Effects of Soil Moisture and Planting Depth on the Growth of 2-year Old Ginseng Plant (Panax ginseng C. A. Meyer) (토양수분(土讓水分) 및 재식심도(栽植深度)가 저년근(低年根) 고려인삼(高麗人蔘)의 생육(生育)에 미치는 영향(影響))

  • This experiment was conducted to determine the effect of soil moisture content and planting depth on the growth of 2-year old ginseng plant. The results obtained are summarized as follows; 1. When the ginseng seedlings were planted in soil by 4 to 5cm in depth, the length of leaflet and stem and the number of branch roots were significantly decreased but the stem diameter was increased. 2. Highly significant quadratic regressions were shown between soil moisture content and the growth of the stem, leaf and root of the ginseng plant. 3. Estimated amount of soil moisture for the maximum growth of the stem was 75% of field capacity, and that for length and width of the leaflet was about 65 to 66% of field capacity. Estimated soil moisture for the maximum growth of the root was about 56 to 58% of field capacity and that for increase in root weight was about 60 to 61% of field capacity. 4. Estimated soil moisture content for best growth of ginseng roots was 1 to 5% lower when the seedling was planted in 3cm depth compared with 2cm in depth. And when the amount of soil moisture was 31% of field capacity, the deep planting was adequate for good root growth. 5. Significant correlations were resulted between the dry weight of roots and the leaf length, the leaf width and the dry weight of stem and leaves. And also, significant correlations were obtained between the ratio of root dry weight to root fresh weight and the stem length, the leaf length, the leaf width and the dry weight of stem and leaves.

  • 본 연구는 고품질 산양삼 생산을 위한 표준재배기술 개발을 위해 산양삼의 생육특성과 재배지의 입지환경 간의 상관관계를 구명하고자 하였다. 전국 9지역의 산양삼 재배지를 선정하여 13년생 산양삼의 생육특성을 조사하고, 각 재배지의 임상 및 토양 이화학성을 분석하여 입지환경과 산양삼 생육특성과의 상관관계를 확인하였다. 총 9개의 재배지 중에서 산양삼의 생육특성은 거창과 함양 재배지에서 다른 재배지에 비해 유의적으로 높은 것으로 나타났다. 임산과의 상관관계를 확인한 결과, 산양삼의 부피는 재배지 임상의 흉고직경과 유의적인 정의 상관관계를 보였고, 활엽수의 비율과 유의적인 부의 상관관계, 침엽수의 비율과는 유의적인 정의 상관관계를 보였다. 침엽수의 비율이 100%로 확인된 영월 재배지의 산양삼 생육특성이 다른 재배지에 비해 유의적으로 높지 않은 것으로 확인되어, 이러한 결과는 침활혼효림에서 침엽수의 비율과 상관관계가 있는 것으로 판단된다. 토양 특성과의 상관관계 분석에서는 재배지 토양의 pH, K, Ca, Mg과 생육특성 간의 유의적인 부의 상관관계를 확인할 수 있었다. 이와 같이 산양삼의 생육특성과 입지환경 간의 상관관계를 분석한 결과, 산양삼의 생육은 재배지 임상의 흉고직경이 크고, 침활혼효림에서 침엽수의 비율이 높으며, 토양의 pH, K, Ca, Mg 함량이 높지 않은 재배지에서 적합하다는 것을 확인할 수 있었다. 본 연구의 결과를 통해 산양삼 재배에 적합한 재배적지의 입지환경 조건을 확인할 수 있었고, 산양삼은 7~20년 동안 무농약, 무비료의 청정환경에서 생산하는 것이 원칙이기 때문에 본 연구의 결과를 활용하여 향후 산양삼의 예상재배지를 선정하는데 있어 도움을 줄 수 있을 것으로 사료된다.

