The Effects of Hyangsayukgunja-tang Extract on Indomethacin-Induced Gatric Mucosal Lesions (향사육군자탕(香砂六君子湯)의 Indomethacin 유발 위점막 손상에 대한 효과)

  • Objective: This study was carried out to investigate the effects of Hyangsayukgunja-tang extract on indomethacin-induced gastric mucosal lesions of mice. Methods: To evaluate the effects of Hyangsayukgunja-tang extract and Misoprostol, the morphology of gastric mucosa, and the distribution of mucose cells, PNA(Peanut Agglutinin), ICAM(intercellular adhesion molecule), and apoptotic cells were observed. Hyangsayukgunja-tang extract and Misoprostol were intragastric injected to the test groups at hour 72 before and just before indomethacin treatment(HYT-J, HYT-72, M-J, M-72), while the INDO group was injected only with indomethacin and the control group was subcutaneously injected only with saline. Results: The gastric mucosal lesions incresed in the fundus and body of INDO group, but softened in HYT group and M group, the effects were more excellent in the HYT-72, M-72 groups than the HYT-J, M-J groups and in the HYT group than M group. The disappearance of surface and neck mucose cells were shown in INDO group, but softened in HYT group and M group. The mucosal configuration of HYT-72 group was the same as control group. The numerical increase of PNA positive reaction in cytoplasm of perietal cells were appeared in INDO group. The PNA positive reaction in HYT group and Miso-group were shown in surface mucous cells and microvilli of apical surface in chief cells as control group, and were the same as control group in all mucosa of HYT-72 group. The distribution of ICAM positive cells, increased in INDO group, but decreased in M-72 group, and were the same as control group in HYT-72 group. The apoptotic cells, increased noticeably in gastric mucosa of INDO group, decreased in HYT group and M group, and decreased noticeably in HYT-72 group. Conclusions: Hyangsayukgunja-tang extract had excellent effects on indomethacin-induced gastric mucosal lesions.

Neurologic Study of SP6(Sanyinjiao, 三陰交) Acupuncture on Bladder parasympathetic Nerve, Tibial Nerve and Blood Pressure in Rats (삼음교(三陰交) 침자(鍼刺)가 부교감신경(副交感神經)의 방광분지(膀胱分枝), 경골신경(脛骨神經) 및 혈압(血壓)에 미치는 영향(影響))

  • Objective : The effects of SP6(Sanyinjiao, 三陰交) acupuncture stimulation on bladder parasympathetic nerve activity (PNA), tibial nerve activity (TNA) as well as blood pressure were examined in anesthetized rats and the results are as follows. Methods : The parasympathetic nerve leading to the bladder was exposed retroperitoneally, and the tibial nerve in the hindlimb was exposed through inguinal area. PNA and TNA were recorded using a bipolar platinum-iridium wire electrode and it was connected to preamplifier. Blood pressure was measured using cannula that was inserted into femoral artery and was recorded with blood pressure sensor that is connected with transducer amplifier. The mean value of all parameters measured for 30 seconds before the stimulation was expressed as 100%. An acupuncture needle was inserted into the SP6 region to a depth of 4 mm. Results : Under intact state (IS), acupuncture stimulation for 120 seconds induced significant increase of PNA which was accompanied with decrease of blood pressure. At the same time, TNA showed temporary increase only with acupuncture stimulation. In 4 mg/kg of naloxone-administrated state (NAS), significant decrease of PNA was seen and it was accompanied with increase of blood pressure. TNA also showed temporary increase only with acupuncture stimulation under the NAS just like the IS. Conclusion : consider that SP6(Sanyinjiao, 三陰交) acupuncture stimulation activated parasympathetic nerve and caused vasodilation to lead into the diuresis, but naloxone reversed the effect of acupuncture for antidiuresis.

