Properties of Active Sites of D-Xylanase and $\beta$-Xylosidase from Penicillium verruculosum (Penicillium verruculosum의 D-Xylanase와 $\beta$-Xylosidase의 활성부위 특성)

  • To investigate the characteristics of active sites of the D-xylanase and $\beta$-xylosidase purified from Penicillium verruculosum, effects of various chemicals on the enzyme activity were analyzed. The D-xylanase was activated by Cua), however it was inhibited by metal ions, Hg2+ and Mna+, by chemicals, N-bromosuccinimide, iodine, diethylpyrocarbonate, and 2,3-butanedione. These results suggested that the D-xylanase from Penicillium verruculosum contained tyrosine, histidine, arginine and tryptophan at the active center. The $\beta$-xylosidase was inhibited by Hg2+, N-bromosuccinimide and sodium dodecyl sulfate, however it was not effected by Mn2+ and Cu2). It was suggested that the enzyme contained tryptophan at the active center.

New Records of Four Species Belonging to Eurotiales from Soil and Freshwater in Korea

  • Four strains of Penicillium and Talaromyces species are described and illustrated in an inventory of fungal species belonging to Eurotiales. The strains, CNUFC-DDS17-1, CNUFC-DDS27-1, CNUFC-PTM72-1, and CNUFC-YJW3-31, were isolated from soil and freshwater samples from South Korea. Based on their morphological characteristics and sequence analyses by the combined b-tubulin and calmodulin gene, the CNUFC-DDS17-1, CNUFC-DDS27-1, CNUFC-PTM72-1, and CNUFC-YJW3-31 isolates were identified as Penicillium pasqualense, Penicillium sanguifluum, Talaromyces apiculatus, and Talaromyces liani, respectively. The designated strains were found to represent a previously undescribed species of Korean fungal biota. In this study, detailed morphological descriptions and phylogenetic relationships of these species are provided.

Diversity of Aspergillus, Penicillium, and Talaromyces Species Isolated from Freshwater Environments in Korea

  • In order to elucidate the fungal diversity and community structure in freshwater environments, numerous fungal strains were isolated from freshwater, submerged soils, twigs, dead insects, etc. Among them, the present study has focused specifically on Aspergillus, Penicillium, and Talaromyces species, which produce diverse useful metabolites in general. Twelve strains of Aspergillus isolated were identified as A. japonicus (n = 5), A. tubingensis (3), A. niger (2), and A. flavus (2), 10 strains of which belong to Aspergillus section Nigri, named black Aspergillus. Eight strains of Penicillium were identified as P. brasilianim (n = 3), P. oxalicum (2), P. crustosum (1), P. expansum (1), and P. piscarium (1). Two different strains of Talaromyces were identified as T. pinophilus and T. versatilis. Thus far, Penicillium piscarium and Talaromyces versatilis have been unrecorded in Korea, for which we provide detailed morphological and molecular characteristics.

Biosynthesis of Skyrin in Penicillium islandicum (Penicillium islandicum에 의한 Skyrin의 생합성)

  • Skyrin was extracted from Penicillium islandicum cultivated on synthetic Czapek-Dox medium. A preliminary experiment in Penicillium islandium using $[^{14}C]$-acetate proved the incorporation of $[^{14}C]$ into skyrin. In the cell-free system using the fungus $[^3H]$-emodin and $[^3H]$-emodinanthrone were shown to give some reliable evidences far the biosynthesis of skyrin. The incorporation of $[^3H]$-emodin was less significant than that of $[^{3}H]$-emodinanthrone. As a result of the cell-free experiment, the biosynthetic pathway of skyrin by the fungus was suggested as follows: $emodinanthrone{\rightarrow}emodin{\rightarrow}skyrin$.

Six Species of Penicillium Associated with Blue Mold of Grape

  • Grape fruits with blue mold symptoms were collected from house storages in different locations in Korea and were investigated for their association with Penicillium species. A total of 12 isolates of Penicillium were isolated from the collected fruits. Based on morphological and cultural characteristics and ${\beta}-tublin$ gene sequence data analysis, they were identified as P. bialowiezense, P. citrinum, P. echinulatum, P. expansum, P. solitum and unidentified Penicillium species. P. solitum was the predominant followed by P. expansum. P. bialowiezense and P. echinulatum were newly recorded in Korea. ${\beta}-Tubulin$ gene sequences could be used to distinguish each species of Penicillium and the molecular groups were correlated well with the morphological species. The unidentified species was supposed to be a new species, not previously reported in literature.

Detection of Aspergillus and Penicillium genera by Enzyme-Linked Immunosorbent Assay Using a Monoclonal Antibody

  • Enzyme linked-immunosorbent assay (ELISA) for a rapid detection of fungi, Aspergillus and Penicillium genera in food, were developed and their efficiencies were approved by detecting artificially contaminated agricultural commodities. Mice were immunized with partially purified Aspergillus flavus extracellualr polysaccharide (EPS) and lymph node cells of the mice were fused with the myeloma cells for production of monoclonal antibodies. Mab 1G11, one of the antibodies, was selected and purified. A sandwich ELISA was established and its detection limit toward A. flavus EPS was 1mg/ml. Among the 59 strains tested (including 18 species of Aspergillus, 16 of Penicillium, 11 of Fusarium, 1 of Absidia, 2 of Alternaria, 2 of Candida, 2 of Cladosporium, 2 of Geotrichum, 2 of Mucor, 2 of Rhizopus, 1 of Trichoderma), species of Aspergillus and penicillium had a high reactivity with Mab 1G11 even up to 10,000 times dilution of culture broths. The other genera except Cladosporium resinae showed no reactivity, thus Mab 1G11 was specific to the genera of Aspergillus and Penicillium. The epitope of A. flavus EPS against monoclonal Mab 1G11 was on the carbohydrate moiety when 1 to 100$\mu g/g$ A. flavus EPS were put into rice, potato, and mandarin orange, the average recoveries detected by sandwich ELIA were 123, 59, and 76%, respectively. Correlation was found to be linear between the EPS, and mycelium of A. flavus and Penicillium citrinum grown in a liquid medium (r=0.87 and 0.96), and also between the EPS and colony forming unit in solid media of rice of potato (r=0.91-0.99).

