Studies on Enzyme Activities Responsible for Biosynthesis of Putrescine in Ginseng(Panax ginseng C.A. Meyer) (인삼에서 Putrescine 합성에 관련된 효소활성에 관한 연구)

  • Specific activities of ADC and ODC from 2-4 year old ones were higher than that from seedlings whereas those activities were not changed significantly from 2 to 4 years. Generally, activity of ADC was predorminant compared to that of ODC. Free arginine content in roots was much higher than that of leaves. And arginase specific activity from roots was higher than that of leaves. Cumulative results suggest that putrscine formation from ornithine in roots may be more effective than leaves and contribute to putrescine biosynthesis to some extract.

  • The seasonal variation in the activity of sucrose synthase, ADP-glucose pyrophosphorylase and UDP-glucose pyrophosphorylase in roots of Panax ginseng C.A.Meyer have been studied. It was revealed that sucrose synthase and ADP-glucose pyrophosphorylase are adaptive enzymes and can serve as markers of sink strength, while UDP-glucose pyrophosphorylase is the maintenance enzyme. The average day temperature exceeded 24。C appeared to cause the disturbance in refilling process, affecting the starch synthesis. Study on the dependence of oxygen consumption in stele tissue with temperature revealed the sharp accelerating of this process after 24。C.

  • This study was undertaken to examine the effects of fresh ginseng on the physicochemical and microbiological changes in dongchimi juice fermented under various conditions. The pH was somewhat lower in dongchimi juice added with 2.0% and 4.0% of fresh ginseng than that without ginseng, whereas the titratable acidity was higher in dongchimi juice with 2.0% and 4.0% of ginseng addition than the control. The addition of fresh ginseng to dongchimi preparation increased the numbers of total viable bacteria, lactic acid bacteria including Leuconostac mesenterotdes in dongchimi juice during fermentation. The changes in the counts of lactic acid bacteria were similar to those of total viable cells throughout the experiment except the initial stage of fermentation. However, the number of Leucosfastoc mesenternidgs decreased after the palatable stage. Key words : Panax ginseng C.A. Meyer, dongchlmi juice, pH, titratable acidity, microbiological changes.

Root-Rot Development of 2-Year old Ginseng (Panax ginseng C.A. Meyer) Caused by Cylindrocarpon destructans (Zinssm.) Scholten in the Continuous Cultivation Field (Cylindrocarpon destructans (Zinssm.) Scholten에 의한 연작지 2년근 인삼의 근부병 발병 특성)

  • The disease development of root-rot [pathogen:Cylindrocarpon destruction (Zinssm.) Scholten] occurred in 2-year old ginseng (Panax ginseng C.A. Meyer) was investigated in the continuous (the first cultivation: 1978∼1982, 2nd cultivation: 1990∼1993 and abolished in 1993 due to replanting problem) and replanted cultivation (period of cultivation: 1980∼1984). In the continuous cultivation, incidences of root-rot were 0.7% on May 3, 48.6% on May 24, and 95.8% on June 14, respectively. In the replanted cultivation, no symptom was observed on May 3 and disease incidence was generally lower compared with the continuous cultivation. On the aerial part of the ginseng infected by C. desiccates, the end and/or margin of leaves were changed to dark reddish color that appeared for behind the root-rot symptom. In this field, the longitudinal growth of lateral root was more inhibited than in the case of the replanted cultivation by C. destmctans. The inhibition rate of rootlet growth was 37.3% in the continuous cultivation as compared with that of replanted cultivation at June 14. Though lesions of root-rot were formed all over the roots, 61.2% of the lesions was positioned within 6 cm under the rhizome. Properties of Crude $\beta$-amylase from Korean ginseng, Panax ginseng C. A. Meyer (고려인삼(Panax Ginseng C. A. Meyer) 중의 조(組) $\beta$-amylase의 분리와 그 성질)

  • Curde $\beta$-amylase was prepared by frationation of the water extracts from Korean ginseng, Panax Ginseng C.A. Meyer, with 0.2-0.6 saturation of ammonium sulfate. The enzyme showed the typical properties of $\beta$-amylase, producing only maltase from starch. The enzyme preparation also showed no maltase activity. The enzyme was stable at the pH 5-9 and at the temperature below $40^{\circ}C$. The enzyme showed the optimum pH at 5.0 and the optimum temperature at $35^{\circ}C$. Its activities had proportional relations with substrate concentration below 12 mg%, showing Km V slues of 4.76 mg%. The enzyme was inhibited by $Ag^{+}$, $Hg^{++}$, $Cd^{++}$, $Cu^{++}$,$ Al^{3+}$, and $Fe^{3+}$.

