An important factor in the production of organic ginseng is the control of $Alternaria$ blight and anthracnose, which mostly affect the leaves in the summer. We compared the effects of a lime-bordeaux mixture (LBM) and agricultural chemicals on the growth characteristics and ginsenoside content in 4-year-old ginseng plants when they were sprayed at 15-day intervals from mid-June to the end of September. The increases in leaf length, and survive-leaf ratio in plants sprayed with LBM were greater than the increases of the control plants, but less than those of agricultural chemicals treatment. The root weight per plant in the plants sprayed with LBM increased more distinctly than that in the control plants, while it was significantly lower than that in plants sprayed with agricultural chemicals. The root yield in plants sprayed with LBM increased by 21% compared to the root yield in the control plants, but decreased by 7% compared to that in plants sprayed with agricultural chemicals because of the decreases in leaf area and survive-leaf ratio. Spraying of LBM had a significant effect on the ginsenoside contents. The total ginsenoside content was highest in the control plants and lowest in the plants sprayed with agricultural chemicals and total ginsenoside contents was great relative to survive-leaf ratio and root weight.
Lime-bordeaux mixture has been used to prevent diseases in the field of ginseng. The purpose of this study is to investigate the efficacy of lime-bordeaux depending on the concentration and treatment time on major diseases of ginseng such as Alternaria blight and anthracnose, and to evaluate the root growth of ginseng. Lime-bordeaux caused damage on leaf when it was sprayed on ginseng between April and early May. No difference was found in root growth by spraying lime-bordeaux mixture between ratio 4-4 and 8-8 ratio in concentration. Plot of 6-6 raito and 8-8 ratio appeared to be similar efficacy compared to that of practical chemical control. However, the plot of 4-4 ratio showed lower than that of chemical control.
The distribution pattern of mineral nutrients, among various Parts of Korea ginseng (Panax ginseng C.A. Meyer) was investigated to understand ginseng nutrition by simple correlation analysis. Five·year old ginseng plants grown under four different nutritional environments were sampled and separated into leaf, petiole, stem, rhizome, cortex and epidermis of tap foot, central part of tap root, cortex and epidermis of lateral root, central part of lateral root, fine root in the middle of truly, for chemical analysis. Between mineral nutrients in root, N and P showed highly significant positive correlation each other and with Mg and Cu while all other elements (K, Ca, Mg, Fe, Mn, Zn, Cu, B) showed highly significant positive correlation each other. In shoot, number of mineral nutrient pairs haying significant relation was much less than in root. (Negative: P with Ca or B, K with N, Fe, Mn or Cu, Positive: N with Mg, Fe, Mn or Cu, K with Zn, Ca with Mg, Zn, or B, Fe Mn Cu each other, Mn with B.) The number of pairs having significant correlation in whole plant was approximately the same as the number in root but three of them showed significant negative correlation. The distribution pattern similarity of mineral contents among various parts was also discussed in relation to physiological significance in Korea ginseng plant.
To study the optimal cultivation for paddy soil, growth characteristics and ginsenoside content was investigated by both of poor drainage class (PDC) and imperfect drainage class (IDC) in three-year-old ginseng of varieties, Cheonpoong (CP), Yeonpoong (YP), Hwangsookjong (HS), and Jakyeongjong (JK). Root yield of IDC was higher than that of PDC by 3.6 times because stem length, leaf area, and chlorophyll contents were increased, while discolored leaf ratio was decreased. Root yield of HS in PDC was highest among four varieties because chlorophyll contents, leaf area, and survived plant ratio were relatively high. Root yield of CP in IDC was highest among four varieties because of high leaf area and survived plant ratio, and low discolored leaf ratio. Ratio of rusty-colored root showed significant difference by varieties, which was the highest in HS and the lowest in CP among four varieties irrespective of drainage classes. Total ginsenoside contents showed significant difference by drainage classes, which were high in IDC of good growth and low in PDC of poor growth. Total ginsenoside contents were high in JK and CP, while low in HS and YP both of drainage classes.
