To increase thennotolerance of forage crops, transgenic rice plants as a model for transformation of monocots were generated. A cDNA encoding the chloroplast-localized small heat shock protein (small HSP) of rice, Oshsp21, was introduced into rice plants via Agrobacterium-mediated gene transfer system. Calli induced from scutella were co-cultivated with a A. tumefaciens strain EHAlOl canying a plasmid, pIGhsp21. A large number of transgenic plants were regenerated on a medium containing hygromycin. Integration of Oshsp2l gene was confirmed by PCR and Southern blot analyses with genomic DNA. Northern blot and immunoblot analyses revealed that the Oshsp21 gene was constitutively expressed and accumulated as mature protein in transgenic plants. Effects of constitutive expression of the OshspZl on thermotolerance were first probed with the chlorophyll fluorescence. Results indicate that inactivation of electron transport reactions in photosystem I1 (PSII), were mitigated by constitutive expression of the Oshsp21. These results suggest that the chloroplast small HSP plays an important role in protecting photosynthetic machinery during heat stress. (Key words : Thermotolerance, Rice, Transgenic, cDNA)
Forest-grown American ginseng (Panax quinquefolius L.) is exposed to daily and seasonal light variations. Our goal was to determine the effect of understory light changes on the maximum quantum yield of photosystem II, expressed as $F_v/F_m$, and photosynthetic pigment composition of two-year-old plants. Understory light photon flux density and sunfleck durations were characterized using hemispherical canopy photography. Our results showed that understory light significantly affected the $F_v/F_m$ of American ginseng, especially during the initial development of the plants when light levels were the highest, averaging 28 mol $m^{-2}d^{-1}$. Associated with low $F_v/F_m$ during its initial development, American ginseng had the lowest levels of epoxidation state of the xanthophyll cycle of the season, suggesting an active dissipation of excess light energy absorbed by the chlorophyll pigments. As photon flux density decreased after the deployment of the forest canopy to less than 10 mol $m^{-2}d^{-1}$, chlorophyll a/b decreased suggesting a greater investment in light harvesting pigments to reaction centers in order to absorb the fleeting light energy.
The psbC gene, encoding the intrinsic chlorophyll-binding protein of CP43, one of the PS core complex polypeptides, was cloned from the Panax ginseng chloroplast, which is composed of 1,422 nucleotides and the overall nucleotide sequence shows more than 84% identity to those of eukaryotic photosynthetic organisms. The predicted topology of CP43, based on hydropathy analysis, includes six membrane-spanning ${\alpha}-helices$ resulting in three lumenal and four stromal loops. The putative translation start codon for the psbC gene is located at 48 nucleotides upstream from the stop codon of the psbD gene whose product is also a component of the PSII reaction center, implying that the promoter of the psbC gene is possibly located in the middle of the structural gene of the psbD gene. Northern blot analysis of the in vivo accumulation of the psbC transcript from the plants grown under the various growth light intensities (5%, 10%, 20%, and 100%) of daylight indicated that the steady-state level of the psbC transcript was not significantly affected by light intensity.
Plants dissipate excess excitation energy from their photosynthetic apparatus by a process called non-photochemical quenching (NPQ). The major part of NPQ is energy dependent quenching (qE) which is dependent on the thylakoid pH and regulated by xanthophyll cycle carotenoids associated with photosystem (PS) II of higher plants. The acidification of the lumen leads to protonation and thus conformational change of light harvesting complex (LHC) proteins as well as PsbS protein of PSII, which results in the induction of qE. Although physiological importance of qE has been well established, the mechanistic understanding is rather insufficient. However, recent finding of crystal structure of LHCII trimer and identification of qE mutants in higher plants and algae enrich and sharpen our understanding of this process. This review summarizes our current knowledge on the qE mechanism. The nature of quenching sites and components involved in this process, and their contribution and interaction for the generation of qE appeared in the proposed models for the qE mechanism are discussed.

Cucumber leaves subjected to light chilling stress exhibit a preferential inactivation of photosystem(PS) I relative to PSII, resulting in the photoinhibition of photosynthesis. In light chilled cucumber leaves, Cu/Zn-Superoxide dismutase(SOD) is regarded as a primary target of the light chilling stress and its inactivation is closely related to the increased production of reactive oxygen species. In the present study, we further explored that inactivation of PSI in cucumber leaves is not a light chilling specific, but general to various oxidative stresses. Oxidative stress in cucumber leaves was induced by treatment of methylviologen(MV), a producer of reactive oxygen species in chloroplasts. MV treatment decreased the maximal photosynthetic O$_2$ evolution, resulting in the photoinhibition of photosynthesis. The photoinhibition of photosynthesis was attributable to the decline in PSI functionality determined in vivo by monitoring absorption changes around 820 nm. In addition, MV treatment inactivated both antioxidant enzymes Cu-Zn-superoxide dismutase and ascorbate peroxidase known sensitive to reactive oxygen species. From these results, we suggest that chloroplast antioxidant enzymes are the primary targets of photooxidative stress, followed by subsequent inactivation of PSI.