Peptide Nucleic Acid Probe-Based Analysis as a New Detection Method for Clarithromycin Resistance in Helicobacter pylori

  • Background/Aims: Helicobacter pylori eradication rates are decreasing because of increases in clarithromycin resistance. Thus, finding an easy and accurate method of detecting clarithromycin resistance is important. Methods: We evaluated 70 H. pylori isolates from Korean patients. Dual-labeled peptide nucleic acid (PNA) probes were designed to detect resistance associated with point mutations in 23S ribosomal ribonucleic acid gene domain V (A2142G, A2143G, and T2182C). Data were analyzed by probe-based fluorescence melting curve analysis based on probe-target dissociation temperatures and compared with Sanger sequencing. Results: Among 70 H. pylori isolates, 0, 16, and 58 isolates contained A2142G, A2143G, and T2182C mutations, respectively. PNA probe-based analysis exhibited 100.0% positive predictive values for A2142G and A2143G and a 98.3% positive predictive value for T2182C. PNA probe-based analysis results correlated with 98.6% of Sanger sequencing results (${\kappa}$-value=0.990; standard error, 0.010). Conclusions: H. pylori clarithromycin resistance can be easily and accurately assessed by dual-labeled PNA probe-based melting curve analysis if probes are used based on the appropriate resistance-related mutations. This method is fast, simple, accurate, and adaptable for clinical samples. It may help clinicians choose a precise eradication regimen.

Inhibition of Melanosome Transport by Inducing Exon Skipping in Melanophilin

  • Exon skipping is an efficient technique to inhibit specific gene expression induced by a short-sequence peptide nucleic acid (PNA). To date, there has been no study on the effects of PNA on skin pigmentation. In melanocytes, the tripartite complex is responsible for the transport of mature melanosomes from the nucleus to the dendrites. The tripartite complex is composed of Rab27a, Mlph (Melanophilin), and Myosin Va. Defects in the protein Mlph, a melanosome transport-related protein, are known to cause hypopigmentation. Our study shows that Olipass peptide nucleic acid (OPNA), a cell membrane-permeable PNA, targets exon skipping in the Mlph SHD domain, which is involved in Rab27a binding. Our findings demonstrate that OPNA induced exon skipping in melan-a cells, resulting in shortened Mlph mRNA, reduced Mlph protein levels, and melanosome aggregation, as observed by microscopy. Therefore, OPNA inhibits the expression of Mlph by inducing exon skipping within the gene. These results suggest that OPNA, which targets Mlph, may be a potential new whitening agent to inhibit melanosome movement.

  • Mammary epithelial cells contain a subpopulation of cells with a large proliferativ potential which are responsible for the maintenance of glandular cellularity and are the progenitor cells of mammary cancer. These clonogens give rise to multicellular clonal alveolar or ductal units(AU or DU) on transplantation and hormonal stimulation. To isolate putative mammary clonogens, enzymatically monodispersed rat mammary epithelial cells from organoid cultures and from intact glands are sorted by flow cytometry according to their affinity for FITC labeled peanut lectin(PNA) and PE labeled anti-Thy-1.1 antibody(Thy-1.1) into four subpopulations : cells negative to both PNA and Thy-1.1(B-), PNA+cells, Thy-1.1+cells, and cells positive to both reagents(B+). The in vivo transplantation assays indicate that the clonogenic fractions of PNA+cells from out-growths of organoids in primary cultures for three days in complete hormone medium(CHM) are significantly higher than those of cells from other subpopulations derived from cultrues or from intact glands. Extracellular matrix(ECM) is a complex of several proteins that regulated cell function ; its role in cell growth and differentiation and tissue-specific gene expression. It can act as a positive as well as a negative regulator of cellular differentiation depending on the cell type and the genes studied. Regulation by ECM is closely interrelated with the action of other regulators of cellular function, such as growth factors and hormones. Matrigel supports the growth and development of several different multicellular colonies from mammary organoids and from monodispersed epithelial cells in culture. Several types of colonies are observed including stellate colonies, duct-like structures, two- and three-dimensional web structures, squamous organoids, and lobulo-duct colonies. Organoids have the greatest proliferative potential and formation of multi-cellular structures. Phase contrast micrographs demonstrate extensive intracellular lipid accumulation within the web structures and some of duct-like colonies. At the immunocytochemical and electron micrograph level, casein proteins are predominantly localized near the apical surface of the cells or in the lumen of duct-like or lobulo-duct colonies. Squamous colonies are comprised of several layers of squamous epithelium surrounding keratin pearls as is typical fo squamous metaplasia(SM). All-trans retinoic acid(RA) inhibits the growth of SM. The frequency of lobulo-ductal colony formation increased with the augmentation of RA concentration in these culture conditions. The current study models could provide powerful tools not only for understanding cell growth and differentiation of epithelial cells, but also for the isolation and characterization of mammary clonogenic stem cells.