  • Induction and culture of hairy roots from ginseng(Panax ginseng C. A. Meyer) roots discs by A. rhizogenes strain $A_4$ were studied. After 6-12 weeks infected with A. rhizogenes tumor and hairy roots emerged from the root discs. The ratio of hairy root induction on root discs was higher in 5-year old than in 3, 4, and 6-year old ginseng. On treatment with IAA, IBA, 2, 4-D and tryptophan, hairy roots formation showed a significant increase at 15-30mg/1 tryptophan treated. Subsequently, hairy roots were cultured on hormone-free RCM medium(pH 4.5, sucrose 30g/1).

  • Ginseng root explants and calli were cultured on modified Murashine and Skoog's media supplemented with different concentrations of organic or inorganic compounds and plant growth requlators to clarify the effects of chemical compositon and plant growth regulators in the medium on the growth of ginseng calli and the production of ginseng saponin. For optimum growth of ginseng calli, the concentrations of 2, 4-D and sucrose were in the range of 1 to 5 mg/l and 1 to 3%, respectively. And it was clarified that sucrose, nitrogen, phosphate, calcium, magnesium, plant growth regulators and their concentrations influcenced the relative biosynthesis of saponin in tissue cultures of Panax ginseng.

  • Crude invertase was obtained from the water extracts of Korean ginseng, Panax ginseng C. A. Meyer, by fractionation with $0.8{\sim}1.0$ saturation of ammonium sulfate. The properties of the crude invertase were as follows: Crude invertase was stable in the pH range between 5 and 9, and at the temperature below $35^{\circ}C$. Crude invertase showed the optimum pH at 5.0 and the optimum temperature at $50^{\circ}C$. The activity of the crude invertase was inhibited by $Ag^{+}\;Mn^{+}\;Hg^{+}\;Zn^{+},\;and\;Rb^{+}$, while $Ca^{+}\;Cu^{+},\;and\;Fe^{3+}$ demonstrated remarkable increasing effects on the enzyme activity.

  • Panax ginseng C.A. Meyer has been extensively used in the traditional oriental medicine as a restorative, tonic and prophylatic agent. This study was devised to develop a chemopreventive agent from panax ginseng to be able to suppress the genotoxicity and oxidative damage by ractive oxygen species, which are involved with cancer or aging. Ginseng petroleum ether extract (GPE) and one of its fraction, P2, showed an antioxidative effect on the lipid peroxidiphenyl-2-picryl hydrazil (DppH) radical generation. They also showed the suppressive effect of H2O2 or KO2 induced DNA damage by single cell gel electrophoresis (SCGE). Results from our study indicate that GPE and P2 are capable of protecting lipid peroxidation, and oxidative DNA damage. Therefore, GPE and P2 may be useful chempreventive agents which are involved with cancer and aging.

  • Solubility and degree of swelling of the starch were about 15% and 30%, respectively when the starch in agueous solution was heated at 90$^{\circ}C$ for 30minutes. The starch showed no singnificant differences in the degree of swelling and soblubility with a growing period of ginseng. The starch was begun gelatinize at 50-55$^{\circ}C$ and completed at 65-70$^{\circ}C$ by the amy tical methods of X-ray diffraction, disappearance of crystalinity. Brabender angly lographic analysis and amylase digestion test. The maximum and minimum peak viscosities of 10% starch solution were 5.500 B.U and 1960 B.U, respectively. The starch showed no significant differences in pasting temperature, maximum and minimum viscosities on amylogram with a growing period of ginseng. The intrinsic viscosities of the starch and amylose were 0.54 and 1.5-1.9, respectively.