  • 본 연구는 전국 임의의 산양삼 재배지를 선정하여 재배지 내의 토양 특성 및 토양세균군집을 분석하고, 토양 특성, 세균군집 및 산양삼 생육특성 간의 상관관계를 구명하기 위하여 수행되었다. 토양 이화학성 분석은 농촌진흥청의 종합분석실 매뉴얼에 따라 분석하였고, 토양세균군집 분석은 pyrosequencing analysis (Illumina platform)를 이용하였다. 토양세균군집과 생육특성 간의 상관관계는 Spearman's rank correlation을 이용하여 분석하였다. 전국 8개 산양삼 재배지로부터 분리한 토양세균군집은 2개의 cluster로 군집화를 이루는 것을 확인하였다. 모든 토양 샘플에서 Proteobacteria와 Alphaproteobacteria가 각각 평균 상대적 빈도수가 35.4%, 24.4%로 우점종으로 나타났다. 나타났다. 두 개의 cluster 간 토양세균군집의 상대적 빈도수를 비교 분석한 결과, 먼저 Proteobacteria (p = 0.03), Actinobacteria (p = 0.02), Ahlpaproteobacteria (p = 0.029), Betaproteobacteria (p = 0.021)는 cluster 1에서 cluster 2에 비해 상대적 빈도수가 유의적으로 높았고, Fimicutes (p = 0.004), Cyanobacteria (p = 0.004), Acidobacteriia (p = 0.041), Ktedonobacteria (p = 0.019), Gammaproteobacteria (p = 0.034), Bacilli (p = 0.009)은 cluster 2에서 유의적으로 상대적 빈도수가 높은 것으로 나타났다. 토양세균군집 cluster 간 산양삼의 생육특성을 비교 분석한 결과, cluster 2 재배지에서 수집한 산양삼 시료의 지하부 생중량은 cluster 1 재배지에서 수집한 산양삼 시료에 비해 cluster 2에서 유의적 (p = 0.04)으로 높았다. 산양삼 생육특성과 토양세균군집 간의 상관관계를 분석한 결과, 산양삼의 생육은 토양 pH가 낮고 Acidobacteria의 상대적 빈도수가 높은 토양에서 증가하였으며, Acidobacteriia와 Koribacteraceae의 상대적 빈도수는 산양삼의 생육과 유의적인 정의 상관관계를 보이는 것으로 나타났다. 본 연구 결과는 토양미생물군집과 산양삼 생육 간의 상관관계를 구명하는 중요한 자료가 될 것으로 생각되고, 나아가 산양삼 재배적지를 선정하는데 있어 보다 명확한 정보를 제공할 수 있을 것으로 사료된다.

  • 본 연구에서는 7년근과 13년근 산양삼을 선정하여 적변 유무에 따라 분류하고 적변에 따른 산양삼의 생육특성과 진세노사이드 함량을 조사하기 위해 수행되었다. 산양삼 재배지 9개소의 산양삼 지하부의 생육특성을 분석한 결과, 7년근과 13년근 산양삼의 생육특성은 적변에 따른 유의적인 차이는 확인되지 않았다. 적변에 따른 연근별 산양삼의 진세노사이드 함량을 비교 분석한 결과, 7년근 산양삼은 Rb1과 Rg1은 진세노사이드 함량이 일반삼에 비해 적변삼에서 높았고, 진세노사이드 Rc, Rd, Re, Rg2는 적변삼에 비해 일반삼에서 유의적으로 높은 것을 확인하였으나 적변에 따른 13년근 산양삼의 진세노사이드 함량 유의적인 차이는 확인할 수 없었다. 산양삼 생육특성과 진세노사이드 함량 간의 상관관계를 분석한 결과, 일반삼의 경우 진세노사이드 Rb2, Rc, Rd, Rf, Rg1이 산양삼의 지하부 길이와 유의적인 정의 상관관계를 보였고, 적변삼은 진세노사이드 Rd가 지하부 길이와는 유의적인 정의 상관관계, 주근직경과는 유의적인 부의 상관관계를 보였다. 본 연구에서는 7년근과 13년근 모두 적변에 따른 생육특성의 차이는 확인되지 않았고, 진세노사이드 함량은 7년근에서는 적변에 따라 유의적인 차이를 보였지만 13년근에서는 유의적인 차이가 없는 것을 확인하였다. 임가에서 산양삼은 7년근에 비해 13년근이 보다 고가에 거래되는 만큼 본 연구의 결과를 통해 적변삼에 대한 소비자들의 인식을 개선하여 산양삼 재배 농가의 소득 증대에 도움을 주고, 향후 산양삼의 품질규격을 정립하는 데 있어 유용한 정보를 제공할 수 있을 것으로 사료된다.

Mass Culture and Ginsenoside Production of Ginseng Hairy Root by Two-Step Culture Process (2계단 배양방법을 이용한 인삼 모상근의 대량배양과 Ginsenoside 생산)

  • A hairy root clone of Panax ginseng C.A. Meyer, HRB-15 was cultured iu various conditions with 3 L bubble type bioreactor to enhance both growth and ginsenoside production. The hairy roots were more rapidly grown under the dark condition than under the light condition. However, total amount of ginsenoside of hairy roots cultured under the light for 30 days increased 2 folds as compared with the dark condition and was 1.10% based on 6 ginsenosides. Especially, ginsenoside-Re was significantly increased and some ginsenosides except for ginsenoside-Re was slightly reduced. Also, the growth of hairy roots decreased about 30% as compared with the dark condition. In contrast, addition of sodium acetate led to decreased production of ginsenoside and growth of hairy roots under light condition. The influence of potassium dihydrogenphosphate concentration was examined in MS medium and a 1.25 mM concentration was found to be the most appropriate for growth and ginsenoside production under light condition. Two-step process of hairy roots culture with yeast elicitation or without ammonia in culture medium was developed to enhance growth and giusenoside synthesis. $50\;\mu\textrm{g}$ of yeast elicitor per g of fresh weight showed a synergistic effect on the ginsenoside synthesis of hairy roots on 20 days after culture. At that time, the content of total ginsenoside was 1.15%, while the growth of hairy roots decreased 21 % as compared with the dark condition. In addition, when elimination of ammonia on 20 days after culture, the content of total ginsenoside was 1.26% with significant increment of ginsenoside-Rd (0.27%) in addition to ginsenoside-Re and the growth of hairy roots decreased 10% as compared with the dark condition. In this system, we have demonstrated a unique two-step process of hairy root cultures to maximize biomass and secondary metabolites. It has found possibility to enhance ginsenosides production by growing hairy roots in this method.

