Background: There are at least two different subsets of B cells, B-1 and B-2. The characteristic features and function of B-2 cells in addition to the effect of steroids on B-2 cells are well-known. Although B-1 cells have different features and functions from B-2 cells, the effect of steroids on B-1 cells is not completely understood. Therefore, this study examined the effects of dexamethasone on peritoneal (or B-1 cells) and splenic B cells (or B-2 cells). Methods: Purified B cells were obtained from the peritoneal fluid and the spleens of mice. The isolated B cells were cultured in a medium and after adding different concentrations of dexamaethasone. The cell survival rate was measured by flow cytometry using propidium iodide. The expression level of the B cell surface marker was analyzed by flow cytometry. During the culture of these cells, immunoglobulin secreted into the culture supernatants was evaluated by an enzyme-linked immunosorbent assay. Results: The survival rate of peritoneal and splenic B cells decreased with increasing dexamethasone concentration. However, the rate of peritofieal B cell apoptosis was lower than that of splenic B cells. CDS and B7.1 expression in peritoneal B cells and CD23 and sIgM expression in splenic B cells after the dexamethasone treatment were reduced. When B cells were treated with dexamethasone, the spontaneous IgM secretion decreased with increasing dexamethasone concentration. Conclusion: Dexamethasone induces apoptosis in peritoneal and splenic B cells. However, peritoneal B cells are less sensitive to dexamethasone. The dexamethasone suppressed expression of the surface markers in peritoneal B cells is different from those in splenic B cells.
The purpose of this Study was to investigate effects of KaegiBokryengHwan(KBH) on anti-tumor, immunocytes and nitric oxide(NO). This Study estimated the proliferation of L1210 cell lines, HeLa cell lines, SK-OV3 cell lines, MCF-7 cell lines, balb/c mouse 3T3 cell lines, mouse thymocytes and mouse splenocytes and NO production from peritoneal macrophages in vitro. and estimated the proliferation of L1210 cells, mouse thymocytes and splenocytes and NO production from peritoneal macrophages and body weight in L1210 cells-transplanted mice in vivo. The result were obtained as follow ; 1. KBH inhibited significantly SK-OV3 cell lines in vitro. 2. KBH was accelerate significantly the proliferation of balb/c mouse thymocytes in vitro. 3. KBH increased significantly NO production from peritoneal macrophages in vitro. 4. KBH didn't effect the cytotoxicity of L1210 cells in L1210 cells-transplanted mice. 5. KBH was accelerate the proliferation of splenocytes in L1210 cells-transplanted mice. 6. KBH increased NO production from peritoneal macrophages in L1210 cells-transplanted mice. 7. KBH increased the body weight as comparing with control group in L1210 cells-transplanted mice.

TSS accelerated the proliferation of mice thymocytes and splenocytes In L1210 cells-transplanted mice. 6. TSS was increased significantly the nitric oxide production from mice peritoneal macrophages in L1210 cells-transplanted mice. 7. TSS was increased the body weight as comparing with control group in Ll210 cells-transplanted mice.
Background: B-1 cells differ from conventional B-2 cells both phenotypically and functionally. The aim of this study was to investigate the difference between peritoneal B-1 cells and splenic B-2 cells in proliferation. Methods: We obtained sorted B-1 cells from peritoneal fluid and B-2 cells from spleens of mice. During the culture of these cells, immunoglobulin secreted into the culture supernatants was evaluated by enzymelinked immunosorbent assay. Entering of S phase in response to LPS-stimuli was measured by proliferative assay. Results: Spontaneous Immunoglobulin M production occurred in peritoneal B-1 cells but not in splenic B-2 cells. LPS stimulated peritoneal B-1 cells secreted IgM at day 1, but splenic B-2 cells at day 2. In thymidine incorporation, peritoneal B-1 cells entered actively S phase after 24hours LPS-stimulation but splenic B-2 cells entered actively S phase after 48 hours. Conclusion: IgM secretion and S phase entering occurred early in peritoneal B-1 cells compared to splenic B-2 cells.
The objective of this study was to investigate the possible effects of oral administration of single cell products (SCP) of apple on activating peritoneal macrophages. Apples were processed either for cold-pressed juice or SCP, which were produced by incubating sliced apples with a protopectinase, Sumyzyme MC. Both cold-pressed juice and SCP of apple were administered to C57BL/6 mice for 10 days to compare their efficacy, along with the control group, in stimulating peritoneal macrophages. The viability of macrophages was significantly increased by up to 161% of that of the control following the administration of apple SCP, whereas the viability of macrophages was increased to a lesser extent of up to 143% in the apple juice (AJ) administered group. Administration of apple SCP also induced a significantly higher production of $H_2O_2$ from macrophages (317% of the control) than that of cold-pressed AJ (210%). Although nitric oxide (NO) production was not increased by the administration of either AJ or SCP, the latter slightly but significantly increased tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) production from macrophages from 560.4 to 579.8 pg/mL. The results of this study suggest that administering SCP is more efficient than administering AJ to stimulate functions of peritoneal macrophages.
A murine leukemia x LM fibroblast hybrid cell line with immune augmenting properties stimulated resistance to Mycobacterium avium complex (MAC) in mouse peritoneal macrophages, and in immune deficient beige mice (C57BL/6/bgj/bgj). The proliferation of MAC in mouse peritoneal macrophages was inhibited by medium conditioned by the growth of the hybrid cells (hybrid cell-CM). Under similar circumstances, media conditioned by the growth of LM cells (LM cell-CM), a mouse fibroblast cell line used as one parent in forming the hybrid cell, was exhibited no inhibitory effect. Treatment of mouse peritoneal macrophages with hybrid cell-CM, but not with LM cell-CM, stimulated the expression of each of four previously described macrophage activation antigens, suggesting that the hybrid cells formed immunomodulators in addition to those formed by LM cells. Furthermore, the morphology of the macrophages following treatment with hybrid cell-CM was clearly distinguishable from that following exposure of the cells to LM cell-CM. The therapeutic effects of hybrid cells on the progression of MAC-infection were indicated by the prolonged survival of MAC-infected immune-deficient beige mice. One hundred percent of treated animals survived more than 60 days, while untreated animals died in approximately 22 days.

