  • Senegalimassilia sp. KGMB 04484 was isolated from fecal samples obtained from a healthy Korean. The whole-genome sequence of Senegalimassilia sp. KGMB 04484 was analyzed using the PacBio Sequel platform. The genome comprises a 2,748,041 bp chromosome with a G+C content of 61.18%, 2,300 total genes, 2,139 protein-coding gene, 21 rRNA genes, and 51 tRNA genes. Also, we found that strain KGMB 04484 had some genes for hydrolysis enzyme, fatty acid biosynthesis and metabolism in its genome based on the result of genome analysis. Those genes of KGMB 04484 may be related to regulation of human health and digest.

  • The genus of Bacteroides has been isolated from vertebrate animal feces. Bacteroides sp. KGMB 02408 was isolated from fecal samples obtained from a healthy Korean. The wholegenome sequence of Bacteroides sp. KGMB 02408 was analyzed using the PacBio Sequel platform. The genome comprises a 5,771,427 bp chromosome with a G + C content of 39.50%, 5,005 total genes, 18 rRNA genes, and 74 tRNA genes. Furthermore, we found that strain KGMB 02408 had some genes for oxidoreductases and menaquinone biosynthesis in its genome based on the result of genome analysis.

  • Effect of post-annealing treatment times at $200^{\circ}C$ on the peel strength of screen-printed Ag film/polyimide substrate were systematically investigated by $180^{\circ}$ peel test for thermal reliability assessment of printed interconnect. Initial peel strength around 16.7 gf/mm increased up to 29.4 gf/mm after annealing for 24hours, and then sharply decreased to 22.3, 3.6, 0.6, and 0.1 gf/mm after 48, 100, 250, and 500 hours, respectively. Ag-O-C chemical bonding as well as binder organic bridges formations seemed to be responsible for interfacial adhesion improvement after the initial annealing treatment, while excessive Cu oxide formation at Cu/Ag interface seems to be closely related to sharp decrease in peel strength for longer annealing times.

  • Present stadies were carried out to obtain fundermental data for effective management of the soils by investigating the physicochemical characteristics rubber fiber, humification grade and bearing power of peats and muck which were included in Geongdeog series, and Gimje series in Honam area. The results abtained were as follows; 1. Humification grade of peats were about 23, while that of muck soils was about 45-71 and those were higher in Gimje series than Bongnam series. 2. The organic tiers which humification grade was higher were high pH, bulk density, and contents of K, $P_2O_5$, B.S and while were low OM, T-C, T-N, Na, $SiO_2$ and rubbed fiber. 3. In peats and mucks, bulk density, pH, contents of T-N $P_2O_5$ and ash were negatively correlated with rubbed fiber and OM, CEC, T-C, PAC, C/N, C/P were positively correlated with one, while these physicochemical characteristics were oppositely correlated with humification grade. 4. Cone bearing power of each soil tiers was low in every Geongdeog series which had the thick organic tiers and showed poorly drainage and it was the highest in Gimje series which was similar to in organic soils because of its thin organic tiers.

  • To characterize thermo-chemical feature of sugar conversion of woody biomass, poplar wood ($Populus\;alba{\times}glandulosa$) powder was treated with supercritical water system. Supercritical water treatment (SCWT) was performed for 60 seconds at different temperatures (subcritical zone 350; supercritical zone $300,\;400,\;425^{\circ}C$) under two pressures $230{\pm}10atm$ as well as $330{\pm}10atm$, respectively, using flow type system. After separation of solid residues from SCWT products, the monomeric sugars in aqueous part converted from poplar wood powder were quantitatively determined by high performance anionic exchange chromatography [HPAEC] equipped with PAD detector and Carbo Pac PA10 column. As the temperature treated increased, the degradation of poplar wood powder was enhanced and ca 83% of woody biomass was dissolved into the water at $425^{\circ}C$. However, the pressure didn't help the degradation of biomass components. At subcritical temperature range, xylose was first formed by degradation of xylan, which is main hemicellulose component in hardwood species, while cellulose degradation started at the transition zone between sub and supercritical conditions and was remarkably accelerated at the supercritical temperature. In the supercritical water system the maximum yield of monomeric sugars amounts to ca. 7.3% based on oven dried wood weight at $425^{\circ}C$.

Genome Characteristics of Lactobacillus fermentum Strain JDFM216 for Application as Probiotic Bacteria

  • Lactobacillus fermentum strain JDFM216, isolated from a Korean infant feces sample, possesses the ability to enhance the longevity and immune response of a Caenorhabditis elegans host. To explore the characteristics of strain JDFM216 at the genetic level, we performed whole-genome sequencing using the PacBio system. The circular draft genome has a total length of 2,076,427 bp and a total of 2,682 encoding sequences were identified. Five phylogenetically featured genes possibly related to the longevity and immune response of the host were identified in L. fermentum strain JDFM216. These genes encode UDP-N-acetylglucosamine 1-carboxyvinyltransferase (E.C. 2.5.1.7), ErfK/YbiS/YcfS/YnhG family protein, site-specific recombinase XerD, homocysteine S-methyltransferase (E.C. 2.1.1.10), and aspartate-ammonia ligase (E.C. 6.3.1.1), which are involved in peptidoglycan synthesis and amino acid metabolism in the gut environment. Our findings on the genetic background of L. fermentum strain JDFM216 and its potential candidate genes for host longevity and immune response provide new insight for the application of this strain in the food industry as newly isolated functional probiotic.

