Objective: Pluripotent stem cell-derived lymphatic endothelial cells (LECs) show great promise in their therapeutic application in the field of regenerative medicine related to lymphatic vessels. We tested the approach of forced differentiation of mouse embryonal stem cells into LECs using biodegradable poly lactic-co-glycolic acid (PLGA) nanospheres in conjugation with growth factors (vascular endothelial growth factors [VEGF-A and VEGF-C]). Methods: We evaluated the practical use of heparin-conjugated PLGA nanoparticles (molecular weight ~15,000) in conjugation with VEGF-A/C, embryoid body (EB) formation, and LEC differentiation using immunofluorescence staining followed by quantification and quantitative real-time polymerase chain reaction analysis. Results: We showed that formation and differentiation of EB with VEGF-A/C-conjugated PLGA nanospheres, compared to direct supplementation of VEGF-A/C to the EB differentiation media, greatly improved yield of LYVE1(+) LECs. Our analyses revealed that the enhanced potential of LEC differentiation using VEGF-A/C-conjugated PLGA nanospheres was mediated by elevation of expression of the genes that are important for lymphatic vessel formation. Conclusion: Together, we not only established an improved protocol for LEC differentiation using PLGA nanospheres but also provided a platform technology for the mechanistic study of LEC development in mammals.
탈미네랄화된 골분(demineralized bone particle, DBP)은 골/연골 형성의 강력한 유도인자로 사용된다. 본 연구에서는 용매 캐스팅/염 추출법을 이용해 함량별 DBP와 PLGA가 하이브리드화된 다공성 지지체를 실제 디스크 형태와 유사하게 제조하였다. 제조된 지지체의 특성을 분석하기 위하여 다공도, 표면 젖음성 및 물 흡수성을 측정하였다. 디스크 세포인 섬유륜 및 수핵 세포는 토끼로부터 분리하여 제조된 지지체에 각각 파종한 후, 지지체를 재조합하여 배양하였다. 지지체에 파종된 디스크 세포의 생존율과 증식률은 MTT(3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide) 분석 방법을 이용하였고, 면역결핍 쥐의 피하에 삽입하여 이들의 디스크 조직 형성 정도를 확인하였다. 피하에 이식된 지지체를 적출하여 육안으로 관찰하고 모폴로지의 변화를 확인한 후, 조직을 파라핀으로 고정시켜 슬라이드를 제조하여 hematoxylin과 eosin 염색을 수행하였다. 천연/합성 하이브리드 담체로서의 DBP/PLGA 담체가 PLGA 단독으로 사용하였을 때와 비교하여 볼 때 디스크 조직의 형성이 우수하였으며, 특히 20, 40%의 DBP가 함유된 지지체가 세포의 성장과, 디스크 조직화에 유리함을 확인하였다.
본 연구에서는 슈반세포와 다양한 세포 부착인자로 코팅된 고분자 표면과의 상호관계에 대해 연구하였다. 세포 접착인자로 알려진 피브로넥틴, 피브리노겐, 라미닌, 비트로넥틴, 폴리-D-라이신 및 폴리-L-라이신을 PLGA 필름에 코팅하고 물 접촉각 측정과 ESCA 분석을 실시해 표면특성을 평가하구 접착인자로 코팅하거나 하지 않은 PLGA 필름의 표면에 슈반세포를 배양한 후 세포 계수와 SEM 관찰을 통해 세포 부착과 성장을 알아보았다. 세포 계수 결과에서 세포 부착은 PLGA 표면의 단백질 흡착과 관련 있음을 확인할 수 있었으며, 세포의 성장은 배양액의 우태아혈청 함량의 영향을 받는 것으로 나타났다. 이러한 결과를 통해 슈반세포의 접착과 성장이 특정한 세포 접착인자에 의해 영향을 받음을 알 수 있었다. 본 실험의 결과를 통해 조직공학적 신경 재생에 응용하기 위한 신경유도관의 개발에서 세포의 부착과 성장을 향상시키기 위해서는 세포의 종류 및 배양조건에 따라 신경유도관이 적절한 표면환경을 제공해야 함이 필수적임을 알 수 있었다.
