These experiments were conducted to investigate the nematicidal effects of the insecticides, Mocap(O-Ethyl-S, S-dipropyl phosphorodithioate), Carbofuran(2,3-Dihydro-2,2-dimethyl benzofuranyl ethyl carbamate), Terbufos (S-tert-buthylthio methyl O,O-diethyl phosphordithioate) and their mixtures (Mocap+carbofuran, Mocap+Terbufos, Carbofuran+Terbufos) on growth of 2year-old ginseng, Panax ginseng C.A. Meyer, and the control of root-knot nematodes. There was no evidence of plant injury from insecticide treatment of ginseng, although the rate of emergence of the treated ginseng was slightly inhibited. The insecticide treatments showed no of-flavor of ginseng plant. Terbufos and Mocap provided heifer confrol of the root-knot nematodes than carbofuran alone and their mixtures. Mixtures of the insecticides showed antagonisitic effect to the root-knot nematodes.
Background : This study was conducted to determine out the effect of storage temperature on the quality of fresh ginseng (Panax ginseng C. A. Meyer) during distribution. Methods and Results : Fresh ginseng was washed, packed in $30{\mu}m$ low density polyethylene (LDPE) film, then stored at 0, -2 and $-4^{\circ}C$. After 4 weeks of storage, ginseng was then stored at $5^{\circ}C$, as a simulation of the distribution process. Ginseng stored at $-4^{\circ}C$ showed higher respiration rate, ethylene production and electrolyte conductivity during the distribution phase than those stored at 0 and $-2^{\circ}C$. Decay and browning rate rapidly increased following 3 weeks of distribution in samples stored $-4^{\circ}C$. However ginseng stored $-2^{\circ}C$, which is below freezing point, for 4 weeks did not show the physiological change or quality deterioration. Ginsenoside contents decreased during storage for all plant, but did not differ significantly between storage temperatures. Conclusions : Storage at temperatures below $-2^{\circ}C$ can negatively affect respiratory characteristics and electrolyte leakage and increase quality deterioration and decay rates during distribution.
This study was conducted to investigate chemical and physical characteristics of pectin in Korean ginseng plant. The results obtained are as follows: 1, The molecular weight of the pectin in ginseng plant was in the range of 1.1-2.4x 104 and athydrouronic acid content in the pectin was 97.98%. 2. The intrinsic viscosity of pectin as well as apparent visosity of the pectin in the roots were increased with cultural period. 3. The IR spectra of ginseng pectin showed the OH stretch, C-H bending vibration and the vibration of ester group carboxyl. 4. Sugars present in galacturonic acid, glucose, arbinose, xylose, galactose and rhamnose.
백삼 및 백삼에 함유된 페놀성 성분 6종의 산화적 손상에 대한 신경세포보호효과와 LPS로 유도된 대식세포에서의 NO 생성억제효과를 검정한 결과 백삼 추출물은 두가지 모두에서 농도의존적인 효과를 나타내었다. 백삼에 함유된 페놀성 성분 중에는 cinnamic acid 및 maltol이 우수한 신경세포보호활성을 나타내었고 NO 생성억제효과는 ascorbic acid 및 cinnamic acid가 높았다. 또한 페놀성성분의 백삼내 함량을 정량한 결과 ascorbic acid, cinnamic acid, esculetin가 다른 성분들에 비하여 비교적 많이 함유되어 있었으며 이를 종합하여 볼 때 백삼의 신경보호효과 및 항염증효과에는 높은 함량 및 우수한 활성을 보이는 성분인 cinnamic aicd가 크게 기여하는 것으로 판단된다.
We investigated the effect of antioxidants (ascorbate, glutathione, and sodium azide), which efEectively inhibited the chlorophyll bleaching of Panax ginseng CA Meyer under the high light intensity, treated by folilar wiping on the early stage of photosynthesis and transpiration of ginseng in the 5000 $\mu$mol photon.$m^{-2}$.$s^{-1}$. Ascorbate and glutathione, endogenous antioxidant, completely recovered ginseng from the photoinhibition, but sodium azide, synthetic quencher, showed negative effect. We assumed that endogenous antioxidants could be available to the protection of the leaf-burning phenomenon of ginseng.
