Preparation of the Fermented Product by Lactic Acid Bacteria from Cheese whey (치즈훼이를 이용한 유산균 발효제품 제조)

  • The optimum conditions for the development of a lactic acid beverage from the concentrated whey were studied using reverse osmosis system. For lactose hydrolysis rate and acid productivity, the strain mixture of Streptococcus thermophilus and Lactobacillus bulgaricus was more efficient than that of Streptococcus cremoris and Streptococcus lactis. The titratable acidity was increased at higher LCR (lactose concentration ratio) of whey. However, the higher LCR of whey was, the slower the pH decreasing rate was. The amount of sediment was maximum at LCR of 1.0 : 1 whey, hit there was no sediment at LCR 3.0 : 1 whey after 12 hours. Propylene glycol alginate was the best stabilizer and prevented from sedimentation at the concentation of less than 0.1%. Aspartame as a sweetener of yoghurt flavor had the best palatability.

  • This paper describes characteristics of solid-state $NaBH_4$ hydrogen generation and supply system for fuel cell UAV. Flow rate and pressure of the generated hydrogen were dramatically changed during $NaBH_4$ decomposition using acid. Hydrogen supply was stabilized by a self-pressurized reactor, and hydrogen stabilization method was introduced. For hydrogen generation in below zero-temperature, hydrochloric acid was diluted by propylene glycol-water mixtures. Solid-state $NaBH_4$hydrogen generation and supply system was designed. Basic operation experiments was performed to reveal the characteristics of this hydrogen generation system.

  • The production of sorbitol by permeabilized cells of Zymomonas mobilis immobilized in $\kappa$-carrageenan was investigated. Cetyltrimethylammoniumbromide (CTAB) permeabilized cells were treated with glutaraldehyde prior to immobilization for cross-linking of enzymes, glucose-fructose oxidoreductase (GFOR) in cells. Rigidity of the immobilized beads was increased two-fold with 90$\%$ conversion efficiency by the additions of 40$\%$ (w/v) polyols (glycerol 25 g + propylene glycol 15 g) to 60$\%$ (w/v) distilled water containing 2.5$\%$ (w/v) $\kappa$-carrageenan as a final concentration, prior to immobilization. $\kappa$-Carrageenan beads entrapping permeabilized cells were dried to improve bead rigidity and storage stability. During s6mi-batch process for 72 h with dry beads, there was an improvement of the loss of enzyme activity (less than 10$\%$). In batch process, the kinetic results of $K_m.fructose$ value for the free cells, wet beads and dry $\kappa$-carrageenan beads were 71.7, 72.4 and 116.7 g/l, respectively. Higher productivity was obtained with two-stage continuous packed bed reactors with both wet and dry $\kappa$-carrageenan beads at 25.00 and 21.15 g/l/h, respectively, when measured at second stage.

Rat Skin Permeation of Diclofenac and its Prodrugs (디클로페낙 프로드럭들의 흰쥐 피부 투과)

  • Various alkyl ester prodrugs of diclofenac were synthesized in order to investigate the relationship between their skin permeation characteristics and physicochemical properties. Solubility in various vehicles was measured at room temperature. 1-Octanol/water partition coefficients (Log P) and capacity factors (k') were measured to determine the lipophilicity of the prodrugs. Stability of prodrugs in the skin extract and homogenate was also investigated before conducting the skin permeation studies. Increases in the Log P and capacity factor values were observed when alkyl esters of diclofenac were prepared. Since the aqueous solubility of the prodrugs was not high enough, they were saturated in propylene glycol (PG) for skin permeation studies. Prodrugs were rapidly metabolized to diclofenac, both in skin homogenate and in dermal extract of skin. The skin permeation rate of alkyl ester prodrugs was significantly higher than diclofenac with shorter lag time. Moreover, a parabolic relationship was observed between the permeation rate and the log P values of prodrugs, and the maximum flux was achieved at a log P value of around 4.0.

The Effect of Enhancers on the Penetration of Clenbuterol through Hairless Mouse Skin (클렌부테롤의 피부투과에 미치는 경피흡수촉진제의 영향)

  • Clenbuterol, a selective ${\beta}_2-adrenergic$ receptor stimulant, has been introduced as a potent bronchodilator for patients with bronchial asthma, chronic obstructive bronchial disease, chronic bronchitis and pulmonary emphysema. The percutaneous permeation of clenbuterol was investigated in hairless mouse skin after application of 50/50 buffer(pH 10)/propylene glycol solvent mixture. The enhancing effects of various penetration enhancers such as terpenes, non-ionic surfactants, pyrrolidones, fatty acids and some other enhancers on the permeation of clenbuterol were evaluated using Franz diffusion cell. Among terpenes studied, 1,8-cineole was the most effective enhancer, which increased the permeability of clenbuterol approximately 39.33-fold compared with the control without penetration enhancer, followed by menthone with enhancement ratio of 23.57. Nonionic surfactants did not have significant enhancing effects. N-Lauryl-2-pyrrolidone increased the permeability of clenbuterol approximately 4.51-fold compared with the control. Lauric acid increased the permeability of clenbuterol approximately 35.57-fold with decreasing the lag time from 2.64 to 0.52 hr. Oleic acid, linoleic acid, linolenic acid and capric acid showed enhancement ratio of 22.62, 19.60, 17.45 and 16.51, respectively. $Labrafil^{\circledR}$ enhanced the permeability of clenbuterol 9.24-fold compared with that without enhancer.

