Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. This study aims at developing a new seismic resistant method by using precast concrete wall panels for existing low-rise, reinforced concrete beam-column buildings such as school buildings. Three quasi-static hysteresis loading tests were performed on one unreinforced beam-column specimen and two reinforced specimens with U-type precast wall panels. Top shear connection of the PC panel was required to show the composite strength of RC column and PC wall panel. However, the strength of the connection did not influence directly on the ultimate loading capacities of the specimens in the positive loading because the loaded RC column push the side of PC wall panel and it moved horizontally before the shear connector receive the concentrated shear force in the positive loading process. Under the positive loading sequence(push loading), the reinforced concrete column and PC panel showed flexural strength which is larger than 97% of the composite section because of the rigid binding at the top of precast panel. Similar load-deformation relationship and ultimated horizontal load capacities were shown in the test of PR1-LA and PR1-LP specimens because they have same section dimension and detail at the flexural critical section. An average of 4.7 times increase in the positive maximum loading(average 967kN) and 2.7 times increase in the negative maximum loading(average 592.5kN) had resulted from the test of seismic resistant specimens with anchored and welded steel plate connections than that of unreinforced beam-column specimen. The maximum drift ratios were also shown between 1.0% and 1.4%.

This study tackles the development of an improved detail of partially restrained CFT square column-to-beam connection and the evaluation of its mechanical behavior under monotonic loading. The connection is designed to strengthen shearing capacity at the bottom of the connection due to the ultimate behavior of PR-CC by its detail of the bottom connection and simplify the fabrication process. The suggested connection is the welded bottom beam flange connection(M-2) and is compared with the existing PR-CC of bolted seat angle connection(M-1). Two specimens were fabricated in actual size and tested under monotonic loading. Based on the test results, the welded bottom beam flange connection exhibited about 85% of the stiffness of steel beam. It was similar to the bolted seat angle connection and behaved as PR-CC. The specimen of the supposed connection type failed at the shear connection of web but was similar to the bolted seat angle connection until the failure. It obtained sufficient stiffness and capacity through the reinforcingsteel and the capacity and deformational ability equivalent to the full-plastic moment through the anchor inside the steel tube at the web connection. So, it can be said that the suggested connection exhibits sufficient ductile behavior.

A sophisticated reliability analysis method is proposed to evaluate the reliability of real nonlinear complicated dynamic structural systems excited by short duration dynamic loadings like earthquake motions by intelligently integrating the response surface method, the finite element method, the first-order reliability method, and the iterative linear interpolation scheme. The method explicitly considers all major sources of nonlinearity and uncertainty in the load and resistance-related random variables. The unique feature of the technique is that the seismic loading is applied in the time domain, providing an alternative to the classical random vibration approach. The four-parameter Richard model is used to represent the flexibility of connections of real steel frames. Uncertainties in the Richard parameters are also incorporated in the algorithm. The laterally flexible steel frame is then reinforced with reinforced concrete shear walls. The stiffness degradation of shear walls after cracking is also considered. The applicability of the method to estimate the reliability of real structures is demonstrated by considering three examples; a laterally flexible steel frame with fully restrained connections, the same steel frame with partially restrained connections with different rigidities, and a steel frame reinforced with concrete shear walls.

Background: Risk of developing breast cancer increases with short breastfeeding and the use of hormones. The prognosis of breast cancer is better if the tumours are hormone receptor positive. Since breast feeding affects estrogen and progesterone receptors, we wanted to investigate how such reproductive factors as breastfeeding and the use of hormones interact with known prognostic markers and specific tumour characteristics in women with breast cancer. Materials and Methods: A total of 250 women treated for breast cancer from a larger cohort completed a questionnaire on breastfeeding, number and age at births and use of hormones. A logistic regression analysis was made to search for connections between known prognostic markers on the one hand (type of cancer, grade, tumor size, estrogen receptor and progesterone receptor, lymphovascular invasion and DNA-ploidy) and reproductive data, breastfeeding, and hormone use on the other. Results and Conclusions: Hormone use, but not breastfeeding, was significantly associated, also on multivariate analysis, with the prognostic variable lymphovascular invasion, connected to a worse prognosis. No other hormone use or breast feeding correlations with prognostic variables were found.

Seismic resistant test of anchored and welded steel plate connections manifested an average of 2.8 times increase in the maximum loading (average 591.8 kN) in comparison to unreinforced beam-column specimen. The maximum drift ratios were also shown between 1.4% and 2.7%. An analytical study was performed while assuming the RC column on the right side and the vertical element of the reinforced PC panel to behave in completely composite manner and the RC column on the left side and PC panel to behave in completely non-composite manner when loading was exerted from upper right end of RC frame of specimen to its left side. It was found with the assumptions that the overall flexural behavior in principle agreed with the experimental result.
Background: Aberrant promoter hypermethylation has been recognized in human breast carcinogenesis as a frequent molecular alteration associated with the loss of expression of a number of key regulatory genes and may serve as a biomarker. The E-cadherin gene (CDH1), mapping at chromosome 16q22, is an intercellular adhesion molecule in epithelial cells, which plays an important role in establishing and maintaining intercellular connections. The aim of our study was to assess the methylation pattern of CDH1 and to correlate it with the expression of E-cadherin, clinicopathological parameters and hormone receptor status in breast cancer patients of Kashmir. Materials and Methods: Methylation specific PCR (MSP) was used to determine the methylation status of CDH1 in 128 invasive ductal carcinomas (IDCs) paired with the corresponding normal tissue samples. Immunohistochemistry was used to study the expression of E-cadherin, ER and PR. Results: CDH1 hypermethylation was detected in 57.8% of cases and 14.8% of normal adjacent controls. Reduced levels of E-cadherin protein were observed in 71.9% of our samples. Loss of E-cadherin expression was significantly associated with the CDH1 promoter region methylation (p<0.05, OR=3.48, CI: 1.55-7.79). Hypermethylation of CDH1 was significantly associated with age at diagnosis (p=0.030), tumor size (p=0.008), tumor grade (p=0.024) and rate of node positivity or metastasis (p=0.043). Conclusions: Our preliminary findings suggest that abnormal CDH1 methylation occurs in high frequencies in infiltrating breast cancers associated with a decrease in E-cadherin expression. We found significant differences in tumor-related CDH1 gene methylation patterns relevant to tumor grade, tumor size, nodal involvement and age at diagnosis of breast tumors, which could be extended in future to provide diagnostic and prognostic information.
This paper analyses the decision process of the Gilhoe Line construction of the Manchurian Railway and consignment process of the North Korea Line to the South Manchuria Railways as part of the 12-year Chosun railway plan. It was found that these two events were connected and could be explained by Japan's consistent continental policy, which had expanded its territory toward China. This paper explicates Japan's railroad policy and the characteristics of the Chosun railway. Moreover, evidence to identify connections among Japan's national railway, the Manchurian railway, and the Chosun railway has been scrutinized. One territory policy between Chosun and Manchuria is also examined. Comparison of new resources on the Gilhoe Line construction of the Manchurian railway and the 12-year plan of the Chosun railway, and their correlations, is expounded. This paper elucidates that the Gilhoe Line construction and the North Korea line consignment operation by the South Manchuria Railways had the intention of obtaining Japanese military and economic supremacy over East Asia.
