Studies on the Improvement of Precision in Early Pregnancy Diagnosis by Milk Progesterone Test(EIA) in Dairy Cows (Milk Progesterone Test(EIA)에 의한 소의 임신조기판단 정확도 향상에 관한 연구)

  • These experiment was carried out to improve the precision of early pregnancy diagnosis in dairy cattle. Changes in progesterone concentration of milk were measured by Enzyme Immunoassay(EIA) in 73 cows up to 21 days after insemination. The average concentraton of progesterone in milk was 1.9ng/ml at eatrus ; it increased to 17.8ng/ml on day 14, and thereafter it declined to 4.3ng/ml on day 21 in nonpregnant cows. Whereas in pregnant animals, it was maintained and elevated further to 22.2ng/ml on day 21. The accuracy of the pregnancy diagnosis for animals classified as positive (pregnant ; over 10ng/ml and decreasing rate<1.5) and negative (non-pregnant ; under 7ng/ml and decreasing rate>1.5) were 95% and 100% respectively. The samples ranging between 7ng/ml and 10ng/ml were classified as positive (decreasing rate<1.5) and negative(>1.5), which accuracy was 54.5% and 100% respectively. However this range appears to be of the most interest for the veterinary practitioner since cows in proestrus or early interestrus tend to have milk progesterone levels within this value. Causes for the insufficient precision of false pregnancy diagnosis are discussed.

Spatio-temporal Expression and Regulation of Dermatopontin in the Early Pregnant Mouse Uterus

  • During endometrial differentiation the extracellular matrix (ECM) changes dramatically to prepare for implantation of the embryo. However, the genes regulating the ECM build-up in the uterine endometrium during early pregnancy are not well known. Using the PCR-select cDNA subtraction method, dermatopontin was identified in the uterus of a pregnant mouse on day 4 of gestation. Dermatopontin mRNA increased dramatically on day 3, and was at its highest level at the time of implantation. Administration of RU 486 significantly inhibited mRNA expression by day 4 of gestation, but ICI 182,780 did not. Progesterone markedly induced dermatopontin expression in ovariectomized uteri within 4 h of administration, whereas estrogen had little effect. In silico analysis revealed progesterone receptor binding sites in the dermatopontin promoter region. Decidualization did not induce expression of dermatopontin; instead dermatopontin mRNA became strongly localized at the interimplantation site. In situ hybridization revealed that expression gradually decreased in the luminal epithelial cells as pregnancy progressed, whereas it increased in the stromal cells. The pattern of localization and the changes of intensity of dermatopontin mRNA coincided with those of collagen. Collectively, these results strongly suggest that dermatopontin expression is steroid-dependent. They also suggest that, at the time of implantation, dermatopontin expression is primarily regulated spatio-temporally by progesterone via progesterone receptors, and is modulated by the decidual response during implantation. Dermatopontin may be one of the regulators used to remodel the uterine ECM for pregnancy.

Effects of Administration of GnRH Alone and Concomitant with PMSG on the Fetus in Pregnant Rats (GnRH를 단독으로 또는 PMSG와 동시에 투여했을 때 임신 랫드의 태아에 미치는 영향)

  • The effect of GnRH alone and concomitant with PMSG on the prevention of implantation. termination of pregnancy, and concentration of plasma progesterone were studied in pregnant rats. GnRH 50, 100 or 200 ug alone and concomitant with PMSG 25 or 50 IU were administered once on day 2 or 9 of gestation, respectively. Rats were autopsied on days 7 or 20. Administration of GnRH on day 2 did not result in the prevention of implantation and termination of pregnancy but resulted in termination of pregnancy administering on day 9. Administration of GnRM concomitant with PMSG on day 2 or 9 resulted in prevention of implantation and termination of pregnancy, but injection of GnRH 50 ug concomitant with PMSG 25 IU on day 9 had only one live fetus. Administration of GnRH alone and concomitant with PMSG on day 2 had no effect on the concentration of plasma progesterone determining on day 7. Administration of GnRH concomitant with PMSG on day 2 resulted in decrease of progesterone level determining on day 20 but GnRH alone was normal level. Administration of GnRH alone and concomitant with PMSG on day 9 resulted in decrease of the concentration of progesterone but was normal concentration administering GnRH 50 ug concomitant with PMSG 25 IU.

