An Effective Method of Diagnosis of Potato Leafroll Virus by RT-PCR (RT-PCR 방법을 이용한 효과적인 감자 잎말림 바이러스의 검정)

  • 감자 잎말림 바이러스를 검정하기 위하여 ELISA 및 전자현미경에 의해 바이러스 감염이 확인된 기내 배양중인 감장의 줄기로부터 RT-PCR 분석을 수행하였다. 분리된 총 RNA들로부터 바이러스 cDNA를 합성하고 감자 잎말림 바이러스 외피단백질의 일부인 465bp를 특이하게 증폭하도록 고안한 두 primer를 사용하여 PCR 반응을 하였다. 증폭된 465pb의 DNA 절편의 염기서열을 분석한 결과 역시 감자 잎말림 바이러스임을 확인하였다. 바이러스 검정에 있어서 EL-ISA 방법과 RT-PCR 방법간의 민감도를 조사한 결과 RT-PCR 방법간의 민감도를 조사한 결과 RT-PCR 방법이 ELISA 방법보다 감자 잎말림 바이러스검정에 있어서보다 정확한 방법인 것으로 사료된다.

  • Tempting to further understand the molecular mechanism of pharmacological action of ethanol, we evaluated effects of phosphatidylethanol (PET) on inositol 1,4,5-triphosphate (IP3) level and protein kinase C (PKC) activity in cultured NG108-15 cells. PET increased intracellular concentration of IP$_3$. PET incorporation into membranes of NG108-15 cells had no effect on the phosphorylation of the PKC-specific substrate MBP$\_$4-14/, thus indicates that PET does not affect PKC activity in this system.

  • In this study, we evaluated the anti-nociceptive activities of Sorbus alnifolia. To investigate the anti-nociceptive properties of the methanolic extract of Sorbus alnifolia (MSA), we conducted several tests using various experimental mouse pain models. Herein, MSA significantly delayed the latency time and writhing motion in the hotplate test and acetic acid test, respectively. These result indicated that MSA has an ability to manage both peripheral and central nociception. We could further confirm the analgesic effects of MSA by performing formalin test. In combination test using naloxone, a non-selective opioid receptor antagonist, analgesic activity of MSA was partly antagonized by naloxone, but not completely, indicating that the MSA acts as a partial opioid receptor agonist. Out results suggest that the S. alnifolia may be possibly used as valuable anti-nociceptive agent.

  • It is over 100 years after Lee Je Ma has proposed concept of the Sasang constitutional medicine in his book DongEuiSooSeBoWon in 1894. As well known, this concept describes four constitution of Taeyang, Taeum, Soyang, and Soeum which are deals with physical body status and shape, desease and symptoms, personal characters, and organ function. In these days there are growing needs to elucidate this Sasang concept by the scientific manner, especially by biological tools. However there are no genes revealed related to the Sasang and moreover it is not easy to give a biological evidence for the Sasang constitution. Here are some considerations about the brief history of biological research on the Sasang constitution, and some prospectives of how to do to find genes of Sasang, and which to be done for what is about Sasang.

  • D-class cyclins play important roles in controlling the cell cycle in development and in response to external signals by forming the regulatory subunit of cyclin-dependent kinase (CDK) complexes. To evaluate the effects of D-class cyclins in transgenic rice plants, Arabidopsis cyclin D2 gene (CycD2) was linked to the maize ubiquitin1 promoter (Ubi1) and introduced into rice by the Agrobacterium-mediated transformation method. Genomic deoxyribonucleic acid (DNA), ribonucleic acid (RNA), and Western blot hybridizations of the Ubi1:-CycD2 plants revealed copy number of transgene and its increased expression in leaf and callus cells at messenger RNA (mRNA) and/or protein levels. The H1 kinase assay using the immunoprecipitates of protein extracts from the Ubi1:CycD2 plants and nontransgenic controls demonstrated that the introduced Arabidopsis CycD2 forms a functional CycD2/CDK complex with an unidentified CDK of rice. Shoot and root growth was enhanced in the Ubi1:CycD2 seedlings compared with nontransgenic controls, together, suggesting that Arabidopsis cyclin D2 interacts with a rice cyclin-dependent kinase, consequently enhancing seedling growth.