EXTEMPORANEOUS MICELLAR SOLUBILIZATION OF TITRATED EXTRACT OF CENTELLA ASIATICA IN AQUEOUS MEDIA

  • Titrated Extract of Centella asiatica (TECA) is a poorly water-soluble extract from the Centella asiatica. Despite excellent wound preparation causes pain due to a low aqueous solubility and high hypertonicity and therefore, the patient's compliance of this drug is low. The objective of this study is to design a formulation of TECA with an improved therapeutic applicability via an adequate solubilization. A mixture of propylene glycol and ethoxylated hydrogenated caster oil achieved an acceptable solubilization of TECA (i.e. 10%) via a formulation of micelle. A preparation of extemporaneous TECA micelle was achieved by a dilution of the original micellar formulation with either water or saline. An estimated distribution of particle size was between 15.9 and 32.6 ㎜. The osmotic pressure of the formulation was found to be much lower than that found In a commercially available injectable (i.e. Madecassole). The erthrocytic hemolysis of micellar solution was lower than that with the conventional dosage form, consistent with the osmotic pressure data. Pain score after an injection of the micellar solution was assessed by the hind-paw writhing test using ICR mice and compared with that found with the conventional injectable. The data indicated that the injection of the micellar solution was a significantly less painful. These results indicated that a micellar solubilization, followed by an extemporaneous dilution, is a novel formulation of TECA with an improved therapeutic applicability.

The Whitening Effect of Angelicae dahuricae Radix Water Extract in Brown Guinea Pigs (Brown guinea pig에서 백지 열수추출물의 미백효과)

  • The purpose of this study was to investigate the whitening effect of Angelicae dahuricae Radix water extract (ADWE) by its application on artificial tanning spots which were induced by 1,500 $mJ/cm^2$ of UVB radiation on the backs of brown guinea pigs weighing approximately 450~500 g. Thirty ${\mu}l$ of ADWE, at each application, were applied twice a day, 5 days a week, for 8 weeks to the guinea pigs. The artificial tanning spots were divided into 3 groups which were vehicle control group [propylene glycol: ethanol: water (5:3:2)], positive control group (2% hydroquinone) and experimental group (2% ADWE). The visible whitening effect and changes in melanin index were evaluated once a week. On completion of the experiment, the animals were sacrificed under anesthetization, and the artificial tanning spots were excised by biopsy punch and stained with Hematoxylin and Eosin (H&E) to observe histological change and also stained with Fontana-Masson's silver (F-M) and S-100 to observe melanin pigmentation and melanocytes. In the gross observation, the experimental group showed higher pigmentation than the positive control group but lower pigmentation than the vehicle control group. Histological observation confirmed that ADWE had a positive whitening effect by showing a lower distribution of melanin and melanocytes in the epidermis of experimental group than in the vehicle control group.

Increase of Permanent Wave Efficacy and Decrease of Hair Damage by using Enhancer of Permanent Wave Lotion (흡수촉진제를 이용한 펌제의 웨이브 효율 증가 및 모발손상 억제)

  • Human hair could be damaged by various physicochemical conditions and treatment. Permanent and decoloring treatment were the most serious factor on hair damage. The new permanent wave lotion containing Permeation enhancers such as Cremophor EL, Transcutol and propylene glycol based on cysteine permanent wave lotion were prepared. Efficiency of permanent wave and hair damage following pH of permanent wave lotion and addition of permeation enhancer were investigated. PH of solution, wave efficiency, loss of protein from hair, morphology of hair by SEM and solubility of alkaline solution were evaluated. The addition of Cremophor EL and Transcutol with ethanol increased permanent wave efficacy and decreased hair damage effectively. They diminished permanent wave lotion's pH and augmented permanent wave lotion's penetration compare to cysteine permanent wave lotion. new permanent wave lotion containing permeation enhancers such as Cremophor EL could be a good candidate for a new permanent wave lotion.

  • Because omeprazole(OMP) is very unstable in aqueous condition, $OMP-{\beta}-CD$, the inclusion complex of OMP and ${\beta}-cyclodextrin({\beta}-CD)$ was made and physicochemical properties of it were compared with those of OMP. Skin permeability of OMP and $OMP-{\beta}-CD$ in propylene glycol vehicle and the reciprocal action of ${\beta}-CD$ with various enhancers were examined through hairless mouse. Adhesive patches were prepared with polyisobutylene and the skin permeability and stability of OMP were investigated. The inclusion complex showed higher solubility and lower partition coefficient than OMP itself. DMSO, 1-methyl 2-pyrrolidone and sodium cholate had an enhancing effect. However ethanol and polysorbate 80 hardly showed the enhancing effect of OMP. When sodium lauryl sulfate and sodium cholate as enhancer were added in patch, the former case showed higher permeability of OMP.

Controlled Release Properties of Ketoprofen from Methacrylate Polymer Gels (메타크릴레이트 폴리머로 제조한 겔 제제로부터 케토프로펜의 제어 방출특성)

  • Hydrogels containing ketoprofen were prepared by adding NaOH or $Ca(OH)_2$ solution to Eudragit L, S and Eudispert hv at various concentration. And xerogels were prepared by drying hydrogels. On the other hand, organogels containing ketoprofen were prepared by mixing Eudragit L or S and propylene glycol. Effects of polymer content and base on drug release were investigated using KP V dissolution method. The release rate of ketoprofen from Eudragit L & S hydrogel decreased with increasing in polymer content. And the drug release rate from cal. hydroxide based gels were more decreased than that from sod. hydroxide based gels. At pH 7.2 dissolution medium, e release of ketoprofen from Edispert hv hydrogel followed apparent zero order kinetics. The release of ketoprofen from xerogel involved in simultaneous absorption of water and desorption of ketoprofen via a pH-dependant swelling controlled mechanism. The release of ketoprofen from Eudragit S organogels followed apparent zero order kinetics, providing strong evidence for a surface erosion mechanism.
