Studies on the Pectolytic Enzymes from Byssochlamys fulva I (Byssochlamys fulva가 생성하는 펙틴질 분해효소에 관한 연구 I)

  • Effects of carbon sources, incubation time, incubation temperature, initial incubation pH, and vitamin B complex on the polygalacturonase activity of Byssochlamys fulva were studied to confirm the optimum conditions for the production of that enzyme. When pectin was used as carbon source, polygalacturonase activity reached to the maximum value of 0.50 units/ml. After 5 days of incubation, polygalacuturonase activity reached to its maximum of 0.48units/ml. Polygalacturonase activities were similar between $25^{\circ}C\;and\;45^{\circ}C$, however, decreased dramatically in the outside of this range. Polygalacturonase activity was not signicicantly influenced by the variation of initial incubation pH. However, at pH5.0, polygalacturonase activity was slightly through the addition of thiamine and riboflavin, and the optimum concentrations were $10^{-2}M$ in case of thiamine and $10^{-3}M$ in riboflavin.

Colonization of Retama raetam Seeds by Fungi and Their Significance in Seed Germination

  • Examination by scanning electron microscopy and potato-dextrose agar medium showed that the dry seeds of R. raetam were externally free of fungi. When planted in sandy loam soil, the seeds become colonized with eleven soilborne fungal species. The fungi were isolated on cellulose agar, pectin agar and lignin agar media. Aspergillus flavus, A. niger, Penicillium capsulatum and Fusarium oxysporum had broad occurrence and recovered on the three media. The production of hydrolytic enzymes by the isolated fungi depends on the substrate and species. P. capsulatum, P. spinulosum and A. niger had wide enzymatic amplitude and they were able to produce cellulolytic, pectolytic and lignolytic activities on corresponding substrates as well as on seed coat containing media. The lignolytic activities of the isolated species except Chaetomium bostrychods and Trichoderma viride were enhanced on applying the seed coat materials as C-source rather than using lignin. Soaking R. raetam seeds in culture filtrates of the most fungi grown on seed coat supplemented media induced pronounced and distinct stimulating effect on seed germination. The most effective filtrates were those of P. capsulatum, P. spinulosum and Sporotrichum pulverulentum.

Cell Wall Localization of Two DUF642 Proteins, BIIDXI and TEEBE, during Meloidogyne incognita Early Inoculation

  • The root-knot nematode Meloidogyne incognita infects a variety of plants, including Arabidopsis thaliana. During migration, root-knot nematodes secrete different proteins to modify cell walls, which include pectolytic enzymes. However, the contribution of host cell wall proteins has not been described during this process. The function of two DUF642 cell wall proteins, BIIDXI (BDX, At4g32460) and TEEBE (TEB, At2g41800), in plant development could be related to the regulation of pectin methyl esterification status in the cell walls of different tissues. Accordingly, the expression of these two genes is up-regulated by auxin. BDX and TEB were highly induced during early M. incognita inoculation. Moreover, cell wall localization of the proteins was also induced. The cell wall localization of BDX and TEB DUF642 proteins during M. incognita early inoculation suggested that these two proteins could be involved in the regulation of the degree of pectin methylation during cell separation.

Production of Pectolytic enzymes by Alternaria mali Roberts (Alternaria mali Roberts에 의(依)한 Pectin질(質) 분해효소(分解酵素)의 생산(生産))

  • Isoates with changed pathogenicity were selected among iprodione-resistant Alternaria mali to investigate any relationship between their pectolytie enzyme activity and pathogenicity. In artificial medium, strongly pathogenic isolates $S_1$, and $R_3$ showed higher enzyme activity than weakly pathogenic isolate $R_8$.Activity of endo-polymethylgalacturonase and end-o-polygalacturonase was more than 3 times. But activity of pectin methylesterase and pectin Iyase by isolaste $S_1$ was higher than those by $R_3$ and $R_8$ isolates. In apple medium dialyzed against distilled water, activity of enzyme by each isolate was increased but growth of each isolate was reduced. When iprodione was added to the medium, enzyme activity and growth of isolate $S_1$, were reduced but strongly pathogenic isolate .$R_3$ among iprodione-resistant ones showed increased enzyme activity except for exo-polygalacturonase in dialyzed apple medium.
