Development of Gene-based Markers for the Allelic Selection of the Restorer-of-fertility Gene, Rfo, in Radish (Raphanus sativus)

  • Cytoplasmic male sterility (CMS) and fertility restoration have been utilized as valuable tools for $F_1$-hybrid seed production in many crops despite laborious breeding processes. Molecular markers for the selection of CMS-related genes help reduce the expenses and breeding times. A previously reported genomic region containing the Ppr-B gene, which is responsible for restoration of fertility and corresponds to the Rfo locus, was used to develop gene-based or so-called "functional" markers for allelic selection of the restorer-of-fertility gene (Rfo) in $F_1$-hybrid breeding of radish (Raphanus sativus L.) Polymorphic sequences among Rfo alleles of diverse breeding lines of radish were examined by sequencing the Ppr-B alleles. However, presence of Ppr-B homolog, designated as Ppr-D, interferes on specific PCR amplification of Ppr-B in certain breeding lines. The organization of Ppr-D, resolved by genome walking, revealed extended homology with Ppr-B even in the promoter region. Interestingly, PCR amplification of Ppr-D was repeatedly unsuccessful in certain breeding lines implying the lack of Ppr-D in these radishes. Ppr-B could only be successfully amplified for analysis through designing primers based on the sequences unique to Ppr-B that exclude interference from Ppr-D gene. Four variants of Rfo alleles were identified from 20 breeding lines. A combination of three molecular markers was developed in order to genotype the Rfo locus based on polymorphisms among four different variants. These markers will be useful in facilitating $F_1$-hybrid cultivar development in radish.

  • To achieve rapid developments and cost savings in manufacturing industries including automotive die shops, new paradigm and its supporting systems of information managements through total product life cycle are needed for concurrent and collaborative engineering. For manufacturing of automotive press dies, integrated and efficient managements of PPR information including product, manufacturing process and resource are essential. In this paper, we introduce a PLM approach to achieve engineering collaborations in product development and production of automotive dies. To prove concepts and benefits of PPR information managements, we implement new business workflow and detail procedures, PPR information management system and other related applications. By PPR information managements in PLM, improvements in quality of engineering results and savings in time from design to production of dies are possible.

Evaluating the role of vaccine to combat peste des petits ruminants outbreaks in endemic disease situation

  • Among the main intimidation to the sheep and goat population, PPR outbreaks are causing huge losses especially in endemic areas. During recent times, six outbreaks of PPR were confirmed at semi-organized goat farms/herds in various regions of Punjab province and Islamabad capital territory (ICT), Pakistan. The disease started after introduction of new animals at these farms with no history of previous PPR vaccination. The clinical signs appeared affecting respiratory and enteric systems and spread quickly. Disease caused mortality of 10-20% and morbidity of 20-40% within a time period of four weeks. Morbidity and mortality rates were 30.38% (86/283) and 15.55% (44/283), respectively. Three treatment regimes were executed to demonstrate the role of vaccination during outbreak at these farms. First was to use only the broad spectrum antibiotics (Penicillin & Streptomycin and/or Trimethoprim and Sulfadiazine) at two farms (Texilla and Attock). Second treatment regime was to use the same broad spectrum antibiotic along with extensive fluid therapy (Farms at ICT-1 and ICT-2). The third regime was to use of broad spectrum antibiotic plus fluid therapy along with vaccinating the herd against PPR during first week of outbreak (ICT-3 and ICT-4). The third scheme of treatment gave the better results as there was no mortality in third week post-outbreak. Therefore, it is suggested to give proper importance to PPR vaccination along with conventional symptomatic treatment when dealing the PPR outbreaks in endemic disease conditions.

Enhancement of Lysine Production in Recombinant Corynebacterium glutamicum through Expression of Deinococcus radiodurans pprM and dr1558 Genes (Deinococcus radiodurans 유래 DR1558과 PprM에 의한 Corynebacterium glutamicum의 라이신 생산 향상 연구)

  • The expression of Deinococcus radiodurans dr1558 and pprM genes was examined for enhanced lysine production in recombinant Corynebacterium glutamicum. These genes are known to confer high tolerance to pH and osmotic shock in Escherichia coli. D. radiodurans dr1558 and pprM genes were expressed in C. glutamicum by using 6 synthetic promoters of different strengths, to evaluate the effect of expression efficiency on lysine production. Recombinant C. glutamicum expressing DR1558 under the L26 and I64 promoters showed higher lysine production than that expressing DR1558 under other promoters. Similarly, recombinant C. glutamicum expressing PprM under same promoters (L26 and I64) showed a higher increase in lysine production compared to that expressing PprM under other promoters. In the absence of $CaCO_3$ in the medium, the expression of DR1558 or PprM also increased lysine concentration in C. glutamicum depending on the promoter used. Together, these results suggest that genes involved in radiation tolerance in D. radiodurans can be used to enhance production of amino acids and their derivatives.

