Korean Red Ginseng and Portulaca oleracea Extracts Attenuate Lipopolysaccharide-induced Inflammation via Downregulation of Nuclear Factor Kappa-B and the Mitogen-activated Protein Kinase Signaling Pathway in Macrophage Cell Line RAW 264.7

  • Korean red ginseng (Panax ginseng Meyer) is a well-known traditional medicine, with numerous biological functions in the body. Portulaca oleracea (P. ole) belongs to the Portulacaceae family and has bioactive potential as a traditional medicine. This study aimed to determine the anti-inflammatory effects of Korean red ginseng extract (RGE) and P. ole extract on lipopolysaccharide (LPS)-treated RAW 264.7 cells. The combination of RGE (50 ㎍/mL) and P. ole (6.25 ㎍/mL) extracts significantly suppressed LPS-induced nitric oxide synthesis. The expression of proinflammatory mediators, including inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), and proinflammatory cytokines, including interleukin-1β, interleukin-6, and tumor necrosis factor-α, were markedly decreased by the combined treatment with RGE (50 ㎍/mL) and P. ole (6.25 ㎍/mL). Moreover, iNOS and COX-2 protein expression levels were also significantly reduced in the combined treatment compared to the LPS-stimulated group. In addition, the nuclear translocation of phosphorylated nuclear factor kappa-B was suppressed by the treatment with RGE and P. ole. Moreover, the mitogen-activated protein kinase pathway was also partially inhibited by the combination treatment with RGE and P. ole. Our results demonstrate that the treatment mixture with RGE and P. ole could be used as functional food and therapeutic herbal medicine in various inflammatory diseases.

  • This study proposes a monitoring system that effectively watches surroundings by cooperating the various sensor information including image information on a sensor network system. The monitoring system proposed in this paper is developed to watch certain intruders to the internal spaces through the interested region for exceptional time by installing cameras, PIR(Pyroelectric Infrared Ray) sensor and body detectors in such interested regions. Moreover the monitering system is implemented based on the SenWeaver plateform which is a integrated development tools for building wireless sensor network system. In the results of the test that was applied to a practically experimental environment by implementing some interfaces for the proposed system, it was considered that it is possible to watch surroundings effectively using the image information obtained from cameras and multiple sensor information acquisited from sensor nodes.

  • 생명과학 기술의 급속한 발달로 인류 복지 증진에 많은 기여를 하였지만 아직도 각종 질병 등으로 많은 사람들이 고통 받고 있으며, 이를 극복하기 위한 연구 및 기술개발은 세계 각처에서 계속되고 있다. 이러한 연구 및 기술개발의 결과로 산출되는 생명정보 데이터의 양은 기하급수적으로 증가하고 있기에 이런 방대한 양의 생명정보 데이터를 분석하고 분석된 데이터에서 인류 복지에 유용한 정보를 얻기 위한 생명정보학(Bioinformatics)이 등장하게 되었다. 이에, 한국과학기술정보연구원(KISTI)은 IT 기반 생명정보 인프라 구축의 중심기관으로 CCBB(Center for Conputationa Biology & Bioinformatics) 웹사이트를 운영하고 있다. CCBB는 전산학적인 기술을 이용한 생명현상 연구를 지원하기 위하여 21종의 생명정보 콘텐트(DB 및 분석도구)를 수집 분석 구축 제공하고 있다. 이 중에서 GenBank, PDB, PIR, Swiss-prot 등의 데이터베이스는 KISTI에서 개발한 KRISTAL 검색엔진을 통하여 국내에서도 빠르고 쉽게 검색 가능하도록 자체 구축하고 있으며, 이와 더불어 BLAST, FASTA, ClustalW 등의 주요 분석 도구 또한 제공하고 있다. 본 논문에서는 CCBB에서 제공중인 21종의 콘텐트 중에서 GenBank, REBASE, GeneCards, InterProScan 등 4종의 대용량 고효율 생명정보 콘텐트의 소개 및 업데이트 방법에 관한 내용을 기술하고자 한다.