The peritoneal sheath, epithelium of the follicle and cucticle have the maximum thickness at prepupa stage and afterthen gradually they began to degenerate. The epithelium of the follicle which formed by invagination of the peritoneal sheath is differently differentiationed from the peritoneal sheath and it is similar to the adult's from the pupa 2 days. The cell organelles begin to increase in the cytoplasm of the cyst cell enclosed the sperm bundles at pupa 3 hrs. The electron densed granules which observed in the peritoneal sheath and epithelium of the follicle at prepupa seem to be related with fusion of the testis.
The unripe fruit of Poncirus trifoliata Raf has been used for the treatment of allergic disease. Recently it was reported that the fruit inhibits passive cutaneous anaphylaxis and histamine release at mast cell. Type I immediate hypersensitivity of anaphylactic type is caused by released mediate chemical at mast cell. Histamine is also known as one of potent mediate chemical. Also release of mediate chemical is affected by specific stimulation of IgE combined with mast cell. Activation of mast cell is known to be stimulated by compound 48/80 and inhibited by increase of cAMP. In this experiment, the effect of water extract of Poncirus trifoliata Raf fruit (PT) on a histamine release, cAMP concentration and IgE production was measured. Compound 48/80 was administrated to the mouse peritoneal cell which was pretreated with PT. PT dosedependently inhibited histamine release at peritoneal mast cell and the serum level of histamine induced by compound 48/80. PT also instantly increased cAMP level of peritoneal mast cell right after it was added and the level gradually decreased. Production of IgE induced by antigens at mouse peritoneal cell was inhibited by PT. The IgE synthesis is induced by IL-4 and it is known that lipopolysaccharide(LPS) plus IL-4 cause an increase in IgE secretion by murine B cells. The effects of PT inhibited the production of IgE activated by LPS plus IL-4 at human U266B1 cells. These results indicate that PT has antiallergic activity by Inhibition of IgE production from B cells and histamine release by increase of cAMP.

Vitro 배양(培養)의 배양액(培養液)으로서 이용(利用)할수 있는지를 검토(檢討)하기 위하여 실시(實施)하였다. 그 결과(結果)를 요약(要約)하면 다음과 같다. 2) 2세포(細胞), 4세포(細胞), 8세포(細胞), 16세포기(細胞期)의 가토난자(家兎卵子)를 복수(腹水)를 써서 $37^{\circ}C$에서 48시간(時間) 배양(培養)한 결과 분할(分割)된 난자(卵子)의 정도(程度)는 각각(各各) 78.1%, 83.3%, 91.7% 및 93.6%로서 이들의 성적(成績)은 가토혈청(家兎血淸)으로 배양(培養)했을 경우보다도 어느 것이나 좋았다. 2) 2세포(細胞)의 난자(卵子)를 복수(腹水)로 96시간(時間) 배양(培養)했을 때 배양난자(培養卵子)의 88%가 상실기이상(桑實期以上)으로 발달(發達)되고 24%가 배반포(胚盤胞)에 이르고 있으며 가토혈청(家兎血淸)으로 배양(培養)한 성적(成績)과 거의 같은 경향(傾向)을 보였다. 이상(以上)과 같은 점(點)에서 가토난자(家兎卵子)의 배양액(培養液)으로서 가토(家兎)의 복수(腹水)가 유효(有效)하라는 것이 입증(立證) 되었다.
Several studies have shown that extracts from yellow-green vegetables reveal antitumor activities. In the present study we investigated the effect of phytol in order to elucidate the immunological mechanism of antitumor activity of this substance. The results obtained from the experiment as follows: 1) Phytol showed cytotoxic effect on sarcoma 180 cells in vitro. 2) When phytol was injected into the peritoneal cavity of mice transplanted with sarcoma 180 cells, the average survival time (24.0 days) tended to increase as compared with the nontreated control (19.2 days). 3) When sarcoma 180 cells were injected subcutaneously into the right groin of mice, and then phytol was injected into the peritoneal cavity, the tumor inhibition ratio was 33%. 4) The natural killer(NK) cell activity was significantly augmented by phytol in vitro and in vivo. Similar augmentations of NK cell activity were obtained with culture supernatants of phytol exposed spleen cells and peripheral blood mononuiclear cells. 5) Phytol on the macrophage from peritoneal cavity showed a higher effectiveness in vivo than in vitro. These results indicate that phytol shows the inhibitory effect for growth of sarcoma 180 cells in vitro, also it can augment macrophage and NK cell activities in vivo.
