The spectrum of 5p deletion in Korean 20 patients with Cri du chat syndrome (한국인 묘성증후군 20명 환자에서의 5p 결실 양상 분석)

  • Purpose : Cri-du-Chat syndrome (CdCs) is a rare but clinically recongnizable condition with an estimated incidence of 1:50,000 live births. The clinical characteristics of the syndrome include severe psychomotor and mental retardation, microcephaly, hypertelorism, hypotonia, and slow growth. Also the size of the chromosome 5p deletion ranges were known from the region 5p13 to the terminal region. In this study, we report the spectrum of 5p deletion in Korean 20 pts. with CdCs and genotype-phenotype associations in CdCs. Methods : In order to delineate genotype-phenotype correlation, molecular cytogenetic studies including GTG banding and clinical characterization were performed on Korean 20 pts with CdCs including parents. CGH array and Fluorescence in situ hybridization (FISH) analysis were used to confirm a terminal deletion karyotype and map more precisely the location of the deletion breakpoint. Results : Molecular analysis of the spectrum of 5p deletion revealed 9 pts (45%) with a del (5)(p14), 7 pts. (35%) a del (5)(p13), 3 pts. (15%) a del (5)(p15.1) and 1 pt. (5%) a del (5)(p15.2) in 20 pts with CdCs. 4(20%)pts were identified to have additional chromosome abnormalites of deficiency and duplication involving chromosomes of 6, 8, 18, & 22. Parental study identified 3 familial case (2 paternal and 1 maternal origin) showing parents being a balanced translocation carrier. And the comparison study of the deletion break points among these 20 pts. with their phenotype has showed the varying clinical pheno-types in the CdCs critical region. Conclusion : The characterization of 5p deletion including parental study may help to delineate the genotypephenotype correlation in CdCs. Also these molecular cytogenetic analyses will be able to offer better information for accurate genetic diagnosis in CdCs and further make possible useful genetic counseling in pts. and family.

Nucleotide Sequence and Characterization of ptsG Gene Encoding Glucose-specific Enzyme II of Phosphotransferase System from Brevibacterium flavum

  • Nucleotide sequence of Brevibacterium flavum ptsG gene capable of complementing Escherichia coli ZSC113 mutations defective to glucose permease activity of phosphotransferase system was completely determined, and the gene product was compared with other glucose-specific enzyme II ($EII^{Glc}$). A ptsG gene of B. flavum consisted of open reading frame of 2,025 nucleotides putatively encoding polypeptide of 675 amino acid residues and TAA stop codon. Deduced amino acid sequence of B. flavum ($EII^{Glc}$) had high homology with ($EIIs^{Glc}$) of Corynebacterium glutamicum, C. efficiens, and B. lactofermentum. Arrangement of structural domains, IIBCA, of B. flanum ($EII^{Glc}$) protein was identical to that of EIIs belonging to glucose-phosphotransferase system.

  • The purpose of this study is to research and develop conducting polymer(CP) composite electrode for supercapacitor. Supercapactior cell of CP composite electrode with 1M LiClO$_4$/PC brings out good capacitor performance below 4V. The radius of semicircle of CP composite cell with PAn composite electrode adding 15Wt% SP270(PAnS15) and PT composite electrode adding 50%wt% SP270(PTS50) was absolutely small. The total resistance of supercapacitor cell mainly depended on internal resistance of the electrode. The discharge capacitance of supercapacitor cell with PTS50(+)/PAnS15(-) in 1st and 20th cycles was 38F/g and 28F/g at current density of 1mA/$\textrm{cm}^2$, respectively.

Comparison of vessel failure probabilities during PTS for Korean nuclear power plants

  • Plant-specific analyses of 5 types of domestic reactors in Korea are performed to assure the structural integrity of the reactor pressure vessel (RPV) during transients which are expected to initiate pressurized thermal shock (PTS) events. The failure probability of the RPV due to PTS is obtained by performing probabilistic fracture mechanics analysis. The through-wall cracking frequency is calculated and compared to the acceptance criterion. Considering the fluence at the end of life expected by surveillance test, the sufficient safety margin is expected for the structural integrity of all reactor pressure vessels except for the oldest one during the pressurized thermal shock events. If the flaw with aspect ratio of 1/12 is considered to eliminate the conservatism, the acceptance criteria is not exceeded for all plants until the fluence level of $8{\times}10^{19}\;n/cm^2$, generating sufficient margin beyond the design life.

  • Fracture mechanics is one of the major areas of the pressurized thermal shock (PTS) evaluation. To evaluate the reactor pressure vessel integrity associated with PTS, PFM methodology demands precise calculation of temperature, stress, and stress intensity factor for the variety of PTS transients. However, the existence of stainless steel cladding, with different thermal, physical, and mechanical property, at the inner surface of reactor pressure vessel complicates the fracture mechanics analysis. In this paper, treatment schemes to evaluate stress and resulting stress intensity factor for RPV with stainless steel clad are introduced. For a reference transient, the effects of clad thermal conductivity and thermal expansion coefficients on deterministic fracture mechanics analysis are examined.

Dong-ku1 Taejon 300-100, Korea - The nucleotide sequence of approximately 2.4 kb immediately adjacent to ptsG gene coding for the glucose permease of Brevibacterium ammoniagenes was detennined. A putative open reading frame (ORP) of 1.467 nucleotides encoding a polypeptide of 489 amino acid residues and a TAA stop codon was identified. The deduced amino acid sequence of the ORF product has a high homology with the 30S ribosomal protein S 1 of Mycohacteriwn tuberculosis (83 % ). M leprae (74%), Streptomyces coelicola (77%), and Escherichia coli (40%). suggesting that the predicted product of ORF is a ribosomal protein S 1. The ORF is located at a distance of 266 nucleotides upstream from ptsC gene with a same translational direction.

