Replacement value of cassava for maize in broiler chicken diets supplemented with enzymes

  • Objective: Pellet durability, particle size distribution, growth response, tibia bone characteristics and energy retention were measured to evaluate cassava as an alternative energy source to replace maize in broiler diets with or without Ronozyme (A+VP) enzyme composites. Methods: A total of 480 one-day broiler chicks were randomly assigned to 8 treatments in a 4×2 factorial arrangement. Four levels of cassava: (0%, 25%, 50%, 75%) and 2 levels of enzymes (0 and 500 g/tonne) were used. Each treatment was replicated six times, with ten birds per replicate. Results: The particle size distribution in the diets showed an increasing trend of small particles with increase in cassava level. Pellet durability decreased (p<0.05) with cassava inclusion. Feed intake was highest in birds fed diets with medium cassava level at 1 to 24 d and 1 to 35 d of age. The body weight gain of birds reduced (p<0.037) as cassava level increased, but it increased (p<0.017 when enzymes were added. The feed conversion ratio was high (p<0.05) when cassava level was increased, but it reduced (p<0.05) when enzymes were added. The dressing percentage (DP), and weight of drumsticks reduced (p<0.05) with increasing cassava level. Enzyme supplementation increased (p<0.05) DP, and weight of breast, thighs and drumsticks. Ash content, weight, length, width, and bone strength decreased (p<0.05) when cassava level was increased, however, they were increased with enzyme addition. The contents of Ca, K, and Zn were raised (p<0.001) with increasing cassava level. Enzyme inclusion increased (p<0.001) all mineral contents in tibia bones. Body fat and energy retained as fat decreased (p<0.001) as cassava level increased. Enzyme inclusion increased (p<0.05) body protein content and energy retained as protein. Conclusion: Although broiler performance was depressed by high levels of cassava inclusion, it was not affected by low levels, which further improved by enzyme supplementation.

Antibacterial Effect of Bacteria Isolated from the Korean Traditional Foods against Pathogenic Bacteria (한국전통식품으로부터 분리 된 세균의 항균활성 효과)

  • Aquaculture continues to be an ever-growing sector. However, high-density farming increases disease outbreaks due to deteriorating water quality and internal stress. To prevent disease, the most common method chemotherapy is using antibiotic administration. In this study, probiotic bacteria were isolated from Korean traditional foods, such a Gochu pickle and cutlassfish salted seafood. Various bacteria were isolated, and their 16S rDNA sequences were analyzed. The antimicrobial activities of four isolates from Gochu pickle and seven isolates from cutlassfish salted seafood were assayed, in addition to the antibacterial activity of culture pellet and supernatant. The antibacterial activity of the pellet was higher than that of the supernatant. Isolate JKM-2 showed the highest antibacterial activity against Streptococcus iniae (43 mm), S. parauberis (40 mm), S. mutans (35 mm), and Vibrio vuinificus (26.5 mm). The sequences of the isolated strains were compared with those of Bacillus subtilis (97.71%), B. tequilensis (97.71%), Brevibacterium halotolerans (97.71%), B. subtilis (97.63%), B. subtilis (97.63%), B. mojavensis (97.54%), B. vallismortis (97.46%), B. nanillea (97.45%), B. methylotrophicus (97.37%), and B. ssiamensis (97.37%). Future through analysis and new strains confirmed the bacterial cell material investigation of JKM-3, and to ensure sufficient stability, it is desired to verify the utility value as a substitute material for antibiotics by application to the form of the industry.

Effects of the Different Fish Meal and Wheat Flour Content in Extruded Pellet on the Growth and Apparent Digestibility for Olive Flounder Paralichthys olivaceus (어분 종류 및 소맥분 함량이 다른 배합사료가 넙치 (Paralichthys olivaceus)의 성장과 소화율에 미치는 영향)

