PRODUCTION OF GINSENOSIDES THROUGH IN VITRO CULTURE OF GINSENG(Panax ginseng C.A. MEYER)

  • Ginseng root explants and calli induced from selected cell lines were cultured on modified Murashige and Skoog's media supplemented with different concentrations of organic or inorganic compounds and plant growth requlators to clarify the effects of chemical composition and plant growth regulators in the medium on the growth of ginseng calli and the production of ginseng saponin. For optimum growth of calli, the concentrations of 2, 4-D and sucrose were the range of 1 to 3 mg/${\ell}$l and 1 to $3\%,$ respectively. And it was clarified that sucrose, nitrogen, phosphate, calcium, magmesian plant growth regulators and their concentrations influcenced the relative biosynthesis of saponin in tissue cultures of Panax ginseng. The patterns of ginsenosides, pharmacologically useful component, were different among the cell lines and contents of ginsenosides were much higher in selected cell lines than in original cell line.

Changes of Growth Characteristics and Ginsenoside Content by Growth Stages and Different Planting Position in Panax ginseng C.A. Meyer (인삼의 재식위치와 생육시기에 따른 생육특성 및 진세노사이드 함량 변화)

  • This study was conducted to investigate the effect of planting position on the growth characteristics, yield and ginsenoside content in Panax ginseng C.A. Meyer at different growth stages. Referring to shoot growth characteristics, stem length, stem diameter and leave area were higher at front than rear, increasing as the proceeding of growth stages. But a lower chlorophyll contents was caused at front compared to rear and decreased as the proceeding of growth stages contrarily. According to root characteristics, root length and main body length were higher at front, with a positive correlation to growth stages, which was also shown on fresh root weight and dry root weight with the maximum in August. Meanwhile, the effect of planting position on ginsenoside content could also be definite by the highest content at front showing high light intensity, increasing as the proceeding of growth stages as well.

Suppressive effect of Petroleum Ether Extract of Panax ginseng against Benzo(a)pyrene induced Micronuclei in Mice (인삼 석유에텔 추출물이 흰쥐에서 Benzo(a)pyrene에 의해 유도된 소핵생성의 억제효과)

  • Petroleum ether extracts of panax ginseng C.A. Meyer (GPEE) were tested for the evaluation of anticlastogenic effects against benzo(a)pyrene-induced micronucleated polychromatic erythrocytes using mouse bone marrow micronucleus test. When the GPEE was singly administered before benzo(a)pyrene injection, GPEE showed significant anticlastogenic effect at $50{\sim}200\;mg/kg$. When the GPEE was multiply administered for 5 consecutive days before benzo(a)pyrene injection, GPEE showed potent anticlastogenic effect, even at the low doses, $5{\sim}50\;mg/kg/day$. As a control experiment, GPEE was administered without benzo(a)pyrene injection to demonstrate a clastogenic effect of this extract. When the range of $1{\sim}200\;mg/kg/day$ for 5 consecutive days was administered to mice, it was found that there was no increase of MNPCEs frequency.

Effects of Root Diameter Within Different Root Parts on Ginsenoside Composition of Yunpoong Cultivar in Panax ginseng C. A. Meyer (인삼 연풍의 근 부위별 직경이 진세노사이드 함량에 미치는 영향)

  • This study was carried out to investigate the correlation between root diameter and ginsenoside composition of Panax ginseng C. A. Meyer cultivar Yunpoong. Dry matter ratio of main root was a little higher than that of lateral root and fine root, and that was higher by the increase of root diameter in the same root parts. Total ginsenosides composition of main and lateral roots increased by the decrease of root diameter, especially in lateral root. Similar resulted in fine root, but there was no significant difference where root diameter was below 2.5 mm. Except for ginsenoside-$Rg_1$, other ginsenosides component, PDs, PTs and total ginsenosides had highly negative correlation with the root diameter within whole root, main root+lateral root and lateral root+fine root, while $Rg_1$ had positive correlation with the root diameter.