Hairy roots were induced from Korean ginseng (Panax ginseng C. A. Meyer) root explants and studied for their gene expression. A total of 3,000 ESTs (expressed sequence tags) from ginseng hairy root were determined and about 2,700 ESTs have a length of readable sequence, which result in 1,352 unique ESTs sequences. The 879 ESTs showed significant similarities to known nucleotide or amino acid sequences in other plant species, which were divided into eleven categories depending upon gene function. The remaining 473 sequences showed no significant matches, which are likely to be transcripts or to be matched to other organisms. The results indicated that the analysis of the ginseng hairy root ESTs by partial sequencing of random cDNA clones may be an efficient approach to isolate genes that are functional in ginseng root in a large scale. Our extensive EST analysis of genes expressed in ginseng hairy root not only contributes to the understanding of the dynamics of genome expression patterns in root organ but also adds data to the repertoire of all genomic genes.
This study was conducted to investigate the characteristics of nonstarchy polysaccharides in Korean ginseng, (Panax ginseng C.A. Meyer). The results obtained are as follows. 1. The total sugar content of ginseng roots were decreased with increasing the cultural period. On the other hand, the crude fiber content was increased with that of the ginseng leaves or stems. But the crude fiber in root was much less than that of leaves and stems. 2. The dietary fiber content of ginseng root on 5 years old was 14.20% as neutral detergent fiber, 9.08% as acid detergent fiber, hemicellulose 5.12%, cellulose 7.98% and lignin of 1.10%, respectively. 3. Much more pectin was found in ginseng roots which was cultivated for shooter Period. And it was contained much more in the root than in the leaves and stems. 4. ginseng hemicellulose content in root was 5% to 10%. It was decreased with increasing: cultivated period. Hemicellulose was constituted of xylose, arabinose, glucose, rhamnose and xylose of these sugars was the predominant. 5. X-ray diffraction Pattern of ginseng cellulose showed maximum intensity at tile interplanar angle of 4.1$^{\circ}$.
Cultures of hairy root induced from ginseng(Panax C.A. Meyer) transformed with Agrobacterium rhizogenes (strain A4, ATCC 15834) were established and morphologically two different hairy root strains (HB1, HB2) were obtained. To determine the optimum growth rate, the hairy root (HB2) was cultured in various liquid medium supplemented with or without plant growth hormone. The growth rate of hairy root cultured on MS medium was 1.3-3.1 times higher than those cultured on other media, and the optimum sucrose concentration and pH were 3-6%, 5.5-6.5, respectively. Also, the growth rate of hairy root was increased when 0.02 M ammonium nitrate, 1.2 mM potassium phosphate (monobasic) and 0.5 mg/l IBA were supplied to liquid medium. The saponin patterns and contents of hairy root (HB2) were determined by TLC and HPLC. The crude saponin contents were 4.67% and the total saponin contents were 1.0%, on dry weight basis.
1. To know the site of saponin synthesis in this plant, 4-years old Panax ginseng C.A. Meyer was administered with 1, 2-l4C-acetate (Na salt, 10 ucilplant) by stem injection and was continued to grow for 3 weeks and the distribution of the radioactivity in leaf, stem and root part was identified. The percentage of radioactivity recovered was about 3.99%. 2. The sliced roots or leaf discs (2g) were bathed in the reaction mixture containing sugar, ATP, NADPH, and the distribution of the radioactivity of the fractions (sugar, saponin, sapogenin) was identified. 3. It seemed that major synthesized saponins in roots and leaves are dial and triol-type, respectively. Although both types of saponins are synthesized in roots, the main saponins seemed to be dial saponins and a significant portion of triol saponins are supplied from leaves through stem.
Calli and leaflets of ginseng (Panax ginseng C.A. Meyer) were cultured on 1/2MS media supplement. with kinetin, 2 iP, NAA, 2,4-D and IBA to assess their capacity to regenerate embryoids and organs. Root calli produced numerous embryoids and shoots in 1/2MS medium supplemented with 2 mg/l NAA and 2mg/s 2iP, and the combination of 2 iP and NAA was more effective than the combination of kinetin and NAA in induction of embryoid and shoot from root calli. Culture of leaflet in the medium supplemented with IBA resulted in profuse root regeneration.