  • A feeding trial was carried out to investigate the effect of different fish meal and wheat flour contents in extruded pellets on the growth and apparent nutrient digestibility of olive flounder Paralichthys olivaceus. Four extruded pellets (HB1, HB1T, HB2 and HB2T) were prepared using two kinds of high quality brown fish meal and low quality tuna fish meal. Two diets (HB1 and HB2) contained two different kinds of brown fish meal, respectively; in HB1T and HB2T, we substituted 50 % of the brown fish meal with 10 % wheat flour. Three replicate groups of fish (initial body weight of 82 g) were fed the experimental pellets to visual satiation for 15 weeks. The highest weight gain was observed in fish fed HB1, and weight gain of fish fed HB2 was significantly higher than that of fish fed HB2T (P<0.05). Feeding efficiency for fish fed HB1 was significantly higher than that of fish fed HB1T and HB2T (P<0.05), but was not significantly different from fish fed HB2. Apparent dry matter digestibility in HB1 and HB2 was significantly higher than in HB1T and HB2T (P<0.05). The highest crude protein digestibility was observed in HB1, and protein digestibility in HB1T and HB2 was higher than in HB2T (P<0.05). The results of this study suggest that fish meal quality considerably affects growth and nutrient digestibility of olive flounder fed extruded pellets, and the use of HB1 is recommended over HB1T, HB2 and HB2T for efficient olive flounder production.

  • Although flounder is one of the most important marine fish for aquaculture in Korea, feeding the flounder in commercial farms depends mainly on moist pellet in which over 70% frozen fishes (e.g. frozen horse mackerel) are incorporated in its formulation. Therefore, for further expansion of flounder farming, it is essential to employ practical formulated feeds that can support reasonable growth. Development of nutritionally balanced and cost-effective feeds is dependant on the information about nutritional requirement and feed utilization of the species. Nutrient and energy source in feed are needed for the growth and maintenance of fish. Protein is probably the most important nutrient affecting fish growth and feed cost. Therefore, it is essential to determine the optimum dietary protein level for the growth of fish, both its high proportion in the feed and because it is the main factor in determining feed cost. Dietary energy level is also critical because protein source in the feed is utilized as an energy source when the feed deficient in energy is fed to fish, whereas when the feed excess in energy is fed to fish, feed consumption decreased and resulted in growth reduction due to lack of other necessary nutrients for normal growth. Improper dietary protein, energy levels and/or their ratio will lead to an increase of fish production cost and deterioration of water quality resulting from wasted feed; thus, they are important in formulating commercial feed. Dietary lipids play important roles in providing energy and essential fatty acid for normal growth and survival of fish. Although carbohydrates are not essential nutrients for carnivorous fish, these compounds play important roles as a low-cost energy source for protein sparing and also as a feed binder. Nutrition researches for flounder have identified its requirements of protein, lipid and essential fatty acid, vitamin, and minerals for normal growth. Other studies have also been carried out to investigate the utilization of the protein, lipid and carbohydrate sources. Based on these nutritional information obtained, practical feed formulations have been studied for improve aquaculture production of flounder. The results of the researches on utilization of dietary protein, lipid and carbohydrate by flounder are discussed in this review.

Stimulation of Ovarian Development in a Tropical Damselfish by Prolonged Photoperiod using Pellets Containing Long-afterglow Phosphorescent Pigment

  • The present study examined whether light emitted by long-afterglow phosphorescent pigments (LumiNova) would stimulate gonadal development in fish during the nonbreeding season. Pellets containing LumiNova powder (treatment group) were prepared and placed on the calvaria of specimens of the sapphire devil Chrysiptera cyanea, a reef-associated damselfish that requires long days for gonadal recrudescence. A pellet without LumiNova powder was placed on the calvaria of the control fish (control group). Fish were reared at $26^{\circ}C$ under a light-dark cycle (12 h photophase, 12 h scotophase; LD 12:12) for 4 weeks. No difference in the gonadosomatic index (GSI) or ovarian histology was observed among the control, sham-operation, and treatment groups 1 week after the start of the experiment. After 4 weeks, the GSI of the control and sham-operation groups remained at low levels, and ovaries contained immature oocytes at the perinucleolus stage. In contrast, the treatment group exhibited significantly higher values of GSI as well as developed ovaries with fully vitellogenic oocytes. These results demonstrated that long-day conditions were produced by light emitted from the LumiNova pellets, thus stimulating ovarian development in the damselfish. Therefore, long-afterglow phosphorescent pigments can be used as an alternative to standard light sources for purposes of artificial stimulation of gonadal development in fish.

Effect of light, ultrasonication and liquid smoke on germination of proso millet (Panicum miliaceum L.) seeds

  • High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.

Evaluation and modification of alkaline lysis plasmid preparation method from Lactobacillus spp.

