  • This study was investigated and analyzed the side of phenomenological of the chlorophyll bleaching phenomenon on the leaf burning-disease of the Ginseng (Panax ginseng C.A. Meyer) leaf. Red light (660-700 nm) was confirmed as one which induced the bleaching phenomenon and blue light (400-500 nm) did not at all. Temperature as 1 environmental factor had not any influence on chlorophyll bleaching phenomenon at all. Therefore, simple burning (thermal damage) hypothesis was perfectly ruled out by the result of this study. And, low pH accelerated chlorophyll bleaching velocity. A primary factor of chlorophyll bleaching phenomenon may be peculiar structural difference of the Ginseng leaf compared with other plant.

Effect of Soil Properties and Soil Bacterial Community on Early Growth Characteristics of Wild-simulated Ginseng (Panax ginseng C. A. Meyer) in Coniferous and Mixed Forest (침엽수림과 혼효림에서 토양특성과 토양세균 군집이 산양삼 초기 생육특성에 미치는 영향)

  • Background: This study investigated the effect of soil properties and soil bacterial community on early growth characteristics of wild-simulated ginseng (Panax ginseng C. A. Meyer) in coniferous and mixed forest experimental fields. Methods and Results: The soil bacterial community was analyzed using a high throughput sequencing technique (Illumina MiSeq sequencing). The relationship between the soil bacterial community, soil properties, and growth characteristics of wild-simulated ginseng were analyzed using principal coordinate analysis (PCoA) and the Pearson's correlation analysis. Soil properties and soil bacterial community showed significant difference with forest physiognomy. Results of Pearson's correlation analysis and PCoA showed that the soil properties (soil pH, organic matter, total nitrogen, and cation exchange capacity) and soil bacterial community had significant correlation with tree species ratio and early growth characteristics of wild-simulated ginseng. Conclusions: This study clearly demonstrated the effect of soil properties and soil bacterial community on early growth characteristics of wild-simulated ginseng in coniferous and mixed forest. Moreover, these results will help in the selection of suitable cultivation sites for wild-simulated ginseng.

Effects of Growth Regulators on the Formation of Somatic Embryo and Adventitious Bud from the Cotyledon of Korean Ginseng (Panax ginseng C.A. Meyer) (고려인삼(Panax ginseng C.A. Meyer)의 자엽으로부터 체세포배 및 부정아의 발생에 미치는 식물호르몬의 영향)

  • Cotyledon explants of Korean ginseng (Panax ginseng C.A. Meyer), a perennial medicinal plant, produced direct somatic embryos at a high frequency on MS medium without growth regulators. Cytokinin highly suppressed the somatic embryogenesis but stimulated direct fomlation of adventitious buds. BAP was more effective than kinetin for the formation of adventitious bud. IBA combination with cytokinin enhanced the frequency of adventitious bud formation. The highest frequency of adventitious bud formation were $40\%$ at 0.05 mg/l IBA and 5 mg/l BAP. Adventitious buds were mainly formed near the distal portion of cotyledon, while somatic embryos were only formed near the proximal portion of cotyledon. Adventitious buds were covered with sheath similar to axillary buds in the zygotic embryos, and then leaf-like epicotyls were developed.

Pharmacognostical Studies on Ginseng Folium (인삼잎의 생약학적 연구)

  • Ginseng Folium has been used to cure acute gastritis, alcoholism and anemia. The morphological and anatomical characteristics of the leaf and stem of Panax ginseng C.A. Meyer cultivated in Korea for 1 to 6 years are described. It is found that the vessels increase in number and diameter, and also the vascular bundles increase in number, as the plant becomes old. The result showed that the commercial &In Sam Ip& samples on the markets of Pusan, Kumsan, Taegu and Seoul have been derived from the leaf and stem of 4 to 6 years old P. ginseng plants, most of them being 5-6 years old.

  • Two lignanes, Comp.-I, mp 108-1$0^{\circ}C$ and Comp.-II 50-52$^{\circ}$were isolated from Korea ginseng extract by repeated column chromatographic purification. Comp.-I was identified as gomisin-N and Comp.-II as gomisin-A by spectrometric analysis, both of which have already been described as the anti-hepatotoxic lignan components of Schizandra chinesis Bail.

  • Ginseng zygotic embryos, seedlings, and exised cotyledonary nodes were cultured on Murashinge and Skoog's(MS) medium, supplemented with 6-benzyladenine(BA) and gibberellic acid(GA3) to induce flower buds. As the concenteration of nitrogen compounds in MS medium was reduced to half of its strength, the flowering frequency of zygotic embryos increased up to 90%. The optimum concentration of sucrose in the medium for flowering of seedlings was 30-60 g/1. In all cases flower buds were formed on elongated axillary branches from the cotyledonary nodes, while the apices remained vegetative. When zygotic embryos and excised cotyledonary nodes were cultured on the medium, supplemented with all possible combinations of BA, GA3, and abscisic acid(ABA) of 5 $\mu$M indole-3-acetic acid(IAA) in the above combinations did not affect flowering. These results suggest that cytokinins, gibberellins, and inhibitors play primary, permissive, and preventive roles, respectively, in the induction of flowering of ginseng.

  • We studied a assay method on the measurement of superoxide dismutase (SOD Superoxide : superoxide oxidoreductase, EC. 1. 15. 1. 1) activity with photoreduced flavin and nitroblue tetrazolium (NBT) as superoxide (${O_2}^{-}$) source and detector, respectively. The $\Delta$E (1000 ng SOD$.$$min.)^{-1}$ of photoreduced flavin-NBT system was 0.08, whereas that of xanthine-xanthine-cytochrome system used broadly in experiments was 0.014. Therefore, the new method was regarded more simple and utilizable than xanthine-xanthine cytochrome system method. In the present paper, we also carried out to investigate the SOD activity and isozyme pattern for the parpose of study of leaf-burning disease in ginseng (Panax ginseng C.A. Meyer) leaves.

  • 1. To know the site of saponin synthesis in this plant, 4-years old Panax ginseng C.A. Meyer was administered with 1, 2-l4C-acetate (Na salt, 10 ucilplant) by stem injection and was continued to grow for 3 weeks and the distribution of the radioactivity in leaf, stem and root part was identified. The percentage of radioactivity recovered was about 3.99%. 2. The sliced roots or leaf discs (2g) were bathed in the reaction mixture containing sugar, ATP, NADPH, and the distribution of the radioactivity of the fractions (sugar, saponin, sapogenin) was identified. 3. It seemed that major synthesized saponins in roots and leaves are dial and triol-type, respectively. Although both types of saponins are synthesized in roots, the main saponins seemed to be dial saponins and a significant portion of triol saponins are supplied from leaves through stem.
