Endophytes are microorganisms that live in the internal tissues of plants without harming the host plants. In this symbiotic relationship, the host plants provide nutrients and shelter to the endophytes, in turn, endophytes can promote the growth of host plants and act as a biological control agents against plant pathogens. Plant-microbe interactions like this are noted for natural methods for sustainable agriculture and environmental conservation. However, in spite of the infinite potential, there are only a few reports on the endophytes present in ginseng. In this study, we isolated and identified the endophytes from Panax ginseng seeds and evaluated the biological activities (IAA production ability, nitrogen fixation ability, phosphate solubilization capacity, siderophore production ability, and antifungal activities) of the endophyte isolates. Eight different endophytes were identified by 16S rRNA sequencing. Most of the endophytes have antibiotic and plant growth promoting (PGP) activities. Particularly, PgSEB5-37E have the highest antibiotic activity, both PgSEB5-37B and PgSEB5-37H have high PGP traits such as an abilities to produce IAA, solubilize phosphate and fix nitrogen. These results indicated that the endophytes from P. ginseng seeds may have applicable value to many industries. In order to use the isolated endophytes, quantitative analysis and field tests are needed to be performed.

Background: Wild ginseng, Panax ginseng Meyer, is an endangered species of medicinal plants. In the present study, we analyzed variations within the ribosomal DNA (rDNA) cluster to gain insight into the genetic diversity of the Oriental ginseng, P. ginseng, at artificial plant cultivation. Methods: The roots of wild P. ginseng plants were sampled from a nonprotected natural population of the Russian Far East. The slides were prepared from leaf tissues using the squash technique for cytogenetic analysis. The 18S rDNA sequences were cloned and sequenced. The distribution of nucleotide diversity, recombination events, and interspecific phylogenies for the total 18S rDNA sequence data set was also examined. Results: In mesophyll cells, mononucleolar nuclei were estimated to be dominant (75.7%), while the remaining nuclei contained two to four nucleoli. Among the analyzed 18S rDNA clones, 20% were identical to the 18S rDNA sequence of P. ginseng from Japan, and other clones differed in one to six substitutions. The nucleotide polymorphism was more expressed at the positions 440-640 bp, and distributed in variable regions, expansion segments, and conservative elements of core structure. The phylogenetic analysis confirmed conspecificity of ginseng plants cultivated in different regions, with two fixed mutations between P. ginseng and other species. Conclusion: This study identified the evidences of the intragenomic nucleotide polymorphism in the 18S rDNA sequences of P. ginseng. These data suggest that, in cultivated plants, the observed genome instability may influence the synthesis of biologically active compounds, which are widely used in traditional medicine.
An efficient in vitro protocol has been established for somatic embryogenesis and plantlet conversion of Korean wild ginseng (Panax ginseng Meyer). Wild-type and mutant adventitious roots derived from the ginseng produced calluses on Murashige and Skoog (MS) medium supplemented with 0.5 mg/L 2,4-dichlorophenoxyacetic acid and 0.3 mg/L kinetin; 53.3% of the explants formed callus. Embryogenic callus proliferation and somatic embryo induction occurred on MS medium containing 0.5 mg/L 2,4-dichlorophenoxyacetic acid. The induced somatic embryos further developed to maturity on MS medium with 5 mg/L gibberellic acid, and 85% of them germinated. The germinated embryos were developed to shoots and elongated on MS medium with 5 mg/L gibberellic acid. The shoots developed into plants with well-developed taproots on one-third strength Schenk and Hildebrandt basal medium supplemented with 0.25 mg/L 1-naphthaleneacetic acid. When the plants were transferred to soil, about 30% of the regenerated plants developed into normal plants.