인삼(人蔘)의 근(根) 및 지상부(地上部) 사포닌을 얇은 막 크로마토그래피로 동정(同定)한 결과(結果) 인삼근(人蔘根)에 함유된 사포닌 중 $ginsenoside-Re,\;-Rg_1,\;-Rc,\;-Rf,\;-Rb_2$ 및 $-Rb_1$ 은 각각 엽(葉)과 경(莖)에서도 동정(同定)되었으며 이외에도 엽(葉)에서 10개과 경(莖)에서 9개의 unknown spot를 동정(同定)할 수 있었다. 또한 부위별(部位別) 총(總)사포닌을 50% 초산으로 온화한 조건에서 산(酸) 가수분해(加水分解)하여 생성(生成)된 prosapogenin의 조사결과(調査結果), panaxadiol계 사포닌 및 ginsenoside-Re의 산(酸) 가수분해물(加水分解物)인 $ginsenoside-Rg_3$ 및 $ginsenoside-Rg_2$가 각각 동정(同定)되었다. 한편 근(根), 엽(葉) 및 경(莖)의 총사포닌을 황산 가수분해(加水分解)하여 sapogenin 조사결과, 근(根)에서는 panaxadiol, panax atriol 및 oleanolic acid가 검출되었고, 엽(葉)과 경(莖)에서는 panaxatriol 및 panaxadiol만이 동정(同定)되었다.
Ginseng has been used as a general tonic agent to invigorate the human body as an adaptogenic agent. In a previous report, we have shown that ginseng contains a novel glycolipoprotein called gintonin. The main function of gintonin is to transiently enhance intracellular free $Ca^{2+}$$[Ca^{2+}]_i$ levels in animal cells. The previous method for gintonin isolation included multiple steps using organic solvents. In the present report, we developed a simple method for the preparation of crude gintonin from ginseng root as well as stem and leaf, which produced a higher yield of gintonin than the previous one. The yield of gintonin was 0.20%, 0.29%, and 0.81% from ginseng root, stem, and leaf, respectively. The apparent molecular weight of gintonin isolated from stem and leaf through sodium dodecyl sulfate polyacrylamide gel electrophoresis was almost same as that from root but the compositions of amino acids, carbohydrates or lipids differed slightly between them. We also examined the effects of crude gintonin from ginseng root, stem, and leaf on endogenous $Ca^{2+}$-activated $Cl^-$ channel (CaCC) activity of Xenopus oocytes through mobilization of $[Ca^{2+}]_i$. We found that the order of potency for the activation of CaCC was ginseng root > stem > leaf. The $ED_{50}$ was $1.4{\pm}1.4$, $4.5{\pm}5.9$, and $3.9{\pm}1.1$ mg/mL for root, stem and leaf, respectively. In the present study, we demonstrated for the first time that in addition to ginseng root, ginseng stem and leaf also contain gintonin. Gintonin can be prepared from a simple method with higher yield of gintonin from ginseng root, stem, and leaf. Finally, these results demonstrate the possibility that ginseng stem and leaf could also be utilized for ginstonin preparation after a simple procedure, rather than being discarded.
This study was devised to observe the nutritional effects of the diets supplemented with the leaf or trunk in rats. The male albino rats (110 heads), Sprague-Dowley strain weighing 75g to 79g, were used as the experimental aninl mils. The animals were divided into twelve diet groups and maintained with corresponding diet for 40 days, and then sacrificed. The growth rate, the consumption and efficiency ratios of the food and protein, the absorption rates of carbohydrate, lipid and protein, and the utilization rates of energy were determined during the feeding term. The results obtained are summarized as follows: 1) The growth rate, the consumption and efficiency ratios of the food and protein in each experimental diet group added ginseng steamed leaf or trunk were higher than those in experimental group A which has the diet supplemented with ginseng raw leaf of trunk. 2) The absorption rates of the carbohydrate were above 96%, higher than those or the control group, But those of fat, protein and the utilization rate of energy in each experimental diet group were generally lower than those in the control group.
