흑토란의 식품재료화를 위한 숙성 조건에 따른 토란의 특성 및 관능 최적화 (Characteristics and Sensory Optimization of Taro (Colocasia esculenta) under Different Aging Conditions for Food Application of Black Taro)

  • 토란은 풍부한 영양 성분과 다양한 효능을 가지고 있으나 활용도가 높지 않고, 옥살산 등의 독성 성분과 높은 수분 함량을 가지며 갈변 현상이 쉽게 일어나기 때문에 상품화를 위해서는 새로운 가공 방법에 대한 연구가 필요하다. 본 연구에서는 토란을 증자, 숙성, 건조하는 과정을 통해 차로 음용할 뿐만 아니라 다양한 식품 재료로 활용할 수 있는 새로운 가공 방법을 개발하고자 하였으며, 이를 흑토란이라 명명하였다. 흑토란을 제조하기 위하여 증자($95{\pm}3^{\circ}C-1\;h$) 후 다양한 조건(85, 90, $95^{\circ}C-20$, 40, 60 h)에서 숙성시켰으며, 이를 건조($60^{\circ}C-24h$)하여 분말화하였다. 그 후, 이화학적 특성, 산화방지 활성을 분석하였으며, 반응표면분석을 이용하여 관능적 최적점을 탐색하였다. 토란의 일반성분 분석 결과, 생토란은 87.69%의 수분 함량을 보였고 증자 시 90.70%으로 증가하였다가 흑토란에서 4.87-8.56%로 감소하였다. 흑토란은 생토란과 증자 토란에 비해 탄수화물, 조단백, 조지방, 조회분 함량이 모두 증가하였으며, 조섬유는 생토란에서 0.45%, 흑토란에서 5.23-7.86%의 함량을 나타내었다. 무기질 성분 분석 결과, 모든 시료에서 포타슘 함량이 가장 높았다. 토란의 총당과 함량 결과, 생토란은 112.53 mg/g, 증자 토란은 70.51 mg/g을 나타냈으며, 흑토란은 숙성 시간에 따라 유의적으로 증가하였고 54.94-120.03 mg/g의 함량을 보였다. 환원당은 42.48 mg/g에서 증자 후 12.29 mg/g로 감소하였으며, 흑토란에서 12.73-24.14 mg/g의 함량을 나타내었다. 분말색도는 생토란을 열처리함에 따라 L값이 84.57에서 증자 토란의 71.05, 그리고 흑토란에서 47.91-55.28으로 감소하였으며, a값은 증가하고 b값은 감소하는 경향을 나타내었다. 갈색도는 생토란에서 0.08의 값을 나타냈고, 증자 후 0.02로 감소하였다가 흑토란에서 급격하게 증가하였으며 $90^{\circ}C$에서 40시간 숙성한 토란에서 0.88의 가장 높은 값을 보였다. 옥살산칼슘 함량 측정 결과, 생토란에서 59.07 mg/100 g의 함량을 보였으며, 증자 후 37.20 mg/100 g으로 감소하였고, 숙성 과정을 거치며 시간에 따라 유의적으로 감소하여 $90^{\circ}C$에서 60시간 숙성시킨 토란에서 11.46mg/100 g의 가장 낮은 값을 보였다. 흑토란의 총 폴리페놀 함량을 측정한 결과, 20.61-28.30 mg GAE/g의 값을 나타냈으며, 숙성 온도에 따라서 폴리페놀 함량이 유의적으로 증가함을 확인하였다. 흑토란의 산화방지 활성을 DPPH, ABTS 자유라디칼 소거능 및 FRAP 활성으로 측정하였다. DPPH와 ABTS 라디칼 소거능 모두 숙성 온도가 높아짐에 따라 증가하는 경향을 보였다. DPPH 라디칼 소거능은 3.67-7.52 mg VCE/g의 값을, ABTS 라디칼 소거능은 9.63-20.32 VCE/g의 값을 나타냈으며, 두 소거능 모두 $95^{\circ}C$에서 60시간 숙성시킨 시료에서 가장 높았다. FRAP 활성 또한 유사한 경향을 보였고, 3.43-6.79 mg VCE/g의 값을 나타내었다. 흑토란의 관능 최적 숙성 조건을 탐색하기 위하여 색, 맛, 향, 전체적인 기호도에 대하여 반응표면분석을 실시하였다. 색($R^2=0.8471$), 맛($R^2=0.8244$), 전반적인 기호도($R^2=0.7480$)에 대하여 유의한 반응 표면을 이끌어낼 수 있었으며, 맛과 전반적인 기호도 항목에서 실험 범위 내의 최적점을 이끌어낼 수 있었다(맛: $88.73^{\circ}C$, 39.50시간, 전반적인 기호도: $88.82^{\circ}C$, 42.60시간). 관능 특성간의 상관 분석 결과, 전반적인 기호도에는 색, 맛, 향 모두 영향을 미치는 것으로 나타났으며, 맛(r=0.884)의 영향이 가장 큰 것으로 나타났다. 본 연구를 통해 토란의 숙성 조건에 따른 이화학적 특성 및 산화방지 활성의 차이를 확인하였으며, 관능적으로 최적이 되는 토란의 숙성 조건을 확인하였다. 실험 결과를 종합해 보았을 때, 생토란에 비해 흑토란에서 섬유소 함량의 증가, 옥살산칼슘의 유의적인 감소, 그리고 갈변 반응으로 인한 생리활성 성분의 증가 효과를 기대할 수 있다. 높은 산화방지 효과를 위해서는 높은 온도에서 오랜 시간 숙성이 필요하며, $88.73-88.82^{\circ}C$의 온도에서 39.50-42.60시간 동안 숙성시켰을 시 관능적으로 우수한 흑토란을 생산할 수 있을 것으로 사료된다. 본 연구는 토란의 이용성을 증대시키며 흑토란 분말의 다양한 식품재료로서의 활용 가능성을 탐색하기 위한 기초 자료가 될 것으로 기대된다.