The productivities of Chlorella ovalis and Dunaliella parva were influenced by the rates of medium compositions obtained from the fermented animal wastewater (BM: bacteria mineral water) including a natural substitute chelator for EDTA (etylenediaminetetraacetic acid). The most favorable medium was -E+50 adding 50% BM in f/2 medium instead of EDTA, a chemical chelator, which increased more 19-fold of cell density in C. ovalis and 7-fold in D. parva than cells cultured on f/2 medium as well as the enhancements of chlorophyll a (f/2-E: 0.26 g L–1, -E+50: 1.5 g L–1 in C. ovalis; f/2-E: 2.7 g L–1, -E+50: 15 g L–1 in D. parva) and the increase of maximal PSII quantum yields. These results were verified that the BM could play an important part as a natural chelator substituted for EDTA. In the fields of biotechnology, food organisms in fishery and eco-industries of CO2 sequestration in air and nutrient removal in water, the natural chelator of BM could be applied to enhance the biomass of the other microalgae.

SCM415 alloy was implanted with nitrogen ions using plasma source ion implantation (PSII), at a dose range of $1{\times}10^{17}\;to\;6{\times}10^{17}\;N^+cm^{-2}$ Auger electron spectrometry (AES) was used to investigate the depth profile of the implanted layer. Friction and wear tests were carried out on a block-on-ring wear tester. Scanning electron microscopy (SEM) was used to observe the micro-morphology of the worn surface. The results revealed that after being implanted with nitrogen ions, the frictional coefficient of the surface layer decreased, and the wear resistance increased with the nitrogen dose. The tribological mechanism was mainly adhesive, and the adhesive wear tended to become weaker oxidative wear with the increase in the nitrogen dose. The effects were mainly attributed to the formation of a hard nitride precipitate and a supersaturated solid solution of nitrogen in the surface layer.
Dunaliella salina and Dunaliella bardawil are well known for carotenogenesis, the overproduction of carotenoids, under stress conditions. The effect of high light (HL) and low light (LL) on the growth, morphology, photosynthetic efficiency, and the ${\beta}$-carotene and zeaxanthin production of D. salina CCAP 19/18 and D. bardawil was investigated and compared. Both strains showed similar growth kinetics under LL growth condition, but D. salina CCAP 19/18 was faster. As the light intensity increased, D. salina CCAP 19/18 cells were elongated and D. bardawil cells became larger. Both strains showed decrease of the maximum quantum yield of PSII ($F_v/F_m$) and election transport rate (ETR) under HL growth condition and D. salina CCAP 19/18 was less liable to the light stress. Both strains had about 1.8 and 5 times difference in the $O_2$ evolution rate at LL and HL conditions, respectively. The ${\beta}$-carotene and zeaxanthin production were increased as the light intensity increased in both strains. D. bardawil was more sensitive to light intensity than D. salina CCAP 19/18. The possible application of D. salina CCAP 19/18 as a carotenogenic strain will be discussed.
Energy is vital to global prosperity, yet dependence on fossil fuels as our primary energy source contributes to global climate change environmental degradation, and health problems. Hydrogen $(H_2)$ offers tremendous potential as a clean renewable energy currency. Hydrogen has the highest gravimetric energy density of any known fuel and is compatible with electrochemical and combustion processes for energy conversion without producing carbon-based emission that contribute to environmental pollution and climate change. Numerous methodologies have been developed for effective hydrogen production. Among them, the biological hydrogen production has gained attention, because hydrogen can be produced by cellular metabolismunder the presence of water and sunlight. The green alga Chlamydomonas reinhardtii is capable of sustained $H_2$ photoproduction when grown under sulfur deprived condition. Under sulfur deprived conditions, PSII and photosynthetic $O_2$ evolution are inactivated, resulting in shift from aerobic to anaerobic condition in the culture. After anaerobiosis, sulfur deprived algal cells induce a reversible hydrogenase and start to evolve $H_2$ gas in the light. According to above principle, we investigated the effect of induction parameters such as cell age, cell density. light intensity, and sulfate concentration under sulfur deprived condition We also developed continuous hydrogen production system by sulfate re-addition under sulfur deprived condition.