Background: Panax ginseng Meyer has widely been used as a traditional herbal medicine because of its diverse health benefits. Amounts of ginseng compounds, mainly ginsenosides, vary according to seasons, varieties, geographical regions, and age of ginseng plants. However, no study has comprehensively determined perturbations of various metabolites in ginseng plants including roots and leaves as they grow. Methods: Nuclear magnetic resonance ($^1H$ NMR)-based metabolomics was applied to better understand the metabolic physiology of ginseng plants and their association with climate through global profiling of ginseng metabolites in roots and leaves during whole growing periods. Results: The results revealed that all metabolites including carbohydrates, amino acids, organic acids, and ginsenosides in ginseng roots and leaves were clearly dependent on growing seasons from March to October. In particular, ginsenosides, arginine, sterols, fatty acids, and uracil diphosphate glucose-sugars were markedly synthesized from March until May, together with accelerated sucrose catabolism, possibly associated with climatic changes such as sun exposure time and rainfall. Conclusion: This study highlights the intrinsic metabolic characteristics of ginseng plants and their associations with climate changes during their growth. It provides important information not only for better understanding of the metabolic phenotype of ginseng but also for quality improvement of ginseng through modification of cultivation.
1. Panax ginseng의 재배년수에 따른 내부형태적 차이를 검토한 결과, 소엽신(小葉身)의 주맥부(主脈部), 소엽병(小葉柄), 엽병(葉柄)의 횡절면(橫切面)에 있어서 각각의 특징에 의하여 발육년수(年數)를 판단할 수가 있었다. (Tabel II) 2. 내부형태를 비교검토한 결과, 서울, 부산, 대구, 금산의 $\ulcorner$인삼잎$\lrcorner$은 원식물(原植物)이 Panax ginseng으로서 대부분이 5년 및 6년생이었으며 부분적으로 4년생이 함유되어 있었다.
종간, 속간의 잡종에서는 흔히 불임현상이 일어나는데 인삼의 경우도 예외가 아니어서 고려인삼(panax ginseng: P.G.)과 미국인삼(panax quinquefolium: P. Q.) 그리고 죽절인삼(Panax japonicus: P.J) 간의 교잡육종에서 P.G.${\times}$P.J.에서만 아주 희귀하게 종자를 얻을수 있고 다른 종합에서는 전혀 잡종 제이세대를 얻을 수가 없었다고 하므로 본실험에서는 P.G.${\times}$P.Q.와 P.G.${\times}$P.J.의 교잡종의 화기 및 종자형성중의 유리아미노산의 소장을 추구함으로서 불임현상과 물질대사와의 관련성의 일단을 밝히고저 한다. 1. 두 교잡종의 시료에서 검출된 유리아미노산의 Chromatogram은 서로 유사한 pattern을 나타냈으며 그들의 양친이 나타내는 pattern과 유사하나 spot의 종류나 크기 정색도는 여러 가지 점에서 특이한 변화가 있다. P.G.${\times}$P.Q.에서 19종, P.G.${\times}$P.J.에서는 21종의 spot을 검출하였는데 전자에서 Alanine, Valine, Leucine, Phenyl alanine, Proline, Hydroxy proline, Serine, Threonine, Tyrosine, Aspartic acid, Glutamic acid, Lysine, Arginine, ${\beta}$-Alanine, Cysteic acid, Tryptophan, Asparagine, Glutamine, ${\gamma}$-Amino butyric acid를 확인하고 후자의 경우는 상기한 것 외에 Methionine과 한 개의 미지 spot를 얻었다. 2. Alanine, Aspartic acid, Glutamic acid, Cysteic acid와 Asparagine이 전체적으로 보아 주체를 이루고 있는 것은 P.G. P.Q. P.J에서와 같으나 Asparagine이 소포자기와 화분성숙기에 최다량으로 나타나는 것은 이들의 양친의 경우에는 보지 못했던 특이한 일이다. 3. 홍숙기에 Cysteic acid가 감소되는 것은 P.Q. P.J.의 경우와는 유사하나 P.G의 경우와는 반대되는 현상이며 P.G.${\times}$P.J.에서 Methionine이 검출된 것도 특이하다. 4. Proline은 그들의 양친의 경우와는 현저한 차이가 있었는데 소포자기에서 아주 미약한 정색을 나타낼뿐이며 이 때에 Asparagine의 spot가 가장 크고 강한 정색을 나타내었는데 이제까지의 여러 연구 결과를 종합해 보면 Proline의 양과 화분의 임성과는 아주 밀접한 관계가 있으며 이런 관계가 인삼 종자 형성에서도 나타나고 있다. 또한 Proline이 결핍될 때 Asparagine의 축적이 있다는 많은 보고도 있다. 5. 교잡종에서의 결실불능의 원인을 Proline의 과소에 의한 것으로 보며 Proline이 약의 화분퇴화와 중요한 관계를 가지고 있을 것으로 추찰된다. 6. 그밖에 여러 아미노산의 소장이 P.G. P.Q. P.J. 등의 자식계와 그들과의 잡종사이에 있어서 상당한 차이가 생기는 것은 교잡에 의한 Gene-action system의 변혁에 따른 결과라고 추찰되는 것으로 Proline의 대사 및 생리적 영향과 더불어 보다 깊은 조사가 필요한 것으로 생각된다.