항암제인 독소루비신의 지속적인 방출을 위하여 PLGA와 독소루비신의 미립구를 수중유형(O/W) 용매증발 방법을 이용하여 약물의 농도와 고분자의 분자량의 변화에 따른 방출거동의 차이를 확인하였다. 이중층 미립구내의 독소루비신의 방출을 분석하기 위하여 형광 분광 광도계를 이용하여 5주 동안 독소루비신의 방출을 보았으며 주사전자현미경을 이용하여 이중층미립구의 단면과 표면을 확인하였다. 제조된 이중층 미립구는 외부층이 전체적으로 매끄러운 표면과 구형을 나타내고 있었고 이중층 미립구의 단면을 잘라 계면층을 중심으로 하여 내부형태와 외부형태를 구분 지을 수 있었다. 또한 제조된 이중층 PLGA 독소루비신 미립구는 방출 결과를 통하여 저분자량의 고분자를 이용할수록 방출이 빠르다는 것을 확인할수 있었다. 따라서 미립구를 제조하는데 있어서 고분자의 분자량을 조절함으로써 방출거동을 조절할 수 있다는 것을 확인하였다.
골 결손 치료를 위해 생분해성 고분자인 PLGA에 골다공증 치료제인 이프리플라본(IP)을 함유한 미립구를 O/W 유화 용매 증발법으로 제조하였으며, 생체외 방출실험에서 IP 방출량은 HPLC로 분석하였다. SEM을 이용하여 미립구에 부착된 세포의 거동을 확인하였으며, IP가 세포에 미치는 독성평가는 CCK-8 분석방법을 이용하여 측정하였다. 골 형성 지표인 ALP 활성도를 측정하였으며, 배양된 BMSCS의 골세포로의 표현형을 확인하기 위하여 RT-PCR을 수행하였다. 방출결과에서 IP 방출은 거의 40일 이상으로 지속적이었으며, IP를 함유한 미립구에서의 세포의 부착, 성장 등이 잘 이루어짐을 확인하였고, ALP 활성도 및 RT-PCR 분석결과에서도 단독의 PLGA 미립구보다 IP를 함유한 미립구에서의 값이 더 높은 것을 확인할 수 있었다. 본 실험결과를 바탕으로 향후 서방성 제제화에 있어서 IP/PLGA 미립구를 응용하게 되면 국소지향성 골분화 주사용 지지체로서 중요한 역할을 할 것으로 보인다.
본 연구는 신경손상 모델에서 신경재생을 유도하는 슈반세포(SC)의 배양에 케라틴이 미치는 영향을 확인하기 위한 실험으로써, 친수성 아미노산으로 구성된 케라틴과 PLGA를 혼합하여 케라틴/PLGA 필름을 0, 10, 20, 그리고 50 wt%가 되도록 제조하여, 케라틴 안에 존재하는 다양한 단백질 및 신호전달물질이 슈반세포의 증식, 부착형태 그리고 표현형 유지에 미치는 영향을 확인하였다. 세포의 배양 방법은 손쉽고 순수한 세포 분리가 가능한 Morrissey의 방법을 이용하였고 필름의 젖음성 확인을 위하여 접촉각 측정을 실시하였으며, 정해진 시간에 세포를 계수하여 케라틴 함량에 따른 세포 성장차이를 비교하였다. 케라틴/PLGA 필름에서의 세포의 부착 거동 및 세포 형태를 SEM을 통하여 확인하였고 슈반세포의 표현형 유지를 확인하기 위해 RT-PCR을 수행하였다. 실험 결과, 다른 함유량과 비교 시 케라틴 10 또는 20 wt%가 함유된 케라틴/PLGA 필름이 SC 성장 및 표현형 유지에 긍정적인 영향을 미침을 확인하였다.
Pioglitazone을 봉입한 poly(lactide-co-glycolide)(PLGA) 나노입자를 emulsion-evaporation 방법을 이용하여 제조하여 최적의 나노입자와 봉입률을 조절하였다. 제조된 나노입자의 크기는 125~170 nm이었으며 30% pioglitazone이 봉입된 나노입자(3% PVA)의 봉입률은 85% 이상이었다. 이러한 나노입자들은 40일 동안 일정하게 용출이 되었다. 당뇨병 모델을 이용한 동물실험에서 글루코오스 농도를 저하시켰을 뿐만 아니라, 조직검사에서는 낮은 독성을 가지고 있는 것을 확인하였다. 이러한 결과는 pioglitazone 경구투여를 위한 약물전달을 위한 운반체로 사용될 수 있음을 확인하였다.