Effect of Vinegar Made with Hydroponic-cultured Panax ginseng C. A. Meyer on Body Weight and Lipid Metabolism in High-Fat Diet-Fed Mice (수경재배 인삼으로 제조한 식초가 고지방 식이를 섭취한 마우스의 체중 및 지질대사에 미치는 영향)

  • The aim of this study was to develop hydroponic-cultured ginseng vinegar (HGV) containing ginsenoside Rg2 in order to its anti-obesity and anti-hyperlipidemic effects in C57BL/6J mice. HGV was prepared by two-stage fermentation. The ginsenoside Rg2 contained in acetic acid-fermented HGV increased by 4.0 times compared to that in pre-fermented HGV. To measure the anti-obesity effect of HGV, thirty two mice were randomly divided into four groups: normal diet group (ND), high-fat diet group (HFD), high-fat diet-supplemented with HGV group (HGV), and high-fat diet-supplemented with green tea extract group (GT). Body weight, fat weight, and liver weight decreased in the HGV group. The HGV group also showed lower plasma levels of low-density lipoprotein (LDL)-cholesterol and triglycerides, and higher levels of high-density lipoprotein (HDL)-cholesterol compared to the corresponding levels in the HFD group. Furthermore, there were significant decreases in plasma aspartase aminotransferase (AST) and alanine aminotransferase (ALT) levels in the HGV group compared to the corresponding levels in the HFD group. These results suggest that HGV can be used as an anti-obesity therapeutic agent or functional ingredient.

Memory Enhancing and Neuroprotective Effects of Selected Ginsenosides

  • The effects of ginsenosides Rg$_3$(R) , Rg$_3$(S) and Rg$_5$/Rk$_1$ (a mixture of Rg$_5$ and Rk$_1$ 1:1, w/w), which are components isolated from processed Panax ginseng C.A. Meyer (Araliaceae), on memory dysfunction were examined in mice using a passive avoidance test. The ginsenosides Rg3(R), Rg3(S) or Rg$_5$/Rk$_1$, when orally administered for 4 days, significantly ameliorated the memory impairment induced by the single oral administration of ethanol. The memory impairment induced by the intraperitoneal injection of scopolamine was also significantly recovered by ginsenosides Rg3(S) and Rg$_5$/Rk$_1$. Among the three ginsenosides tested in this study, Rg$_5$/Rk$_1$ enhanced the memory function of mice most effectively in both the ethanol­and scopolamine-induced amnesia models. Moreover, the latency period of the Rg$_5$/Rk$_1$­treated mice was 1.2 times longer than that of the control (no amnesia) group in both models, implying that Rg$_5$/Rk$_1$ may also exert beneficial effects in the normal brain. We also evaluated the effects of these ginsenosides on the excitotoxic and oxidative stress-induced neuronal cell damage in primary cultured rat cortical cells. The excitotoxicity induced by glutamate or N­methyl-D-aspartate (NMDA) was dramatically inhibited by the three ginsenosides. Rg$_3$(S) and Rg$_5$/Rk$_1$ exhibited a more potent inhibition of excitotoxicity than did Rg$_3$(R). In contrast, these ginsenosides were all ineffective against the H$_2$O$_2$- or xanthine/xanthine oxidase-induced oxidative neuronal damage. Taken together, these results indicate that ginsenosides Rg$_3$(S) and Rg$_5$/Rk$_1$ significantly reversed the memory dysfunction induced by ethanol or scopolamine, and their neuroprotective actions against excitotoxicity may be attributed to their memory enhancing effects.

  • In asthma, T helper 2 (TH2)-type cytokines such as interleukin (IL)-4, IL-5, and IL-13 are produced by activated $CD^{4+}$ T cells. Dendritic cells played an important role in determining the fate of naive T cells into either $T_H1$ or $T_H2$ cells. We determined whether RG-II regulates the $T_H1/T_H2$ immune response by using an ovalbumin-induced murine model of asthma. RG-II reduced IL-4 production but increased interferon-gamma production, and inhibited GATA-3 gene expression. RG-II also inhibited asthmatic reactions including an increase in the number of eosinophils in bronchoalveolar lavage fluid, an increase in inflammatory cell infiltration in lung tissues, airway luminal narrowing, and airway hyperresponsiveness. This study provides evidence that RG-II plays a critical role in ameliorating the pathogenic process of asthmatic inflammation in mice. These findings provide new insights into the immunotherapeutic role of RG-II in terms of its effects in a murine model of asthma.