Streptozotocin유발 당뇨쥐에 있어서 Metallothionein 합성에 미치는 비타민 E의 영향 (Effects of Vitamin E on the Metallothionein Synthesis in Streptozotocin-induced Diabetic Rats)

  • 식이내 비타민 E 함량을 0mg vit. E/kg diet(STZ-OE군), 40mg vit. E/kg diet(STZ-40E군) 그리고 40mg vit. E/kg diet(STZ-400E군)로 달리하여 4주간 사육한 후 streptozotocin으로 당뇨병을 유발한 후 4일째에 희생시켜, 당뇨병 쥐에 있어서 MT합성에 미치는 비타민 E의 영향을 관찰한 결과는 다음과 같다. 체중증가는 STZ을 투여하기 전까지는 대조군에 비해 STZ-0E군만 낮았으나 STZ투여한 후에는 STZ투여군들 모두가 현저하게 감소하였다. 실험군간을 비교해 보면 STZ-400E군이 STZ-0E, STZ-40E군에 비해 STZ을 투여한 후 감소율이 낮았다. 혈당량은 당뇨군들이 대조군에 비해 약 3배 정도 높았으나, 당뇨군들 중에서 비타민 E의 영향은 없었다. 간조직 중의 비타민 E 함량은 STZ-0E 및 STZ-40E군에서는 대조군 보다 각각 50, 36%씩 크게 감소되었으나 STZ-400E군은 차이가 없었다. Glutathione(GSH) 함량은 비타민 E의 다량 첨가군(STZ-400E)에서 GSH 함량과 GSH/GSSG비가 증가되었다. 간조직 중의 지질과산화가는 STZ-0E군과 STZ-40E군이 대조군 보다 5.6, 2.5배씩 높았으나 STZ-400E군은 유의적인 차이가 없었다. Piasma insulin level은 대조군에 비해 당뇨유발군 모두가 현저하게 감소하였고, 비타민 E의 효과는 관찰되지 않았다. Piasma cortisol level은 대조군에 비해 STZ-0E, STZ-40E군이 증가하였으나 STZ-400E군은 이들 두군에 비해 유의적으로 감소하여 대조군 수준이었다. 간장, 신장, 소장조직의 MT 함량은 대조군에 비해 당뇨유발군이 각각 5.3, 2.8, 16배 정도 현저히 증가되었는데 STZ-400E군은 STZ-0E, STZ-40E 이들 두군에 비해 현저하게 감소되었다. Sephadex G-75를 사용하여 간조직 중의 Zn-MT를 분리한 결과 당뇨 유발군에서 대조군 보다 Zn-MT peak가 증가되었으나 Cu-MT는 변화가 없었다. Zn-MT는 DEAE A-25를 사용하여 MTI, MTII peak를 확인할 수 있었다. 이상의 결과에서 STZ유발 당뇨쥐는 산화적 stress에 더욱 감수성이 높으므로 지질과 산화 반응이 정상군에 비해 촉진되었으며, 식이 중 공급되는 비타민 E 함량이 부족시는 더욱 가속화되었다. 더불어 간조직 중의 MT합성도 같은 양상으로 증가되어 당뇨병이 free radical 생성으로 인해 MT를 유도하는 하나의 새로운 인자인 것으로 확인하였다. 그러나 식이 중 비타민 E의 다량 급여시는 생체내 항산화계 효소들이 정상군에 가깝게 활성화되고, 생리적 항산화물질이 증가되었다. 따라서 식이비타민 E가 당뇨병으로 인한 지질과산화에 대한 항산화적 방어기구를 강화시키므로써 조직의 과산화적 손상을 환화시키는데 크게 기여함을 알 수 있었다. 또한 비타민 E 다량 투여로 간조직과 신장, 소장조직내의 total MT 함량이 감소되고 분리정제한 Zn-MT의 peak가 감소하는 것으로 보아 MT가 free radical scavenger로서 작용한 것이라고 생각된다.