  • Lactic acid bacteria (LAB) has been regarded as a useful microorganism and tried to manipulate plasmid DNA for increasing the usefulness. Although several methods have been developed to isolate plasmid DNA from Escherichia coli (E. coli), these methods were not sufficient to apply to LAB with exception of O'Sullivan's lysis method. So, we evaluated plasmid DNA extraction from LAB using general E. coli preparation methods and tried to improve the extraction yield and DNA purity by modifying O'Sullivan's alkaline lysis method. To improve the extraction yield, salt and carrier were added to precipitant and those were incubated at $-70{^{\circ}C}$. Only incubation at $-70{^{\circ}C}$ was the effective method of those modifications. Purity of plasmid DNA was improved by two times of each centrifugation and phenol/chloroform extraction. However, DNA was damaged by twice extraction with phenol/chloroform. Also, exclusion of ethidium bromide showed negative effect to purity. Additionally, it was recommended that improvement of the extraction yield may be due to centrifugation at high speed for more time and to dissolving complete DNA pellet before addition of 7.5 M ammonium acetate. Extraction using this modification produced higher quality of plasmid DNA.

Quality Characteristics of Puffed Snacks (ppeongtuigi) with Purple Sweet Potato Flours Using Different Puffing Conditions (자색 고구마 분말을 첨가한 팽화과자(뻥튀기)의 팽화 조건에 따른 품질 특성)

  • The effect of puffing conditions on the physical properties and sensory evaluations of a puffed snack (ppeongtuigi) made of purple sweet potato flour was investigated. The snacks consisted of coated artificial rice with purple sweet potato flour with water, tempered to 7%, 9% and 11% moisture. The coated material was puffed at 236, 241, or $246^{\circ}C$ for 4, 5, or 6 s. The $L^*$ value decreased as the heating time increased. The $a^*$ and $b^*$ values, specific volume, and hardness increased with increasing heating temperature and heating time. Puffed snacks produced under conditions of higher moisture (7%), higher heating temperature ($246^{\circ}C$), or longer heating time (5 s) showed high scores for appearance, color, taste, and overall acceptability in sensory evaluations.

Management of Earthworm Casts using Pesticides and Tea Tree (Camellia oliefera) Extracts in Turfgrass of Golf Courses (골프장에서 차나무류 추출물과 화학 방제제를 이용한 지렁이 분변토 발생 관리)

  • Earthworm cast is a harmful factor which reduces the uniformity of management of turfgrass and play quality in golf courses and playground. This research was conducted to carry out the efficacy of some pesticide (bifenthrin GR + clothianidin GR, ethoprophos GR, imidacloprid SC, picoxystrobin WG, thiophanate-methyl WP) and tee tree (Camellia oliefera) extracts against reducing earthworm cast in golf courses. All tested materials were acted reducing earthworm cast, thiophanate-methyl among them showed the highest effect reducing earthworm casts in golf courses. Tea tree extracts was occurred similar efficacy among tested chemical pesticides. However, pellet type byproduct of tea seed extract and liquid formulation of tea tree extract were better than granule formulation of them. Byproduct of tea seed and tea tree extract can be used for alternative biological management material for reducing earthworm cast in heavily casting occurrence area.

  • Wood biomass including forest residues, waste wood, and construction residuals has been widely generated in Korea, but forest biomass from the National Forest Management Operation Project plays a big role in generating wood biomass. Unfortunately the promotion policy of woody energy organized by the Forest Service in Korea concentrates more on demand creation rather than on supply expansion. Therefore, in order to utilize insufficient wood resources effectively, it is greatly required to develop uses for maximizing their added value. In particular, more attention to the use of the second generation biomass has been paid in foreign countries because there is a threshold that the first generation biomass cannot produce enough biofuel without threatening food supplies and biodiversity. In Korea, wood pellets are regarded as the alternative clean fuels to oils and coals that emit green house gases into the atmosphere. However, using wood as pellet raw materials can not be an economic way because the value of wood disappears right after burning in the boiler in spite of its contribution to the decrease of carbon emission. Differently from wood pellets, kraft pulping process using woody biomass produces black liquor as a by-product which can be used to generate electricity, bioenergy and biochemicals through gasification. Thus, it can be more economical to make a torrefaction of lignocellulosic biomass such as low-quality wood and agricultural leftovers as raw materials of pellets.
