Regulatory B Subunits of Protein Phosphatase 2A Are Involved in Site-specific Regulation of Tau Protein Phosphorylation

  • Overexpression of amyloid precursor protein with the Swedish mutation causes abnormal hyperphosphorylation of the microtubule-associated protein tau. Hyperphosphorylated isoforms of tau are major components of neurofibrillary tangles, which are histopathological hallmarks of Alzheimer's disease. Protein phosphatase 2A (PP2A), a major tau protein phosphatase, consists of a structural A subunit, catalytic C subunit, and a variety of regulatory B subunits. The B subunits have been reported to modulate function of the PP2A holoenzyme by regulating substrate binding, enzyme activity, and subcellular localization. In the current study, we characterized regulatory B subunit-specific regulation of tau protein phosphorylation. We showed that the PP2A B subunit PPP2R2A mediated dephosphorylation of tau protein at Ser-199, Ser-202/Thr-205, Thr-231, Ser-262, and Ser-422. Down-regulation of PPP2R5D expression decreased tau phosphorylation at Ser-202/Thr-205, Thr-231, and Ser-422, which indicates activation of the tau kinase glycogen synthase kinase 3 beta ($GSK3{\beta}$) by PP2A with PPP2R5D subunit. The level of activating phosphorylation of the $GSK3{\beta}$ kinase Akt at Thr-308 and Ser-473 were both increased by PPP2R5D knockdown. We also characterized B subunit-specific phosphorylation sites in tau using mass spectrometric analysis. Liquid chromatography-mass spectrometry revealed that the phosphorylation status of the tau protein may be affected by PP2A, depending on the specific B subunits. These studies further our understanding of the function of various B subunits in mediating site-specific regulation of tau protein phosphorylation.

  • The preparation methods are to be used as "melting method" and "absorption method", respectively. The reaction mixture in the reaction container was heating up the reaction mixture to $200^{\circ}C$ for 2 hour. The mixing time of lab scale preparation should be provided quit long, instead of the short working time in a compounder vessel. And The PP-PCM mixture in the reaction container was heating up the mixture around $60-80^{\circ}C$ for 2 hour. A melting method of frozen gel with 50/50 weight ratio of polypropylene-normal octadecane was prepared by adding PP chip and normal octadecane. An absorption method of frozen gel with 70/30 weight ratio of PP 4.8-normal n-octadecane was prepared by adding PP powder and normal octadecnae.

  • The TOL plasmid, pWWO, conjugally transferred from Pseudomonas putida mt-2 was dissociated into TOL* and TOL $\Delta$A in P. putidu PpGl carrying CAM plasmid. The TOL* was integrated into the CAM plasmid, and the resulting plasmid was designated as CAM::TOL*. The introduction of NAH plasmid, belonging to Inc P9 incompatibility group, into P. putida CSTBA carrying CAM::TOLt plasmid and TOL A plasmid did not affect m-toluate catabolism, but resulted in expelling the TOL $\Delta$ plasmid.

Lifetime characteristics of flexible organic light emitting diodes on PET substrate with plasma polymer barrier layers

  • Plasma polymerized para-xylene ($PP_PX$) deposited by plasma-enhanced chemical vapor deposition (PECVD) was used as the barrier layer on the polyethylene terephthalate (PET) substrate to improve lifetime of the flexible organic light-emitting diodes (FOLEDs). The $PP_PX$ barrier layer deposited on top of the PET substrate with plasma power of 30 W at deposition pressure of 0.2 torr showed transmittance spectra good enough to be applied in FOLED on PET substrates. FOLEDs with the $PP_PX$ barrier layer (barrier-FOLEDs) showed similar I-V and B-V characteristics to FOLEDs without the $PP_PX$ layer (control-FOLEDs). The lifetime of barrier-FOLED was two times longer than that of the control-FOLED. With $PP_PX$ passivation layers, lifetimes of both control and barrier-FOLEDs were improved by more than 4 times. These results show that PECVD deposited $PP_PX$ layers can be used as barrier layers for FOLEDs on plastic substrates as well as passivation layers for general OLEDs.

A Study of the Optimization of the Compounded PP Using the DOE (실험계획법을 이용한 복합 폴리프로필렌의 최적화 연구)

  • In order to formulate the compounded polypropylene(C-PP) which is suitable to an automotive door trim panel, 9 sorts of properties were measured after manufacturing the C-PP using an extruder and an injection machine with polypropylene(PP), ethylene-octene rubber(EOR) and talc. Mixture design, especially extreme vertices design, in DOE with MINITAB - commercial software was used to analyze the data. The relations between each property and each component, for example, $y=0.00907222x_1+0.00870556x_2+0.0155722x_3$ for specific gravity, were found out by the regression analysis and the variance analysis. The optimized formulation of the C-PP for an automotive door trim panel was acquired at PP(77.6962), EOR(11.0238) and talc(10.2800) by use of the response optimizer(mixture) in MINITAB.