  • Information technology is being applied to the development of ubiquitous healthcare system, which provides both efficient patient care and convenient treatment regardless of patient's location. However, the increasing number of users and medical information give rise to the problem of user management and the infringement of privacy. In order to address this problem we propose a user access scheme based on the RBAC (Role Based Access Control) model. The preceding trust management model for Grid security, FAS(Federation Agent Server), was analyzed and extended to provide supplementary functions for role-based access control in u-Healthcare system. The RBAC model provides efficient user management and access control, but very vulnerable in case when one with valid role tries to leak confidential inner medical information. In order to resolve this problem, a RBAC-WS (Work Status with RBAC) model has been additionally developed which allows only qualified staffs to access the system while on duty. Th proposed RBAC and RBAC-WS model have been merged together and applied to the PACS (Picture Archiving and Communication System).

  • Given a domain X and a collection H of functions h : X → {0, 1}, the Vapnik-Chervonenkis (VC) dimension of H measures its complexity in an appropriate sense. In particular, the fundamental theorem of statistical learning says that a hypothesis class with finite VC-dimension is PAC learnable. Recent work by Fitzpatrick, Wyman, the fourth and seventh named authors studied the VC-dimension of a natural family of functions ℋ'2t(E) : 𝔽2q → {0, 1}, corresponding to indicator functions of circles centered at points in a subset E ⊆ 𝔽2q. They showed that when |E| is large enough, the VC-dimension of ℋ'2t(E) is the same as in the case that E = 𝔽2q. We study a related hypothesis class, ℋdt(E), corresponding to intersections of spheres in 𝔽dq, and ask how large E ⊆ 𝔽dq needs to be to ensure the maximum possible VC-dimension. We resolve this problem in all dimensions, proving that whenever |E| ≥ Cdqd-1/(d-1) for d ≥ 3, the VC-dimension of ℋdt(E) is as large as possible. We get a slightly stronger result if d = 3: this result holds as long as |E| ≥ C3q7/3. Furthermore, when d = 2 the result holds when |E| ≥ C2q7/4.

Purification of the Glycosylated Polyphenol Oxidase from Potato Tuber

  • Glycosylated polyphenol oxidase was purified from potato tuber using ammonium sulfate fractionation, Sephadex G-100, and concanavalin A Sepharose column chromatography. Two or three types of polyphenol oxidase were separated on concanavalin A Sepharose. Type I and II polyphenol oxidases did not bind to concanavalin A Sepharose. Type I seemed to be an aggregated form of polyphenol oxidase. Type III polyphenol oxidase, which is presumed to be glycosylated because it was bound to concanavalin A Sepharose and eluted with $\alpha$-D-methyl glucopyranoside, was further purified by chromatography on Econo-Pac Q and Superose 12. Glycosylated polyphenol oxidase was purified 130-fold from the dissolved ammonium sulfate pellet resulting in about $6\;{\mu}g$ of the enzyme from 100 g of potato tuber periderm. The molecular weight of the glycosylated enzyme determined by SDS-polyacrylamide gel electrophoresis was about 64,000. Optimum temperature and pH of both II and type III potato polyphenol oxidases were $20^{\circ}C$ and pH 7.0, respectively. Glycosylated form of polyphenol oxidase (type III) preferred catechol to catechin as a substrate, whereas type II enzyme showed the reverse substrate preference.

A Basic Helix-Loop-Helix Transcription Factor Regulates Cell Elongation and Seed Germination

  • Plants are sessile and rely on a wide variety of growth hormones to adjust growth and development in response to internal and external stimuli. We have identified a gene, designated NAN, encoding a basic helix-loop-helix (bHLH) transcription factor that regulates cell elongation and seed germination in plants. NAN has an HLH motif in its C-terminal region but does not have any other discernible homologies to bHLH proteins. A bipartite nuclear localization signal is located close to the HLH motif. An Arabidopsis mutant, nan-1D, in which NAN is activated by the insertion of the 35S enhancer, exhibits growth retardation with short hypocotyls and curled leaves. It is also characterized by reduced seed germination and apical hook formation, symptomatic of GA deficiency or disrupted GA signaling. The phenotypic effects of nan-1D were increased by treatment with paclobutrazol (PAC), an inhibitor of gibberellic acid (GA) biosynthesis. NAN is constitutively expressed throughout the life cycle. Our observations indicate that NAN has a housekeeping role in plant growth and development, particularly in seed germination and cell elongation, and that it may modulate GA signaling.