Breeding Process and Characteristics of KG101, a Superior Line of Panax ginseng C.A. Meyer (인삼 우수계통 KG101의 육성경과 및 생육특성)

  • To develop a new ginseng varieties with good quality and high yielding, a lot of individual ginseng plants were selected in the farmer's fields in 1972. Among them, a promising line, 7259-3-1, has been developed through comparative cultivation of several lines selected with pure line separation of local races in Korea Ginseng '||'&'||' Tobacco Research Institute. Preliminary and advanced yield trials were performed for 8 years. It was then designated as "KG 101" and tested in the regional yield and adaptation trials for 10 years (1981-1990). KG101 has a green stem with light violet and orange-yellow fruit and flowers 3-7 days later than local race, Takyungjong. Taproot of KG101 was longer than local race Jakyungjong, and root yield of KG101 was 9% higher than local race Jakyungjong. In red ginseng quality, the rates of Chun-Jeesam (Chun and Jee means 1st and 2nd grade, respectively) were 22.3% and 9.4% for KG101 and Jakyungjong, respectively. In these results, it was clarified that KG101 was superior ginseng line with good quality.y.

Effects of Antioxidants on the Photosynsthesis and Carbohydrates/Saponin Contents in Panax ginseng Leaves (인삼잎의 광합성과 탄수화물.사포닌 함량에 미치는 항산화제의 효과)

  • We studied the folilar wiping effects of antioxidants (ascorbate, glutathione and sodium azide), which effectively inhibited the chlorophyll bleaching or completely recorved the early stage of photosynthesis of Panax ginseng C.A. Meyer, on photosynthesis, stomatal resistance, free sugar, starch, and total saponin contents of ginseng under the excess light intensity (45 kLux) during 6 days. Ascorbate and glutathione, endogenous antioxidant, recovered photosynehtsis and stomatal resistance, and reduced the photoinhibition by the excess light intensity (45 kLux) on free sugar, starch and total saponin contents. But sodium azide, exogenous $^{1}O_2$ quencher, showed negative effect. Therefore, we assumed that carbohydrates and saponin metabolisms of ginseng by antioxidants (ascorbate, glutathione) were normal. For the reduction of inhibition by excess light in ginseng a program for the higher activation of antioxidants and antioxidative enzymes in ginseng leaf will be desirable. Panax ginseng: a candidate herbal medicine for autoimmune disease

  • Panax ginseng Meyer (P. ginseng; Korean ginseng) is well known for its medicinal properties. It can alleviate pathological symptoms, promote health, and prevent potential diseases via its anti-inflammatory, antioxidant, homeostatic, and other positive effects on biological metabolism. Although many studies have determined effects of P. ginseng on various diseases, such as cardiovascular, neurological, and immunological diseases, little is known about the effect of P. ginseng on autoimmune diseases. Here, we review a few reports about effects of P. ginseng on autoimmune diseases (e.g., multiple sclerosis, Crohn's disease, ulcerative colitis, atopic dermatitis, and rheumatoid arthritis) and suggest the possibility of P. ginseng as a candidate herbal medicine to prevent and treat autoimmune diseases as well as the need to study it.

Studies on the Volatile Flavor Components of Fresh Ginseng (수삼중 휘발성 향기성분에 관한 연구)

  • Volatile flavor components of fresh ginseng (Panax ginseng C.A. Meyer.) were studied. Steam distillate of fresh ginseng was extracted with ethyl ether and the extract was separated into four fraction: neutral, phenolic, acidic and basis fractions. The ethyl ether concentrates and neutral fraction were analyzed by a combination of SE-54 fused silica capillary gas chromatography and mass spectrometry. Major flavor components of fresh ginseng were predominantly mono(n +2) and sesquiterpenes(n +3) in over two hundred constituents. Of these, 28 were newly identified in volatile flavor components of fresh ginseng by GC-MS.
