Purification and Characterization of Polyphenol Oxidase from Oyster Mushroom (Pleurotus ostreatus) (느타리버섯(Pleurotus ostreatus)의 Polyphenol Oxidase 분리 정제 및 특성 조사)

  • Polyphenol oxidase (PPO) isoforms were partially purified from oyster mushroom (Pleurotus ostreatus) using various chromatography techniques, and their characteristics of heat stability, substrate affinity, optimum pH, and optimum temperature were investigated. Three PPO isoforms named PO-I, PO-II-1, and PO-II-2 were partially purified from oyster mushroom. The molecular weight of PO-II-1 was 70 kDa and PO-I and PO-II-2 were less than 6 kDa each. Characterization was carried out using a PPO isoform partially purified by hydrophobic interaction chromatography. Optimum temperature was $55^{\circ}C$ and optimum pH 5.0. However, the PPO was inactivated at neutral pH or by heating at $80^{\circ}C$ for 30 min, while the 40% PPO still remained active after heating at $60^{\circ}C$ for 45 min. The PPO isoform showed the highest substrate affinity to chlorogenic acid and pyrogallol, in which KM values were 1.01 and 2.06 mM, respectively. Therefore, these results suggested that the mushrooms should be stored at a pH higher than 7.0 and at a low temperature to prevent enzymatic browning.

Subcellular Distribution and Characteristics of Polyphenol Oxidase from Perilla frutescens Leaves (들깨잎 polyphenol oxidase의 세포내 분포 및 특성)

  • Polyphenol oxidase (PPO) activity in 200×g (cell wall), 4,000×g (plastid), 100,000×g (mitochondrial) and soluble fractions of the perilla leaves was monitored in the upper, middle and lower sections of the plant. In the course of plant growth, PPO activities in plastid and mitochondrial fractions were decreased, while those in cell wall fraction were maintained. During growing process, specific activities and PPO activities of each fraction were decreased, while total phenol content were decreased in middle (middle) and then increased in later stage (lower). Cell wall, plastid, mitochondrial (pellet) and soluble fraction had slightly different pH optima and substrate specificities. Isoenzyme patterns were identical in two bands for PPO activity in different subcellular fractions. Their molecular weights were 37KD and 48KD respectively.

Identification of highly pathogenic Beauveria bassiana strain against Pieris rapae larvae

  • Seven different strains of Beauveria bassiana were used in a bioassay on Pieris rapae larvae. The results showed that an B. bassiana strain showed relatively high pathogenicity towards P. rapae larvae. The adjusted mortality rate was 92.86 %, and the infection rate was 85.71 % in 10 days post inoculation. Molecular identification was performed to identify the unknown strain. Internal Transcribed Spacer sequence analysis showed that the polymerase chain reaction amplicon length of the unknown strain of Beauveria sp. was 573 bp, and sequence similarity to the known B. bassiana sequences in the NCBI database was 99 %. The B. bassiana strain was named Bb01. The changes of proteins and PPO of P. rapae larvae infected by B. bassiana Bb01 strain at different times was determined. The activity of PPO increased in 1-6 d and decreased in 7 d again after inoculation. The B. bassiana invaded into the insect body affected the balance of the proteins and PPO.

Effects of $\beta$Carotene on the Browning of Phenolics Extracted from Potato and Polyphenol Oxidase Characteristics (감자에서 추출한 phenolics의 갈변화에 미치는 $\beta$carotene의 영향과 polyphenol oxidase 특성)

  • The effects of synthetic and carrot carotene on the browning of polyphenolics extracted from potato for eliminating the effects of other components in potato which inhibit the browning of potato juice and the potato polyphenol oxidase (PPO) activity were investigated. Total phenolics content in potato sample was 1.854mg/g D.W. and PPO activity was 100 unit. Delta 'L'value of polyphenolics extracted from potato decreased makedly, but that of potato with carrot decreased little by little. Potato with P-carotene showed a little decrease after 50 min. At the same time, polyphenolics extracted from potato were mixed with carotene extracted from carrot or with syntetic $\beta$-carotene. As the results, the delta 'L'value of the former increased but decreased similarly to the latter after 1 hour. The effects of enzyme and substrate concentration on the browning of PPO extracted from potato were investigated. Optimum enzyme concentration was 10% and optimum substrate concentration was 13.3% $\beta$-carotene concentration did not appear to influence significantly on PPO activity.

Differential Responses of Antioxidant Enzymes on Chilling and Drought Stress in Tomato Seedlings (Lycopersicon esculentum L.) (토마토 유묘에 있어서 저온과 수분 스트레스에 대한 항산화효소의 활성 차이)

  • Responses of antioxidant enzymes on chilling and drought stress in tomato seedlings were investigated. Growing patterns of tomato based on fresh weight of tomato seedlings were severely affected by chilling and drought stress. Fresh weight of tomato seedlings were reduced by 69.5% in chilling stress and 50.6% in drought stress compared to those in the unstressed control seedlings after 12 days of stress. The specific and gel activity of SOD and POD in the leaves, shoots, and roots of tomato seedlings were significantly increased by chilling and drought stress. Activation of SOD and POD activity by chilling stress were higher in the roots than those of drought stress. However, activation of SOD and POD activity by drought stress were higher in the leaves and shoots than those of chilling stress. The specific and gel activity of GR in the leaves, shoots, and roots of tomato seedlings were also significantly increased by chilling and drought stress. When the seedlings were treated with chilling or drought stress, one GR isozyme band (GR-3) was newly expressed in the leaves of tomato seedlings. The specific and gel activity of PPO was significantly increased in the roots and shoots of tomato seedlings by chilling and drought stress, respectively. However, the specific and gel activity of PPO in the leaves is no difference between stressed and controlled tomato seedlings.