Characterization of ptsHI Operon from Leuconostoc mesenteroides SY1, a Strain Isolated from Kimchi

  • The ptsHI operon from Leuconostoc mesenteroides ssp. mesenteroides SY1 (L. mesenteroides SY1), a strain isolated from kimchi, was cloned and characterized. The ptsH open reading frame (ORF) was 273 bp in size, which can encode a protein of 90 amino acid residues with a molecular weight of 9,212 Da. The pfsI ORF was 1,719 bp in size, which was capable of encoding a protein of 572 amino acids with a molecular mass of 62,549 Da. ptsH and pfsI genes were transcribed as a single transcript of 2.0 kb in size regardless of carbon sources, supporting the operon structure. Although the deduced amino acid sequences of the HPr and EI were highly homologous with those of other Gram-positive bacteria, an additional amino acid (glutamine at the $3^{rd}$ amino acid) was present in HPr from L. mesenteroides SY1. Phosphorylation sites of HPr included the histidine residue ($16^{th}$) and serine residue ($47^{th}$). Mutant HPrs, in which each phosphorylation site was mutated into alanine, were obtained, and phosphorylation with HPr and mutated HPrs showed that HPr was phosphorylated at the serine residue ($47^{th}$) by HPr kinaseiphosphorylase (HPr K/P).

Influence of Ginseng Saponins on the Isolated Aortic Contractile Response of the Spontaneously Hypertensive Rat

  • The present study was attempted to investigate the effects of total ginseng saponin (G75), panaxadiol-type (PDS) and panaxatriol-type saponin (PTS) on contractile responses of vasoconstrictors in aortic smooth muscle stripes of normotensive (NR) and spontaneous hypertensive rats (SHR). Phenylephrine (an adrenergic $\alpha$$\_$1/-receptor agonist) and high potassium (a membrane depolarizing agent) caused greatly contractile responses in both NR and AHR aorta, respectively. Phenylephrine- and high potassium-induced contractile responses were greater in NA than those in SHR aortic smooth muscle stripes. In NR, the contractile responses of high potassium (5.6$\times$10$\^$-2/ M) were not affected in the presence of GTS (300 $\mu$g/ml), PDS (300 $\mu$g/ml), and PTS (300 $\mu$g/ml), respectively whereas phenylephrine (10$\^$-6/ M)-induced contractile responses were markedly inhibited. In SHR, the contractile responses of high potassium (5.6$\times$10$\^$-2/ M) were not affected in the presence of GTS (300 $\mu$g/ml), PDS (300 $\mu$g/ml), and moderate doses of PTS (150-300 $\mu$g/ml), respectively but greatly blocked by high concentration of PTS (600 $\mu$g/ml). Phenylephrine (10$\^$-6/ M)-induced contractile responses were inhibited in a dose dependent fashion (150-600 $\mu$g/ml) by the pretreatment with PTS while not altered in the presence of GTS (300 $\mu$g/ml) and PDS (300 $\mu$g/ml), respectively. Taken together, these experimental results suggest that ginseng saponins cause vascular relaxation through blockade of adrenergic $\alpha$$\_$1/-receptors and some unknown mechanisms, and that there is some difference in sensitivity of vascular smooth muscle between NR and SHR in responses to ginseng saponins. It seems that panaxatriol type of some ginseng saponins has the greatest potency in vascular relaxation.

A New PAPR Reduction Method in the OFDM System using GD and Radix-2 DIF IFFT (OFDM 시스템에서의 GD방식과 Radix-2 DIF IFFT를 이용한 효과적인 PAPR 감소 방식)

  • Many methods have been developed to overcome the PAPR(peak-to-average power ratio) problem. Selective mapping(SLM), partial transmit sequence(PTS), subblock phase weighting(SPW) and gradient descent(GD) are used widely to reduce the PAPR. In this paper, we present an effective PAPR reduction method that decreases the number of calculations through Radix-2 DIF IFFT procedure and GD method that transmits selected data sequence. The data sequence is constructed by choosing elements that satisfy threshold value as one part of the sequence and the rest elements of each sequence are chosen to have the lower papr operating, which yields performance improvement.

Testing the Reliability of a Smartphone-Based Travel Survey: An Experiment in Seoul (스마트폰 기반 통행 행태 조사 자료 신뢰성 검증: 서울에서 수집된 자료를 바탕으로)

  • With programmable applications that utilize sensors, such as global positioning systems and accelerometers, smartphones provide an unprecedented opportunity to collect behavioral data in an unobtrusive and cost-effective manner. This paper assesses the relative accuracy and reliability of the Future Mobility Sensing (FMS), a smartphone-based prompted-recall travel survey. We compared the data extracted from FMS with the data collected from the Korea Passenger Trip Survey (PTS), a traditional self-reported, paper-based travel survey. In total, 46 undergraduate students completed the PTS for seven consecutive days, while also carrying their smartphones with the activated FMS applications for the same time span. After completing the PTS, the participants validated their FMS data on the web-based prompted recall surveys. We then matched the validated FMS data with the PTS-based records. The FMS turns out to be superior in detecting short trips, which are usually under-reported in self-reported travel surveys. The reported PTS travel times are longer than for the FMS, suggesting that participants tend to overestimate their travel time in the PTS. This study contributes to the ongoing development of smartphone-based travel behavior data collecting methods.