Goat Meat Does Not Cause Increased Blood Pressure

  • While there are persistent rumors that the consumption of goat meat dishes increases blood pressure, there is no scientific evidence to support this. Two experiments were conducted to clarify whether or not blood pressure increases in conjunction with the consumption of goat meat dishes. In experiment 1, 24 Dahl/Iwai rats (15 weeks old, body weight $309.3{\pm}11.1$ g) were evenly separated into 4 groups. The control group (CP) was fed a diet containing 20% chicken and 0.3% salt on a dry matter basis. The goat meat group (GM) was fed a diet containing 20% goat meat and 0.3% salt. The goat meat/salt group (GS) was fed a diet containing 20% goat meant and 3% to 4% salt. The Okinawan mugwort (Artemisia Princeps Pampan)/salt group (GY) was fed a diet containing 20% goat meat, 3% to 4% salt and 5% of freeze-dried mugwort powder. The experiment 1 ran for a period of 14 weeks during which time the blood pressure of the animals was recorded. The GS, and GY groups consumed significantly more water (p<0.01) than the CP and GM groups despite the fact that their diet consumption levels were similar. The body weight of animals in the CP, GM, and GS groups was similar while the animals in the GY group were significantly smaller (p<0.01). The blood pressure in the GM group was virtually the same as the CP group throughout the course of the experiment. In contrast, while the blood pressure of the animals in the GS and GY group from 15 to 19 weeks old was the same as the CP group, their blood pressures were significantly higher (p<0.01) after 20 weeks of age. The GY group tended to have lower blood pressure than the GS group. In experiment 2, in order to clarify whether or not the increase in blood pressure in the GS group and the GY group in experiment 1 was caused by an excessive intake of salt, the effects on blood pressure of a reduction of salt in diet were investigated. When amount of salt in the diet of the GS and GY group was reduced from 4% to 0.3%, the animal's blood pressure returned to normotensive. These results indicate that, as in the case of chicken consumption, prolonged consumption of goat meat does not cause increased blood pressure, rather the large amount of salt used in the preparation of goat meat dishes is responsible for the increase in blood pressure.

Des-Gamma-Carboxyprothrombin for Early Identification and Prognosis of Hepatocellular Carcinoma - A Case Control Study from Western Nepal

  • Objective: To assess the diagnostic and prognostic value of AFP and des-gamma-carboxyprothrombin (DCP) in combination and alone for hepatocellular carcinoma. Materials and Methods: A case control study carried out in the Department of Biochemistry of Manipal College of Medical Sciences, Pokhara, Nepal between $1^{st}$ January 2010 and $31^{st}$ December 2011. The variables collected were age, gender, BMI, total proteins, albumin, AST, ALT, total bilirubin, DCP, AFP. Approval for the study was obtained from the institutional research ethical committee. Estimation of AFP was performed by ELISA reader for all cases. Analysis was done using descriptive statistics and confidence interval (CI). The data was analyzed using Excel 2003, R 2.8.0 Statistical Package for the Social Sciences (SPSS) for Windows Version 16.0 (SPSS Inc; Chicago, IL, USA) and the EPI Info 3.5.1 Windows Version. Results:The mean age of HCC cases was $53.6{\pm}14.93$ yrs. The percentage of females was less than males in both cases (23%) and controls (29%). The specificity of DCP reached 100% when its values was equal or greater than 150 (MAU/ml) for 0, 3, 6, 9, 12 months preceding the diagnosis of HCC. Similarly, the specificity for AFP was also nearly 100% when its value was equal or greater than 200 ng/ml 0, 3, 6, 9, 12 months earlier to the finding of HCC. The specificity of DCP (${\geq}40MAU/mL$) and AFP(${\geq}20$ ng/mL) in combination was 93%, 97%, 95%, 96%, 97% in respect to 0, 3, 6, 9, 12 months prior to the diagnosis of HCC. Conclusion: The combination of both DCP and AFP will improve the finding of initial HCC and the sensitivity of these markers was utmost at the time of HCC identification and noticeably lesser at former time points.

Effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs

  • Weaning is the most stressful event for nursery pigs because they are moved from familiar to unfamiliar environments. In addition, weaned pigs have immature digestive and immune systems. This situation makes weaned pigs susceptible to diseases and makes the absorption of nutrients from diets difficult. A feed approach, such as dietary enzyme supplementation, can be considered a solution. This study investigated the effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; combination of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Incidence of diarrhea, packed cell volume (PCV), white blood cells (WBC) count, and immunoglobulin content were measured. A significantly lower incidence of diarrhea (p < 0.05) was observed in the Cocktail group as compared with the CON group. The Cocktail group also showed a decreased PCV (p < 0.1) on d 3 after weaning than the CON group. However, no differences were observed for number of WBC and contents of immunoglobulin G, M, and A between the Cocktail and CON groups. Consequently, inclusion of an enzyme cocktail in diets for weaned pigs had a positive influence on gut health by reducing the incidence of diarrhea in the present study.

  • Soybean, one of most widely used swine feed component in the world, contains non-starch polysaccharides (NSP). The digestive system of weaned pigs is not yet fully developed, and thus weaned pigs cannot easily digest diets based on corn and soybean meal. Dietary exogenous enzymes supplementation has been intensively investigated to assist digestion of anti-nutritional factors, such as NSP. This experiment was conducted to investigate the effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; mixture of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Growth performance, morphology of ileum, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter, crude protein, and energy of weaned pigs were measured. No significant differences (p > 0.05) were observed for growth performance for the duration of the experimental period, and morphology of ileum, and nutrient digestibility between CON and Cocktail treatment groups. Therefore, the results from the current study indicated that enzyme cocktail supplementation in diets had no influence on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.

다시마와 효소처리 고등어육을 이용한 조미소재의 제조 조건 (Processing Condition of Seasoning Material of the Mixture of Laminaria and Enzyme-Treated Mackerel Meat)

  • 다시마와 효소 처리한 고등어육으로 조미료 소재를 제조하기 위해 가공조건을 검토한 결과는 다음과 같다. 적절한 자숙고등어육을 얻기위해 papain, bromelain, potease-A의 처리 결과 효소의 첨가농도는 0.03%, 효소처리시간은 90분정도가 적당하였다. 건조에 따른 효소처리 고등어육의 영향은 온도가 높을수록 건조 속도가 빨랐으며, 적절한 건조 조건은 $100^{\circ}C$에서 4시간 처리였다. 자숙고등어육을 효소처리 하였을 때의 protease-A 처리구의 총유리아미노산 함량이 5,007.3 mg/100 g으로 가장 높았고, glutamic acid, glycine, alanine, arginine 등의 정미성 아미노산이 약 33.0%를 차지하였다. Bromelain처리구와 papain처리구는 histidine, glutamic acid, aspartic acid, leucine, arginine 등이 주요아미노산이였다. 다시마의 총유리아미노산의 함량은 5281.8 mg/100 g으로, 주요 유리아미노산으로 glutamic acid, aspartic acid이었다. 시제품의 관능검사 및 탁도실험에서 제품 C가 멸치나 제품 A와 B보다 종합평가에서 우수하였으나, flavor에서 약간의 이취가 느껴져 이에 대한 보완이 필요하다고 생각되었다.

배출계수 개발 및 배출량 산정 체계 고도화를 통한 건설기계의 연식, 출력 및 기종별 대기오염물질 배출량 산정 (An Estimation of Age-, Power-, and Type-Specific Emission Inventories for Construction Equipments Using Improved Methodologies and Emission Factors)

  • The construction equipment is one of the major sources for hazardous air pollutants in Korea, and the its management has been of great concern recently. The objective of this study was to estimate each contribution of emission of construction equipments according to their production year, electric power consumption and type. To achieve this goal, we developed pollutant emission factors for the machineries manufactured after 2009, which are excluded from the present framework of Korean air pollutants inventory, CAPSS. More than 800 data obtained from emission investigations were utilized for the estimation. Compared with the previous estimation, the scheme used this study was modified to incorporate new emission factors as well as to include the corresponding activity data. Such improvement allow us to gain more detailed emission informations which are better characterized by specifications of construction equipments. The total amount of pollutants emitted from construction equipments in 2011 were estimated as 126.8, 7.0, 58.3, and 17.0 kton for $NO_x$, PM, CO, and VOC, respectively. The estimation results indicate that the increase in the emission of equipments is significantly related to their age and power consumption. The emissions of the older ones manufactured from 1992~1996 were estimated to be the contribution ranged from 23.7% to 26.8%, whereas the newer ones (2009~2011) showed the attributions of 11.3~21.5%. In addition, the results show that the emission of each equipment was increased with the increase in the electric power consumption of engine, probably due to their average output power. Among the nine types of machinery compared, excavators and forklifts were investigated to contribute relatively higher emissions in the level of 39.8~44.0% and 32.0~34.2%, respectively.