Results of karyological study of intact plants and some callus lines of Panax ginseng are presented. In the native plants of P. ginseng the nucleus with 1 nucleolus (90%) dominate, and nucleus with 2 nucleoli is rare. One nucleolar nucleus also dominate in interphase nuclei of cells of cultivated P. ginseng (from 2006), but we also found nucleus with 2 to 3 nucleoli in the same cell lines. Interphase nuclei of P. ginseng in long cultivated lines (from 1988) contain 1 to 9 nucleoli, with a predominance of nuclei containing from 3 to 4 nucleoli. It was shown that long-time cells (cultivated since 1988) had cytogenetic changes such as increase level of polyploid and aneuploid cells, increase of nucleoli number into interphase nucleus and decrease of nuclei/nucleoli ratio. These long-time cultivated cells had very low ginsenoside content.
Forest-grown American ginseng (Panax quinquefolius L.) is exposed to daily and seasonal light variations. Our goal was to determine the effect of understory light changes on the maximum quantum yield of photosystem II, expressed as $F_v/F_m$, and photosynthetic pigment composition of two-year-old plants. Understory light photon flux density and sunfleck durations were characterized using hemispherical canopy photography. Our results showed that understory light significantly affected the $F_v/F_m$ of American ginseng, especially during the initial development of the plants when light levels were the highest, averaging 28 mol $m^{-2}d^{-1}$. Associated with low $F_v/F_m$ during its initial development, American ginseng had the lowest levels of epoxidation state of the xanthophyll cycle of the season, suggesting an active dissipation of excess light energy absorbed by the chlorophyll pigments. As photon flux density decreased after the deployment of the forest canopy to less than 10 mol $m^{-2}d^{-1}$, chlorophyll a/b decreased suggesting a greater investment in light harvesting pigments to reaction centers in order to absorb the fleeting light energy.

Somatic embryogenesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology such as medicinally important plants. Single embryos develop into normal plantlets with shoots and roots. Therefore, direct single embryogenesis derived from single cells is highly important for normal plant regeneration. Here we demonstrate that the cyclic secondary somatic embryogenesis in Panax ginseng Meyer is a permanent source of embryogenic material that can be used for genetic manipulations. Secondary somatic embryos were originated directly from the primary somatic embryos on hormone-free Murashige and Skoog medium, and proliferated further in a cyclic manner. EM medium (one third of modified MS medium [MS medium containing half amount of NH4NO3 and KNO3] with 2% to 3% sucrose) favored further development of proliferated secondary somatic embryos into plantlets with root system. The plantlets developed into plants with well-developed taproots in half-strength Schenk and Hildebrandt basal medium supplemented with 0.5% activated charcoal.
This study was performed to investigate the effects of enhanced light transmission on plant growth, photosynthetic ability, and disease tolerance to leaf blight, anthracnose in ginseng (Panax ginseng C. A. Meyer, Araliacease family) during the early growth stage (April to June). The photosynthetic ratio, stomatal conductance, and stem diameter of plants grown under a shade net with 15% light transmission rate showed an increasing trend compared to the control plants (5% light transmission rate) although the growth of the aerial parts were not influenced significantly. Plant height, stem length, and leaf length of treated plants were not significantly different from those of the control plants. Root parameters, such as root length, diameter, and weight of treated plants increased significantly compared to the control. Yield performance ($187.4kg{\cdot}10a^{-1}$) of treated plants was 55.5% higher than that of the control ($150.4kg{\cdot}10a^{-1}$). Additionally, disease severity scores of treated plants were lower than those of the control plants, revealing higher survival rates. To retain high yield potential and enhance the level of disease tolerance in ginseng, we suggest the increase of light transmission rate during the early growth stage.