  • The fracture mechanics investigation of the thermoplastic vulcanizates(TPV) from EPDM and PP/Ionomer ternary blends was performed in terms of the J-integral by measuring fracture energy via the locus method. The TPV from ternary blends consisting of EPDM, PP and ionomer were prepared in a laboratory integral mixer by blending and vulcanizing simultaneously. Vulcanization was performed with dicumyl peroxide (DCP) and the composition of EPDM and PP was fixed at 50/50 by weight. Two kinds of poly(ethylene-co-methacrylic acid) (EMA) lonomers were used. The J-integral values at crack initiation, Jc, of the dynamically vulcanized EPDM and PP/EMA Ionomer ternary blends were affected by the cation types $(Na^+\;or\;Zn^{2+})$ and contents(5-20wt%) of the added EMA Ionomers. The ternary blend containing 20wt% zinc-neutralized EMA Ionomer and 1.0phr DCP showed the highest Jc values of the blends.

A Study on the Mechanical, Thermal, Morphological, and Water Absorption Properties of Wood Plastic Composites (WPCs) Filled with Talc and Environmentally-Friendly Flame Retardants (친환경 난연제와 탈크를 첨가한 목재·플라스틱 복합재의 기계적, 열적, 형태학적 및 수분흡수 특성에 관한 연구)

  • Wood plastic composite (WPC) is a green composite made of wood flour and thermoplastics to provide better performance by removing the defects of both wood and plastics. However, relatively low thermal stability and poor fire resistance of wood and plastics included in WPC have been still issues in using WPC as a building material for interior applications. This study investigated the effect of environmentally-friendly flame retardants (EFFRs) on the mechanical, thermal, morphological, and water absorption properties of wood flour (WF)/talc/polypropylene (PP) composites in comparison with neat PP. The whole EFFRs-filled WF/talc/PP composites showed higher values in flexural strength, flexural modulus, and impact strength compared to neat PP. In thermal properties, aluminum hydroxide (AH)-filled composite showed a $36^{\circ}C$ reduction in maximum thermal decomposition temperature ($T_{max}$) compared to neat PP, but magnesium hydroxide (MH) played an important role in improving thermal stability of filled composite by showing the highest $T_{max}$. From this research, it can be said that MH has potentials in reinforcing PP-based WPCs with improvement of thermal stability.

  • When we apply the LDPC code for high density optical storage channel, it is necessary to make an algorithm that the modulation code decoder must feed the LDPC decoder soft-valued information because LDPC decoder exploits soft values using the soft input. Therefore, we propose the soft-input soft-output run-length limited 17PP decoding algorithm and compare performance of LDPC codes. Consequently, we found that the proposed soft-input soft-output decoding algorithm using 17PP is 0.8dB better than the soft-input soft-output decoding algorithm using (1, 7) RLL.

Preparation of Epoxidized Soft Terpolymers and Their Reactive Compatibilizing Effects on PP/EVOH Blends

  • In this study, we prepared epoxidized poly ethylene-ter-1-decene-ter-divinylbenzene (Epo-PEHV) as a reactive compatibilizer to prevent phase separation phenomenon which occurs upon blending polypropylene (PP) and ethylene-vinyl alcohol copolymer (EVOH). Firstly, PEHV was prepared under high catalyst activity according to content of catalyst and cocatalyst. After then, we modified vinyl group of the terpolymer with epoxy group. We observed that the terpolymer was successfully epoxidized by 1H-NMR and FT-IR analysis. The Epo-PEHV was added by 2, 5, 10% in PP/EVOH blends. The morphologies and mechanical properties of PP/Epo-PEHV/EVOH blends were analyzed by SEM and UTM, respectively. Epo-PEHV enhanced the interfacial adhesion of PP and EVOH blends.

The Association of Ponticulus Posticus & Elongated Styloid Process with Headaches

  • Purpose: The present study was designed to investigate the association of ponticulus posticus (PP) and elongated styloid process (ESP) with headaches. Methods: Analysis of head and neck cone beam computed tomography samples from the archives of the Department of Oral Radiology was done for the presence of partial or complete PP and ESP length, type, thickness, mediolateral angulation, anterioposterior angulation (horizontal & vertical), lateral or medial curvature. This was followed by personal & telephonic questionnaires to the subjects for the evaluation of the presence of headaches & other associated symptoms. Results: Among 134 subjects, 62 subjects (46.3%) presented with headache and 72 subjects (53.7%) did not have any headache. On further analysing the total 62 subjects with headache, it was found out that 31 subjects (50.0%) of them had ESP and PP both, 16 subjects (25.8%) had only ESP, and 15 subjects (24.2%) had only PP. A strong association was present between headache and presence of PP & ESP individually and together. Conclusions: All health care professionals dealing with the head and neck pain disorders should also consider the presence of ESP & PP during diagnosis and treatment.