Red ginseng extract inhibits lipopolysaccharide-induced platelet-leukocyte aggregates in mice

  • Background: Platelet-leukocyte aggregates (PLAs) play important roles in cardiovascular disease and sepsis. Red ginseng extract (RGE) has been well-studied for its antiplatelet and anti-inflammatory activities. However, the potential inhibitory effects of RGE on PLA have not been investigated. Methods: Six-week-old ICR mice were given oral gavage of RGE for 7 days, followed by an intraperitoneal injection of 15 mg/kg of lipopolysaccharide. Mice were euthanized 24 h later, and blood samples were collected for further analysis. Flow cytometry was utilized to sort populations of PLAs and platelet-neutrophil aggregates (PNAs). By using confocal microscopy, PNAs were validated. Morphological changes in platelets and leukocytes were visualized with scanning electron microscopy. Expressions of tissue factor (TF) and platelet factor 4 (PF4) were investigated using enzyme-linked immunosorbent assay. Results: Populations of activated platelets, PLAs and PNAs, were significantly increased with LPS-induction. Treatment with 200 and 400 mg/kg of RGE decreased platelet activation. Moreover, the populations of PLAs and PNAs were reduced. PNAs were visible in the blood of septic mice, and this was attenuated by treatment with 400 mg/kg of RGE. Morphologically, sepsisinduced platelet activation and fibrin formation in the blood. This was reduced with RGE treatment. Sepsis-induced increase in the plasma levels of TF and PF4 was also reduced with RGE treatment. Conclusion: This study shows that RGE is a potential therapeutic that reduces the activation of platelets and targets PLA and PNA formation. Detailed inhibitory mechanisms of RGE should be studied.

Evaluation of different milking practices for optimum production performance in Sahiwal cows

  • The production performance of multiparous lactating Sahiwal cows (n = 24) was evaluated according to both milking frequency and method. Selected animals were randomly divided into four groups containing six animals each under a completely randomized design. Cows in groups A & B were milked by the hand milking method three times per day, respectively. Similarly, cows in groups C & D were milked by the machine milking method two and three times per day, respectively. All animals were maintained under uniform feeding and management conditions. Dry matter intake was high in animal groups milked three times per day, and it remained unchanged between the hand and machine milking methods. Milk yield was higher (P < 0.05) in cows milked three times compared to those milked twice per day, and it did not differ between hand and machine milking methods. Milk fat percentage was higher (P < 0.05) in cows milked twice per day compared to those milked three times using both machine and hand milking methods. The percentage of total solids showed a similar pattern as the fat percentage. However, percentages of protein, lactose, and non-fat solids in milk were not significantly different (P > 0.05) among the treatment groups. Collectively, the results show that milking three times per day instead of twice at 8-hour intervals can enhance milk yield in Sahiwal cows using both hand and machine milking methods.

Diversity of vir Genes in Plasmodium vivax from Endemic Regions in the Republic of Korea: an Initial Evaluation

  • Variant surface antigens (VSAs) encoded by pir families are considered to be the key proteins used by many Plasmodium spp. to escape the host immune system by antigenic variation. This attribute of VSAs is a critical issue in the development of a novel vaccine. In this regard, a population genetic study of vir genes from Plasmodium vivax was performed in the Republic of Korea (ROK). Eighty-five venous blood samples and 4 of the vir genes, namely vir 27, vir 21, vir 12, and vir 4, were selected for study. The number of segregating sites (S), number of haplotypes (H), haplotype diversity (Hd), DNA diversity (${\pi}$ and ${\Theta}_w$), and Tajima's D test value were conducted. Phylogenetic trees of each gene were constructed. The vir 21 (S=143, H=22, Hd=0.827) was the most genetically diverse gene, and the vir 4 (S=6, H=4, Hd=0.556) was the opposite one. Tajima's D values for vir 27 (1.08530, P>0.1), vir 12 (2.89007, P<0.01), and vir 21 (0.40782, P>0.1) were positive, and that of vir 4 (-1.32162, P>0.1) was negative. All phylogenetic trees showed 2 clades with no particular branching according to the geographical differences and cluster. This study is the first survey on the vir genes in ROK, providing information on the genetic level. The sample sequences from vir 4 showed a clear difference to the Sal-1 reference gene sequence, whereas they were very similar to those from Indian isolates.

Descriptive Report on Pattern of Variation in Cancer Cases within Selected Ethnic Groups in Kamrup Urban District of Assam, 2009-2011