Scutellaria baicalensis Extracts as Natural Inhibitors of Food Browning (천연 갈변저해제로서 황금 추출물의 효소적 갈변 저해 효과)

  • This study was designed to develop natural browning inhibitors. The anti-browning effects of distilled water (SBD) and 80% ethanol extracts (SBE) of Scutellaria baicalensis Georgi in apple slices were investigated by L and ${\Delta}E$ values. Both SBD and SBE were effective in reducing the browning of apple slices and were successively fractionated into chloroform ($CHCl_3$), ethyl acetate (EtOAc), and water ($H_2O$) fractions. These extracts were measured for total phenolic content, flavonoid content, anti-oxidative activity (through free radical scavenging activity and the FRAP assay), ferrous ion chelation, and the inhibition of PPO (polyphenol oxidase) activity. Overall, fractions of SBE were better than fractions of SBD in all measurements. The highest total phenolic and flavonoid content were measured in the EtOAc and $CHCl_3$ fractions of SBE. EtOAc and $CHCl_3$ fractions also exhibited the highest anti-oxidative activities (in DPPH and ABTS free radical scavenging and the FRAP assay). Unusually, the highest ferrous ion chelating capacity was found in the $H_2O$ fraction of SBD, but the other fractions showed more than triple the ascorbic acid already in use. Also, $CHCl_3$ fractions showed a stronger inhibition of PPO activity than ascorbic acid. All of these results suggest that EtOAc and $CHCl_3$ fractions from Scutellaria baicalensis can be used as natural anti-browning agents.

Effects of Dipping and Preheating Treatments on Susceptibility to Browning of Potato Slices During Cold Storage (침지용액과 예열 처리가 감자 슬라이스의 냉장 중 갈변정도에 미치는 영향)

  • Potato slices were dipped in solution of 2% CaCl$_2$, and 1% chitosan or preheated in each of these solutions for refrigeration for 4 weeks at 5$^{\circ}C$. Changes in L value, content of total phenol and chlorogenic acid and po lyphenol oxidase activity were determined. During refrigeration, it was found that L values of potato slices treated with CaCl$_2$, (CaPS) and those treated with chitosan (ChPS) increased. Contents of total phenol and chlorogenic acid of potato slices decreased, While potato slices preheated in CaCl, solution (Hcaps) showed much decreases. And polyphenol oxidase (PPO) activity of potato slices decreased except CaPS and decreases in PPO activities were much larger in preheated potato slices. From these results, of those treatments used, chitosan treatment combined with preheating have shown to be effective to control enzymatic browning of potato slices during refrigeration.

Studies of Characteristics of Polyphenol Oxidase in Yan and Antimutagenic effect of Enzymatic Browning Reaction Products (마의 Polyphenol oxidase의 특성과 효소갈변생성물의 항돌연변이 효과)

  • Polyphenol oxidase in Yam was partially purified through ammonium sulfate fractionation, DEAE-cellulose column chromatography. The specific activity of purified PPO was 138.22 unit/mg protein. The optimum pH and temperature of purified PPO were respectively 7.0 and 30$^{\circ}C$. The heat treatment at 80$^{\circ}C$ for 6 min, decreased PPO activity to 50%. The enzyme showed high substrate specificity toward catechol. The Km value for catechol was 5 mM. In the Ames test using S. typhimurium TA98 and TA100 catechol-YEBRP, pyrogallol-YEBRP, chlorogenic-YEBRP showed strong antimutagenicity on sodium azide and MECF excepting hydroquinone-YEBRP showed killing effect on both strains.

Characterization of 'Biuti' Peach Polyphenoloxidase

  • In Brazil canned 'Biuti' peach is a very popular form of this sub-tropical fruit. This production represents an important economic agro-activity in Minas Gerais, Brazil during the summer period, in preparation for the Christmas celebrations. The aim of this work was to characterize the 'Biuti' peach polyphenoloxidase (PPO), since peach products show enzymatic oxidation of the polyphenols by oxidative enzymes, which affects the products during their shelf life. Two different hypothesis for the browning problem in processed peaches were studied: the inadequacy of the blanching treatment and the presence of a latent phenolase in the peaches. The PPO was characterized: pH optimum (5.5) and stability (5.5-6.5); optimum temperature at $20^{\circ}C$ and 80% of the activity retained after 30 min at $15-40^{\circ}C$. The test for the presence of latent PPO in the processed and canned peaches was negative. Ascorbic acid, ${\beta}$-mercaptoethanol, sodium metabisulfite, and cysteine were efficient in inhibiting the PPO.

Thermostability of Polyphenol Oxidase from Potato (Solanum tuberosum L.) (감자 Polyphenol Oxidase의 열안정성)

  • Factors affecting thermostability of polyphenol oxidase (PPO) from potato were studied for the purpose of providing useful information for food processing operations. The enzyme was most stable at pH 7.0 and it was inhibited to 70% after heat treatment at 8$0^{\circ}C$ for 1 min. The z-value for the thermal inactivation of the PPO was 12.17$\pm$0.58$^{\circ}C$. The thermostability of the enzyme was reduced by addition of sodium chloride. And the activity was inhibited by addition of reducing reagents such as 2-mercaptoethanol and dithiothreitol.