The Effects of Pyeongwibunsoeum Extract on Indomethacin-induced Gastric Mucosal lesions (평위분소음의 Indomethacin 유발 위점막 손상에 대한 효과)

  • Objectives : This study was carried out to investigate the anti-ulcer effects of Pyeongwibunsoeum extract and Misoprostol on indomethacin-induced gastric mucosal lesions of mice. Methods : Experimental groups were classified into non-treated group, non-administered group, Misoprostol-administered group and Pyeongwibunsoeum-extract-administered group. This study examined the morphological change, distribution of mast cells and apoptic cells, PNA reaction, COX-1, BrdU, NF-B p50, COX-2, IL-2R-, and ICAM-1of gastric mucosa. Results : 1. The hemorrhagic erosion of gastric mucosa was reduced in the Misoprostol-administered and Pyeongwibunsoeum-extract-administered groups. 2. The Misoprostol-administered group and Pyeongwibunsoeum-extract-administered group showed positive reaction on the PNA distribution. 3. The distribution of apoptic cells, activity of NF-B p50, COX-2, IL-2R-, and ICAM-1 decreased in both the Misoprostol-administered group and Pyeongwibunsoeum-extract-administered group. 4. The Misoprostol-administered and Pyeongwibunsoeum-extract-administered groups showed increase on COX-1, BrdU. Conclusions : As the results indicate, hemorrhagic erosion of gastric mucosa were reduced in both the Misoprostol-administeredand Pyeongwibunsoeum-extract-administered groups, and severe inflammatory reaction of gastric mucosa was reduced. The effects on the Pyeongwibunsoeum-extract-administered group were superior to those on the Misoprostol-administered group. Pyeongwibunsoeum extract can be widely administered for gastric ulcer disease in clinical treatments.

Validity and Reliability of the Korean Version of the Families' Importance in Nursing Care-Pediatric Nurses' Attitudes Instrument

  • Purpose: This study aimed to test the validity and reliability of the Korean version of the Families' Importance in Nursing Care-Nurses' Attitudes (FINC-NA) instrument developed by Saveman et al. Methods: The 222 pediatric nurses' data were collected from 13 hospitals in South Korea and were analyzed using descriptive statistics, exploratory factor analysis, the Pearson correlation coefficient, and the Cronbach's ${\alpha}$ in SPSS software. The AMOS program was used to conduct confirmatory factor analysis of construct validity. Results: Of the 26 initial items, 24 were ultimately selected after evaluating content validity, construct validity, and reliability. The following 6 factors were included in the Korean version of the Families' Importance in Nursing Care-Pediatric Nurses' Attitudes (KFINC-PNA): family as a 'conversational partner', 'participant in care', 'supporter for the nurse', 'burden', 'recipient of empowerment', and 'its own resource'. Conclusion: The KFINC-PNA was partially modified to explain differences in language and culture, but its validity and reliability were verified. Pediatric nurses' attitudes can be assessed using the KFINC-PNA, and adjustments to the care of hospitalized children and their families can be made based on these items. We recommend developing and verifying intervention methods that will improve family-centered care for hospitalized children and their families.

