Mori Cortex Radidus, and total phenolic contents in Mori Cortex Radidus were more than in methanol extracts of Mori Folium and Mori Ramulus. Various kinds of phenolic compounds were observed in chloroform fraction of Mori Cortex Radidus. Conclusion : According to these results, the chloroform fraction of Mori Cortex Radidus could be the most possible resource that has potentials on anti-oxidant agents on cosmoceutical fields.

Quality Characteristics and Antioxidant Potential of Seeds of Native Korean Persimmon Genotypes

  • Persimmon seeds contain considerable amounts of minerals, amino and organic acids, natural antioxidants and phenolic compounds. The objective of this study was to investigate quality characteristics and antioxidant potential of Korean persimmon seeds. The pH (4.88-4.94), color values, contents of minerals, free amino acids, organic acids, and phenolic compounds and DPPH free radical scavenging potentials of persimmon seed extracts significantly (p < 0.05) varied with the genotypes. This study showed that the seeds could be used as a source of different mineral elements (47.14-85.07 mg/kg) without any measureable amount of heavy metals such as arsenic, cadmium, lead and mercury. Similarly, considerable amounts of organic (1550.13-2413.08 mg/kg) and essential amino (50.85-54.03 mg/kg) acids and total phenolic compounds ($1227.91-1307.78{\mu}g$ gallic acid equivalent/g) were also found in the seed extracts, indicating their potential food value as a natural antioxidant. Results of the present study imply that prethanol-A, a food preservative, can be used as an effective extraction to obtain the minerals, organic and free amino acids, and phenolic compounds from the persimmon seeds, which possess a big potential to be commercially used in food, cosmetic and pharmaceutical industries.

Aging Characteristics of Glass Fabric/Phenolic Composites for Tilting Train Using Accelerated Aging Tester (가속노화시험장치를 적용한 틸팅열차용 유리섬유직물/페놀릭 복합재의 노화특성 평가)

  • Aging characteristics of glass fabric/phenolic composites for tilting train subjected to combined environmental aging factors were investigated. A 2.5KW accelerated aging tester with a xenon-arc lamp was used to provide environmental aging factors such as temperature, moisture, and ultraviolet. A series of aging tests were conducted up to 3000 hours and several types of specimens were prepared along the warp direction and the fill direction. Mechanical degradations for tensile, flexural, and shear properties were evaluated as a function of exposure times through a material testing system. Thermal analysis properties such as storage shear modulus, loss shear modulus, and tan 3 were measured through a dynamic mechanical analyzer. Finally exposed surfaces of the composites were examined using a scanning electron microscope. According to the experimental results, mechanical properties and thermal analysis properties of glass fabric/phenolic composites were found to be slightly degraded as a function of exposure times due to combined environmental effects.

  • In this study, the porosity and electrical resistivity of the formed coke produced by sintering the mixture of domestic anthracite and phenolic resin were mainly investigated, when the effect of the amount of binder, the hydration temperature and time, the hardening temperature and time, sintering temperature and time, the particle size of anthracite, the grade of anthracite, and the mixing ratio of phenolic resin on the physical properties of the coke were studied. As a result, It was found that the electrical resistivity and porosity of the formed coke are varied in the range of $0.3\sim1.2\Omega{\cdot}cm$ of $10\sim30%$, respectively, in accordance with the variation of factors.

  • This paper presents the tensile characteristics for carbon/epoxy, carbon/phenolic and silica/phenolic composites under elevated temperature, which are considered for vehicle structure or thermal protection materials. The tensile test was conducted with servo-hydraulic testing machine and high temperature furnace, and the mechanical properties such as tensile strength, elastic modulus and Poisson's ratio were evaluated by using high temperature strain gages. Also, they were compared each other with respect to fiber orientation and temperature effect. These test results were used for designing and analyzing some airframe structures with these composites.

Changes in Polyphenol Oxidase Activity, Phenol Concentration and Browning Degree of Potato Slices with Different Cultivars during Cold Storage (품종별 한국산 감자 슬라이스의 냉장 중 Polyphenol oxidase, 페놀함량 및 갈변정도의 변화)

  • Three potato cultivars, Sumi,Daejima and Namjak, were prepared as slices. They were dipped in distilled water for 20 seconds. The potato slices were packed in polyethylene bags and stored at $5^{\circ}C$. Browning degree, total phenol and chlorogenic acid contents and polyphenol oxidase (PPO) activity were measured. And the correlation analysis of browning parameters were conducted. The results were as follows. There were increase in browning degree, phenolic content and PPO activity during cold storage of potato slices with different cultivars. Among three cultivars, Sumi showed the highest browning degree, phenolic content and PPO activity and also showed the highest % increse of browning and PPO activity during cold storage. On the contrary, Daejima was the lowest. But Daejima showed the highest % increase in phenolic contents during cold storage. With Sumi, browning degree was significantly correlated with PPO activity and phenolic contents (p<0.05). With Daejima and Namjak, a significant correlation was found between browning degree and PPO activity (p<0.05). From the above results, enzymatic browning reactions of potato slices and factors affecting them were dependent on cultivar. Among the tested three cultivars, Daejima showed the lowest browning degree during cold storage and thus seems to be desirable for minimal processing.

The Total Phenolic Contents and DPPH Radical Scavenging Activities of Korean Potatoes according to Physical Characteristics and Cooking Methods (한국산 감자의 기관별, 품종별, 중량별, 분포별 및 조리 방법에 따른 총 페놀 함량과 DPPH 라디칼 소거능에 관한 연구)

  • This study was conducted to measure the total polyphenol contents and DPPH radical scavenging activities of different potato plants parts, varieties, and grades, and by distribution and different cooking conditions. The results were as follows. For the plant parts, total phenolic content and DPPH radical scavenging activity were highest in the flowers followed by leaves and stems, respectively. Among 5 potato varieties, 'Jasim' had the highest DPPH radical scavenging activity and the activity of its pulp was lower than that of its cortex(peel). Regardless of potato grade, the cortexes(peel) of samples had two-fold higher DPPH radical scavenging activity than pulp, and the Grade SS potato had the highest phenolic content. It was also found that the bud ends and stem ends had comparably larger amounts of phenolic compounds in horizontally cut potatoes. Finally, the descending order for DPPH radical scavenging activity, according to different cooking conditions, was as follows: gas oven range-baking, frying, microwave-heating, sauteing, boiling in 1% saline solution, steaming, and boiling in 3% saline solution.

Degradation of Phenolic Resin, Resole by Microbial Consortia (미생물 컨소시엄에 의한 페놀수지 Resole의 분해)

  • Three microbial consortia were screened for their ability to degrade phenolic resin, resole as a sole carbon source. These microbial consortia were derived from soil samples collected from a phenolic resin manufacturing plant site. Among the consortia, the test consortium, designated as MS2, displayed approximately 70% degradation of the substrate, 100 mg of resole per liter, within the fist twelve days of incubation but the degradation was inhibited. During the incubation period, pH was decreased from 7.0 to 2.7, and the resole degradation became inhibited under the conditions. UV-scans of spent culture showed that the wavelength of maximum absorption was 261 nm for resole.

  • Cell growth and production of phenolic compounds by callus cultures of Panax ginseng C. A. Meyer were investigated under various phytohormones concentrations and inoculum size. The results indicated that the cell growt was improved by a MS medium supplemented with 2 mg/L of CPA. The maximum cell yield was obtained at inoculum size of 1 g/flasd. The production of phenolic compounds in the callus cultures was higher than those in the ginseng root. Especially, one cell line (20601) showed the highest content of phenolic compounds and antioxidant activity.