Isolation and Identification of Filamentous Fungi from Indoor Air of a Sogokju Traditional Rice Wine Factory (소곡주 공장의 공기로부터 곰팡이의 분리 및 동정)

  • To investigate the mycoflora of indoor air in a Sogokju, traditional rice wine, factory, fungi were sampled and analyzed from the air of several rooms in the factory using an Anderson air sampler and from two kinds of Nuruk. Twelve fungal species belonging to the genera of Aspergillus, Penicillium, Gibberella, Cladosporium, and Talaromyces were isolated. Species belonging to Aspergillus and Penicillium genera were the major species. Seven different species of Penicillium were isolated from each different room of the factory. The Aspergillus species found from indoor air of the factory was also found from Nuruk. Rhizopus sp. was commonly isolated from Nuruk but not from indoor air of the factory. This is first report of fungi present in indoor air of a traditional rice wine factory in Korea.

  • This study was conducted to investigate the major mycotoxins occurred during storage from apple, peat, citrus and grape. Analyses of the mycotoxins were conducted by TLC and HPLC. Patulin was only detected from apple, pear, citrus and grape infected by penicillium mycotoxins, but citrinin did not detected. The detected amount except grape ranged from $5.68{\sim}46.81\;{\mu}g/g$ in apple, $3.48{\sim}84.7\;{\mu}g/g$ in pear, $0.16{\sim}0.27\;{\mu}g/g$ in citrus, respectively. When compared to the effect of control on penicillium mycotoxins during storage in apple, pear and citrus, sodium hypochloride gas and heat at $37^{\circ}C$ treatment exhibited effective control. Especially, heat at $37^{\circ}C$ treatment exhibited 100% effective control without any injuring by treatment. In pear and citrus sodium hypochloride gas treatment caused injury by treatment.

  • The effect of the Penicillium sp. PS-113 with high phosphate solubilizing activity and Lactobacillus sp. was studied on cucumber growing test in green house. Inoculation of Pemicillium sp. Ps113 $5.0{\times}10^6$cfu/ml and Lactobacillus sp. $6{\times}10^6$cfu/ml were longer than control about 3cm length of cucumber. The average weight was also increased 89.2g more than control. Total product of cucumber was 10,681.7g and it was increased 4517.3g as compared to control. Thus, Inoculations of high phosphate solubilizing microorganism increased in productivity on cucumber.

Protective Effect of Iminoctadine tris(albesilate) and Kresoxim-methyl Fungicides to Citrus Postharvest Diseases caused by Penicillium spp. (저장 감귤의 부패에 관여하는 Penicillium spp.에 대한 Iminoctadine tris(albesilate)와 Kresoxym-methyl의 방제 효과)

  • The biological effects of the iminoctadine tris (albesilate) and kresoxim-methyl for the protection of citrus postharvest diseases caused by penicillium spp. were assayed. In vitro tests, $EC_{50}$ values of iminoctadine tris(albesilate) were $0.01{\sim}0.02\;and\;0.01{\mu}g$ a.i./mL against mycelial growth of P. italicum and P. digitatum, respectively, but iminoctadine tris(albesilate) at $0.64{\mu}g$ a.i. /mL inhibited a little mycelial growth of unknown Penicillium sp. which produced another symptom different to blue and green mold caused by P. italicum and P. digitatum, respectively. And against germination and growth of germ tube of P. italicum and P. digitatum, $EC_{50}$ value of iminoctadine tris(albesilate) was $0.0013{\sim}0.0025{\mu}g$ a.i./mL. But spore germination of unknown Penicillium spp. was not nearly inhibited at $0.2{\mu}g$ a.i./mL. $EC_{50}$ values of kresoxim-methyl were $0.08{\sim}0.16$, 0.04 and $0.16{\mu}g$ a.i./mL against mycelial growth of P. italicum, P. digitatum and unknown Penicillium sp., respectively, and $0.04{\sim}0.08{\mu}g$ a.i./mL and $0.01{\sim}0.02{\mu}g$ a.i./mL against germination and growth of germ tube of P. italicum and unknown Penicillium sp., and P. digitatum, respectively. Iminoctadine tris(albesilate) and kresoxim-methyl were markedly effective to control the postharvest disease by 7 days spray prior to harvest. When the fruits were sprayed with iminoctadine-tris(albesilate) ($200{\mu}g$ a.i./mL) and kresoxim-methyl ($155{\mu}g$ a.i./mL) 7 days prior to harvest and subsequently stored for 90 days, the percentage of diseased fruit by Penicillium spp. was $3.6{\pm}1.8%$ in treatment of kresoxim-methyl and $5.9{\pm}1.8%$ in iminoctadine-tris(albesilate), respectively. On the other hand, tile percentage of diseased fruit was relatively high, $20.3{\pm}10.0%\;and\;19.5{\pm}9.6%$ in thiophanate-methyl ($700{\mu}g$ a.i./mL) and non-treatment, respectively. Maximum residue amount (ppm) among fruits (flesh and peel) assayed 0, 30, 60 and 90 days after storage was 0.45 and 0.10 ppm in treatment of kresoxim-methyl and iminoctadine, respectively.