The Effects of High Temperature High Pressure Steam Sterilization on Woohwangchungsimwon (고온고압증기멸균이 우황청심원에 미치는 영향)

  • Objectives To check marker content for appropriate quality control of Woohwangchungsimwon sterilized to ensure microbiological safety and to observe antioxidant activity for any changes in efficacy. Methods To measure any effects of sterilization on the effective compounds, 8 ingredients of Woohwangchungsimwon were screened for any changes in marker content using HPLC-DAD. Using the colorimetric method on the microplate reader any changes in total phenolic compound and flavonoid levels were observed. Antioxidant activity was measured using the DPPH, ABTS, and FRAP. Results Of the ingredients of Woohwangchungsimwon, 8 were subject to quantitative analysis before and after sterilization. 21.6 mg and 1.93 mg of Glycyrrhizin was found in Glycyrrhiza uralensis Fischer pre and post sterilization, respectively. Decursin found in Angelica gigas Nakai increased from 0.16 mg to 0.29 mg after sterilization. Bilirubin found in Gallstone of Bostaurusvar. domesticus increased from 0.24 mg to 0.33 mg. Cinnamic acid found in Cinnamomum cassia Blume increased from 0.02 mg to 0.05 mg. Ginsenoside Rb1 found in Panax ginseng C. A. Meyer decreased from 0.02 mg to 0.14 mg. Paeoniflorin found in Paeonia lactiflora Pallas increased from 1.05 mg to 1.13 mg. Amygdalin found in Armeniacae Amarum Semen increased from 2.68 mg to 2.83 mg. L-muscone found in Musk increased from 0.63 mg to 0.76 mg. As for total phenolic compound and total flavonoid content, there was a 1.22 and 4.15-fold increase. DPPH and ABTS increased by 20.45% and 20.69%, respectively. FRAP activity was 2.78 times more active post stabilization. Conclusions This study confirmed that high temperature high pressure steam sterilization, a method used to ensure microbiological safety of Woohwangchungsimwon, does not affect marker content; in other words, does not affect quality of the Woohwangchungsimwon. It could also be seen that total phenolic compound and flavonoid content increased after sterilization. An antioxidant activity test showed that there was significantly increased activity of antioxidants.

Experimental Studies on the Effect of Ginsenoside Rg1 Mixtures in an Isolated Rat Heart after Ischemic Arrest and Reperfusion (흰 쥐 적출 심장에서 비작업성 관류 회로를 이용한 인삼 성분 Ginsenoside Rg1 Mixtures의 심근 보호 효과에 관한 실험적 연구)

  • Panax Ginseng C.A. Meyer has been known for hundreds of years as the most valuable drug having mysterious effects among all the herbal medicines and plants in Korea. Also, many experimental studies have been performed recently that the various effects were identified and applied clinically. So we attempted an experimental study on the effect of ginsenoside Rg1 mixtures in an isolated rat heart with the use of the Langendorff model. The objective of this study was to determine whether this ginsenoside Rg1 mixtures would protect the myocardial injury after ischemic arrest and reperfusion. Isolated rat hearts were allowed to equilibrate for 20 minutes and were then subjected to 15 minutes of normothermic ischemia. After this ischemic period, isolated rat hearts were allowed to reperfusion for 10 minutes(Ischemic Group). In other group , isolated rat hearts were perfused for 60 minutes continuously with normothermia( Normothermic Group). Hemodynamic and biochemical parameters such as heart rate, left ventricular pressure, +dp/dt max, coronary blood flow and cardiac enzymes were measured during initial perfusion, ischemia, reperfusion period (Ischemic group) and 20, 40 and 60 minutes after continuous perfusion(Normothermic group). After completion of the experiment, this data was evaluated and the following results were obtained. 1. Heart rates showed an increase in both ischemic and normothermic experimental groups, but statistically significant differences were not identified. 2. LVP(Left Ventricular Pressure) showed statistically significant differences in both ischemic and normothermic experimental groups(p<0.005, p<0.01). 3. +dp/dt max showed statistically significant differences in both ischemic and normothermic experimental groups(p<0.01, p<0.01). 4. There were no statistically significant differences in coronary blood flow and cardiac cenzymes in all groups, but experimental groups seemed to have better protection and recovery. These results suggest that ginsenoside Rg1 mixtures has a protective effect on the myocardial injury after ischemia and reperfusion.