치주 질환으로 인하여 소실된 치주조직을 재생시키려는 여러 술식이 많이 연구되고있다. 그 중 bioactive factor의 적용은 치주조직의 재생에 있어서 우수한 치료법으로 평가되고 있으며, 이를 수용부에 적절히 적용하기 위한 운반체로 생체친화적인 중합체가 이용되고 있다. 본 연구의 목적은 PLGA를 Inorganic filler에 혼합시킨 재료를 성견의 일벽성 골내낭에 적용하여 이 재료의 생체 친화성과 생체 흡수도를 보고자 하는 것이다. 5마리의 비글견에서 제 3 소구치를 모두 발치한 뒤, 8주간의 치유기간이 지나고 제 2 소구치 원심면과 제 4 소구치 근심면에 5mm 깊이, 4mm폭의 일벽성 골내낭을 형성하였다. 좌측 defect에는 PLGA/inorganic filler matrix를 이식하였고 우측에는 아무것도 이식하지 않은 대조군으로 나누어 술 후 8주에 희생하여 치유 결과를 조직학적으로 비교 관찰하였다. 조직학적 분석 결과, 모든 결손부에서 염증의 소견이 관찰되지 않았으며 치근흡수와 유착은 발견되지 않았다. 백악질과 치조골, 치주인대를 포함한 치주조직의 재생에 있어서 대조군, 실험군 간에 조직학적으로 치유양상에 있어 차이를 많이 보이지 않았으며 PLGA/inorganic filler matrix는 8주 내에 완전히 흡수되어 결합조직이나 신생골내에서 그 흔적을 발견할 수 없었다. 이러한 결과는 PLGA/inorganic filler matrix는 생체친화성 및 생체흡수성이 우수한 재료로서 치주 조직의 재생 치료에 있어서 신체활성인자의 scaffold로 사용될 수 있는 가능성을 보여주었다.

In order to fabricate new sustained delivery device of nerve growth factor (NGF), we developed NGF-loaded biodegradable poly(L-lactide-co-glycolide) (PLGA, the mole ratio of lactide to glycolide 75:25, molecular weight: 83,000 and 43,000 g/mole, respectively) film by novel and simple sandwich solvent casting method for the possibility of the application of neural tissue engineering. PLGA was copolymerized by direct condensation reaction and the molecular weight was controlled by reaction time. Released behavior of NGF from NGF-loaded films was characterized by enzyme linked immunosorbent assay (ELISA) and degradation characteristics were observed by scanning electron microscopy (SEM) and gel permeation chromatography (GPC). The bioactivity of released NGF was identified using a rat pheochromocytoma (PC-12) cell based bioassay. The release of NGF from the NGF-loaded PLGA films was prolonged over 35 days with zero-order rate of 0.5-0.8 ng NGF/day without initial burst and could be controlled by the variations of molecular weight and NGF loading amount. After 7 days NGF released in phosphate buffered saline and PC-12 cell cultured on the NGF-loaded PLGA film for 3 days. The released NGF stimulated neurite sprouting in cultured PC-12 cells, that is to say, the remained NGF in the NGF/PLGA film at 37 $^{\circ}C$ for 7 days was still bioactive. This study suggested that NGF-loaded PLGA sandwich film is released the desired period in delivery system and useful neuronal growth culture as nerve contact guidance tube for the application of neural tissue engineering.
Background: Nano-therapy has the potential to revolutionize cancer therapy. Chrysin, a natural flavonoid, was recently recognized as having important biological roles in chemical defenses and nitrogen fixation, with anti-inflammatory and anti-oxidant effects but the poor water solubility of flavonoids limitstheir bioavailability and biomedical applications. Objective: Chrysin loaded PLGA-PEG-PLGA was assessed for improvement of solubility, drug tolerance and adverse effects and accumulation in a gastric cancer cell line (AGS). Materials and Methods: Chrysin loaded PLGA-PEG copolymers were prepared using the double emulsion method (W/O/W). The morphology and size distributions of the prepared PLGA-PEG nanospheres were investigated by 1H NMR, FT-IR and SEM. The in vitro cytotoxicity of pure and nano-chrysin was tested by MTT assay and miR-34a was measured by real-time PCR. Results: 1H NMR, FT-IR and SEM confirmed the PLGA-PEG structure and chrysin loaded on nanoparticles. The MTT results for different concentrations of chrysin at different times for the treatment of AGS cell line showed IC50 values of 68.2, 56.2 and $42.3{\mu}M$ and 58.2, 44.2, $36.8{\mu}M$ after 24, 48, and 72 hours of treatment, respectively for chrysin itslef and chrysin-loaded nanoparticles. The results of real time PCR showed that expression of miR-34a was upregulated to a greater extent via nano chrysin rather than free chrysin. Conclusions: Our study demonstrates chrysin loaded PLGA-PEG promises a natural and efficient system for anticancer drug delivery to fight gastric cancer.
