천연(天然)쿠마린의 생리활성(生理活性) (Biological Activity of Natural Coumarins)

  • The naturally occurring coumarins are likely to be a large and important group of plant products which are not only metabolic products of the living cells but possess varied physiological and pharmacological actions. Investigations concerning various biological activities of natural coumarins have been undertaken by many investigators in continuing effort. The authors describe on experimental results of biological activities of coumarin derivatives with particular emphasis on 10 different activities that are considered to be of importance in view of the development of new drugs from natural resources.

  • Pomegranate, a small tree originating in Orient, belongs to Punicaceae family. The seeds contain an oil of which about 80% is rare trans 18 carbon fatty acid (punicic acid), and have highest botanical concentration of a sex steroid, estrone. Pharmacological properties of pomegranate extract have been studied, with anti-microbial, anti-parasitic, anti-viral. and anti-cancer effects. (omitted)

  • A mixture of several bacterial metabolites $(Sterodin{\circledR})$ was used to study its effect on major immunocytes, in vivo and in vitro. This mixture of bacterial metabolites increased number of macrophages and neutrophils and their phagocytic activity in experimental animals for a transient period. BSA induced antibody production was found to be higher in the drug treated group. These results indicated that the bacterial metabolites probably acted through non-specific defence mechanism against invading organisms and the chance of reinfection was reduced.

  • Since ampicillin, one of the important semi-synthetic derivatives of penicillins, is restricted in use because of its unfavorable physico-chemical and pharmacological properties, the author synthesized a new derivative, methampicillin lysinate and its physical and characteristics were determined. The antibacterial activity of this compound against ten species of Gram positive and negative bacteria in vitro showed that it was more active to Gram negative bactria than the positive and that was three times more potent than ampicillin trihydrate.

  • Capsaicin [N-(4-hydroxy-3-methoxybenzyl)-trans-8-methyl -6-nonenamide], a pungent principle of red pepper. is known to induce analgesia. Though it has shown remarkable pharmacological activity, the toxicity and side effects inhibited its wider application. The interest has been renewed by the emergence of its agonists, resineferatoxin and ruthenium red. Recently, few successful capsaicinoids including 01vanil (NE-19550), NE-28345, and NE-21610 were reported.

  • Antitumor and antimicrobial activities of water soluble fractions of Duchesnea indica (Andr.) and Focke (Rosaceae), have been studied. The contents of total polysaccharide were 22% and its composition was identified by G.C.: L-arabinose(Rt: 7.71), D-xylose (Rt. 9.91), D-glucuronic acid (Rt. 12.77), D-fructose (Rt. 13.63), D-galactose (Rt. 14.68), D-glucose (Rt. 15.42). The antimicrobial activity of Duchesnea indica against four bacterial species, S. aureus, S. paratyphi, S. dysenteriae and Ps. aeruginosa was observed.

  • The influence of Panax ginseng on alcohol-induced hyperuricemia were observed. Changes of uric acid blood levels and hepatic xanthine oxidase activities were studied by means of treating alcohol intoxication with ginseng. It was found that a single dose (4 mg/Kg) of ginseng saponin administered intraperitoneally significantly inhibits the hepatic xanthine oxidase activities and decrease urate blood levels in ethanol-induced hyperuricemic mice. It was also observed that there were some difference in pharmacological aspect between Panax ginseng and allopurinol which is a potent inhibitor of xanthine oxidase from any sources.

  • There have been various inflammatory skin disorders in humans including atopic dermatitis, eczema and psoriasis. Although some drugs have been used for these disorders, there is an urgent need for safer and more effective topical anti-inflammatory agents. Plant flavonoids possess anti-inflammatory activity and some of them have multiple pharmacological mechanisms, inhibition of eicosanoid metabolizing enzymes, histamine release and/or down-regulation of pro inflammatory gene expression. These properties of flavonoids may be suitable for treating chronic skin inflammatory disorders. Especially, wogonin, some prenylated flavonoids and biflavonoids have a strong potential as new anti-inflammatory agents by topical application.

  • Excipients of the drug products can sometimes affect the rate and extent of drug absorption. The changes in components or composition of them can also affect the pharmacological activity.