  • Background: The global burden of cancer is continuously increasing. According to recent report of the National Cancer Registry Programme (NCRP) on time trends it is estimated that future burden of cancer cases for India in 2020 will be 1,320,928. It is well known that knowledge of the incidence of cancer is a fundamental requirement of rational planning and monitoring of cancer control programs. It would help health planners to formulate public health policy if relevant ethnic groups were considered. North East-India alone contains over 160 Scheduled Tribes and 400 other sub-tribal communities and groups, whose cancer incidence rates are high compared to mainland India. As since no previous study was done focusing on ethnicity, the present investigation was performed. Materials and Methods: In this paper PBCR-Guwahati data on all cancer registrations from January 2009 to December 2011 for residents of the Kamrup Urban District, comprising an area of 261.8 sq. km with a total population of 900,518, including individual records with information on sex, age, ethnicity and cancer site are provided. Descriptive statistics including age adjusted rates (AARs) were taken as provided by NCRP. For comparison of proportional incidence ratios (PIR) the Student's t test was used, with p<0.05 considered as statistically significant. Results and Conclusions: Differences in leading sites of Kamrup Urban District since from the beginning of the PBCR-Guwahati were revealed among different ethnic groups by this study. The results should help policy makers to formulate different strategies to control the level of burden as well as for treatment planning. This study also suggests that age is an important factor of cancer among different ethnic populations as well as for overall population of Kamrup District of Assam.

Investigation of Compressive Strength and Foaming Characteristics of Acid Anhydride Epoxy Foam by Foaming Agent (발포제에 따른 산무수물계 에폭시 폼의 압축강도 및 포밍특성 분석)

  • Polymer foams were used to fill the void in the structure in addition to flame retardant and heat insulation. Polymer foams such as polyurethane, polyisocyanurate, poly(vinyl chloride), polyethylene terephthalate were used to weight lighting materials. In this study, epoxy foam was used to improve mechanical properties of polymer foam. Acid anhydride type hardener reacts with polyol. Using this phenomenon, if blowing agent was added into epoxy resin using acid anhydride type hardener, formation and compressive properties of epoxy foam was studied. Formation of polymer foam was compared with type of blowing agent and concentration of blowing agent via compressive test. As these results, optimized condition of epoxy foam was found and epoxy foam had better compressive property than other polymer foam.

Present Status and Future Management Strategies for Sugarcane Yellow Leaf Virus: A Major Constraint to the Global Sugarcane Production

  • Sugarcane yellow leaf virus (SCYLV) is a distinct member of the Polerovirus genus of the Luteoviridae family. SCYLV is the major limitation to sugarcane production worldwide and presently occurring in most of the sugarcane growing countries. SCYLV having high genetic diversity within the species and presently ten genotypes are known to occur based on the complete genome sequence information. SCYLV is present in almost all the states of India where sugarcane is grown. Virion comprises of 180 coat protein units and are 24-29 nm in diameter. The genome of SCYLV is a monopartite and comprised of single-stranded (ss) positive-sense (+) linear RNA of about 6 kb in size. Virus genome consists of six open reading frames (ORFs) that are expressed by sub-genomic RNAs. The SCYLV is phloem-limited and transmitted by sugarcane aphid Melanaphis sacchari in a circulative and non-propagative manner. The other aphid species namely, Ceratovacuna lanigera, Rhopalosiphum rufiabdominalis, and R. maidis also been reported to transmit the virus. The virus is not transmitted mechanically, therefore, its transmission by M. sacchari has been studied in different countries. SCYLV has a limited natural host range and mainly infect sugarcane (Sachharum hybrid), grain sorghum (Sorghum bicolor), and Columbus grass (Sorghum almum). Recent insights in the protein-protein interactions of Polerovirus through protein interaction reporter (PIR) technology enable us to understand viral encoded proteins during virus replication, assembly, plant defence mechanism, short and long-distance travel of the virus. This review presents the recent understandings on virus biology, diagnosis, genetic diversity, virus-vector and host-virus interactions and conventional and next generation management approaches.

Privacy Preserving Keyword Search with Access Control based on DTLS (프라이버시를 보호하는 접근제어가 가능한 키워드 검색 기법)

  • To protect sensitive personal information, data will be stored in encrypted form. However in order to retrieve these encrypted data without decryption, there need efficient search methods to enable the retrieval of the encrypted data. Until now, a number of searchable encryption schemes have been proposed but these schemes are not suitable when dynamic users who have the permission to access the data share the encrypted data. Since, in previous searchable encryption schemes, only specific user who is the data owner in symmetric key settings or has the secret key corresponding to the public key for the encrypted data in asymmetric key settings can access to the encrypted data. To solve this problem, Stephen S. Yau et al. firstly proposed the controlled privacy preserving keyword search scheme which can control the search capabilities of users according to access policies of the data provider. However, this scheme has the problem that the privacy of the data retrievers can be breached. In this paper, we firstly analyze the weakness of Stephen S. Yau et al.'s scheme and propose privacy preserving keyword search with access control. Our proposed scheme preserves the privacy of data retrievers.
