This study describes an efficient and widely applicable droplet-vitrification following cryopreservation for shoot tips of strawberry (Fragaria × ananassa Duch.) cvs. 'Wonkyo3114' and 'Gurumi40'. The shoot tips were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.5M). Precultured explants were osmoprotected with loading solution (LS, C4) containing 20% glycerol and 20% sucrose for 40 min and exposed to dehydration solution (B5) containing 40% glycerol and 40% sucrose for 40 min at 25℃, Subsequently, the explants were transferred onto droplets containing 2.5 μL PVS3 on sterilized aluminum foils (4 cm × 0.5 cm) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regrowth rate (%) in both the cultivars was obtained when the shoot tips were precultured with MS + 0.3M sucrose for 40 h at 25℃. The cryopreserved shoots tips exhibited 55% regrowth rate by culturing in NH4NO3-free MS medium supplemented with 3% sucrose, 1.0 g/L casein, 1.0 mg/L GA3, and 0.5 mg/L BA for 5 weeks and in MS medium supplemented with 0.5 mg/L GA3 for 8 weeks. This result shows that droplet-vitrification could be employed as a promising method for cryostorage of strawberry germplasm.
Autophagy is a process of cytoplasmic degradation of endogenous proteins and organelles. Although its primary role is protective, it can also contribute to cell death. Recently, autophagy was found to play a role in the activation of host defense against intracellular pathogens. The aims of our study was to investigate whether host cell autophagy influences Toxoplasma gondii proliferation and whether autophagy inhibitors modulate cell survival. HeLa cells were infected with T. gondii with and without rapamycin treatment to induce autophagy. Lactate dehydrogenase assays showed that cell death was extensive at 36-48 hr after infection in cells treated with T. gondii with or without rapamycin. The autophagic markers, LC3 II and Beclin 1, were strongly expressed at 18-24 hr after exposure as shown by Western blotting and RT-PCR. However, the subsequent T. gondii proliferation suppressed autophagy at 36 hr post-infection. Pre-treatment with the autophagy inhibitor, 3-methyladenine (3-MA), down-regulated LC3 II and Beclin 1. The latter was also down-regulated by calpeptin, a calpain inhibitor. Monodansyl cadaverine (MDC) staining detected numerous autophagic vacuoles (AVs) at 18 hr post-infection. Ultrastructural observations showed T. gondii proliferation in parasitophorous vacuoles (PVs) coinciding with a decline in the numbers of AVs by 18 hr. FACS analysis failed to confirm the presence of cell apoptosis after exposure to T. gondii and rapamycin. We concluded that T. gondii proliferation may inhibit host cell autophagy and has an impact on cell survival.

The shoot tips were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7M). Precultured explants were osmoprotected with loading solution (LS, C4) containing 17.5% glycerol and 17.5% sucrose for 40 min and exposed to dehydration solution (B1) containing 50% glycerol and 50% sucrose for 40 min at 25oC. Subsequently, the explants were transferred onto droplets containing 2.5 µL PVS3 on sterilized aluminum foils (4 cm× 0.5 cm) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regrowth rate (%) in both the cultivars was obtained when the shoot tips were precultured with 0.3M sucrose for 30 h + 0.5M sucrose for 16 h at 25oC. The cryopreserved shoots tips exhibited 57.8 % recovery rate by culturing in NH4NO3-free MS medium supplemented with 3% sucrose, 1.0 g/L casein, 1.0mg/L GA3, and 0.5 mg/L BA for 5 weeks and in MS medium supplemented with 0.5 mg/L GA3 for 8 weeks. Variation was not observed in both of ploidy analysis and morphological investigation on plantlets of two accessions cryopreserved under variable preculture conditions.