  • Rhizopus nigricans produces 11.alpha.-hydroxyprogesternoe with a unidentified byproduct, which is hardly separated. Results of chromatography, IR and NMR spectroscopy identified the byproduct to be 11.alpha.-hydroxy-allopregnane-3,20-dione. R. nigricans was found to transform progesternoe into a monoform intermediate, 11.alpha.-hydroxyprogesterone, from which 11.alpha.-hydroxy-allopregnane-3,20-dione and 6.betha., 11.alpha. - dihydroxyprogesterone were formed respectively by 5.alpha.-reduction and 6.betha.-hydroxylation.

  • Progesterone level in the seminal plasma and serum were measured by radioimmunoassay. The average level of seminal plasma progesterone was 0.76$\pm$0.15 ng/ml, similar to that of serum: 0.64$\pm$0.08 ng/ml. Progesteron levels in the seminal plasma were higher in the breeding season than in the nonbreeding season.

  • A study was undertaken to determine the incidence of short oestrus cycles and associated serum progesterone levels in Nili-Ravi buffaloes. Forty lactating buffaloes were kept under observation for oestrus activity for a period of sixty days. Blood samples from jugular vein from twenty buffaloes were used to determine associated serum progesterone levels during short oestrus cycles. Incidence of short oestrus cycles was 15.5 percent (Nine out of fifty eight oestrus cycles) and most of the short oestrus cycles occurred between first and second oestrus after calving. Lengths of short oestrus cycles ranged from 6 to 14 days. The most frequent length was 11 days. Serum progesterone levels were lower at oestrus and increased gradually attaining peak levels of 1.3 and 1.8 ng/ml on 7th day of 9 and 11 day short oestrus cycles. One buffalos exhibiting a short oestrus cycle of 12 days conceived which indicated that ovulation took place at the oestrus of short cycle.

  • The effect of supplementary feeding on postpartum ovarian activity, reproductive performance and plasma progesterone concentrations in Cheju native cows was studies. The group with adequate nutrition had greater mean body weight than did the underfed group before and after calving. First ovulation occurred 19.5 days and first oestrus at 37.8 days postpartum in the standard group, however, there was delay in first ovulation and oestrus in the restricted group (73.4 days and 111.8 days). Progesterone concentraitons of full fed animals gradually increased from 6 weeks(0.5ng/ml) to 12 weeks(3.3ng/ml) after calving. However, progesterone concentrations of underfed animals had a slight increase in 10 weeks after dalving(0.80ng/ml).

  • The purpose of this study was to measure the serum content of progesterone, estradiol 17${\beta}$ and cortisol. Blood samples collected from day 20 prepartum to day 20 postpartum in 8 sows. Progesterone and estradiol-17${\beta}$ were assayed by radioimmunoassay methods and cortisol was determined by competitive protein-binding methods. Progesterone levels began to decline on day -4, reached 1.9ng/ml by day +2 and remained quite constant thereafter. Progesterone levels remained fairly constant(18.4 to 20.0 ng/ml) from 20 to 6 days before parturition. Estrdiol-17${\beta}$ increased from 205 pg/ml at day 6 prepartum to 425 pg/ml at the time of parturition. Cortisol reached a peak level of 86.5 ng/ml at day 0.

  • Serum progesterone concentrations of pregnant and non-pregnant Hanwoo were compared to evaluate the possibility of early pregnancy prediction. Twenty five female Hanwoo were devided into two groups. Eighteen heads were injected with 980$\mu\textrm{g}$ tiaprost trometamol salt(Iliren:PGF2$\alpha$ analogue) for estrous synchronization and fifteen heads of them were inseminated artifically and seven heads were non-treated. Blood samples were collected from the jugular vein before PGF2$\alpha$ injection and on day 0, 18, 21 after estrous induction. Pregnancy of cattle was confirmed by rectal palpaton after 60 days of artificial insemination. The serum progesterone concentrations at heat showed no significant differences between pregnant and non-pregnant while on day 18 and 21 after estrous induction showed significant differences(p<0.05). The accuracy of pregnancy diagnosis on day 18 and 21 after estrous induction was 85.7 and 71.4% respectively while that of non-pregnancy were 71.4 and 100.0% respectively.

  • The effects of estradiol-17$\beta$ and progesterone on the spontaneous motility of pig oviductal isthmic smooth muscle were investigated. The motility of the Isolated smooth muscle was recorded by using physiological recording system. The results were summarized as follows; 1. The amplitude and frequency of spontaneous motility in pig isthmic smooth muscle were 0.251$\pm$0.023 g and 15.380$\pm$0.935/min in follicular stage, and 0.201$\pm$0.027g and 14.520$\pm$1.382/min in luteal stage. 2. The spontaneous motility of pig isthmic smooth muscle was excited by estradiol-17$\beta$ but was not by progesterone in follicular and luteal stage.