A Study of Standard Head Model for Korean Adults by 3D Measurement (한국 성인의 3차원 표준 머리모형)

  • The purpose of this study were to analyse of craniofacial shape by 3D and to develope of head models for Korean adults with gender and age groups. The 3D measurement technique adapted in this study was a novel approach compared that the same technique has been commonly used in measuring human bodies. The data and the model of head analysis can be used as a basic reference in developing various head related items such as hat, helmet, gas mask, ear phone, and etc. In this study, heads of 836 Korean adults were measured in 3D, analyzed by statistical methods, and modelized in 3D by gender and age groups. From the basic statistical data analysis, vertex-tragion and the length between the pupils were the longest in their twenties for both men and women, and grew shorter in elderly groups. In all categories, a significant difference appeared between men and women in their 20's, but the differences were less noticeable in elderly groups. Compared to the one size standard head model of the Korea Occupational Safety and Health Agency, the above three-dimensional standard head model would provide a more through fit because gender and age groups were sub-divided and analyzed in 3D.

A Study on the Consciousness of Mortgage Loan and Related Factors of Prospective Home-Buying Households (주택구매예정가구의 모기지론에 대한 의식과 관련변인)

  • The study analyzed the consciousness of mortgage loan of the prospective home-buying households using self-administered questionnaire surveys. The sample of the survey was chosen by convenience sampling method to be 366 prospective home-buying households in Ulsan, with the households head's age being younger than 50. These are the results. First, approximately 80% of the respondents had plans to buy a house through self-support and loan. Second, the consciousness of mortgage loan was relatively low, but the willingness to use it was very high. Third, the need for mortgage loan was relatively high, especially the need for specialists to facilitate the information circulation. Lastly, the awareness and need for mortgage loan were significantly influenced by the family and housing characteristics of households including family life cycle stages, the structure of dwelling, tenure type and monthly household income. It is necessary to provide potential house buyers with appropriate education and information on housing financing, the change of interest rate, and the effects of various financing packages.

  • A series of experiments have been performed in a vertical tube of 9.4 mm inner diameter using the Freon, HFC-134a as working fluid medium under the supercritical pressure range. Two kinds of experiments, i.e. steady-state and pressure transient, have been carried out. As for the steady-state heat transfer experiment, the mass flux was in the range between 600 and $2000\;kg/m^2s$ and the maximum heat flux was $160\;kW/m^2$. The selected pressures were 4.1, 4.3 and 4.5 MPa which correspond to 1.01, 1.06 and 1.11 times the critical pressure, respectively. In the pressure transient experiments, the inlet pressures were varied from 3.8 to 4.5 MPa and vice versa in the pressure transient simulations. In this study, heat transfer correlation and criterion for the heat transfer deterioration are suggested under the supercritical pressures. And also heat transfer characteristics during the pressure transient are examined.

Effects of Anemarrhenae Rhizoma on LPS-stimulated Expression of COX-2 and iNOS in mouse BV2 microglial cells

  • Anemarrhenae Rhizoma (AR) has been widely used for the treatment of various diseases in Oriental medicine. To investigate whether AR possesses anti-inflammatory effects against lipopolysaccharide (LPS)-induced inflammation in the BV2 microglial cells, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide) assay. reverse transcription -polymerase chain reaction (RT-PCR), and prostaglandin E2 (PGE2) assay, and nitric oxide (NO) detection assay were performed. From the present results, AR was shown to suppress PGE2 synthesis and NO production by inhibiting the LPS-stimulated enhancement of cyclooxygenase-2 and inducible nitric oxide synthase expression in BV2 microglial cells. These results suggest that AR may offer a valuable means of therapy in the treatment of inflammatory diseases by attenuating LPS-induced PGE2 synthesis and NO production.

  • An ionization chamber is still widely used in many fields by virtue of its' simple operational characteristics and the possibility of its' various shapes. A parallel type of an ionization chamber for a steel sheet thickness measurement was designed and fabricated. High pure xenon gas, which was pressurized up to 6 atm, was chosen as a filling gas to increase the current response and sensitivity for a radiation. A high pressure gas system was also constructed. The active volume and the incident window size of the fabricated ionization chamber were $30\;cm^3\;and\;12\;cm^2$, respectively. Preliminary tests with a 25 mCi $^{241}Am$ gamma-ray source and evaluation tests in a standard X-ray field were performed. The optimal operation voltage was set from the results of the collection efficiency calculation by using an experimental two-voltage method. Linearity for a variation of the steel sheet thickness, which is the most important factor for an application during a steel sheet thickness measurement, was 0.989 in this study.