Fractionation of Exopeptidase from Viscera of Argentina Shortfin Squid, Illex argentinus (원양산 오징어(Illex argentinus) 내장으로부터 Exopeptidase의 분획)

  • For the effective use of exopeptidase from squid viscera as food processing aids, the viscera of Argentina shortfin squid (Illex argentinus) were fractionated by various methods such as acetone treatment, ammonium sulfate treatment, anion exchange chromatography, and gel filtration. The positive exopeptidase fractions were obtained from the fraction II treated by cold acetone ($30{\sim}40%$, w/w), the fraction V by ammonium sulfate ($60{\sim}70%$ saturation), the fraction II (0.2 M NaCl) by anion exchange chromatography, and the fraction I ($30{\sim}50\;kDa$) by gel filtration. The specific activities of positive fractions from viscera of I llex argentinus against substrates were higher to LeuPNA than to ArgPNA. Total activity and recovery against LeuPNA of positive fraction by gel filtration were 1,867 U and 30.69%, respectively, which were the highest among those of positive fraction. The results suggested that the gel filtration chromatography method was the most efficient method for the fractionation of exopeptidase from viscera of Illex argentinus.

All-trans Retinoic Acid Induced Differentiation of Rat Mammary Epithelial Cells Cultured in Serum-free Medium

  • Retinoids are applied to not only cancer prevention but also cancer chemotherapy by stimulating differentiation of cells. We studied differentiation inducing effect of all-trans retinoic acid (ATRA) by studying proportion of high dense fractions of stem-like cells and the size of S phase fraction in cell cycle. From mammary organoids obtained from 7- to 8-week old F344 female rat mammary gland, we cultured rat mammary epithelial cells (RMEC) and treated physiological doses of $10^{-6}$, $10^{-7}$, and $10^{-8}$ M ATRA from the first day and then cultured for 4, 7, and 14 days. After that, immunostaining was performed using peanut agglutinin (PNA) and anti-Thy-1.1 monoclonal antibody (Thy-1.1) that can be used as markers of differentiation. We separated four different cell subpopulations by flow cytometry: cells negative to both reagents (B-), PNA-positive cells (PNA+), Thy-1.1-positive cells (Thy-1.1+), and cells positive to both reagents (B+). We observed continuous decreases of high dense fractions of stem-like cells (PNA+ subpopulations) for 14 days and as much decreases as high doses of ATRA, which were thought to be proportional to doses of ATRA. We labeled RMEC with bromodeoxyuridine and investigated cell cycle fractions that went through S phase. We observed a tendency of decrease of S phase fraction with time in culture, which, is thought to be related to continuous decreases of PNA+ subpopulations and inhibitory role of ATRA on cell cycle. These results suggest that physiological doses of ATRA could stimulate differentiation of RMEC and convert stem-like RMEC to differentiated cells in SFM for a relatively long period of 14 days.

  • There have been a total tenth FMD outbreaks in Korea and for the first time, type O and A were detected simultaneously in 2017, which led to difficulties in FMD control. For the effective prevention of FMD, the importance of discrimination of serotypes became greater. Therefore, the most urgent requirement in case of FMD outbreak is differential diagnosis of serotypes. In this study, we developed a PNA probe-mediated multiplex real-time reverse transcription-polymerase chain reaction (rRT-PCR) assay using the peptide nucleic acid (PNA) probe, which is known to be stable to nucleotide mutation and that could specifically detect the all FMDV serotype A, FMDVA Yeoncheon strain which was occurred in Korea in 2017, and FMDV A viruses shown 96% similarity with FMDVA/Yeoncheon strain, at the same time. Therefore, It is believed that the newly introduced FMDVA will be effectively diagnosed using the PNA probe multiplex RT-PCR developed in this study, and ultimately contribute to the prevention of FMD.