본 연구에서는 레트로 바이러스를 이용하여 형질전환 돼지를 생산하기 위한 기초 연구로써 유전자 도입 효율을 증진시키기 위해 sucrose와 poly-brene을 사용하여 그 효과를 조사하였다. 유전자(hGH) 전달체로는 vesicular stomatitis virus gly-coprotein pseudotyped retroviral vector (VSV-G)를 사용하였다. 체외에서 성숙된 돼지 난자는 매우 협소한 위란강내 공간을 가지며 따라서 위란강내 공간의 확장은 필수조건이다. sucrose 처리는 수정율및 배발달에는 영향을 미치지 않았으며 sucrose 처리를 통해 돼지난자는 위란강의 공간이 확장되어 충분한 양의 바이러스 벡터의 주입이 가능하였다. PCR 분석 결과 8.3 %의 유전자 도입율을 나타낸 무처리군에 비해 0.5, 1, 2, 3 % sucrose 처리군에서 39.3, 43.3, 35.7, 40.7 %의 높은 유전자 도입율을 나타내었다. 추가적으로 돼지난자는 sucrose 처리에 의해 다정자 침입의 억제 효과를 나타내었다. 일반적으로 레트로 바이러스를 이용한 세포에의 높은 유전자 도입 효과를 나타내는 것으로 알려진 polybrene을 바이러스와 공동 주입한 결과 0.5, 5, 50$\mu\textrm{g}$/$m\ell$의 농도에서 각각 56.5, 50.0, 57.1 %의 도입율을 나타냄으로써 바이러스 단독 주입군의 34.6%보다 높은 효율을 나타내었다. 그러나 50$\mu\textrm{g}$/$m\ell$의 농도를 주입하였을 경우, 분할율과 배반포발달율이 43.3 및 4.6%를 나타냄으로써 바이러스 단독주입군의 72.0, 17.3%보다 유의적으로 낮았다. 이상의 결과를 종합할 때, sucrose의 처리는 돼지 난자의 발달에 영향을 주지 않으며 유전자 도입효율을 증진시킬 수 있었으며, 추가적으로 다정자 침입억제효과를 보였다. 또한 polybrene의 사용은 낮은 농도에서 유전자 도입효율을 증진시켰다. 이러한 결과들은 형질 전환 돼지의 생산 가능성을 높이는데 이용될 수 있다고 사료된다.

The shoot tips of Chrysanthemum were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7 M). Precultured explants were treated with loading solution (LS, C6) containing glycerol 20% and sucrose 20% for 30 min and exposed to dehydration solution (B5) containing 40% of glycerol and 40% of sucrose for 60 min at $25^{\circ}C$, and then transferred onto droplets containing $2.5{\mu}l$ PVS3 on sterilized aluminum foils ($4cm{\times}0.5cm$) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regeneration rate (%) was obtained when shoot tips were precultured with treatment-2 (exposing of shoot tips to MS + 0.3M sucrose for 30 h and then treated with MS+0.5 M sucrose for 16 h) at $25^{\circ}C$ in both the cultivars. The viability of cooled samples, followed by culturing on $NH_4NO_3$-free MS medium for first 5 days was increased to two-fold (80.7%) regrowth rate over those cultured on normal MS medium or MS medium containing plant growth regulators. This result shows droplet-vitrification would be a promising method for cryobanking chrysanthemum germplasm.

Potatoes are the world's 4th major food crop after maize, rice, and wheat and also are a staple food for 1.3 billion people. Due to their wide adaptability to various environmental conditions, their yeild capacity, and high commercial value, potatoes have contributed to global food security. Many potato germplasms are commonly preserved as whole plants in fields or in storage to maintain their particular genetic combinations. However, field maintenance is expensive and has the risk of potential losses from diseases, pests, plant ageing and climate change. Over the past four decades, meaningful efforts have been made toward the safe long-term conservation of potatoes through cryopreservation methods such as droplet-vitrification. In this study, we tested 4 Korean potato varieties('Golden Egg', 'Golden Ball', 'Ja-Young' and 'Ha-Ryeong') with the modified potato droplet -vitrification protocol. Potato shoot tips are precultured in a sucrose-enriched medium(0.3 and 0.7M for 7 and 17hrs, respectively) and submitted to a loading step with C4 solution for osmoprotection. The treated explants were dehydrated with Plant Vitrification Solution(PVS)2 which is 80% A3 solution in ice for 30 minutes. Thawing and unloading steps were performed with 0.8M sucrose solution for 30 sec(40℃) followed by 30min(25℃, room temperature). In a potato post-culture medium(MS+0.1 mg·L-1 GA3+0.1 mg·L-1 kinetin), we obtained a survival rates of post-thawed explants ranging 16.1-82.2%. The results suggest that modified and optimized protocols are required dependinig on every cultivar, genetic and ecological types. To achieve higher survival and regeneration rates, each step within the cryoprocedure must be carefully optimized.
In this study, cryopreservation by droplet-vitrification was applied to pear (Pyrus spp.) germplasm. We focused on the development and assessment of various strategies for the selection of suitable tissue, osmoprotection, and dehydration. We also evaluated post-thaw recovery of cryopreserved explants by droplet-vitrification. Preferentially, we tested the effects of preculture and loading treatments to determine which tissues were more suitable, either the apical shoot tips or the axillary buds. Apical shoot tips showed the better regrowth rate than in vitro axillary buds. The most effective techniques for cryopreservation were as follows. Shoots from in vitro seedlings which had been cultured for about 5-6 weeks were cold-hardened at 4℃ for one week, excised shoot tips were precultured on liquid MS medium including 0.3 M sucrose for 31 hours and 0.7 M sucrose for 17 hours, osmoprotected in loading solution (LS) for 40 min, and then cryoprotected in dehydration solution (PVS3) for 90 min. In addition, we found that regrowth rates of explants on regrowth medium after exposure to liquid nitrogen (LN) were higher than those on MS medium. Results indicated that the highest regrowth percentage was 95.6% for 'Bartlett' cultivar and 68.9% for 'BaeYun No.3' cultivar. Consequently, apical shoot tips of two pear cultivars, 'Bartlett' (P. communis) and 'BaeYun No.3' (P. pyrifolia), were successfully cryopreserved by droplet-vitrification. Results of this study show that the enhanced droplet-vitrification method described in the present study could be used as an effective means for long-term storage of pear genetic resources.