Management of small yellow croaker and hairtail in Korean waters using the length-based production value-per-recruit (PPR) analysis (체장기반 가입당생산액 분석에 의한 한국 연근해 참조기 Larimichthys polyactis 자원과 갈치 Trichiurus lepturus 자원의 관리)

  • Yield-per-recruit (YPR) analysis is used to provide management guidance for the efficient use of a fish cohort. However, the individual fish price per unit weight of small yellow croaker (Larimichthys polyactis) or hairtail (Trichiurus lepturus) increases dramatically by size in Korea. Therefore, age-based production value-per-recruit (PPR) analysis has recently been developed (Zhang et al., 2014). Since age determination requires a substantial amount of money and time and it is even impossible for some fish species, it is difficult to obtain age information to apply the age-based PPR model. Thus, we attempted to develop an alternative method, which uses length data rather than age information, called the length-based PPR analysis. The results revealed that length-based PPR analysis was much more conservative for stock management than the YPR analysis. Furthermore, the PPR analysis was more economically beneficial than the YPR analysis, which can prevent the fish stock from the economic overfishing. In conclusion, the length-based PPR analysis could be a proper approach for stock assessment in the case that the individual fish price per unit weight increases dramatically by size, and this analysis is useful to obtain vital management parameters under data-deficient situation when traditional stock assessment methods are not applicable.

Monoclonal antibody-based enzyme immuno-slide assay (EISA) in the rapid diagnosis of Peste des petits ruminants of goats

  • Monoclonal antibody (mAb)-based enzyme immune-slide assay (EISA) was used for the detection of Peste des petits ruminants (PPR) virus from field samples collected from a natural outbreak. The clinicopathological study was undertaken to diagnose the case primarily of PPR. Antigen was detected from discharges and faeces of infected goats and swabs of postmortem lesions prepared on glass slide or glass plate using acetone fixation. Nasal discharge collected at the early stage of disease course or lung is an appropriate ante- or postmortem sample for this technique, respectively. Convalescent polyclonal sera collected from recovered animals which were diagnosed as PPR by EISA showed high antibody titer against PPR by C-ELISA, demonstrating the satisfactory specificity of the test. Therefore, EISA is a sensitive and specific assay to confirm PPR infection both in field and laboratory conditions and especially suitable for developing country.

Does Process Quality of Inpatient Care Serve as a Guide to Reduce Potentially Preventable Readmission (PPR)? (의료서비스의 과정적 질과 잠재적으로 예방 가능한 재입원율과의 관계)

  • Objective: The objective of this study is to examine the association between process quality of inpatient care and risk-adjusted, thirty-day potentially preventable hospital readmission (PPR) rates. Data Sources/Study Setting: This was an observational cross-sectional study of nonfederal acute-care hospitals located in two states California and Florida, discharging Medicare patients with a principal discharge diagnosis of heart failure, acute myocardial infarction, or pneumonia January through December 31, 2007. Data were obtained from the Healthcare Cost and Utilization Project State Inpatient Database of the Agency for Healthcare Research and Quality, Centers for Medicare and Medicaid Services Hospital Compare database, and the American Hospital Association Annual Survey of Hospitals. Study Design: The dependent variable of this study is condition-specific, risk-adjusted, thirty-day potentially preventable hospital readmission (PPR). 3M's PPR software was utilized to determine whether a readmission was potentially preventable. The independent variable of this study is hospital performance for process quality of inpatient care, measured by hospital adherence to recommended processes of care. We used multivariate hierarchical logistic models, clustered by hospitals, to examine the relationship between condition-specific, risk-adjusted, thirty-day PPR rates and process quality of inpatient care, after taking clinical and socio-demographic characteristics of patients and structural and operational characteristics of hospitals into account. Findings: Better performance on the process quality metrics was associated with better patient outcome (i.e., low thirty-day PPR rates) in pneumonia, but not generally in two cardiovascular conditions (i.e., heart failure and acute myocardial infarction). Practical Implication: Adherence to the process quality metrics currently in use by CMS is associated with risk-adjusted, thirty-day PPR rates for patients with pneumonia, but not with cardiovascular conditions. More evidence-based process quality metrics closely linked to 30-day PPR rates, particularly for cardiovascular conditions, need to be developed to serve as a guideline to reduce potentially preventable readmissions.

  • In this paper, the high precision position control of AC Servo motor for semiconductor equipment is studied. The control system was implemented using TI DSP TMS320F2812 which has 150 MIPS speed for next generation motor control. The controlled 100W PMSM motor has 2,500 ppr optical incremental encoder. The control system has speed controller and current controller to control the motor position. The encoder pulses are divided into 4 times, which has 10,000 ppr and the motor system has the position accuracy of 1/10,000. If the resolution of the encoder is increased, the resolution of the position control will be increased.

Finding Top-k Answers in Node Proximity Search Using Distribution State Transition Graph

  • Considerable attention has been given to processing graph data in recent years. An efficient method for computing the node proximity is one of the most challenging problems for many applications such as recommendation systems and social networks. Regarding large-scale, mutable datasets and user queries, top-k query processing has gained significant interest. This paper presents a novel method to find top-k answers in a node proximity search based on the well-known measure, Personalized PageRank (PPR). First, we introduce a distribution state transition graph (DSTG) to depict iterative steps for solving the PPR equation. Second, we propose a weight distribution model of a DSTG to capture the states of intermediate PPR scores and their distribution. Using a DSTG, we can selectively follow and compare multiple random paths with different lengths to find the most promising nodes. Moreover, we prove that the results of our method are equivalent to the PPR results. Comparative performance studies using two real datasets clearly show that our method is practical and accurate.