Background: Panax ginseng Meyer is one of the most valuable medicinal plants which is enriched in anti-microbe secondary metabolites and widely used in traditional medicine. Botrytis cinerea is a necrotrophic fungus that causes gray mold disease in a broad range of hosts. B. cinerea could overcome the ginseng defense and cause serious leaf and root diseases with unknown mechanism. Methods: We conducted simultaneous transcriptomic and metabolomic analysis of the host to investigate the defense response of ginseng affected by B. cinerea. The gene deletion and replacement were then performed to study the pathogenic gene in B. cinerea during ginseng - fungi interaction. Results: Upon B. cinerea infection, ginseng defense responses were switched from the activation to repression, thus the expression of many defense genes decreased and the biosynthesis of antifungal metabolites were reduced. Particularly, ginseng metabolites like kaempferol, quercetin and luteolin which could inhibit fungi growth were decreased after B. cinerea infection. B. cinerea quercetin dioxygenase (Qdo) involved in catalyzing flavonoids degradation and ∆BcQdo mutants showed increased substrates accumulation and reduced disease development. Conclusion: This work indicates the flavonoids play a role in ginseng defense and BcQdo involves in B. cinerea virulence towards the P. ginseng. B. cinerea promotes disease development in ginseng by suppressing of defense related genes expression and reduction of antifungal metabolites biosynthesis.

Endophytic fungi were isolated from various tissues (root, stem, petiole, leaf, and flower stalk) of 3- and 4-year-old ginseng plants (Panax ginseng Meyer) cultivated in Korea. The isolated endophytic fungi were identified based on the sequence analysis of the internal transcribed spacer (ITS), 1-5.8-ITS 2. A morphological characterization was also conducted using microscopic observations. According to the identification, 127 fungal isolates were assigned to 27 taxa. The genera of Phoma, Alternaria and Colletotrichum were the most frequent isolates, followed by Fusarium, Entrophospora and Xylaria. Although 19 of the 27 taxa were identified at the species level, the remainder were classified at the genus level (6 isolates), phylum level (Ascomycota, 1 isolate), and unknown fungal species (1 isolate). Endophytic fungi of 13 and 19 species were isolated from 3- and 4-year-old ginseng plants, respectively, and Phoma radicina and Fusarium solani were the most frequently isolated species colonizing the tissues of the 3- and 4-year-old ginseng plants, respectively. The colonization frequency (CF%) was dependant on the age and tissue examined: the CFs of the roots and stems in the 3-year-old ginseng were higher than the CF of tissues in the 4-year-old plants. In contrast, higher CFs were observed in the leaves and petioles of 4-year-old plants, and endophytic fungi in the flower stalks were only detected in the 4-year-old plants. In conclusion, we detected diverse endophytic fungi in ginseng plants, which were distributed differently depending on the age and tissue examined.
본 연구는 장뇌삼(Panax ginseng)의 효율적인 증식과 성공적인 토양 순화의 최적 조건을 구명하기 위해 수행되었다. 장뇌삼의 자엽을 시료로 사용하여 체세포배를 유도한 후, 유도된 체세포배를 발아시켜 얻은 자엽을 다시 시료로 사용하여 2차 체세포배를 유도함으로써 효율성을 증가시켰다. 2차 체세포배의 형성율은 MS 배지의 경우 약 53%이고, WPM 배지의 경우 약 48%인 것으로 나타나 1차 체세포 배의 형성율인 MS 배지와 WPM 배지의 각각 83%, 73%에 비해 약간 떨어지는 것으로 나타났다. 형성된 체세포배는 $GA_3$가 첨가된 WPM 배지에서 발아시키고, 무기염류의 양을 반감시키고, $GA_3$가 첨가된 WPM 배지에서 신장시킨 후, 무기염류를 줄인 SH 배지에서 대기 순화와 무당배양의 처리에 따른 줄기와 뿌리길이, 생존율 차이를 조사하였다. 기내에서 대기 순화와 무당배양 혼용처리 시 가장 정상적인 유식물체로 성장 시킬 수 있었고, 뿌리길이도 가장 길었으며, 생존율 또한 75%로써 가장 높은 생존율을 보였다.