The highest regeneration rate (%) was obtained when shoot tips were precultured with treatment-2 (exposing of shoot tips to MS + 0.3M Sucrose for 30 h and then treated with MS+0.5 M sucrose for 16 h) at $25^{\circ}C$ in both the cultivars. The viability of cooled samples, followed by culturing on MS medium for 4 weeks was 77.8% and 60.0% for 'Derunoka' and 'Jumbo pure berry', respectively. This result shows droplet-vitrification would be a promising method for cryobanking strawberry germplasm.
The impact of midlatitude synoptic system (upper-level trough) on typhoon intensity change was investigated by analyzing the spatial and temporal characteristics of vertical wind shear (VWS), relative eddy momentum flux convergence (REFC), and potential vorticity (PV). These variables were computed over the radial mean $300{\sim}1,000km$ from the typhoon center by using GDAPS (Global Data Assimilation and Prediction System) data provided by the Korea Meteorological Administration (KMA). The selected cases in this study are typhoons Rusa (0215) and Maemi (0314), causing much damage in life and property in Korea. Results show that the threshold value of VWS indicating typhoon intensity change (typhoon to severe tropical storm) is approximately 15 m/s and of REFC ranges 6 to 6.5 $ms^{-1}day^{-1}$ in both cases, respectively. During the period with the intensity of typhoon class, PVs with 3 to 3.5 PVU are present in 360K surface-PV field in the cases. In addition, there is a time-lag of 24 hours between central pressure of typhoon and minimum value of VWS, meaning that the midlatitude upper-level trough interacts with the edge of typhoon with a horizontal distance less than 2,000 km between trough and typhoon. That is, strong midlatitude upper-level divergence above the edge of the typhoon provides a good condition for strengthening the vertical circulation associated with the typhoons. In particular, when the distance between typhoon and midlatitude upper-level trough is less than 1,000 km, the typhoons tend to weaken to STS (Severe Tropical Storm). It might be mentioned that midlatitude synoptic system affects the intensity change of typhoons Rusa (0215) and Maemi (0314) while they moves northward. Thus, these variables are useful for diagnosing the intensity change of typhoon approaching to the Korean peninsula.
숙성기간에 따른 감식초의 휘발성 성분을 알아보기 위하여 3-25년간 숙성한 감식초를 SPME-GC-MS로 분석하였다. 숙성기간에 따라 휘발성 성분 profile은 차이가 있었지만 22년 숙성한 감식초(PV-22)와 25년 숙성한 감식초(PV-25)는 휘발성 성분의 종류 및 농도가 매우 유사하였다. 총 90종의 휘발성 성분이 동정되었으며, 전반적으로 산류와 에스터류의 함량이 높았으며 그 다음으로 케톤류와 알코올류의 함량이 높았다. 산류 중 acetic acid는 숙성기간에 따라 8,925.0-26,132.3 ng/mL 범위로 검출되었다. 에스터류 중 ethyl acetate가 32.5-50,681.7 ng/mL 범위로 존재하였고, 알코올류 중 ethanol은 3년 숙성한 감식초(PV-3)에서만 9,578.5 ng/mL 농도로 존재하였다. 숙성기간에 따른 감식초의 휘발성 성분을 비교해 보면, 숙성기간이 짧았던 감식초 (PV-3)에서 휘발성 성분의 함량이 가장 높았는데 이 중 acetic acid, ethanol과 ethyl acetate의 함량이 휘발성 성분의 대부분을 차지하였다. 이와 대조적으로 숙성기간이 길었던 감식초(PV-15, PV-22와 PV-25)에서는 ethanol이 검출되지 않았으며, ethyl acetate도 낮은 함량으로 검출되었다. 휘발성 성분의 조성을 살펴보면 비교적 숙성기간이 길었던 감식초(PV-15, PV-22와 PV-25)에서는 산류의 함량이 가장 높았으며 그 다음으로 에스터류와 케톤류의 함량이 높았다. 이와 달리 숙성기간이 짧았던 감식초(PV-3)에서는 에스터류의 함량이 가장 높았으며 그 다음으로 산류와 알코올류의 함량이 높았다. 숙성기간이 증가할수록 산류, 알코올류와 에스터류의 함량이 감소한 반면, 알데하이드류와 케톤류의 함량은 증가하였다.