Effect of Sources of Supplementary Protein on Intake, Digestion and Efficiency of Energy Utilization in Buffaloes Fed Wheat Straw Based Diets

  • Sixteen adult male buffaloes (average body weight $443{\pm}14kg$) were equally distributed into four groups in an experiment to study the effect of supplementary protein sources on energy utilization efficiency in buffaloes fed a wheat straw-based diet. The animals in the control group were offered a basal diet composed of 700 g deoiled ground nut cake and ad libitum wheat straw. Animals of other groups were offered 1.8 kg of soyabean meal (SBM), linseed meal (LSM) or mustard cake (MC) along with the basal diet. Protein supplementation increased the digestibility of DM (p<0.01), OM (p<0.01) CP (p<0.01) and CF (p<0.05). Maximum CP digestibility was observed on SBM, followed by LSM and MC when compared to the control. Total DMI and DOMI was significantly (p<0.01) higher in protein supplemented groups with no differences between treatment groups. Digestible crude protein (DCP) intake and N balance were significantly (p<0.01) different between the groups; maximum response was obtained with SBM supplementation, followed by LSM and MC. Faecal energy was significantly (p<0.01) lower in SBM and LSM groups in comparison to other groups. Methane production (% DEI) was significantly (p<0.05) lower on the SBM treatment. Metabolizable energy (ME) intake increased significantly due to protein supplementation. Metabolizable energy intake (MEI) of animals in the MC group was less than LSM and SBM. Energy balance was increased significantly (p<0.01) due to protein supplementation and within supplement variation was also significant with maximum balance in SBM followed by LSM and MC groups. Protein supplementation significantly (p<0.05) increased the digestibility and metabolizability of energy from whole ration. Metabolizable energy (ME) content (Mcal/kg DM) of SBM, LSM and MC was 4.49, 3.56 and 2.56, respectively. It was concluded that protein supplementation of wheat straw increased intake, digestibility and metabolizability of energy and maximum response could be obtained when soybean meal was used as a supplement.

  • Two Gram-staining-negative, red-pinkish, coccus-shaped, non-motile, and aerobic bacterial strains, designated $Ant21^T$ and Ant22, were isolated from the Antarctic coastal sea water. Strains $Ant21^T$ and Ant22 showed UVC and gamma radiation resistance. Phylogenetic analyses based on 16S rRNA gene sequences determined that these strains belong to the genus Deinococcus. Through the analyses of the 16S rRNA gene sequences, strains $Ant21^T$ and Ant22 were found to have 97.7% and 97.8% similarity to Deinococcus marmoris DSM $12784^T$ and 97.0% and 97.2% similarity to Deinococcus saxicola AA-$1444^T$, respectively. The sequence similarity with the type strains of other Deinococcus species was less than 96.9% for both strains. Strains $Ant21^T$ and Ant22 shared relatively high 16S rRNA gene sequence similarity (99.3%) and had a closely related DNA reassociation value of $84{\pm}0.5%$. Meanwhile, they showed a low level of DNA-DNA hybridization (<30%) with other closely related species of the genus Deinococcus. The two strains also showed typical chemotaxonomic features for the genus Deinococcus, in terms of the major polar lipid (phosphoglycolipid) and the major fatty acids ($C_{16:0}$, $C_{16:1}$ ${\omega}6c/{\omega}7c$, $iso-C_{17:0}$, and $iso-C_{15:0}$). They grew at temperatures between $4^{\circ}C$ and $30^{\circ}C$ and at pH values of 6.0-8.0. Based on the physiological characteristics, the 16S rRNA gene sequence analysis results, and the low DNA-DNA reassociation level with Deionococcus marmoris, strains $Ant21^T$ ($=KEMB\;9004-167^T$ $=JCM\;31436^T$) and Ant22 (KEMB 9004-168 =JCM 31437) represent novel species belonging to the genus Deinococcus, for which the name Deinococcus rubrus is proposed.
